Obesity as an epigenetic continuum: developmental roots, adipose remodeling, and the limits of reversibility.

Obesity arises from the complex interplay between genetic and epigenetic factors, with developmental programming shaping adipose fate, energy homeostasis, and treatment responses later in life. Findings from the developmental origins of health and disease framework reveal that prenatal factors-such as maternal nutrition, adiposity, hyperglycemia, smoking, and stress-and early exposures imprint dynamic epigenetic marks-DNA methylation, histone modifications, and noncoding RNAs-on key tissues (adipose and hypothalamic circuits), which are associated with both childhood and adult adiposity. These

Transgenerational somatic mutations and epigenetic imprints on selected DNA repair gene promoters in Arabidopsis thaliana due to pesticides.

Pesticides are known to induce genome instability. Reports on eukaryotes, including humans, mostly covers chromosomal aberrations and DNA strand breaks. Studies on scoring of the rates of loci-specific somatic mutations such as point mutations (PM), somatic homologous recombination (SHR), frameshift mutation (FSM), and transpositions due to pesticides are mostly confined to prokaryotes, using Ames test. Analogous eukaryotic model systems though are available for Drosophila, mouse and certain cell lines, reports using these are meagre, possibly due to the lethality of commercially-used pesticid

Reproductive toxicity of benzo[α]pyrene: Effects, mechanisms, and perspectives.

Polycyclic aromatic hydrocarbons are pervasive environmental pollutants. Benzo[α]pyrene (B[α]P), a prototypical polycyclic aromatic hydrocarbon, elicits increasing apprehension due to its endocrine-disrupting properties and reproductive toxicity in animals; however, the toxicological effects and underlying mechanisms have not been thoroughly compiled and elucidated. This research delineates B[α]P toxicity in the reproductive systems of multiple species, including mammals, amphibians, fish, and invertebrates, alongside the relevant molecular and epigenetic evidence. In males, exposure to B[α]P

Antiviral innate immune memory in alveolar macrophages following SARS-CoV-2 infection ameliorates secondary influenza A virus disease.

Pathogen encounter can result in epigenetic remodeling that shapes disease caused by heterologous pathogens. Here, we examined innate immune memory in the context of commonly circulating respiratory viruses. Single-cell analyses of airway-resident immune cells in a disease-relevant murine model of SARS-CoV-2 recovery revealed epigenetic reprogramming in alveolar macrophages following infection. Post-COVID-19 human monocytes exhibited similar epigenetic signatures. In airway-resident macrophages, past SARS-CoV-2 infection increased activity of type I interferon (IFN-I)-related transcription fac

Testicular origin of epigenetic inheritance independent of sperm mitochondrial DNA and epididymal exposure.

Paternal epigenetic inheritance remains mechanistically unresolved. Recent studies propose that environmental exposures induce mitochondrial DNA (mtDNA)-dependent transcription in sperm during epididymal transit, altering small RNA content and offspring phenotypes. Here, we show that mature murine sperm are effectively devoid of mtDNA, precluding mtDNA-dependent transcription, and that sperm-borne mitochondrial RNAs originate during spermatogenesis. Testicular sperm transmitted diet-induced metabolic traits as efficiently as, and in most cohorts more efficiently than, epididymal sperm. These f

Metabolic imprinting drives epithelial memory during mucosal fungal infection.

Epithelial cells at barrier sites are emerging as active participants in innate immune memory, yet the underlying metabolic and epigenetic mechanisms remain unclear. Here, we uncover a previously unrecognized form of trained immunity in oral epithelial cells that enhances protection against fungal infection. Using a mouse model, we show that mucosal exposure to Candida albicans confers sustained protective memory that is independent of adaptive immunity and myeloid cells. Mechanistically, mucosal memory is driven by proline catabolism via proline dehydrogenase (Prodh) in epithelial cells, whic

Developmental exposure to cigarette smoke particulate matter induces cross-generational DNA methylation changes in zebrafish.

