Phase behavior study of amphiphilic drugs: effect of pharmaceutical excipients.

Effect of various additives, like conventional as well as gemini cationic surfactants, non-ionic surfactants, hydrotropes, bile salts, fatty acid salts and β-cyclodextrin on the cloud point (CP) of two amphiphilic drugs promethazine (PMT) and promazine (PMZ) hydrochlorides was investigated. These additives are generally used as pharmaceutical excipients. The CP variation with the additive concentration has been determined and the energetic parameters of the process have been estimated. All the surfactants increase the CP, while hydrotropes, bile salts and fatty acid salts, when added in low co

Carbon nanotubes as functional excipients for nanomedicines: I. Pharmaceutical properties.

Carbon nanotubes (CNTs) are considered potential biomedical materials because of their flexible structure and propensity for chemical functionalization. In the first part of this two-part review we focus on the properties and use of CNTs as building blocks for novel drug delivery systems, dosage forms, and biomedical substrates. Pharmaceutical excipients have been regarded as inert or nonactive components of dosage forms, but they are essential and necessary components of pharmaceutical preparations. Here we present a collection of data to initiate the description of CNTs as pharmaceutical exc

Effect of pharmaceutical excipients on aqueous stability of rabeprazole sodium.

The chemical stability of a proton-pump inhibitor, rabeprazole sodium, was evaluated in simulated intestinal fluid (pH 6.8) containing various 'Generally Recognized As Safe (GRAS)'-listed excipients, including Brij 58, Poloxamer 188, Cremophor RH40, Gelucire 44/14 and PEG 6000. After incubation at 37 and 60 degrees C, the amounts of rabeprazole and its degradation product, thioether-rabeprazole, were quantitated by HPLC analysis. The main degradation product was separated and characterized by LC/MS. The degradation of rabeprazole followed first-order kinetics. In the absence of any excipients,

[Control of pulmonary absorption of drugs by various pharmaceutical excipients].

In general, drugs are well absorbed from the lung, and the pulmonary absorption of therapeutic protein and peptide drugs, which are poorly absorbed from the gastrointestinal tract, was observed. However, locally acting drugs including antiasthmatic agents, bronchodilators, and expectorants should be localized for a long period in the lung tissues. In this study, the effects of various viscous vehicles on the absorption of theophylline and fluticasone propionate after intrapulmonary administration were examined in rats. Carrageenans were effective in regulating the absorption rate of these drug

Micro-mechanical properties of drying material bridges of pharmaceutical excipients.

This work is part of a larger research effort to elucidate the properties and morphology of pharmaceutical granules produced by wet-granulation. In this work, we measure forces exerted by the drying interparticle bridges. The bridges were formed from aqueous solutions of common pharmaceutical excipients both non-polymeric (lactose, mannitol) and polymeric (hydroxypropyl cellulose (HPC), hydroxypropyl methylcellulose (HPMC), polyvinylpyrrolidone (povidone) (PVP)). We also study the morphology, microstructure and crystalline structure of solidifying bridges. We find that the solidifying behavior

Sweeteners, dyes, and other excipients in vitamin and mineral preparations.

Multivitamins and mineral preparations are widely used for infants and children. All of these preparations contain a variety of excipients ("inert ingredients"). Excipients are generally safe; however, adverse effects have been attributed to them. Complete information about the excipients in various preparations is not readily available. The information about sweeteners, dyes, and other excipients (flavorings, preservatives, stabilizers, and fillers) for 41 chewable/liquid multivitamin and mineral preparations was obtained and tabulated. Sucrose was present in 63% (26/41) of preparations follo

Excipients in pharmaceuticals: mechanisms of hypersensitivity and the role of global pharmacovigilance.

Excipients are important inactive components in drug formulations that ensure stability, bioavailability, and patient compliance. However, emerging evidence suggests that certain excipients, once considered inert, can cause hypersensitivity reactions in certain individuals. Such reactions include mild erythema due to systemic anaphylaxis and create clinical challenges that are difficult to handle. This review presents a systematic review of the existing literature on excipient hypersensitivity, with specific attention paid to commonly implicated excipients such as polyethylene glycol (PEG), pa

Polyenylphosphatidylcholine as bioactive excipient in tablets for the treatment of liver fibrosis.

Liver fibrosis is a condition characterized by the accumulation of extracellular matrix (ECM) arising from the myofibroblastic transdifferentiation of hepatic stellate cells (HSCs) occurring as the natural response to liver damage. To date, no pharmacological treatments have been specifically approved for liver fibrosis. We recently reported a beneficial effect of polyenylphosphatidylcholines (PPCs)-rich formulations in reverting fibrogenic features of HSCs. However, unsaturated phospholipids' properties pose a constant challenge to the development of tablets as preferred patient-centric dosag

Bile and excipient interactions directing drug pharmacokinetics in rats.

