Primary genetic mitochondrial diseases (GMDs) are a clinically and genetically diverse group of diseases estimated to impact over 1 in 4,000 individuals. Leigh syndrome (LS) is the most common pediatric presentation of GMD. LS typically presents within the first years of life and is a severe progressive multi-system disorder. Symmetric progressive inflammatory brain lesions are a defining feature of the disease. Patients can also present with seizures, metabolic dysfunction, muscle weakness, and other symptoms. No effective clinical treatments currently exist. Recent data from the Ndufs4(-/-)

The photocatalytic degradation of pollutants in water remains a significant challenge due to the rapid recombination of photogenerated electron-hole pairs, which hinders photocatalytic efficiency. To address this issue, we successfully constructed a Z-scheme ZnFe2O4/MoO3 composite to activate peroxymonosulfate (PMS) for the efficient degradation of enrofloxacin (ENR) under visible light. By constructing a Z-scheme heterojunction, we significantly improved the average lifetime of photogenerated carriers and effectively enhanced the separation efficiency of e-/h+ pairs. Activating PMS under visi

The use of antimicrobial drugs in food-producing animals contributes to the selection pressure on pathogenic and commensal bacteria to become resistant. This study aims to evaluate the existence of trade-offs between treatment effectiveness, cost, and the dynamics of resistance in gut commensal bacteria. We developed a within-host ordinary differential equation model to track the dynamics of antimicrobial drug concentrations and bacterial populations in the site of infection (lung) and the gut. The model was parameterized to represent enrofloxacin treatment for bovine respiratory disease (BRD)

The combined pollution of antibiotics and heavy metals in aqueous environments increases the risk of aquatic ecosystem disruption and the complication of pollutant management. Here, a fungus Cladosporium cladosporioides 11 (CC11) isolated from aquaculture pond sediments possessed effective capacities to biotransform the combined pollution of enrofloxacin (ENR) and copper ion (Cu). ENR and Cu were considerably abated by CC11, and the presence of Cu (30 mg/L) promoted the biotransformation efficiency of ENR. The biotranformation of ENR in ENR-Cu co-contamination was associated to ligninolytic en

Polymyxins are increasingly used as a last-resort class of antibiotics against extensively drug-resistant (XDR) Gram-negative bacteria. However, resistance to polymyxins can emerge with monotherapy. As nephrotoxicity is the major dose-limiting factor for polymyxin monotherapy, dose escalation to suppress the emergence of polymyxin resistance is not a viable option. Therefore, novel approaches are needed to preserve this last-line class of antibiotics. This study aimed to investigate the antimicrobial synergy of polymyxin B combined with enrofloxacin against Pseudomonas aeruginosa Static time-k

Four cynomolgus macaques housed at our facility became acutely ill with dysenteric symptoms. Enteric isolates established an etiologic diagnosis of Shigella flexneri. Enrofloxacin antimicrobial therapy cleared the infection with no perceptible bacterial shedding or clinical signs of disease. High-dose methyl-prednisolone therapy was administered to the four monkeys for 5 weeks. The animals were monitored for signs of shigellosis and bacterial shedding weekly throughout the study, for a total of 7 weeks. Although methylprednisolone therapy induced marked cellular immunosuppression in all four a

The residues of enrofloxacin (ENR) in animal-derived food and water environments have emerged as a growing public health concern in recent years. Herein, a tri-mode biosensor was designed by employing CuS-CDs@MnO2 as the signal probe and glutathione (GSH)-loaded liposomes for amplification. The manganese dioxide (MnO2) shell of the nanocomposite quenched the fluorescence of CuS-CDs, reduced its photothermal performance and also endowed it with oxidase-like activity. In the presence of ENR, a large amount of GSH was released after aptamer recognition and magnetic separation. Then, the shell of

Dynamic impacts of short-term enrofloxacin (ENR) exposure on juvenile marine fish are not well understood, and the underlying mechanisms remain unclear. We therefore investigated the accumulation and elimination of ENR in the liver of juvenile black seabream Acanthopagrus schlegelii. Meanwhile, the dynamic alterations of biochemical parameters and liver transcriptomes after short-term bath immersion and withdrawal treatment were explored. The results indicated that the contents of ENR in the liver were significantly increased after bath administration for 24 h, and then quickly declined to ver

