Molecular detection and phylogenetic analysis of Hepatozoon spp. in questing Ixodes ricinus ticks and rodents from Slovakia and Czech Republic.

By amplification and sequencing of 18S rRNA gene fragments, Hepatozoon spp. DNA was detected in 0.08 % (4/5057) and 0.04 % (1/2473) of questing Ixodes ricinus ticks from Slovakia and Czech Republic, respectively. Hepatozoon spp. DNA was also detected in spleen and/or lungs of 4.45 % (27/606) of rodents from Slovakia. Prevalence of infection was significantly higher in Myodes glareolus (11.45 %) than in Apodemus spp. (0.28 %) (P < 0.001). Sequencing of 18S rRNA Hepatozoon spp. gene amplicons from I. ricinus showed 100 % identity with Hepatozoon canis isolates from red foxes or dogs in Europe. P

Persistence of Hepatozoon americanum (Apicomplexa: Adeleorina) in a naturally infected dog.

To determine the persistence of Hepatozoon americanum in a naturally infected dog, skeletal muscle biopsies were performed at approximately 6-mo intervals over a period of 5.5 yr, and the samples were examined for presence of lesions of American canine hepatozoonosis (ACH). Nymphal Amblyomma maculatum (Gulf Coast tick) were allowed to feed to repletion on the dog periodically over the 5.5-yr period, and adult ticks were dissected and examined for presence of H. americanum oocysts. With 3 exceptions, the biopsied muscle contained lesions characteristic of ACH; no evidence of infection was found

Canine hepatozoonosis: comparison of lesions and parasites in skeletal muscle of dogs experimentally or naturally infected with Hepatozoon americanum.

We report previously undescribed, early lesions in skeletal muscle of dogs experimentally infected with Hepatozoon americanum by ingestion of laboratory-reared, infected Amblyomma maculatum. The earliest muscle lesion was recognized at the first interval of examination 3 weeks following exposure. The lesion consisted of a large, modified host cell whose cytoplasm frequently contained a demonstrable parasite. In skeletal muscle, the cell was consistently located between muscle fibers or in loose connective tissue adjacent to those fibers. Evidence suggesting that the parasite arrives in muscle

Uniform staining of Cyclospora oocysts in fecal smears by a modified safranin technique with microwave heating.

Cyclospora, a coccidian protist, is increasingly being identified as an important, newly emerging parasite that causes diarrhea, flatulence, fatigue, and abdominal pain leading to weight loss in immunocompetent persons with or without a recent travel history as well as in patients with AIDS. Modified Kinyoun's acid-fast stain is the most commonly used stain to identify the oocyst of this parasite in fecal smears. Oocysts of Cyclospora stain variably by the modified acid-fast procedure, resulting in the possible misidentification of this parasite. We examined fecal smears stained by six differe

First autochthonous case of clinical Hepatozoon felis infection in a domestic cat in Central Europe.

Three different Hepatozoon (Apicomplexa, Hepatozoidae) species have been described infecting domestic cats in Europe (i.e. H. felis, H. canis and H. silvestris), however, reports on clinical hepatozoonosis are uncommon and treatment protocols are not clearly defined. A six-year-old male European short-hair cat from Austria presented poor general condition, lethargy, anorexia, icterus, a painful abdomen, fever, ruffled hair and a tick infestation, and it had never left Austria. Laboratory tests revealed leukopenia, thrombocytopenia and increased serum levels of symmetric dimethylarginine (SDMA)

First report of Babesia gibsoni in Central America and survey for vector-borne infections in dogs from Nicaragua.

BACKGROUND: Although many vector-borne diseases are important causes of morbidity and mortality in dogs in tropical areas and potential zoonoses, there is little information on these conditions in Central America. METHODS: Seven qPCRs for vector-borne pathogens were performed on a Roche LightCycler PCR Instrument to investigate their prevalence in a convenience sample of whole blood samples from apparently healthy dogs in Nicaragua. Also, a qPCR targeting the canine hydroxymethylbilane synthase (HMBS) gene was used as an endogenous internal control and verified the quality and quantity of DNA

Mass spectrometry-based serum peptidomic profiling reveals potential biomarker for canine hepatozoonosis.

Canine hepatozoonosis, caused by Hepatozoon canis, is a significant canine vector-borne disease (CVBD) with a complex life cycle and diverse clinical manifestations, ranging from asymptomatic to severe illness. While several diagnostic methods are available, no studies have yet applied serum peptidomic analysis to this disease. This study aimed to identify peptide mass fingerprints (PMFs), serum peptidome clusters, and potential biomarker peptides in infected dogs. Blood and serum samples were collected from two groups: H. canis-infected dogs (n = 15) and healthy controls (n = 20). Serum pepti

Experimental evidence against transmission of Hepatozoon canis by Ixodes ricinus.

Hepatozoon canis is among the most widespread tick-borne protozoa infecting domestic and wild carnivores. Its distribution is related to the occurrence of its major vector, the brown dog tick Rhipicephalus sanguineus. However, the role of Ixodes ricinus as a vector of H. canis has been hypothesized. In the present study, the development of H. canis was investigated in I. ricinus and R. sanguineus nymphs collected from a naturally infested dog. All I. ricinus ticks examined (n=133) were negative by cytological examination at days 20, 30, and 90 post collection, although H. canis DNA was detecte

Epidemiological and clinicopathological factors associated with infection by multiple pathogens transmitted by Rhipicephalus sanguineus sensu lato in naturally infected dogs in the Semiarid area of Northeastern Brazil.

Domestic dogs are frequently exposed to tick-borne pathogens such as Babesia vogeli, Hepatozoon canis, Anaplasma platys and Ehrlichia canis, which can cause a wide range of clinical manifestations, although little is known about the epidemiological and clinicopathological profiles of mono- and co-infections. This study aimed to characterize infection patterns and identify clinical and epidemiological factors associated with mono-infections, and co-infections with two or more pathogens in naturally infected dogs. We analyzed 181 dogs from a hospital population with suspected hemoparasitic infec

Molecular survey on vector-borne pathogens in urban and peri-urban rodents in Spain.

Rodents are key reservoirs and spillover hosts of major pathogens at the wildlife-domestic-human interface, playing a pivotal role in the eco-epidemiology of vector-borne diseases. Nevertheless, data on rodent infection by pathogens of animal and public health relevance remain limited, particularly in anthropized landscapes. This study aimed to evaluate the circulation of Leishmania infantum, Bartonella spp., and Hepatozoon spp. in rodent populations from anthropized areas in Spain, and to identify potential risk factors associated with infection by these pathogens. Spleen samples from 345 rod