Bile solubilization plays a major role in the absorption of poorly water-soluble drugs. Excipients used in oral drug formulations impact bile-colloidal properties and their molecular interactions. Polymer-induced changes of bile colloids, e.g., by Eudragit E, reduced the flux of the bile interacting drug Perphenazine whereas bile non-interacting Metoprolol was not impacted. This study corroborates these in vitro findings in rats. Eudragit E significantly reduced systemic availability of Perphenazine but not Metoprolol compared to the oral administrations without polymer. This study confirms th

Excipient lung disease secondary to intravenous heroin use.

We present a case of a woman who had progressive shortness of breath and wheezing with a mild restrictive pulmonary function pattern. She was initially diagnosed with eosinophilic granulomatosis with polyangiitis on the basis of peripheral eosinophilia, bronchoalveolar lavage eosinophilia (47%) and surgical lung biopsy findings. Six months following her diagnosis, the patient returned because of persistent symptoms, and a second review of the lung biopsy revealed thrombotic lesions in the pulmonary vessels with polarisable foreign body materials, associated giant cell reactions and numerous eo

Excipient-mediated supersaturation stabilization in human intestinal fluids.

It was the purpose of this study to investigate excipient-mediated precipitation inhibition upon induction of supersaturation of poorly water-soluble drugs in aspirated human intestinal fluids (HIF) representing both the fasted and fed state. Etravirine, ritonavir, loviride, danazol and fenofibrate were selected as model compounds. For comparative purposes, precipitation inhibition was also evaluated in simple aqueous buffer, and in intestinal simulation media representative for the fasted and fed state (FaSSIF and FeSSIF, respectively). Supersaturation was induced in the test media containing

A perspective on testing of existing pharmaceutical excipients for genotoxic impurities.

Guidance recommendations by the Committee for Medicinal Products for Human Use (CHMP) and Pharmaceutical Research and Manufacturers of America (PhRMA) acknowledge the presence of potential toxic impurities in some pharmaceutical ingredients and have proposed setting limits on impurities with genotoxic activity as a means to protect patients in clinical trials and for marketing of the approved products. Recently, there have been suggestions that drug excipients, including existing products, should also be subjected to the same testing procedures and intake limits as proposed for active ingredie

Generation of formaldehyde by pharmaceutical excipients and its absorption by meglumine.

Formaldehyde is a well-known air impurity. The possibility was investigated in this study that pharmaceutical excipients commonly used in oral solid dosage forms might also be sources of formaldehyde. The results showed that formaldehyde is generated by the excipients lactose, D-mannitol, microcrystalline cellulose, low-substituted hydroxypropylcellulose, magnesium stearate and light anhydrous silicic acid. Since the quality and safety of pharmaceutical products can be significantly affected by the presence of formaldehyde, various amines were then investigated for their ability to decrease le

Quantifying molecular specificity in excipient-driven antibody solubilization.

Excipients are widely used to suppress the self-association of therapeutic proteins, yet their mechanisms of action are not well understood and often assumed to be nonspecific. Here we show that excipient-mediated solubilization of therapeutic antibodies is markedly molecularly specific. Using a high-throughput combinatorial droplet microfluidic platform, we systematically quantify the effects of common pharmaceutical excipients across a diverse panel of monoclonal antibodies (mAbs). Although all studied excipients enhance solubility, their effects can vary significantly between antibodies, sp

The Role of Polymers and Excipients for Better Gastric Retention of Captopril.

Captopril is an angiotensin-converting enzyme (ACE) inhibitor that prevents angiotensin I (ATI) from being converted to angiotensin II (ATII). However, it offers certain limitations like instability, dose dumping and burst release due to its usage in the native state. In the last two decades, different polymers and excipients have been used to make captopril more accessible and well-accepted. The present work discusses the efforts made by various scientists so far to make the oral administration of captopril more acceptable by overcoming its limitations. The different factors affecting gastric

Excipient variability and its impact on dosage form functionality.

Pharmaceutical excipients are essential components of most modern dosage forms. Although defined as pharmacologically inert, excipients can be thought of as the true enablers of drug product performance. Unintentional variability in the properties of the excipients may be unavoidable, albeit minimizable. The variability may originate from the source, the excipient-manufacturing process, or during the manufacturing of dosage forms. Excipient variability may have a range of influences on their functionality and performance in the dosage form. A better understanding of these influences on the cri