BACKGROUND: Staphylococcus aureus is one of the main causative agents of mastitis in small ruminants. Antimicrobial use is the major treatment, but there are many flaws linked to resistance, tolerance or persistence. This study aimed to verify changes in resistance, virulence and clonal profiles of S. aureus isolated from persistent mastitis goat milk before and after enrofloxacin treatment.
RESULTS: MIC increased to at least one antimicrobial in S. aureus isolates after enrofloxacin treatment compared to before. The most detected resistance genes before and after treatment were tetK, tetM, a

Enrofloxacin (ENRO) is widely used as an antimicrobial drug for treatment of uncomplicated and complicated infections in veterinary medicine. Its bitter taste limits its clinical applications in veterinary. To mask the bitter taste of this drug, double-coated taste-masking microparticles of ENRO (DTME) were prepared through stearic acid solid dispersion and chitosan-alginate microparticle coating technologies. The taste-masking effect was evaluated by pig feeding experiment. Results showed that DTME exhibited a spherical-like shape (170490μm). DTME yielded a drug loading rate of DTME 20.3% and

SIMPLE, RAPID, AND EXTRACTIVE SPECTROPHOTOMETRIC METHODS WERE DEVELOPED FOR THE DETERMINATION OF SOME FLUOROQUINOLONES ANTIBIOTICS: gemifloxacin mesylate (GMF), moxifloxacin hydrochloride (MXF), and enrofloxacin (ENF) in pure forms and pharmaceutical formulations. These methods are based on the formation of ion-pair complexes between the basic drugs and acid dyes, namely, bromocresol green (BCG), bromocresol purple (BCP), bromophenol blue (BPB), bromothymol blue (BTB), and methyl orange (MO) in acidic buffer solutions. The formed complexes were extracted with chloroform and measured at 420, 40

Antibiotic residues are commonly found in food. The effect of dietary exposure to veterinary antibiotics on the transmission of antibiotic-resistant bacteria and antibiotic resistance genes (ARGs) from food to humans is unknown. We found that dietary exposure to enrofloxacin reduced microbial diversity, interactions, and the immune responses; weakened the colonization resistance of the resident microbiota; and promoted the colonization of exogenous Escherichia coli K-12 MG1655 in the simulated human intestine both in vitro and in vivo experiments in mice. In addition to the growth advantages f

UNLABELLED: Campylobacter is a leading cause of foodborne illness and is commonly present in the poultry gut. Fluoroquinolone (FQ)-resistant Campylobacter quickly emerges in chickens infected with FQ-susceptible Campylobacter and treated with an FQ antibiotic via drinking water due to selection and expansion of FQ-resistant mutants. However, it is unclear whether FQ treatment alters the gut microbiota and metabolome, and consequently provides a favorable environment for FQ-resistant Campylobacter to rapidly expand in the intestine. Here, we investigated cecal microbiota and metabolome profiles

Copper oxide nanoparticles (CuO-NPs) have been suggested as effective catalysts to degrade many persistent organic contaminants. In parallel, CuO-NPs are considered toxic to soil microorganisms, plants and human cells, possibly because they induce oxidative stress and generation of reactive oxygen species (ROS). However, the mechanism of the catalytic process and the generated ROS are poorly understood. Here we discuss the reaction mechanism of CuO-NPs during the catalytic degradation of enrofloxacin - an antibiotic pharmaceutical used in this study as a representative persistent organic compo

The effect of a competitive exclusion product (Aviguard®) on the selection of fluoroquinolone resistance in poultry was assessed in vivo in the absence or presence of fluoroquinolone treatment. Two experiments using a controlled seeder-sentinel animal model (2 seeders: 4 sentinels per group) with one-day-old chicks were used. For both experiments, as soon as the chicks were hatched, the birds of two groups were administered Aviguard® and two groups were left untreated. Three days later, all groups were inoculated with an enrofloxacin-susceptible commensal E. coli strain. Five days after hatchi

The effect of treatment with enrofloxacin was studied on the postvaccinal immune response in pigs. Forty pigs were used (control not vaccinated (C), control vaccinated (CV), vaccinated, received enrofloxacin (ENRO)). From day -1 to day 3 pigs from ENRO group received enrofloxacin at the recommended dose. Pigs from ENRO and CV groups were vaccinated twice against Aujeszky's disease virus (ADV). There was a significant delay in the production of humoral response of enrofloxacin dosed pigs when compared with CV group. Moreover, in ENRO group the significant decrease in IFN-γ production and signif
