BACKGROUND: Maffucci's syndrome (MS) is distinguished by the enigmatic association of benign cartilaginous bone tumors and soft tissue hemangiomas.
METHODS: This study was conducted to define the distribution of nerves and neuropeptides around these tumors. Results were measured by quantitative image analysis of immunohistochemical staining. Metkephamid acetate, a newly synthetized analogue of methionine enkephalin, suppressed plasma vasopressin levels from 2.4 pg/ml to 0.8 pg/ml after intramuscular administration to 4 normal human volunteers. This suppression occurred in the absence of significant changes in either plasma osmolality or blood pressure. Thus, metkephamid acetate, like many other opiates which bind to the mu subtype of opiate receptors, has a direct suppressive effect on plasma vasopressin levels.

The distribution of vasoactive intestinal polypeptide-like structures in the olfactory bulb of the rainbow trout was studied using an indirect-immunoperoxidase technique. Olfactory fibres were very strongly labelled, whereas the fibres or cell bodies in the remaining strata of the olfactory bulb showed no immunoreactivity. In addition, the olfactory nerve fibres were not immunoreactive for methionine- and leucine-enkephalins, motilin, neuropeptide Y, substance P, cholecystokinin-8 and tyrosine-hydroxylase.

The effects of methionine(met)-enkephalin, leucine(leu)-enkephalin, beta-endorphin and blocking substances upon renal function were studied in conscious goats. The synthetic opiate D-Alanine-methionine enkephalin (D-Ala) affects neuronal activity of the fascia dentata (FD). D-Ala causes a depression of the population spike evoked by stimulation of the perforant path, which is antagonized by naloxone. In so called "Tandem" experiments it was shown that D-Ala had opposite effects on the CA1 pyramidal and FD granule cells. It is likely that in the FD the site of action of D-Ala is at the afferent synapses.
