Clinicopathological and ultrastructural study in two cases of chordoid glioma.

Chordoid glioma is a rare benign neoplasm of uncertain histogenesis occurring in the third ventricle/ /suprasellar region. Recently, data have emerged suggesting that chordoid glioma is a variant of ependymoma related to a specialised ependyma of the subcommisural organ or the lamina terminalis area. In this study, we report clinicopathological and ultrastructural findings in two chordoid glioma cases. In case 1, a tumour (1.5 cm in diameter) in a 62-year-old man invaded the anterior-basal part of the third ventricle in the lamina terminalis region. In case 2, a large tumour in a 51-year-old w

Effects of brain ventricular shape on periventricular biomechanics: a finite-element analysis.

OBJECTIVE: A computer simulation based on the finite-element method was used to study the biomechanics of acute obstructive hydrocephalus and, in particular, to define why periventricular edema is most prominent in the anterior and posterior horns. METHODS: Brain parenchyma was modeled as a two-phase material composed of a porous elastic matrix saturated by interstitial fluid. The effects of the cerebrovascular system were not included in this model. The change in the shape of the ventricles as they enlarged was described by two variables, i.e., the stretch of the ependyma and the concavity o

Margins of control: ventricular brain borders as architects in central nervous system autoimmunity.

While multiple sclerosis research has long centered on focal demyelinating lesions, it is increasingly recognized that there are distinct, surface-in gradients of diffuse pathology that exist in regions abutting the cerebrospinal fluid (CSF), such as the subpial cortex and periventricular parenchyma. While subpial pathology is thought to possess a relationship with tertiary lymphoid tissues (TLTs) in the meninges, less is known about the pathogenesis of diffuse periventricular pathology. Emerging evidence suggests that periventricular damage may reflect more than passive exposure to inflammato

Fetal brain damage in congenital hydrocephalus.

BACKGROUND: Congenital hydrocephalus (HCP) is a developmental brain disorder characterized by the abnormal accumulation of cerebrospinal fluid within the ventricles. It is caused by genetic and acquired factors that start during early embryogenesis with disruption of the neurogerminal areas. As might be expected, early-onset hydrocephalus alters the process of brain development leading to irreparable neurological deficit. A primary alteration of the ependyma/neural stem cells (affecting vesicle trafficking and abnormal cell junctions) leads to its loss or denudation and translocation of neural

Neuroependymal denudation is in progress in full-term human foetal spina bifida aperta.

In human spina bifida aperta (SBA), cerebral pathogenesis [hydrocephalus, Sylvius aqueduct (SA) stenosis and heterotopias] is poorly understood. In animal models, loss of ventricular lining (ependymal denudation) causes SA stenosis and hydrocephalus. We aimed to investigate whether ependymal denudation also takes place in human foetal SBA. Considering that ependymal denudation would be related to alterations in junction proteins, sections through SA of five SBA and six control foetuses (gestational ages ranged between 37 and 40 weeks) were immunostained for markers of ependyma (caveolin 1, βIV

Ependymal rests and subcutaneous sacrococcygeal ependymoma.

A 2 cm subcutaneous ependymoma was resected from the natal cleft of a 4-yr-old girl. The coccyx was not removed and there has been no recurrence in 14 years. A study of the post-coccygeal region in 15 control infants revealed subcutaneous islands of ependyma, unconnected with the cauda equina, in 4/4 infants with post-anal dimples and in 6/11 infants with no external post-coccygeal anomaly.

Glutathione Conjugation at the Blood-CSF Barrier Efficiently Prevents Exposure of the Developing Brain Fluid Environment to Blood-Borne Reactive Electrophilic Substances.

Exposure of the developing brain to toxins, drugs, or deleterious endogenous compounds during the perinatal period can trigger alterations in cell division, migration, differentiation, and synaptogenesis, leading to lifelong neurological impairment. The brain is protected by cellular barriers acting through multiple mechanisms, some of which are still poorly explored. We used a combination of enzymatic assays, live tissue fluorescence microscopy, and an in vitro cellular model of the blood-CSF barrier to investigate an enzymatic detoxification pathway in the developing male and female rat brai

Transcriptome analysis of the ependymal barrier during murine neurocysticercosis.

BACKGROUND: Central nervous system (CNS) barriers play a pivotal role in the protection and homeostasis of the CNS by enabling the exchange of metabolites while restricting the entry of xenobiotics, blood cells and blood-borne macromolecules. While the blood-brain barrier and blood-cerebrospinal fluid barrier (CSF) control the interface between the blood and CNS, the ependyma acts as a barrier between the CSF and parenchyma, and regulates hydrocephalic pressure and metabolic toxicity. Neurocysticercosis (NCC) is an infection of the CNS caused by the metacestode (larva) of Taenia solium and a m

Ciliated cell domains with locally coordinated ciliary motion generate a mosaic of microflows in the brain's lateral ventricles.

Circulation of cerebrospinal fluid (CSF) through the brain's ventricles is essential for maintaining brain homeostasis and supporting neurogenesis. CSF flow is supported by the structural polarization of multiciliated cells, which align with the flow direction. However, it remains unclear how the organization of tissue-wide polarity across the ciliary epithelium comprised of thousands of cells, determines the trajectory of the flow and efficient distribution of the CSF. Here, we used new approaches to analyze the organization of translational polarity across extensive areas of the lateral vent

Hydrocephalus due to multiple ependymal malformations is caused by mutations in the MPDZ gene.

