Diseases of the central nervous system (CNS) have historically been among the most difficult to treat using conventional pharmacological approaches. This is due to a confluence of factors, including the limited regenerative capacity and overall complexity of the brain, problems associated with repeated drug administration, and difficulties delivering drugs across the blood-brain barrier (BBB). Viral-mediated gene transfer represents an attractive alternative for the delivery of therapeutic cargo to the nervous system. Crucially, it usually requires only a single injection, whether that be a ge

Degenerative musculoskeletal disorders, including osteoarthritis (OA), intervertebral disc degeneration (IVDD), and osteoporosis (OP), are increasingly viewed as disorders of mitochondrial quality control (MQC) rather than passive mechanical wear. The MQC network, comprising mitochondrial dynamics, mitophagy-biogenesis coupling, and inter-organelle communication, organizes the aging-inflammation-metabolism axis. Its collapse releases mtDNA fragments that act as damage-associated molecular patterns (DAMPs) activating innate immunity via cGAS-STING, and metabolic intermediates that remodel the e

Pituitary neuroendocrine tumors (PitNETs), which were formerly known as pituitary adenomas, are classified in 5th Edition of the WHO Classification of Endocrine and Neuroendocrine Tumors. Since thyrotroph PitNETs are rare PitNETs, most previous studies about former thyroid stimulating hormone (TSH)-secreting pituitary adenoma have focused on a small number of cases. However, the diagnostic rate of thyrotroph PitNET has increased because of increased sensitivity of serum TSH measurement and widespread recognition that thyrotroph PitNETs are the cause of syndrome of inappropriate secretion of TS

Bromodomain and extraterminal domain inhibitors (BETi) represent promising therapeutic agents for metastatic melanoma, yet their mechanism of action remains unclear. Here we interrogated the transcriptional effects of BETi and identified AMIGO2, a transmembrane molecule, as a BET target gene essential for melanoma cell survival. AMIGO2 is upregulated in melanoma cells and tissues compared to human melanocytes and nevi, and AMIGO2 silencing in melanoma cells induces G1/S arrest followed by apoptosis. We identified the pseudokinase PTK7 as an AMIGO2 interactor whose function is regulated by AMIG

Plant tissue culture is a crucial part of biotechnology that supports crop improvement, plant conservation, and other related fields. Although tissue culture has been successfully used in many plants, low regeneration and transformation rates still exist in some species. Agrobacterium-mediated transformation is commonly used in genetic engineering, but its effectiveness depends heavily on establishing a reliable in vitro regeneration system through organogenesis or somatic embryogenesis. Over the past decade, substantial progress has been made in understanding the molecular basis of regenerati

UNLABELLED: The strongest type 2 diabetes signal in Indigenous Americans from Arizona is in intron 15 of KCNQ1. We previously identified a functional region in this intron that contained four type 2 diabetes-associated single nucleotide polymorphisms (SNPs) (rs2299620, rs2237896, rs2237897, and rs74046911) and demonstrated functionality of this fragment using Indigenous American induced pluripotent stem cell (iPSC)-derived pancreatic islets (SC-islets). In the current study, we used isogenic iPSCs with targeted edits at these four SNPs, and three additional SNPs (rs2237895, rs60808706, and rs2

Ischemic stroke (IS) remains a leading cause of disability and mortality in aging populations. Recovery trajectories are shaped not only by the acute vascular insult but also by pre-existing comorbidities, genetic predisposition, and age-dependent molecular remodeling. Common vascular risk factors, such as diabetes mellitus, atrial fibrillation, hypertension, and dyslipidemia, sustain systemic and cerebral inflammation, promote endothelial dysfunction, disrupt blood-brain barrier integrity, and impair neuroplasticity, collectively limiting neurovascular repair and recovery potential. Inhibitors of the menin-KMT2A interaction are promising agents for the treatment of KMT2A-rearranged leukemias. We evaluated menin inhibition in patient-derived xenografts of KMT2A-rearranged leukemias with high-risk features. Three acute myeloid leukemias with high-risk fusion partners (mixed-lineage leukemia-10 [MLLT10] and mixed-lineage leukemia-4 [MLLT4]) and two infant acute lymphocytic leukemia (ALL) samples were sensitive to menin inhibition. We also evaluated serial samples from two patients with multiply relapsed ALL. We found that highly pretreated KMT2A::AFF1 ALL samples were much l

Flowering time is a critical developmental process that directly influences reproductive success, environmental adaptation, and agricultural productivity in Brassica crops. Recent climate instability, including drought, temperature fluctuations, salinity, and irregular photoperiods, has intensified the need to understand the molecular mechanisms regulating flowering adaptation. WRKY transcription factors are key integrators of flowering time. They connect environmental cues, hormone networks, and circadian rhythms to floral regulator genes. This review summarizes the structural, evolutionary,

