[Ethanol Instillation for Mediastinal Cyst].

Mediastinal cyst rarely becomes symptomatic due to compression of adjacent organs. In such case, treatment has to be considered. Either surgical extirpation or percutaneous decompression is the treatment of choice. A 53 years old woman who had been diagnosed with intractable epilepsy, mental retardation and Chilaiditi syndrome was admitted to our hospital because of dyspnea and difficulty in conversation. Chest computed tomography (CT) revealed a mediastinal cystic lesion which expanded to the right chest cavity and occupied most of the right chest cavity. That cyst was treated by percutaneous

Social Factors in Ethanol Sensitization.

Behavioral sensitization is a neuroadaptive process characterized by an increase in a particular behavior after repeated exposure to drugs or other stimuli, such as stress. Sensitization can also be extended to neurochemical and neuroendocrine sensitization. Several factors can influence sensitization to the effects of ethanol. For instance, stress is an important component in addiction that can strengthen ethanol-induced behaviors. In animal models, stressful situations can be induced by alterations in social aspects of the animal's environment, such as maternal separation, social conflicts,

Does ethanol enhance cocaine toxicity?

OBJECTIVES: Results of in vitro and animal studies suggest that ethanol enhances cocaine toxicity. If so, then that this implies that in ethanol users, postmortem blood cocaine concentrations should be lower, or anatomic evidence demonstrable. METHODS: Drug concentrations and autopsy findings were compared in a sample of 72 accidental deaths, where only cocaine, cocaine metabolites, and ethanol were detected. Findings in ethanol positive (E+) and negative (E-) deaths were compared using multiple Student's t-tests and chi2 testing for categorical variables. RESULTS: There were 47 E-decedents

First pass metabolism of ethanol.

The human stomach has both low and high K(m) ADH isozymes, resulting in significant ethanol metabolism in gastric cells in vitro, and decreased bioavailability of ethanol (first pass metabolism: FPM) in vivo. Intraduodenal or intraportal infusion of amounts of ethanol equivalent to those emptied into the duodenum or disappearing from pylorus-ligated stomachs produced significantly higher blood levels than intragastric administration, whereas portal ligation had no effect, documenting the role of gastric ethanol metabolism in vivo. This "protective barrier" against the systemic effects of ethan

Genetic determinants of ethanol reinforcement.

In this paper, we present examples of some of the several behaviors which have been taken to indicate the reinforcing efficacy of drugs, including ethanol. Efforts to identify the genetic determinants of these behaviors have employed diverse pharmacogenetic methods. For example, we have used selective breeding to develop mice selected for severe or attenuated ethanol withdrawal and have found that Withdrawal Seizure Prone mice show a greater conditioned preference for ethanol-associated locations than the selected Withdrawal Seizure Resistant line. Similarly, HOT mice, selected for insensitivi

Genetic components of ethanol responses.

A powerful technique for determining the role of a particular neurotransmitter in mediating a response to ethanol (EtOH) is the analysis of selectively bred lines of animals. Lines selected for sensitivity and resistance to an EtOH effect differ principally in gene frequencies for genes affecting the selected response. Hence, other differences between the lines are likely due to pleiotropic actions of those genes. We discuss behavioral pharmacological experiments in two sets of selected lines. Withdrawal Seizure-Prone (WSP) and -Resistant (WSR) mouse lines were selected for severe and minimal

Urinary ethanol and diabetes mellitus.

Ethanol concentrations were assayed in urine samples obtained from 10 newly presenting diabetic patients (aged 50-69 years) with glycosuria and symptoms suggesting genito-urinary candidiasis. No alcohol was detected initially with diabetic urine samples, but when stored at room temperature 8 out of 10 samples showed increasing levels of ethanol from day 3 to day 20. Peak levels were more than 10,000 mg/l in three samples and more than 1500 mg/l in six samples. Studies were also performed using normal urine and urine to which was added glucose and/or Candida. Samples were kept at either room te

Ethanol abuse and heart disease.

The toxic effects of chronic ethanol abuse on cerebral and hepatic function have long been recognized. The role of ethanol abuse as an etiologic factor in heart disease is less clear and is often attributed to coexistent malnutrition. However, malnutrition has been dissociated from ethanol use in many patients with congestive cardiomyopathy. Studies in various animals provide major support for the role of ethanol as a toxic agent when used in large amounts for a prolonged period. Abnormalities that result from ethanol in test animals include depression of left ventricular performance and metab

S-Allyl-L-cysteine sulfoxide, a garlic odor precursor, suppresses elevation in blood ethanol concentration by accelerating ethanol metabolism and preventing ethanol absorption from gut.

Alcoholic beverages are enjoyed together with meals worldwide, but their excessive intake is associated with an increased risk of various diseases. We investigated whether S-allyl-L-cysteine sulfoxide (ACSO), a sulfuric odor precursor of garlic, suppresses elevation in plasma ethanol concentration by accelerating ethanol metabolism and preventing ethanol absorption from the gut in rats. ACSO and garlic extract with a high ACSO content (Garlic-H) suppressed elevation in concentrations of ethanol and acetaldehyde in plasma and promoted the activities of alcohol dehydrogenase and aldehyde dehydro

Effects of pregnanolone and dehydroepiandrosterone on ethanol intake in rats administered ethanol or saline during adolescence.

