Phenotypic, molecular and pathological characterization of motile aeromonads isolated from diseased fishes cultured in Uruguay.

Information about motile aeromonads from aquaculture systems of the Neotropical region is scarce. The aim of this study was to characterize motile Aeromonas isolated from ornamental and consumable fishes cultured in Uruguay. Biochemical and molecular methods were used for species identification. Antimicrobial susceptibility and the presence of virulence genes were evaluated. Genetic diversity was analysed by rep-PCR, and virulence of the most representative isolates was determined by calculating the fifty lethal dose in experimentally challenged fish (Australoheros facetus). Aeromonas hydrophi

Pathogenesis of acute viral disease induced in fish by carp interstitial nephritis and gill necrosis virus.

A lethal disease of koi and common carp (species Cyprinus carpio) has afflicted many fish farms worldwide since 1998, causing severe financial losses. Morbidity and mortality are restricted to common carp and koi and appear in spring and autumn, when water temperatures are 18 to 28 degrees C. We have isolated the virus causing the disease from sick fish, propagated it in koi fin cell culture, and shown that virus from a single clone causes lethal disease in carp and koi upon infection. Intraperitoneal virus injection or bathing the fish in virus-containing water kills 85 to 100% of the fish wi

[Lecithin: cholesterol acyltransferase (LCAT)--the genetic analysis of familial LCAT deficiency and fish eye disease].

Lecithin: cholesterol acyltransferase (LCAT) is the enzyme that catalyze the esterification of free cholesterol in plasma proteins. The functional abnormalities of LCAT are known to cause two diseases characterized by severe corneal opacity; familial LCAT deficiency that is accompanied with anemia and frequently, though not invariably, renal failure, and fish eye disease (FED) without any other severe symptoms. Notably, it has been shown that the clinical symptoms and biochemical features are highly variable in each patient with the diseases. Recent genetic studies on the cases of LCAT deficie

Fish oil-induced yellow fat disease in rats. IV. Functional studies of the reticuloendothelial system.

In rats with "stage S/E" yellow fat disease an injection of colloidal carbon resulted in a marked reduction in the number of circulating platelets. The death rate of rats with experimental Listeria monocytogenes infection, the number of bacteria in their spleens and the decrease of bacteria in their spleens on the days after infection were the same in rats with yellow fat disease as in controls. The fact that the rats died during the first few days after infection also may indicate that their immunological resistance to L. monocytogenes was not altered by yellow fat disease.

Black spot diseases in seven commercial fish species from the English Channel and the North Sea: infestation levels, identification and population genetics of Cryptocotyle spp.

Fish are often speckled with "black spots" caused by metacercarial trematode infection, inducing a host response. Cryptocotyle spp. (Opisthorchiidae) are among the parasites responsible for this phenomenon. So far, the impact on human health is still unknown. In addition, few publications dealing with black spot recovery, identification, distribution and diversity among commercially important fish are available. Moreover, "black spots" have been observed by fishermen on marine fish, revealing an appreciable but unquantified presence in consumed fish. An epidemiological survey of 1,586 fish fro

A Modelling Framework for Assessing the Risk of Emerging Diseases Associated with the Use of Cleaner Fish to Control Parasitic Sea Lice on Salmon Farms.

Sea lice are the most damaging parasite of marine salmonids, both economically and in terms of potential impacts on wild fish. An increasingly widely applied control is the use of cleaner fish (CF) such as wrasse that eat lice. However, such CF can carry pathogens that may cause disease in salmon, including the potential emergence of new diseases. This is not just a theoretical risk, as demonstrated by a recent outbreak of viral haemorrhagic septicaemia in wrasse held on salmon farms in Shetland. A modelling framework is developed to identify conditions in which emergence might occur, and, fro

Neonatal growth with fish oil or mixed oil lipid emulsions in intestinal failure associated liver disease.

BACKGROUND: Treatment strategies for intestinal failure associated liver disease (IFALD) include use of fish-oil lipid emulsion (FO-ILE) or mixed-oil emulsion containing soy, medium chain triglyceride, olive, and fish oils (SO,MCT,OO,FO-ILE). FO-ILE is limited to 1 g/kg/day, either limiting overall calorie provision or shifting calories toward carbohydrate predominance. The objective of this project was to compare the growth of neonates with IFALD receiving either SO,MCT,OO,FO-ILE or FO-ILE. MATERIAL AND METHODS: A retrospective chart review was conducted of neonates with IFALD receiving SO,M

Effects of UMP, Choline, and Fish Oil on Synaptic Integrity and Motor Coordination in an Alzheimer's Disease Mouse Model.

