Neuropsychiatric disorders such as anxiety and depression have formidable economic and societal impacts. A dysregulation of the hypothalamo-pituitary-adrenal (HPA) axis leading to elevated endogenous glucocorticoid levels is often associated with such disorders. Chronically high glucocorticoid levels may act upon the central nucleus of the amygdala (CeA) to alter normally adaptive responses into those that are maladaptive and detrimental. In addition to glucocorticoids, other steroid hormones such as estradiol and androgens can also modify hormonal and behavioral responses to threatening stimu

The aim of the present study was to investigate differences in the expression of mRNAs for ERalpha, ERbeta and PR in the sow uterus at different stages of the estrous cycle as well as in inseminated sows at estrus and during early pregnancy by use of solution hybridization and in relation to plasma levels of estradiol and progesterone. Uterine samples were collected at different stages of the estrous cycle and after insemination/early pregnancy. In the endometrium, the expression of ERalpha mRNA and PR mRNA was similar for cyclic and early pregnant groups. Both were highest at early diestrus/7

Both epidemiological and experimental findings suggest the possible roles of sex steroids in the pathogenesis and/or development of various human thyroid disorders. In this study, we evaluated the expression of estrogen receptors (ER) alpha and beta in normal thyroid glands (N = 25; female: n = 13, male: n = 10, unknown: n = 2) ranging in age from fetus to adult. Furthermore, using immunohistochemistry, we investigated the expression of ERalpha and beta in 206 cases of thyroid disorders, including 24 adenomatous goiters, 23 follicular adenomas, and 159 thyroid carcinomas. In addition, we also

17β-Estradiol (E2) may influence some of the sex differences in stress-related neuropsychiatric disorders, such as anxiety and depression. E2 may also modulate the hypothalamic-pituitary-adrenal axis function as a cortisol response to stress in women. This study explored the role of E2 (10 and 50 μg/rat) and selective estrogen receptor modulators: diarylpropionitrile (DPN, 10 μg/rat) and propyl pyrazole triol (PPT, 10 μg/rat), in anxiety and depressive behaviors in ovariectomized rats using an animal model. The study also examined the relationship between rats' affective behavior and the plasm

Induction of polyovular follicles (PFs) containing two or more oocytes in a follicle is one example. In this study, the involvement of estrogen receptor subtypes ERalpha and ERbeta in induction of PFs by neonatal treatment with DES was analyzed by using ERalpha knockout (alphaERKO) and ERbeta knockout (betaERKO) mice. Ovaries of mice injected with 3 microg DES for 5 days from the day of birth were examined histologically from 10 to 60 da

A whole cell technique to measure estradiol receptors in cultured rabbit endometrial cells is described. The estradiol receptors measured appear to be composed of at least two components: one component has high affinity but low capacity, while the other component has low affinity but high capacity. Using the assay, the effects of estradiol and progesterone pretreatment were examined on the estradiol receptor levels. It was found that both of the hormones decreased the number of estrogen receptors in the cultured cells. The finding that estradiol decreased its own receptors was unexpected and i

Humans and other vertebrates contain two estrogen receptors (ERs), ERα and ERβ, which mediate the physiological actions of three estrogens: estrone (E1), estradiol (E2) and estriol (E3). Of these three estrogens, in vivo, E2 is the strongest transcriptional activator of ERα and ERβ, E1 is next most active, followed by E3. We studied transcriptional activation of human ERα and ERβ by E2, E1 and E3 in African green monkey kidney (COS-7) cells, which we compared with studies of estrogen stimulation of ER transcription in human embryonic kidney (HEK-293) cells. To our surprise, in COS-7 cells, E3

Female ESR2-null mice (betaERKO) display defects in ovarian function and are subfertile. Follicular maturation is impaired and explains smaller litters, but betaERKO also produce fewer litters, which may be partially due to inadequate ovulatory signals. To test this, the amplitude and timing of the naturally occurring luteinizing hormone (LH) surge was measured in individual intact betaERKO and wild-type (WT) mice. Vaginal cytology was evaluated daily, and blood samples were taken from mice in proestrus. The amplitude of the LH surge was severely blunted in betaERKO mice compared to WT, but pi

Estrogen and glucocorticoids interact in multiple aspects of endocrine regulation by exerting opposing influences on the expression of selective genes. In rats, estrogen receptor (ER)-beta is the predominant form of ER present in the hypothalamic paraventricular (PVN) and supraoptic (SON) nuclei, suggesting its involvement in neuroendocrine regulation. To date, the hormonal regulatory profile of the ERbeta gene in the rat central nervous system has not been closely elucidated. In the present study, we first examined the effects of dexamethasone (DEX) and estradiol benzoate (EB) on the ERbeta p

