A novel cold-adapted esterase FE2495 from Antarctic Flavobacterium ardleyense A2-1: Biochemical characterization and structural basis for cold-adaptation.

Cold-adapted enzymes from microorganisms living in polar zones represent promising biocatalysts for low-temperature applications. In this study, we identified a novel esterase, FE2495, from the Antarctic bacterium Flavobacterium ardleyense A2-1 and investigated underlying mechanisms for its cold-adaptive properties. The enzyme featured a canonical catalytic triad (Ser113, Asp167, His198), with the nucleophilic Ser113 residing within a conserved GFSQG motif. Phylogenetic analysis classified FE2495 as a family VI esterase. Nearly 50% of the enzyme's esterase activity was retained at temperatures

Effectiveness of a new cationic lipid-based nanovaccine for enhancing immersion vaccination against Flavobacterium oreochromis in red tilapia (Oreochromis sp.).

Flavobacterium oreochromis (Fo) poses a significant threat to tilapia, leading to economic losses due to mortality. Immersion vaccines, while practical for small fish, often result in limited antigen absorption. This study aimed to develop a cationic lipid-based nanoparticle vaccine using cetyltrimethylammonium bromide (CTAB) and evaluate its efficacy against F. oreochromis in red tilapia. In the first trial, healthy red tilapia were immersion vaccinated for 30 min, with three groups included in the trial: control (non-vaccinated), formalin-killed sonicated cells (FK-SC), or cationic lipid-bas

Flavobacterium psychrophilum as an Important Pathogen Associated with Overwintering Mortality Syndrome in Grass Carp (Ctenopharyngodon idella): Isolation, Characterization, and Pathogenicity.

Grass carp (Ctenopharyngodon idella), a key species in China's freshwater aquaculture, faces hindered industrial development due to overwintering mortality disease outbreaks. A Gram-negative bacterial strain, designated Hbh25210, was isolated from overwintering mortality-diseased grass carp in a Hubei Province farm. It was identified as Flavobacterium psychrophilum by 16S rRNA gene sequencing, showing 100% homology to reference strains in GenBank. The strain can be successfully cultured at 15 °C within 3 to 5 days. Multilocus sequence typing (MLST) classified it as ST-51 (CC-ST56), while serot

Pleiotropic effects of signal peptide peptidase A (sppA) gene deletion on membrane homeostasis, gliding motility, and virulence in Flavobacterium columnare.

Columnaris disease, caused by Flavobacterium columnare, represents one of the most economically devastating bacterial infections in global freshwater aquaculture. Despite its significant impact, the molecular mechanisms underlying F. columnare pathogenesis remain largely unexplored. Signal peptide peptidase A (SppA) plays a crucial role in bacterial protein secretion by degrading residual signal peptides after protein translocation, yet its function in F. columnare physiology and virulence has not been characterized. Here, we employed a targeted in-frame gene knockout approach to investigate t

Understanding the pathogenesis of Flavobacterium psychrophilum using the rainbow trout monocyte/macrophage-like cell line, RTS11, as an infection model.

The life cycle of Flavobacterium psychrophilum (Fp), the causative agent of bacterial coldwater disease (BCWD) and rainbow trout fry syndrome (RTFS), appears to involve interactions with spleen and head kidney macrophages. To develop an in vitro model for studying this, F. psychrophilum was incubated with a rainbow trout splenic monocyte/macrophage-like cell line (RTS11) and fundamental macrophage functions evaluated. The animal cell basal medium, L15, supplemented with bovine serum (FBS) supports RTS11 maintenance, and surprisingly, L15 with 2% FBS (L15/FBS) also supported F. psychrophilum gr

IL-4/13 expressing CD3γ/δ+ T cells regulate mucosal immunity in response to Flavobacterium columnare infection in grass carp.