Major depressive disorder (MDD) is a highly prevalent and heterogeneous psychiatric disorder shaped by the interplay of genetic, environmental, and epigenetic factors. Histone modifications, particularly acetylation and methylation, have emerged as critical regulators of chromatin dynamics and gene expression in stress adaptation, neuroplasticity, and emotional regulation. This review synthesizes current evidence linking dysregulated histone deacetylases (HDACs) and histone methyltransferases (HMTs) to impaired neuroplasticity, neuroinflammation, mitochondrial dysfunction, and hypothalamic-pit

BackgroundGene silencing is widely recognized as a promising therapeutic approach for dominant monogenic disorders. Current silencing strategies, many of which are transient, utilize RNA interference. Gene silencing may also be achieved through directed epigenetic editing using a CRISPR/dCas9 effector fused to DNA methyltransferase 3A (dCas9-DNMT3A). We used this system to direct DNA methylation to HTT, the causal gene underlying the autosomal dominant neurodegenerative disorder Huntington's disease, to assess the translational potential of this strategy for treating a genetic neurological dis

Chronic pain is sustained by durable yet dynamic changes in gene regulation across sensory neurons, spinal circuits, glial and immune cells, and higher brain regions. This review critically evaluates three related but conceptually distinct regulatory layers: chromatin-associated epigenetic mechanisms, including DNA methylation and hydroxymethylation, histone modifications, and chromatin accessibility; non-coding RNAs that directly interface with chromatin-regulatory machinery; and RNA N6-methyladenosine (m6A), an epitranscriptomic mechanism that acts primarily at the RNA level. Across pain mod

Phytomelatonin has long been viewed as a pleiotropic stress signal that protects plants by scavenging reactive oxygen species (ROS), reinforcing antioxidant capacity, and coordinating hormonal responses. That view remains correct, but it is no longer sufficient. Recent work indicates that phytomelatonin also shapes the regulatory architecture that determines whether stress-responsive genes are accessible, whether transcripts are processed and translated efficiently, and whether stress-induced states can persist after the initial stimulus has disappeared. Here, we synthesize four emerging layer

Autosomal recessive polycystic kidney disease (ARPKD) leads to severe renal cysts and progressive kidney dysfunction, with no approved treatments. The absence of such cystic phenotypes in Pkhd1-/- mice underscores the need for novel models that better recapitulate the human disease. We developed kidney organoid-on-chip models that mimic patients' distal-nephron cysts, identifying RAC1/c-FOS as potential therapeutic targets. However, critical questions remain regarding RAC1 activation during cyst formation, cyst origins, and underlying molecular mechanisms. Using a multifaceted approach, organo

Prader-Willi Syndrome (PWS) is a genetic neurodevelopmental disorder caused by the loss of expression of paternal genes in the 15q11-q13 locus. The same genes on the maternal allele are present but are epigenetically silenced. This genetic alteration causes a spectrum of symptoms associated with hypothalamic dysfunction. Research is increasingly focused on setting strategies aiming to reactivate the maternal locus and restore the expression of that critical gene cluster. Early therapeutic efforts explored broad-spectrum epigenetic drugs, such as DNA- and histone-methyltransferase inhibitors. W

Tuberculosis (TB) continues to be a significant global health issue, with anti-tuberculosis drug-induced liver injury (ATDILI) representing the most frequent and severe adverse event associated with first-line therapy. Despite extensive investigation, the mechanistic basis of ATDILI remains incompletely understood. Recent evidence underscores the critical role of epigenetic regulation, particularly DNA methylation, in modulating the expression of cytochrome P450 (CYP) enzymes and influencing drug metabolism and hepatotoxicity. This review synthesizes current knowledge on the pharmacoepigenetic

The bidirectional relationship between epilepsy and depression illustrates shared neurobiological mechanisms of neuroinflammation, hypothalamic-pituitary-adrenal axis dysregulation, and glutamatergic dysfunction. Depression is present in 20-55% of people with epilepsy, far greater than in the general population, while depression doubles epilepsy risk 2.5-fold, indicating shared pathophysiology. Neuroinflammatory mediators (interleukin-6, tumor necrosis factor alpha, high-mobility group box 1) establish a vicious cycle: seizures exacerbate inflammation and mood disruption, and stress lowers sei

Renal fibrosis is the final common pathway of progressive chronic kidney disease and is maintained by spatially heterogeneous interactions among injured epithelial cells, activated fibroblasts, immune cells, extracellular matrix remodeling, metabolic stress, and persistent profibrotic transcriptional programs. Current therapies slow renal functional decline but do not directly control the regulatory circuits that stabilize maladaptive repair. Photoresponsive renal nanomedicine offers a potential strategy to add external control to anti-fibrotic intervention by combining kidney-directed deliver

BACKGROUND: Quantitative measures of autism spectrum disorder (ASD)-related traits can provide insight into trait presentation across the population. Previous studies have identified epigenomic variation associated with ASD diagnosis, but few have evaluated quantitative traits. We sought to identify DNA methylation patterns in child blood associated with Social Responsiveness Scale score, Second Edition (SRS).
METHODS: We conducted an epigenome-wide association study of SRS in child blood at approximately age 5 in the Study to Explore Early Development, a case-control study of ASD in the Unit