Alzheimer's disease (AD) is an age-related neurodegenerative disorder characterized by progressive synaptic dysfunction, axonal pathology, and cognitive decline, with the hippocampal circuits showing particular vulnerability during disease progression. However, early-life nutritional interventions may influence long-term synaptic resilience. In this study, we investigated the long-term effects of prenatal and lactational supplementation with choline, UMP, and fish oil in the 5XFAD mouse model. To this end, hippocampal synaptic and axonal pathology was assessed at 3, 6, and 9 months using Weste

In vitro and in vivo hemocompatibility study of fish swim bladder-derived biomaterials for the application of chronic venous disease.

Chronic venous disease (CVD) is a prevalent condition affecting the venous system of the lower limbs, characterized by venous hypertension and regurgitation resulting from congenital or acquired venous valve insufficiency. The global morbidity of CVD is as high as 69.94%. Prosthetic venous valve replacement provides a new therapeutic option. However, to mimic natural venous valves, the leaflets of prosthetic venous valves need to be as thin as ~200 μm, and better resistance to thrombosis is required due to the physiological characteristics of the venous system. Application of a Rationally Designed Mucosal Nanovaccine Platform in Bacterial Disease Prophylactics Using Fish as a Model Organism.

The emergence of antimicrobial resistance necessitates innovative vaccine strategies for bacterial disease prevention in aquaculture. Here, we present a rationally designed mucosal nanovaccine platform targeting Vibrio harveyi, a major pathogen in marine fish. Bioinformatics tools identified 15 T-cell and 16 B-cell epitopes from 17 antigenic proteins, which were consolidated into a multiepitope antigen (MAE). This MAE was fused to the self-assembling ferritin H-subunit (HFn) to form MAE@HFn nanoparticles, further functionalized with PEG2000 (MAE@HFn-PEG2k) to enhance mucus penetration. Structu

The impact of helminth infection in fish farms on wild trout populations: host immune regulation of disease risk.

Intensive rearing of farmed fish may risk disease spillover into free-living populations. This study concerns the blood-feeding gill monogenean of salmonids, Discocotyle sagitatta, on the Isle of Man, UK. Heavy infections in 2 fish farms have led to severe disease with periodic mass mortality. Infection levels in rainbow trout (Oncorhynchus mykiss, overall n = 556) increased with age (i.e. years exposed): by year 3, prevalence was 100%, mean intensity c. 100 (maximum 1150) worms/host. Output from farms of many millions of parasite eggs/day has the potential for transmission to downstream popul

Novel pathogenic variant in the LCAT gene in a compound heterozygous patient with fish-eye disease and a mild phenotype.

BACKGROUND AND OBJECTIVE: Low high-density lipoprotein (HDL)-cholesterol and corneal opacity are associated with fish-eye disease (FED), familial lecithin:cholesterol acyltransferase (LCAT) deficiency (FLD), apolipoprotein AI deficiency, and Tangier disease. The differential diagnosis is made by clinical and biochemical tests. Measuring the LCAT activity is the ideal way to distinguish conditions caused by LCAT gene variants (FED and FLD) from the other 2 diseases. However, this is not accessible from all clinics. The cholesteryl ester/total cholesterol (CE/TC) ratio, which is below the refere

Hole-in-the-head disease in discus fish, Symphysodon (Heckel, 1840): Is it a consequence of a dietary Ca/P imbalance?

Hole-in-the-head (HITH) disease-affected fish develop characteristic lesions in the skin above sensory pores of the head and the trunk. This study investigated whether an unfavourable Ca/P ratio in the diet could provoke lesions consistent with HITH disease in discus fish Symphysodon (Heckel, 1840) as a comparable condition to secondary hyperparathyroidism of tetrapod species. Two groups of five fish were fed a plain beef heart diet (Ca/P of 0.03), whereas two other groups were kept on commercial discus feed (Ca/P of 2.73). Each feeding group was submitted to two different water hardness regim

Immune cell-resolved transcriptomics provides insights into the basis for variations of fish genetic resistance to viral disease.

BACKGROUND: The genetic basis of host resistance to viral infections is generally shaped by complex interactions between host genetic variations affecting antiviral immunity and the rapid evolutionary adaptability of viruses. In this study, we investigated two isogenic rainbow trout lines exhibiting extreme resistance or susceptibility to the rhabdovirus VHSV. We compared transcriptomes of the pronephros - a major lymphoid organ in fish - at steady state and following VHSV infection. By integrating bulk tissue RNA-seq with single-cell RNA-seq, we mapped the divergent transcriptomic responses o