Obesity contributes to worse outcomes in breast cancer, particularly in the luminal subtype, where cellular senescence could increase tumor aggressiveness. Parallelly, Estrogen Receptor Beta (ERβ) has emerged as an important mediator in the cellular response to obesity-associated inflammation that metformin could counteract. This study explores metformin's role in targeting senescence to mitigate obesity-induced tumor progression. Using the GSE189757 dataset, differentially expressed genes in obese versus lean luminal breast cancer patients were examined by in silico analysis. T47D, BT474, and

BACKGROUND: Endometriosis is a chronic, estrogen-dependent disease characterized by the presence of endometrial-like tissue growing outside the uterus. The molecular and clinical heterogeneity of endometriosis complicate diagnostic and treatment options -- diagnostic delays of seven to ten years are common and therapies often lack long-term efficacy. Estrogen signaling and estrogen receptor beta (ERβ) expression is thought to be increased in endometriosis, contributing to increased cell proliferation in lesions. The "ERβ dominance hypothesis" is a prevailing hypothesis in the field, setting ER

BACKGROUND: Estrogen controls the pubertal growth spurt, growth plate closure, and accretion of bone mineral density (BMD) of long bones after biding estrogen receptor (ER). There are two subtypes of ER, ERα and ERβ. If each ER subtype has different effects, we may control those actions by manipulating the estrogen binding intensity to each ER subtype and increase the final adult height without markedly reducing BMD or impairing reproductive functions. The purpose of our study was to compare these effects of ERα and ERβ on long bones in ovariectomized rats.
METHODS: Thirty female rats were ov

Estradiol (E(2)) modulates affective and socio-sexual behavior of female rodents. E(2)'s functional effects may involve actions through alpha and beta isoforms of estrogen receptor (ERs). The importance of E(2)'s actions at these isoforms for anxiety (open field, elevated plus maze), depression (forced swim test), and sexual behavior (lordosis) was investigated using an antisense oligonucleotide (AS-ODN) strategy. If ERbeta is required for anti-anxiety and antidepressant-like effects, and ERalpha is required for sexual receptivity, of E(2), then intracerebroventricular administration of AS-ODN

BACKGROUND: The autoimmune thyroid diseases (AITDs), such as Graves' disease (GD) and Hashimoto's thyroiditis (HT), appear to develop as a result of complex interactions between predisposing genes and environmental triggers. Susceptibility to AITDs is conferred by genes in the human leukocyte antigen (HLA) and genes unlinked to HLA, including the CTLA-4 gene. Recently, estrogen receptor (ER) beta, located at human chromosome 14q23-24.1, was identified. We analyzed a dinucleotide (CA)n repeat polymorphism located in the flanking region of ERbeta gene in patients with AITDs and in normal subject

Genetic variation in genes involved in estrogen biosynthesis, metabolism and signal transduction have been suggested to play a role in breast cancer. To determine the possible contribution of genetic variation in the ESR1 (ER-alpha), ESR2 (ER-beta) and AR genes in breast cancer risk the -1174(TA)(7-27), c. 1092+3607(CA)(10-26) and c. 172(CAG)(6-40) repeat variants were studied in a case-control study of 79 women with sporadic breast cancer and 155 controls. No significant difference was observed in the frequency distribution of -1174(TA)(7-27) in the ESR1 gene between patients and controls, wh

The function of estrogen receptor beta (ER-beta) in mammary tissue is not completely understood. While early observations were often conflicting, more recent data suggest an important role as a tumor-suppressor gene. A decrease of ER-beta expression has been observed in ductal carcinoma in situ and invasive carcinoma as compared with benign mammary epithelial cells. The loss of ER-beta resulted in abnormal growth of mammary epithelial cells. We have previously shown that the mRNA expression of the ER-beta gene is almost totally suppressed in breast carcinomas from patients with a poor prognosi

An antibody, GC-17, thoroughly characterized for its specificity for estrogen receptor-beta (ER-beta), was used to immunolocalize the receptor in histologically normal prostate, prostatic intraepithelial neoplasia, primary carcinomas, and in metastases to lymph nodes and bone. Comparisons were made between ER-beta, estrogen receptor-alpha (ER-alpha), and androgen receptor (AR) immunostaining in these tissues. Concurrently, transcript expression of the three steroid hormone receptors was studied by reverse transcriptase-polymerase chain reaction analysis on laser capture-microdissected samples