Prenatal exposure to cigarette smoke has been associated with adverse developmental outcomes and epigenetic alterations; however, the persistence of these effects across generations remains poorly understood. Using zebrafish (Danio rerio), we investigated whether developmental exposure to cigarette smoke total particulate matter (TPM) induces epigenetic changes in exposed larvae (F0) and their unexposed offspring (F1). Embryos were exposed to TPM (0.1, 0.2, or 0.3 μg/mL equi-nicotine units) from 6 to 96 h post fertilization (hpf). Transcript abundance of DNA methyl transferases, global methyla

Continuous DNA methylation deconvolution-based surrogate for B-cell differentiation state in chronic lymphocytic leukemia.

BACKGROUND: Chronic lymphocytic leukemia (CLL) is clinically divided categorically into IGHV mutated (M-CLL) and IGHV unmutated (U-CLL) subtypes. We aimed to generate a continuous metric of CLL epigenetic state to correspond to the many steps of B-cell differentiation from which CLL neoplasms may arise. METHODS: We measured genome-scale DNA methylation in purified CLL samples (n = 89) and utilized reference-based cell deconvolution techniques to develop a continuous metric of epigenetic similarity across a B-naive-like to B-memory-like scale (B-Index). We used B-Index to find epigenome-wide c

A narrative review of the epigenetics of post-traumatic stress disorder and post-traumatic stress disorder treatment.

Epigenetic research in post-traumatic stress disorder (PTSD) is essential, given that environmental stressors and fear play such a crucial role in its development. As such, it may provide a framework for understanding individual differences in the prevalence of the disorder and in treatment response. This paper reviews the epigenetic markers associated with PTSD and its treatment, including candidate genes and epigenome-wide studies. Because the etiopathogenesis of PTSD rests heavily on learning and memory, we also draw upon animal neuroepigenetic research on the acquisition, update and erasur

Continuous DNA Methylation Deconvolution-Based Surrogate for B-Cell Differentiation State in CLL.

Chronic Lymphocytic Leukemia (CLL) is clinically divided into IGHV mutated (M-CLL) and IGHV unmutated (U-CLL) subtypes, which are thought to arise from distinct cells of origin along the B-cell differentiation pathway. We measured genome-scale DNA methylation in purified CLL samples ( n = 89) and utilized reference-based cell deconvolution techniques to develop a continuous metric of epigenetic similarity across a B-naive-like to B-memory-like scale (B-Index). B-Index accurately classifies CLL into clinical subtypes (98.8%), has a stronger epigenetic signal than IGHV gene percent identity, and

Expression GWAS and Differential Methylome Profiling Reveal Upstream Regulators of Drought Memory Genes in Arabidopsis thaliana.

Plants adapt to recurrent drought through transcriptional memory, yet the upstream regulators remain largely unknown. This study integrated expression genome-wide association study (eGWAS) across 115 Arabidopsis thaliana ecotypes with differential methylome profiling to identify these regulators. Focusing on the memory genes LKR, HIS1-3, and DREB1A, eGWAS identified signaling and epigenetic loci involved in ABA/JA responses and DNA methylation. Methylome profiling by whole-genome bisulfite sequencing of ecotypes contrasting in drought tolerance, and in the superinduction of memory genes reveal

Epigenetic Regulation of the Hippocampus, with Special Reference to Radiation Exposure.

The hippocampus is crucial in learning, memory and emotion processing, and is involved in the development of different neurological and neuropsychological disorders. Several epigenetic factors, including DNA methylation, histone modifications and non-coding RNAs, have been shown to regulate the development and function of the hippocampus, and the alteration of epigenetic regulation may play important roles in the development of neurocognitive and neurodegenerative diseases. This review summarizes the epigenetic modifications of various cell types and processes within the hippocampus and their

Altered activity-regulated H3K9 acetylation at TGF-beta signaling genes during egocentric memory in Huntington's disease.

