Differential Methylation Analysis of Hypermelanosis in Chinese Tongue Sole (Cynoglossus semilaevis).

Hypermelanosis on the blind side is a major concern in tongue sole (Cynoglossus semilaevis) aquaculture in China and causes great economic loss for farmers. To investigate the relationship between DNA methylation and hypermelanosis, different skin tissues on both the ocular and blind sides of both normal and hypermelanotic fish were used in this study, and set analysis was utilized to narrow and filter the possible, potential, and direct differentially methylated regions (DMRs). A total of 2278, 1015, and 6740 DMRs were discovered, which belonged to 1786, 908, and 4210 genes, for possible, pot

THIN AND ULTRATHIN STRUCTURE OF ENVELOPES OF STEPHANOSTOMUM BACCATUM (TREMATODA: ACANTHOCOLPIDAE) METACERCARIAE IN YELLOWFIN SOLE, LIMANDA ASPERA.

The structure of the envelopes (capsule and cyst) surrounding metacercariae of Stephanostomum baccatum (Nicoll, 1907) in the second intermediate host, the yellowfin sole Limanda aspera (Pallas 1814), is examined with the methods of light and transmission electron microscopy. The cyst, presumably formed by secretions of the metacercarial tegument, consists of 2 layers: the outer, very thin layer of an electron-dense, finely granular substance and the inner layer composed of loose material of a moderate electron density that includes dense bodies varying in size, shape, and localization. The cap

Cytochemical identification of turbot myeloperoxidase-positive granulocytes by potassium iodide and oxidized pyronine Y staining.

Cytomorphological and cytochemical staining are important methods for the identification of cell types, in particular in fish which often lack biological tools such as specific antibodies. Myeloperoxidase (MPO) is usually used as an intracellular marker of neutrophil accumulation in tissues and a marker of neutrophil activity in plasma. In this study, we reported a potassium iodide and oxidized pyronine Y (KI-PyY) staining method for rapid and highly sensitive detection of MPO-positive cells in turbot blood, peritoneum, and tissues. MPO-positive cells, which mostly represented neutrophils, wer

Phosphorus-potassium fertilizer exposure induces oxidative stress and riboflavin metabolism disruption in juvenile turbot (Scophthalmus maximus).

Intensive agriculture's excessive fertilizer use significantly contributes to coastal nutrient pollution. While phosphorus's ecological impacts are recognized, the toxicity of combined phosphorus nutrients on marine life remains unclear. This study examined phosphorus-potassium fertilizer's (P-K fertilizer) effects on juvenile turbot (Scophthalmus maximus) via a 96-hour acute exposure experiment. Exposure solutions were prepared at environmentally relevant concentrations, resulting in elevated seawater phosphorus and potassium levels by 117.25 mg/L and 152.94 mg/L, respectively. Compound ferti

Early Molecular Immune Responses of Turbot (Scophthalmus maximus L.) Following Infection with Aeromonas salmonicida subsp. salmonicida.

Turbot aquaculture production is an important economic activity in several countries around the world; nonetheless, the incidence of diseases, such furunculosis, caused by the etiological agent A. salmonicida subsp. salmonicida, is responsible for important losses to this industry worldwide. Given this perspective, this study aimed to evaluate early immune responses in turbot (S. maximus L.) following infection with A. salmonicida subsp. salmonicida. For this, 72 fish were individually weighed and randomly distributed into 6 tanks in a circulating seawater system. For the bacterial challenge,

Scuticociliate (Philasterides dicentrarchi) infection cluster in a multispecies marine aquarium system.

Scuticociliatosis, caused by ciliated protozoa of the subclass Scuticociliatia, has been associated with high mortalities in marine fish. Environmental factors such as an increase in water temperature can enhance this disease. The aim of the present report is to describe the occurrence of a cluster of cases of scuticociliatosis in a multispecies marine cold-water system in a public aquarium. Philasterides dicentrarchi was identified by PCR in formalin-fixed tissues of some of the fish showing meningitis or meningoencephalitis, dermatitis and myositis with intralesional protozoa. An increase in

Histopathological and ultrastructural studies on a novel pathological condition in Solea senegalensis.

