Mixtures research at NIEHS: an evolving program.

The National Institute of Environmental Health Sciences (NIEHS) has a rich history in evaluating the toxicity of mixtures. The types of mixtures assessed by the Division of the National Toxicology Program (DNTP) and the extramural community (through the Division of Extramural Research and Training, DERT) have included a broad range of chemicals and toxicants, with each study having a unique set of questions and design considerations. Some examples of the types of mixtures studied include: groundwater contaminants, pesticides/fertilizers, dioxin-like chemicals (assessing the toxic equivalency a

History of US Presidential Assaults on Modern Environmental Health Protection.

The Trump administration has undertaken an assault on the Environmental Protection Agency (EPA), an agency critical to environmental health. This assault has precedents in the administrations of Ronald Reagan and George W. Bush. The early Reagan administration (1981-1983) launched an overt attack on the EPA, combining deregulation with budget and staff cuts, whereas the George W. Bush administration (2001-2008) adopted a subtler approach, undermining science-based policy. The current administration combines both these strategies and operates in a political context more favorable to its designs

From mandatory disclosure data to organisational decision-making guided by planetary boundaries.

Organisations currently lack operational methods to assess how their activities contribute to returning society to within Earth's safe operating space, considering that seven of the nine planetary boundaries are currently exceeded. Although the mandatory Corporate Sustainability Reporting Directive of the European Union generates extensive compliance data, these disclosures remain disconnected from planetary boundary science. In this context, the study addresses two key knowledge gaps: (i) the methodological disconnect between mandatory sustainability reporting requirements and science-based e

Crohn's disease and environmental contamination: Current challenges and perspectives in exposure evaluation.

Although the incidence of Crohn's disease has increased worldwide over the past 30 years, the disorder's exact causes and physiological mechanisms have yet to be determined. Given that genetic determinants alone do not explain the development of Crohn's disease, there is growing interest in "environmental" determinants. In medical science, the term "environment" refers to both the ecological and social surroundings; however, most published studies have focused on the latter. In environmental and exposure sciences, the term "environment" mostly relates to contamination of the biotope. There are

Key scientific issues in the health risk assessment of trichloroethylene.

Trichloroethylene (TCE) is a common environmental contaminant at hazardous waste sites and in ambient and indoor air. Assessing the human health risks of TCE is challenging because of its inherently complex metabolism and toxicity and the widely varying perspectives on a number of critical scientific issues. Because of this complexity, the U.S. Environmental Protection Agency (EPA) drew upon scientific input and expertise from a wide range of groups and individuals in developing its 2001 draft health risk assessment of TCE. This scientific outreach, which was aimed at engaging a diversity of p

A novel approach to scavenging anesthetic gases in rodent surgery.

Laboratory animal procedures using gas anesthetics may amass elevated waste gas concentrations in operating rooms if controls are not implemented for capturing and removing the vapors. Area sampling using an infrared analyzer indicated isoflurane concentrations likely to exceed occupational exposure guidelines. Our study showed environmental concentrations of oxygen as high as 40% and isoflurane concentrations >100 ppm when no controls or merely passive controls were utilized. These extraneous isoflurane emissions were determined to be originating from the pre-procedural induction process as w

Evaluation of a peat moss plus soybean oil (PMSO) technology for reducing explosive residue transport to groundwater at military training ranges under field conditions.

An evaluation of peat moss plus crude soybean oil (PMSO) for mitigation of explosive contamination of soil at military facilities was performed using large soil lysimeters under field conditions. Actual range soils were used, and two PMSO preparations with different ratios of peat moss:soybean oil (1:1, PO1; 1:2, PO2) were compared to a control lysimeter that received no PMSO. PMSO was applied as a 10 cm layer on top of the soil, and Composition B detonation residues from a 55-mm mortar round were applied at the surface of each of the lysimeters. Dissolution of the residues occurred during nat

Recreational scuba divers' aversion to low-frequency underwater sound.

Increasing use of active low-frequency sonar by submarines and ships raises the risk of accidental exposure of recreational divers to low-frequency underwater sound (LFS). This study aimed to characterize the subjective responses of recreational scuba divers to LFS to ascertain the extent to which LFS may impact their enjoyment, comfort, or time spent underwater. Seventeen male and nine female recreational scuba divers participated. Diving was conducted in an acoustically transparent tank located within a larger anechoic pool. Subjects wore scuba gear and were positioned I m below the surface

Environmental health impacts of concentrated animal feeding operations: anticipating hazards--searching for solutions.