Interleukin (IL) 4 and 13 are signature cytokines orchestrating Th2 immune response. Teleost fish have two homologs, termed IL-4/13A and IL-4/13B, and have been functionally characterized. However, what cells express IL-4/13A and IL-4/13B has not been investigated in fish. In this work, the recombinant IL-4/13A and IL-4/13B proteins of grass carp (Ctenopharyngodon idella) were produced in the Escherichia coli (E. coli) cells and purified. Monoclonal antibodies (mAbs) against the recombinant CiIL-4/13A and CiIL-4/13B proteins were prepared and characterized. Western blotting analysis showed tha

PACAP Is Lethal to Flavobacterium psychrophilum Through Either Direct Membrane Permeabilization or Indirectly, by Priming the Immune Response in Rainbow Trout Macrophages.

Pituitary adenylate cyclase-activating polypeptide (PACAP) is a multifunctional neuropeptide that is widely distributed in mammals and is capable of performing roles as a neurotransmitter, neuromodulator, and vasodilator. This polypeptide belongs to the glucagon/secretin superfamily, of which some members have been shown to act as antimicrobial peptides in both mammalian and aquatic organisms. In teleosts, PACAP has been demonstrated to have direct antimicrobial activity against several aquatic pathogens, yet this phenomenon has never been studied throughout a live bacterial challenge. Efficacy of whole-cell-based monovalent and bivalent vaccines against Streptococcus iniae and Flavobacterium covae in fingerling Asian seabass (Lates calcarifer).

Streptococcosis and columnaris caused by Streptococcus spp. and Flavobacterium spp. have been recognized as critical problems in Asian seabass aquaculture development because they cause severe mortality. In this study, we identified various isolates of S. iniae and F. covae from diseased Asian seabass farmed in Thailand for use as candidates for vaccine development. The efficacy of the vaccines was evaluated by challenge tests and immune parameter analyses in fish that received whole-cell-based monovalent and bivalent vaccines containing S. iniae (Sin) and F. covae (Fco) delivered by top-dress

Temporal microbiome changes in axolotl limb regeneration: Stage-specific restructuring of bacterial and fungal communities with a Flavobacterium bloom during blastema proliferation.

The intricate relationship between regeneration and microbiota has recently gained attention, spanning diverse model organisms. Axolotl (Ambystoma mexicanum) is a critically endangered salamander species and a model organism for regenerative and developmental biology. Despite its significance, a noticeable gap exists in understanding the interplay between axolotl regeneration and its microbiome. Here, we analyse in depth bacterial 16S rRNA amplicon dataset that we reported before as data resource and profile fungal community by sequencing ITS amplicons at the critical stages of limb regenerati

Antimicrobial Susceptibility of Flavobacterium psychrophilum from Chilean Salmon Farms and Their Epidemiological Cut-Off Values Using Agar Dilution and Disk Diffusion Methods.

Flavobacterium psychrophilum is the most important bacterial pathogen for freshwater farmed salmonids in Chile. The aims of this study were to determine the susceptibility to antimicrobials used in fish farming of Chilean isolates and to calculate their epidemiological cut-off (COWT) values. A number of 125 Chilean isolates of F. psychrophilum were isolated from reared salmonids presenting clinical symptoms indicative of flavobacteriosis and their identities were confirmed by 16S rRNA polymerase chain reaction. Susceptibility to antibacterials was tested on diluted Mueller-Hinton by using an a

New culture medium enhances the isolation and culture of Flavobacterium psychrophilum, causative agent of bacterial coldwater disease and Rainbow Trout fry syndrome.

OBJECTIVE: Flavobacterium psychrophilum causes bacterial coldwater disease (BCWD) and Rainbow Trout fry syndrome (RTFS) in salmonids, resulting in significant economic losses worldwide. Bacterial culture remains a widely used method for F. psychrophilum diagnostics and is core to BCWD and RTFS research; however, some isolates grow poorly on existing media. The primary goal of this study was to develop a solid culture medium that improves the in vitro recovery of F. psychrophilum across a diverse set of multilocus sequence typing variants. METHODS: Initially, CFU yields of 165 diverse F. psych

Exploring the potential of Flavobacterium covae catalase and DNA starvation/stationary phase protein as recombinant protein vaccines against columnaris disease in channel catfish.