A new parasitic disease affecting cultured sole Solea senegalensis (Kaup, 1858) is characterised by the presence of external protuberances in the skin of the affected fish. These lesions correspond to nodules in the muscular tissue showing an abscess-like aspect. Similar lesions were found in the kidney, heart, liver and digestive tract. Histological sections of these nodules revealed the presence of a large core formed mainly of necrotic tissue surrounded with fibroblasts and macrophages. Round-shaped plasmodial organisms were found in the external layer of the nodules and usually inside macr

The Teleost Thymus in Health and Disease: New Insights from Transcriptomic and Histopathological Analyses of Turbot, Scophthalmus maximus.

The thymus is a primary lymphoid organ that plays a pivotal role in the adaptive immune system. The immunobiology of the thymus in fish is considered to be similar to that of mammals, but it is actually poorly characterized in several cultured teleost species. In particular, while investigations in human and veterinary medicine have highlighted that the thymus can be affected by different pathological conditions, little is known about its response during disease in fish. To better understand the role of the thymus under physiological and pathological conditions, we conducted a study in turbot

[Marine life envenomations: example in New Caledonia].

Marine life in the waters of New Caledonia is extraordinarily rich. However some of the animals inhabiting this wonderland are dangerous including a number of venomous species. A retrospective study conducted at the Territorial Hospital in Noumea for the three-year period between 1995 and 1998 showed that nearly 200 people/year were victims of envenomation by marine animals. Findings also indicated that the incidence of envenomation was rising as the practice of marine activities by the local population and tourists increased. Venomous species can be classified into 4 categories according to t

Unveiling Gene Expression Dynamics during Early Embryogenesis in Cynoglossus semilaevis: A Transcriptomic Perspective.

Commencing with sperm-egg fusion, the early stages of metazoan development include the cleavage and formation of blastula and gastrula. These early embryonic events play a crucial role in ontogeny and are accompanied by a dramatic remodeling of the gene network, particularly encompassing the maternal-to-zygotic transition. Nonetheless, the gene expression dynamics governing early embryogenesis remain unclear in most metazoan lineages. We conducted transcriptomic profiling on two types of gametes (oocytes and sperms) and early embryos (ranging from the four-cell to the gastrula stage) of an eco

Molecular characterization and ontogenetic expression profiles of LPXRFa and its receptor in Japanese flounder (Paralichthys olivaceus).

Investigations concerning the LPXRFa system are rarely conducted in flatfish species. Here, we first identified and characterized lpxrfa and its cognate receptor lpxrfa-r genes in the Japanese flounder (Paralichthys olivaceus). The coding DNA sequence of lpxrfa was 579 bp in length, wich encoded a 192-aa preprohormone that can produce three mature LPXRFa peptides. The open reading frame (ORF) of lpxrfa-r was 1446 bp in size, and encoded a 481-aa LPXRFa-R protein that encompassed seven hydrophobic transmembrane domains. Subsequently, tissue distribution expression profiles of lpxrfa and lpxrfa-

Understanding pseudo-albinism in sole (Solea senegalensis): a transcriptomics and metagenomics approach.

Pseudo-albinism is a pigmentation disorder observed in flatfish aquaculture with a complex, multi-factor aetiology. We tested the hypothesis that pigmentation abnormalities are an overt signal of more generalised modifications in tissue structure and function, using as a model the Senegalese sole and two important innate immune barriers, the skin and intestine, and their microbiomes. Stereological analyses in pseudo-albino sole revealed a significantly increased mucous cell number in skin (P < 0.001) and a significantly thicker muscle layer and lamina propria in gut (P < 0.001). RNA-seq transc

Recombinant phage-display monoclonal antibody against starry flounder (Platichthys stellatus) IgM enables quantitative ELISA for vaccine-induced humoral responses.

Species-matched serological reagents are scarce for many aquaculture species, limiting quantitative assessment of vaccine responses. Here, we developed and validated monoclonal antibodies (MAbs) specific to starry flounder (Platichthys stellatus) immunoglobulin M (IgM) using a recombinant phage-display approach. IgM purified from immune serum was used as the antigen for bio-panning, yielding multiple IgM-reactive scFv candidates, all of which recognized the IgM heavy chain by Western blot analysis. A lead clone (1T1A11) was reformatted as an scFv-Fc minibody, expressed in mammalian cells, puri

The value of multi-year sampling for detecting fine-scale population genetic structure in marine fishes: A case study of juvenile southern flounder.

