Characteristics of the ErmK Protein of Bacillus halodurans C-125.

Bacillus halodurans C-125 is an alkaliphilic microorganism that grows best at pH 10 to 10.5. B. halodurans C-125 harbors the erm (erythromycin resistance methylase) gene as well as the mphB (macrolide phosphotransferase) and putative mef (macrolide efflux) genes, which confer resistance to macrolide, lincosamide, and streptogramin B (MLSB) antibiotics. The Erm protein expressed in B. halodurans C-125 could be classified as ErmK because it shares 66.2% and 61.2% amino acid sequence identity with the closest ErmD and Erm(34), respectively. ErmK can be regarded as a dimethylase, as evidenced by r

Molecular Elucidation of a Urate Oxidase from Deinococcus radiodurans for Hyperuricemia and Gout Therapy.

Urate oxidase initiates the uric acid degradation pathways and is extensively used for protein drug development for gout therapy and serum uric acid diagnosis. We first present the biochemical and structural elucidation of a urate oxidase from the extremophile microorganism Deinococcus radiodurans (DrUox). From enzyme characterization, DrUox showed optimal catalytic ability at 30 °C and pH 9.0 with high stability under physiological conditions. Only the Mg2+ ion moderately elevated its activity, which indicates the characteristic of the cofactor-free urate oxidase family. Of note, DrUox is the

Overexpression of a protein disulfide isomerase-like protein from Methanothermobacter thermoautotrophicum enhances mercury tolerance in transgenic rice.

MTH1745, from thermophilic archaea Methanothermobacter thermoautotrophicum, is a protein disulfide isomerase-like protein (PDIL) with a chaperone function and disulfide isomerase activity. Mercuric cations have a high affinity for sulfhydryl groups and consequently inhibit plant growth. Disulfide compounds (e.g., copper-zinc superoxide dismutase, Cu/Zn SOD) and sulfhydryl compounds (e.g., glutathione, phytochelatins, and metallothioneins) play important roles in mercury (Hg) response. To study the relationship between Hg detoxification and PDILs, we overexpressed MTH1745 in Oryza sativa L. cv.

Cryotolerance in the Antarctic lichen Umbilicaria antarctica is driven by supercooling and structural confinement.

The Antarctic lichen Umbilicaria antarctica exhibits exceptional resilience to extreme cold, enabling survival in harsh polar environments. This study investigates the molecular and structural adaptations underlying its tolerance to subzero temperatures. Nuclear Magnetic Resonance analysis showed that at intermediate hydration level, water undergoes a transition from loosely bound to more strongly bound states between - 17 °C and - 24 °C indicating a non-cooperative water immobilization process rather than conventional freezing. Activation energy and average distances between relaxing proton p

Evaluation of Antibiotic Biodegradation by a Versatile and Highly Active Recombinant Laccase from the Thermoalkaliphilic Bacterium Bacillus sp. FNT.

Laccases are industrially relevant enzymes that have gained great biotechnological importance. To date, most are of fungal and mesophilic origin; however, enzymes from extremophiles possess an even greater potential to withstand industrial conditions. In this study, we evaluate the potential of a recombinant spore-coat laccase from the thermoalkaliphilic bacterium Bacillus sp. FNT (FNTL) to biodegrade antibiotics from the tetracycline, β-lactams, and fluoroquinolone families. This extremozyme was previously characterized as being thermostable and highly active in a wide range of temperatures (

Simplified metabolic pathway design for efficient production in halotolerant microbial systems.

Halotolerant microorganisms are promising cell factories for industrial biomanufacturing because they combine physiological robustness with practical advantages, including contamination-resistant fermentation, reduced sterilization requirements, and compatibility with saline feedstocks. However, the metabolic burden of stress adaptation, together with regulatory redundancy and pathway complexity, often limits production efficiency and genetic stability. Simplified metabolic pathway design has therefore emerged as a key strategy to improve halotolerant cell factories. Here, we review recent adv

Thermorubin Biosynthesis Initiated by a Salicylate Synthase Suggests an Unusual Conversion of Phenols to Pyrones.

Thermorubin is a tetracyclic naphthoisocoumarin natural product that demands investigation due to its novel mechanism of bacterial protein synthesis inhibition and its unusual structural features. In this work, we describe the identification of the biosynthetic cluster responsible for thermorubin from the sequenced Laceyella sacchari producer species and its confirmation via heterologous production in Escherichia coli. Based on an in-depth annotation of the cluster, we propose a biosynthetic pathway that accounts for the formation of the unique, nonterminal pyrone. Additionally, the expression

Consortia of cyanobacteria/microalgae and bacteria in desert soils: an underexplored microbiota.

Desert ecosystem is generally considered as a lifeless habitat with extreme environmental conditions although it is colonized by extremophilic microorganisms. Cyanobacteria, microalgae, and bacteria in these habitats could tolerate harsh and rapidly fluctuating environmental conditions, intense ultraviolet radiation, and lack of water, leading to cell desiccation. They possess valuable metabolites withstanding extreme environmental conditions and make them good candidates for industrial applications. Moreover, most natural microorganisms in these extreme habitats exist as consortia that provid

Regulation of potassium dependent ATPase (kdp) operon of Deinococcus radiodurans.

The genome of D. radiodurans harbors genes for structural and regulatory proteins of Kdp ATPase, in an operon pattern, on Mega plasmid 1. Organization of its two-component regulatory genes is unique. Here we demonstrate that both, the structural as well as regulatory components of the kdp operon of D. radiodurans are expressed quickly as the cells experience potassium limitation but are not expressed upon increase in osmolarity. The cognate DNA binding response regulator (RR) effects the expression of kdp operon during potassium deficiency through specific interaction with the kdp promoter. De

Insights into the radiation and oxidative stress mechanisms in genus Deinococcus.