BACKGROUND: Adolescent alcohol use may contribute to long-term changes in the receptors and neuroactive steroids that may mediate its effects and to subsequent alcohol abuse and dependence as an adult. Therefore, in this study, ethanol preference and intake as an adult were examined after adolescent ethanol or saline administration. In addition, ethanol intake in the same groups was examined after administration of 2 neuroactive steroids with modulatory effects at GABA(A) receptors. METHODS: Two groups of male Long-Evans rats were administered 15 intraperitoneal (i.p.) injections of either et

Long lasting effects of rearing by an ethanol-consuming dam on voluntary ethanol consumption by rats.

For exposure to alcohol early in life to potentiate alcohol abuse in adolescence or adulthood, consequences of early exposure to alcohol must be of considerable duration. In two experiments using Norway rats as subjects, we examined effects of exposure during weaning to a dam consuming ethanol on adolescents' later affinity for ethanol. In a preliminary experiment, we offered rat pups a choice between 8% ethanol and water for 7 days immediately after they were weaned at 26 days of age. Pups whose dam had ingested 8% ethanol for 6 days either immediately or 1 week before we weaned them drank mo

Dpp6 knockout mice exhibit increased ethanol conditioned place preference and acute ethanol-induced anxiolytic behavior.

BACKGROUND: The gene DPP6 has been associated with behavioral phenotypes of alcohol use disorder (AUD) in recent human genome-wide association studies. DPP6 encodes an auxiliary subunit that modulates A-type voltage-gated potassium channels, particularly Kv4.2. METHODS: To further assess the role of this gene in ethanol-related traits, we tested Dpp6 knockout (KO) and wild-type mice for ethanol (EtOH) conditioned place preference (CPP), locomotor activity, and ethanol-induced anxiolysis. RESULTS: Male homozygous KO mice (HOM) showed greater preference for the ethanol-paired (2 g/kg) context

Baicalein blocks ethanol effects on dopamine transmission in the mesolimbic system and ethanol self-administration.

The dopamine (DA) projection from the ventral tegmental area (VTA) to the nucleus accumbens (NAc) is implicated in ethanol (EtOH) reward. While EtOH increases VTA DA neuron firing and basal DA release, it paradoxically reduces evoked DA release in the NAc. This study evaluated whether baicalein, a flavonoid antioxidant from Scutellaria baicalensis, modulates EtOH-induced inhibition of evoked DA release and EtOH reward. Using fast-scan cyclic voltammetry (FSCV), baicalein (10-100 μM) alone did not affect evoked DA release but significantly attenuated EtOH-induced inhibition (80 mM) in mouse NAc

Benefits of minimal ethanol volume and retention without removal in transvaginal ethanol sclerotherapy for endometriomas.

PURPOSE: This study aims to introduce an effective protocol for transvaginal ethanol sclerotherapy, focussing on the aspiration of viscous cystic content and the amount of ethanol instilled in the cystic cavity according to cyst size. METHODS: We retrospectively reviewed the medical records of 90 women who underwent transvaginal ethanol sclerotherapy from 2015 to 2022 at Good Moonhwa Hospital, Busan, Republic of Korea. The volume of ethanol used for sclerotherapy was determined based on the initial volume of aspirated cystic content, with a target limit of 30 cc, and the injected alcohol was

Determination of volatile impurities and ethanol content in ethanol-based hand sanitizers: Compliance and toxicity.

This study aimed to assess volatile impurities and ethanol content in ethanol-based hand sanitizers. A total of 31 different brands of hand sanitizers were analyzed using headspace gas chromatography-mass spectrometry to detect impurities and determine alcohol content for compliance. Volatile impurities were identified through Mass Spectrometry database analysis, and regression analysis was employed to ascertain ethanol percentage. Furthermore, a simulated toxicological analysis was conducted to evaluate the potential toxic effects associated with hand sanitizer usage. The detected impurities

HIV-1 Tat protein exposure potentiates ethanol reward and reinstates extinguished ethanol-conditioned place preference.

Exposure to HIV-1 trans-activator of transcription (Tat) protein potentiates the psychostimulant effects of cocaine, but the functional consequences of the interaction between HIV-1 Tat and other abused drugs is poorly understood. We hypothesized that exposure to HIV-1 Tat would potentiate the rewarding effects of ethanol. GT-tg transgenic mice, where Tat protein is conditionally expressed in brain by a doxycycline-dependent GFAP-linked promoter, were used to test the effects of Tat on ethanol-conditioned place preference (CPP). Compared to uninduced littermates or doxycycline-treated C57BL/6J

Metabolic products of [2-(13) C]ethanol in the rat brain after chronic ethanol exposure.

Most ingested ethanol is metabolized in the liver to acetaldehyde and then to acetate, which can be oxidized by the brain. This project assessed whether chronic exposure to alcohol can increase cerebral oxidation of acetate. Through metabolism, acetate may contribute to long-term adaptation to drinking. Two groups of adult male Sprague-Dawley rats were studied, one treated with ethanol vapor and the other given room air. After 3 weeks the rats received an intravenous infusion of [2-(13) C]ethanol via a lateral tail vein for 2 h. As the liver converts ethanol to [2-(13) C]acetate, some of the a

A 3-day exposure to 10% ethanol with 10% sucrose successfully initiates ethanol self-administration.

The initiation phase of ethanol self-administration is difficult to study using the well-established, sucrose-fading procedure due to the changing concentrations of ethanol in the first few days. The purpose of this experiment was to test whether a modified sucrose-substitution procedure in which rats are initially exposed to high concentrations of ethanol and sucrose for three days would successfully initiate ethanol self-administration. Male Long-Evans rats were trained to lever-press with a 10% sucrose solution in which four or 20 responses allowed 20-min access to the solution. Subsequentl