Columnaris disease, caused by Flavobacterium covae, is a principal cause of mortality in the catfish industry that can lead to several million dollars in annual production losses. This Gram-negative bacterium is an opportunistic pathogen that manifests as a biofilm on the host's mucosal surfaces as the disease progresses. Recent work in our lab identified the upregulation of specific F. covae biofilm genes after stimulation with channel catfish mucus. We have previously demonstrated the effectiveness of using a F. covae recombinant protein for bath immersion vaccination of catfish to protect a

Immersion challenge of three salmonid species (family Salmonidae) with three multilocus sequence typing variants of Flavobacterium psychrophilum provides evidence of differential host specificity.

Bacterial coldwater disease (BCWD), caused by Flavobacterium psychrophilum, results in significant losses among multiple salmonid (family Salmonidae) species. Molecular epidemiology and serotyping studies have suggested that some variants are host specific; however, these associations have not been evaluated by cross-challenging fish species with putatively host-associated F. psychrophilum isolates via more natural (i.e. immersion) exposure routes. To this end, F. psychrophilum isolates US19-COS, US62-ATS and US87-RBT, each originally recovered from diseased coho salmon (Oncorhynchus kisutch),

The regulatory effects of pyridoxine deficiency on the grass carp (Ctenopharyngodon idella) gill barriers immunity, apoptosis, antioxidant, and tight junction challenged with Flavobacterium columnar.

The effects of dietary pyridoxine (PN) on the gill immunity, apoptosis, antioxidant and tight junction of grass cap (Ctenopharyngodon idella) were investigated in this study. Fish were fed semi-purified diets containing graded levels of PN for 10 weeks, and then challenged with Flavobacterium columnare by bath immersion exposure for 3 days. The results indicated that compared with the optimal PN level, PN deficiency resulted in a decline in the antimicrobial compound production of gill. In addition, PN deficiency up-regulated the pro-inflammatory cytokines and down-regulated the anti-inflammat

Investigation of dietary exogenous protease and humic substance on growth, disease resistance to Flavobacterium covae and immune responses in juvenile channel catfish (Ictalurus punctatus).

Many bacterial pathogens impact the US catfish industry, and disease control can be challenging for producers. Columnaris disease in channel catfish, Ictalurus punctatus, is primarily caused by Flavobacterium covae (formerly F. columnare). Immunostimulants may enhance nonspecific immune responses and offer an alternative to antibiotic treatments in catfish. Furthermore, dietary protein sources and inclusions are also essential to fish health and nutrition and may enhance overall fish performance in pond culture. The current project evaluated two immunostimulants: a protease complex (PC) and a

Genetic-biochemical analysis and distribution of the Ambler class A beta-lactamase CME-2, responsible for extended-spectrum cephalosporin resistance in Chryseobacterium (Flavobacterium) meningosepticum.

In vitro synergy between extended-spectrum cephalosporins and either clavulanic acid or cefoxitin was found for Chryseobacterium meningosepticum isolates during a double-disk assay on an agar plate. An extended-spectrum beta-lactamase (ESBL) gene from a C. meningosepticum clinical isolate was cloned and expressed in Escherichia coli DH10B. Its protein conferred resistance to most beta-lactams including extended-spectrum cephalosporins but not to cephamycins or to imipenem. Its activity was strongly inhibited by clavulanic acid, sulbactam, and tazobactam, as well as by cephamycins and imipenem.

Biodegradation of the herbicide bromoxynil (3,5-dibromo-4-hydroxybenzonitrile) by purified pentachlorophenol hydroxylase and whole cells of Flavobacterium sp. strain ATCC 39723 is accompanied by cyanogenesis.

A pentachlorophenol (PCP)-degrading Flavobacterium sp. (strain ATCC 39723) degraded bromoxynil with the production of bromide and cyanide. No aromatic intermediates were detected in the spent culture fluid. The cyanide produced upon bromoxynil metabolism was inhibitory to the Flavobacterium sp. Whole cells degraded PCP more rapidly than they did bromoxynil. Bromoxynil metabolism and PCP metabolism were coinduced, either substrate serving as the inducer. Purified PCP hydroxylase degraded bromoxynil with stoichiometric accumulation of cyanide and without bromide production. A product accumulated