Deinococcus species, noted for their exceptional resistance to DNA-damaging environmental stresses, have piqued scientists' interest for decades. This study dives into the complex mechanisms underpinning radiation resistance in the Deinococcus genus. We have examined the genomes of 82 Deinococcus species and classified radiation-resistance proteins manually into five unique curated categories: DNA repair, oxidative stress defense, Ddr and Ppr proteins, regulatory proteins, and miscellaneous resistance components. This classification reveals important information about the various molecular mec

Comparative Transcriptomic Analysis of the Response of Dunaliella acidophila (Chlorophyta) to Short-Term Cadmium and Chronic Natural Metal-Rich Water Exposures.

Heavy metals are toxic compounds known to cause multiple and severe cellular damage. However, acidophilic extremophiles are able to cope with very high concentrations of heavy metals. This study investigated the stress response under natural environmental heavy metal concentrations in an acidophilic Dunaliella acidophila. We employed Illumina sequencing for a de novo transcriptome assembly and to identify changes in response to high cadmium concentrations and natural metal-rich water. The photosynthetic performance was also estimated by pulse amplitude-modulated (PAM) fluorescence. Transcripto

Cryo-EM study and in vivo chemical mapping of the Methanosarcina acetivorans ribosome and its dimerization via a repurposed enzyme and translation factor.

Despite the overall conservation of ribosomes across all domains of life, differences in their 3D architecture, rRNA sequences, ribosomal protein composition, and translation factor requirements reflect lineage-specific adaptations to environmental niches. In the domain Archaea, structural studies have primarily focused on nonmethanogenic thermophiles and halophiles, leaving it unclear whether these represent the broader Archaea domain. Here, we report the cryo-electron microscopy (cryo-EM) structure of the ribosome from Methanosarcina acetivorans, a previously unreported high-resolution struc

Under the microscope: microbial life at pore scale under extreme deep-environment conditions.

Microcapillaries are shown to be cheap but attractive alternatives to custom micromodels for the investigation of extremophile motility and tropism under reconstituted in situ conditions. We discuss practicalities and pitfalls of their use in anoxic sample preparation and observation. Widefield transmission and fluorescence microscopy, combined with a simple particle tracking algorithm provide insights into motility and tropism. As an example, mono and co-cultures of Thermodesulfatator atlanticus and Methanocaldococcus villosus are examined at 70 °C and 100 bar under H2:CO2 :: 4:1 (v:v), dilut

Genomic insights into novel extremotolerant bacteria isolated from the NASA Phoenix mission spacecraft assembly cleanrooms.

BACKGROUND: Human-designed oligotrophic environments, such as cleanrooms, harbor unique microbial communities shaped by selective pressures like temperature, humidity, nutrient availability, cleaning reagents, and radiation. Maintaining the biological cleanliness of NASA's mission-associated cleanrooms, where spacecraft are assembled and tested, is critical for planetary protection. Even with stringent controls such as regulated airflow, temperature management, and rigorous cleaning, resilient microorganisms can persist in these environments, posing potential risks for space missions. RESULTS

Cytoplasmic CopZ-Like Protein and Periplasmic Rusticyanin and AcoP Proteins as Possible Copper Resistance Determinants in Acidithiobacillus ferrooxidans ATCC 23270.

Acidophilic organisms, such as Acidithiobacillus ferrooxidans, possess high-level resistance to copper and other metals. A. ferrooxidans contains canonical copper resistance determinants present in other bacteria, such as CopA ATPases and RND efflux pumps, but these components do not entirely explain its high metal tolerance. The aim of this study was to find other possible copper resistance determinants in this bacterium. Transcriptional expression of A. ferrooxidans genes coding for a cytoplasmic CopZ-like copper-binding chaperone and the periplasmic copper-binding proteins rusticyanin and A

Hot springs, cool beetles: extraordinary adaptations of a predaceous insect in Yellowstone National Park.

Several metazoans live in extreme environments, but relatively little is known about the adaptations that these extremophiles have evolved to tolerate their conditions. The wetsalts tiger beetle, Cicindelidia hemorrhagica (LeConte) (Coleoptera: Cicindelidae), is found in the western USA, including the active geothermal springs in Yellowstone National Park (YNP). Here, we characterize behavioral, ecophysiological, and morphological traits of adult C. hemorrhagica living on hot springs in YNP compared to adults living in a non-hot spring environment in Idaho. Individuals in YNP behaviorally warm

Diversity, Taxonomic Novelty, and Encoded Functions of Salar de Ascotán Microbiota, as Revealed by Metagenome-Assembled Genomes.

Salar de Ascotán is a high-altitude arsenic-rich salt flat exposed to high ultraviolet radiation in the Atacama Desert, Chile. It hosts unique endemic flora and fauna and is an essential habitat for migratory birds, making it an important site for conservation and protection. However, there is limited information on the resident microbiota's diversity, genomic features, metabolic potential, and molecular mechanisms that enable it to thrive in this extreme environment. We used long- and short-read metagenomics to investigate the microbial communities in Ascotán's water, sediment, and soil. Bact

Spatial Variability of Microbial Communities and Salt Distributions Across a Latitudinal Aridity Gradient in the Atacama Desert.

Over the past 150 million years, the Chilean Atacama Desert has been transformed into one of the most inhospitable landscapes by geophysical changes, which makes it an ideal Mars analog that has been explored for decades. However, a heavy rainfall that occurred in the Atacama in 2017 provides a unique opportunity to study the response of resident extremophiles to rapid environmental change associated with excessive water and salt shock. Here we combine mineral/salt composition measurements, amendment cell culture experiments, and next-generation sequencing analyses to study the variations in s