A scientific conference and workshop was held March 2004 in Iowa City, Iowa, that brought together environmental scientists from North America and Europe to address major environmental health issues associated with concentrated animal feeding operations (CAFOs) in large, industrialized livestock production facilities. After one and a half days of plenary sessions, five expert workgroups convened to consider the most relevant research areas, including respiratory health effects, modeling and monitoring of air toxics, water quality issues, influenza pandemics and antibiotic resistance, and commu

The impact of new technologies on human population studies.

Human population studies involve clinical or epidemiological observations that associate environmental exposures with health endpoints and disease. Clearly, these are the most sought after data to support assessments of human health risk from environmental exposures. However, the foundations of many health risk assessments rest on experimental studies in rodents performed at high doses that elicit adverse outcomes, such as organ toxicity or tumors. Using the results of human studies and animal data, risk assessors define the levels of environmental exposures that may lead to disease in a porti

Antimicrobial pharmaceuticals in the aquatic environment - occurrence and environmental implications.

The environmental occurrence of antimicrobial pharmaceuticals and antibiotic resistant bacteria and antibiotic resistant genes has become a global phenomenon and a multifaceted threat. Integrated actions of many parties are needed to prevent further aggravation of the problem. Well-directed actions require clear understanding of the problem, which can be ensured by frequent revaluation of the existing knowledge and disseminating it among relevant audiences. The goal of this review paper is to discuss the occurrence and abundance of antimicrobial pharmaceuticals in the aquatic environment in co

[Considering the Professionalism of Pharmacists from a Viewpoint of "Regulatory Science"].

Regulatory science underpins scientific regulations, including reflection papers, guidelines, and administrative notices, and is closely related to the quality assurance (QA) of pharmaceuticals, foods, and chemicals in our living environment. Historically, QA has been considered the basis of pharmaceutical science. Therefore, the Pharmaceutical and Medical Device Law specifies that pharmacists, as marketing directors of pharmaceutical products, are responsible for their QA. Furthermore, a pharmacist is responsible for the QA of foods and environmental chemicals by several laws; for example, as

Long-term citizen science data reveals climate influences of tropical tree flowering at the regional scale.

Tropical tree reproductive phenology is sensitive to changing climate, but inter-individual and interannual variability at the regional scale is poorly understood. While large-scale and long-term datasets of environmental variables are available, reproductive phenology needs to be measured in-site, limiting the spatiotemporal scales of the data. We leveraged a unique dataset assembled by SeasonWatch, a citizen-science phenology monitoring programme in India to assess the environmental correlates of flowering in three ubiquitous and economically important tree species - jackfruit, mango and tam

The Use and Misuse of Historical Controls in Regulatory Toxicology: Lessons from the CLARITY-BPA Study.

For many endocrine-disrupting chemicals (EDCs) including Bisphenol A (BPA), animal studies show that environmentally relevant exposures cause harm; human studies are consistent with these findings. Yet, regulatory agencies charged with protecting public health continue to conclude that human exposures to these EDCs pose no risk. One reason for the disconnect between the scientific consensus on EDCs in the endocrinology community and the failure to act in the regulatory community is the dependence of the latter on so-called "guideline studies" to evaluate hazards, and the inability to incorpora

Immune reactions in a changing environment. USA initiatives.

Environmental hazards occurring as an undesirable consequence of economic progress, urbanization and pollution have become a worldwide concern. In the US, this is evident from the campaign against smoking which has focused attention on the lung in part because the lung as a target organ is constantly exposed to many visible environmental hazards. On the other hand, environmental hazards which are not lethal, but cause their effects in an insidious fashion, may be difficult to study and identify. Among the disciplines available to assess adverse health consequences of xenobiotics ('strange' sub

Improving the scientific foundation for mixtures joint toxicity and risk assessment: contributions from the SOT mixtures project--introduction.

Risk assessments are enhanced when policy and other decision-makers have access to experimental science designed to specifically inform key policy questions. Currently, our scientific understanding and science policy for environmental mixtures are based largely on extrapolating from and combining data in the observable range of single chemical toxicity to lower environmental concentrations and composition, i.e., using higher dose data to extrapolate and predict lower dose toxicity. There is a growing consensus that the default assumptions underlying those mixtures risk assessments that are con

Recent advances and public health implications for environmental exposure to Chlamydia abortus: from enzootic to zoonotic disease.

Environmental transmission of Chlamydia abortus as a result of enzootic disease or disease outbreaks and the threats posed by this pathogen has been previously reported, however a state-of-the-science review of these reports and the identification of future research priorities in this area is still lacking. This study provides an overview of the current knowledge of host-pathogen-environment interactions, addressing public health risks and identifying critical questions and research gaps. We performed a systematic PubMed and Web of Science search for publications related to Chlamydia abortus i