Molecular mechanisms underlying cognitive deficits in Huntington's disease (HD), a striatal neurodegenerative disorder, are unknown. Here, we generated ChIPseq, 4Cseq and RNAseq data on striatal tissue of HD and control mice during striatum-dependent egocentric memory process. Multi-omics analyses showed altered activity-dependent epigenetic gene reprogramming of neuronal and glial genes regulating striatal plasticity in HD mice, which correlated with memory deficit. First, our data reveal that spatial chromatin re-organization and transcriptional induction of BDNF-related markers, regulating

Intrauterine heat stress: molecular acclimation and epigenetic transmission in F1 and F2 male mice.

Heat stress (HS) during gestation poses a major physiological challenge to the developing fetus, potentially inducing long-term adaptations that persist across generations. Understanding this process is crucial for reproductive biology, as climate change poses challenges to fertility. This study evaluated the effects of prenatal HS (41 °C, 65% relative humidity) during early, late, or full-term pregnancy on postnatal development and reproductive function of F1 male mice. Additionally, the potential transmission of heat acclimation was evaluated by analyzing gene expression in the testes of F1

Genetic heritage of war: exploring transgenerational inheritance of PTSD and its molecular background.

This review article summarizes the available data about the potential link between war-related trauma and post-traumatic stress disorder (PTSD) and epigenetic alterations that could manifest in future generations. DNA methylation variations in stress-related genes such FKBP5, NR3C1, NR3C2, BDNF, and SLC6A4 have been seen in parents and/or their offspring in populations exposed to genocide, conflict, or combat. Certain results point to timing-dependent or parent-specific patterns, especially when maternal stress occurs during pregnancy. Results, however, are not consistent; some studies have fo

Stability of epigenetic transgenerational inheritance of adult-onset disease and parturition abnormalities.

Previous research on the generational stability of epigenetic transgenerational inheritance was conducted through a ten-generation study of all transgenerational generations in mammals. This study demonstrated both the stability of epigenetic inheritance across generations and demonstrated a generational increase incidence of disease pathology. Building on this research, the present study follows the same lineage of rats with ancestral vinclozolin exposure through twenty generations. The findings offer important insights into long-term mammalian models of epigenetic transgenerational inheritan

In Vivo CRISPR Screening Reveals CHD7 as a Positive Regulator of Short-lived Effector Cells.

CD8+ T cells differentiate into two subpopulations in response to acute viral infection: memory precursor effector cells (MPECs) and short-lived effector cells (SLECs). MPECs and SLECs are epigenetically distinct; however, the epigenetic regulators required for formation of these subpopulations are mostly unknown. In this study, we performed an in vivo CRISPR screen in murine naive CD8+ T cells to identify the epigenetic regulators required for MPEC and SLEC formation, using the acute lymphocytic choriomeningitis virus Armstrong infection model. We identified the ATP-dependent chromatin remode

Altered sperm DNA methylation provides an epigenetic route from overweight and older men to intergenerational disease transmission.

The occurrence of childhood neurodevelopmental disorders has been steadily increasing for decades, yet we have little understanding of modes of inheritance implicated in these diseases. Epidemiology studies have revealed an association between an elevated paternal Body Mass Index (BMI) and age as factors implicated in increased risk for neurodevelopmental disorders in children. Epigenetic regulation of gene expression by DNA methylation at cytosines is susceptible to environmental, diet, and lifestyle changes. Yet, whether a man's BMI and age influence the sperm methylome and potentially impac

BAF53b, a Neuron-Specific Nucleosome Remodeling Factor, Is Induced after Learning and Facilitates Long-Term Memory Consolidation.

Although epigenetic mechanisms of gene expression regulation have recently been implicated in memory consolidation and persistence, the role of nucleosome-remodeling is largely unexplored. Recent studies show that the functional loss of BAF53b, a postmitotic neuron-specific subunit of the BAF nucleosome-remodeling complex, results in the deficit of consolidation of hippocampus-dependent memory and cocaine-associated memory in the rodent brain. However, it is unclear whether BAF53b expression is regulated during memory formation and how BAF53b regulates fear memory in the amygdala, a key brain