Understanding population structure is critical for effective fisheries management in species with complex life histories and variable recruitment. Southern flounder (Paralichthys lethostigma) is a valuable flatfish species with declining populations in the Southeast United States. Improved management may depend on a better understanding of fine-scale and temporal population genetic structure in this region; however, such structure remains poorly characterized. To address our lack of understanding of the spatial and temporal population structure of this important species, we used double digest

Cloning and early expression pattern of two melatonin biosynthesis enzymes in the turbot (Scophthalmus maximus).

Melatonin biosynthesis from serotonin involves the sequential activation of the arylalkylamine N-acetyltransferase (AANAT) and hydroxyindole-O-methyltransferase (HIOMT). Photoperiod synchronizes a daily rhythm in pineal and retinal melatonin secretion through controlling AANAT activity. Teleost fish possess two Aanat, one expressed in the retina (AANAT1) and the other expressed in the pineal gland (AANAT2). We report here the full-length cloning of Aanat1, Aanat2, SmHiomt and Otx5 (orthodenticle homeobox homolog 5) in the turbot (Scophthalmus maximus, Sm), a flatfish belonging to an evolutiona

First Description and Characterisation of Lactococcus garvieae Strains Causing Septicaemic Disease in Farmed Sea Bass (Dicentrarchus labrax; Linnaeus) in Spain.

This study presents the first description and characterisation of Lactococcus garvieae isolated from diseased European sea bass (Dicentrarchus labrax) farmed in Spain. During autumn 2023, two fish farms were affected by infectious outbreaks causing moderate cumulative fish mortality (5%-10%). Diseased fish showed clinical signs of haemorrhagic septicaemia, compatible with lactococcosis. Standardised screening tests revealed the presence of Gram-positive cocci in the kidney, spleen and brain of the diseased fish, and negative results for parasites and viruses. Bacterial cultures recovered from

Outer membrane vesicles (OMVs) from Tenacibaculum maritimum as a potential vaccine against fish tenacibaculosis.

Outer membrane vesicles (OMVs) have been gained increasing attention in vaccinology due to their ability to induce strong protective humoral and cell-mediated immunity. The Gram-negative bacterium Tenacibaculum maritimum, the causative agent of marine tenacibaculosis, poses a significant challenge to the global aquaculture industry due to its difficult prophylaxis. In previous studies, we demonstrated that OMV production is a key virulence mechanism in T. maritimum. Building on this, the present study aimed to evaluate the efficacy of a natural, encapsulated multi-antigen vaccine made from adj

Investigation of sex determination in starry flounder ( Platichthys stellatus) reveals sex chromosome evolution in Pleuronectiformes and identifies a sex-specific marker.

The identification of sex chromosomes is fundamental for exploring the mechanism and evolution of sex determination. Platichthys stellatus, a species exhibiting clear sexual dimorphism and homomorphic chromosome pairs, has received limited research concerning its sex determination mechanisms. Clarifying the sex chromosome of P. stellatus will enhance our understanding of sex chromosome evolution in Pleuronectiformes. This study employed whole-genome resequencing to investigate the sex chromosome and sex determination system in P. stellatus. Notably, Chr23 was identified as the sex chromosome i

Identification and Functional Analysis of foxo Genes in Chinese Tongue Sole (Cynoglossus semilaevis).

The Chinese tongue sole (Cynoglossus semilaevis) is a traditional, precious fish in China. Due to the large growth difference between males and females, the investigation of their sex determination and differentiation mechanisms receives a great deal of attention. Forkhead Box O (FoxO) plays versatile roles in the regulation of sex differentiation and reproduction. Our recent transcriptomic analysis has shown that foxo genes may participate in the male differentiation and spermatogenesis of Chinese tongue sole. In this study, six Csfoxo members (Csfoxo1a, Csfoxo3a, Csfoxo3b, Csfoxo4, Csfoxo6-l