Congenital hydrocephalus is considered as either acquired due to haemorrhage, infection or neoplasia or as of developmental nature and is divided into two subgroups, communicating and obstructive. Congenital hydrocephalus is either syndromic or non-syndromic, and in the latter no cause is found in more than half of the patients. In patients with isolated hydrocephalus, L1CAM mutations represent the most common aetiology. More recently, a founder mutation has also been reported in the MPDZ gene in foetuses presenting massive hydrocephalus, but the neuropathology remains unknown. We describe her

Preliminary electron microscopic observations on ependymal cells in the caudal segment of the filum terminale and the ampulla caudalis of Macaca fascicularis and Cercopithecus griseoviridis (primates, Cercopithecidae).

In a previous paper, the concept of the terminal organ (TO) of the subcommissural complex was forwarded. Functionally this complex is a neuro (glio-) hemal organ which serves to discharge the Reissner's secretory material into the systemic circulation. The TO is characterized by structural specializations that make feasible the discharge and chemical decomposition of the secretory material stowed in the massa caudalis (MC). The TO is probably not only the ampulla caudalis (AC); it may comprise even parts of the filum terminale next to the AC. The boundary of the TO is uncertain as yet. It cann

Cerebral sparganosis in a child with corpus callosum invasion: a case report.

BACKGROUND: Invasion of the corpus callosum by sparganosis is rare in children. After invading the corpus callosum, sparganosis has various migration modes, which can break through the ependyma and enter the ventricles, thus causing secondary migratory brain injury. CASE PRESENTATION: A girl aged 4 years and 7 months presented with left lower limb paralysis for more than 50 days. Blood examination showed that the proportion and absolute number of eosinophils in the peripheral blood were increased. Furthermore, enzyme-linked immunosorbent assay of serum and cerebrospinal fluid samples revealed

Central neurocytoma without intraventricular extension.

A case of central neurocytoma is reported, presenting as a tumor of the occipital lobe, closely related to the ependyma of the occipital horn of the lateral ventricle but not actually invading it. This is a distinctly uncommon location, and only four tumors have been reported so far in a lobar position, without an intraventricular component. The case broadens our knowledge of this recently described tumor and gives an opportunity for discussion of the role of radiotherapy in its management, which currently remains controversial.

Expression of proinflammatory cytokines tumor necrosis factor-alpha and interleukin-1beta in the brain during experimental group B streptococcal meningitis.

We performed mRNA in situ hybridization for TNF-alpha and IL-1beta from infant rats with group B streptococcal meningitis. Induction of both cytokines was seen in the ependyma and the meninges at 4 h. Both cytokines were expressed in the brain parenchyma at 12 h. Induction of IL-1beta mRNA was seen in vessels within the brain cortex. Neutrophilic infiltrate at all time points examined was minimal and could not account for the observed cytokine expression.

Functional herniated choroid plexus in brain parenchyma following VP shunt removal.

We report a 4-year-old male who presented with a blocked ventriculoperitoneal (VP) shunt inserted post excision of a WHO Grade 1 cerebellar pilocytic astrocytoma complicated post-operatively by pseudo meningocoele formation. Imaging revealed choroid plexus that had herniated along the shunt tract. Subsequent MRI showed development of cystic changes around the tract. The ectopic choroid plexus was still in continuity with the ventricular ependyma and was producing CSF in the left parietal lobe.

Dysembryoplastic neuroepithelial tumor located in pericallosal and intraventricular area in a child. Case report.

Dysembryoplastic neuroepithelial tumors (DNETs) are frequently seen in children and young adults with intractable epilepsy, and are typically located in the temporal cortex. Extracortical location of DNET is a very rare occurrence. The authors report on a child with a unique extracortical location of DNET with an extensive involvement to the supracallosal cistern and callosum, septum pellucidum, and lateral ventricle ependyma. The authors discuss the radiological and pathological characteristics of the lesion and reiterate the difficulty in differentiating the histological characteristics of c

Oligodendroglioma arising in heterotopic brain tissue of the soft palate and nasopharynx.

A child with heterotopic brain tissue in her soft palate and nasopharynx is presented. Within the mass there was a small oligodendroglioma. Eighteen previous cases of heterotopic brain tissue in the nasopharynx and soft palate are also reviewed. These differ from nasal gliomas in that the latter do not contain ependyma, choroid plexus, retinal components, or neoplastic tissue. Both nasal gliomas and heterotopic brain tissue in the soft palate and nasopharynx are believed to result from embryonic brain trapped in inappropriate areas during development.

Communicating hydrocephalus in newborn hamsters and cats following vaccinia virus infection.

Newborn hamsters and kittens infected with vaccinia virus developed communicating hydrocephalus without fibrotic changes in the meninges. Following intracerebral viral inoculation, a transient inflammation of the meninges, ependyma, and choroid plexus was found. One month later, 70% of hamsters infected with the WR strain of vaccinia virus and 35% infected with the vaccine strain developed hydrocephalus. The mechanism of hydrocephalus production is unclear, but differs from other models in which it is associated with stenosis of the aqueduct of Sylvius or fibrosis of the subarachnoid space.
