Total Flavones of Choerospondias axillaris Attenuate Cardiac Dysfunction and Myocardial Interstitial Fibrosis by Modulating NF-κB Signaling Pathway.

This study aimed to investigate the effect of total flavonoids of Choerospondias axillaris (TFC) on myocardial infarction (MI)-induced cardiac dysfunction, interstitial fibrosis and inflammatory reaction and further to clarify the potential signaling pathway involved. Rats were subjected to MI via coronary artery occlusion. The model establishment was confirmed by the occurrence of ST-segment elevation in electrocardiogram. Then, TFC was administrated at doses of 75, 150 and 300 mg/kg for 28 consecutive days (gavage). Body weight and heart weight were recorded. Hemodynamics, infarct size and m

Preventive effect of total flavones of Choerospondias axillaries on ischemia/reperfusion-induced myocardial infarction-related MAPK signaling pathway.

Choerospondias axillaris (Guangzao) is a medicinal plant used in Mongolia, and its fruit is commonly used for the treatment of cardiovascular diseases in clinic. The constituents responsible for this effect are always conceded to be the total flavonoids of C. axillaries (TFC). The present study was to evaluate the preventive effect of TFC on acute myocardial infarction induced by ischemia/reperfusion (I/R) in rats and the possible signaling pathway involved. The model of myocardial I/R was caused by occlusion of the left anterior descending coronary artery for 30 min followed by reperfusion fo

Flavones and structurally related 4-chromenones inhibit carbonic anhydrases by a different mechanism of action compared to coumarins.

An inhibition study of several carbonic anhydrase (CA, EC 4.2.1.1) isoforms with flavones and aminoflavones, compounds possessing a rather similar scaffold with the coumarins, recently discovered inhibitors of this enzyme, is reported. The natural product flavone and some of its hydroxylated derivatives did not show time-dependent inhibition of the CAs, sign that they are not hydrolyzed within the enzyme active site as the (thio)coumarins and lactones. These compounds were low micromolar inhibitors of hCA I, II, IX and XII, with K(I)s in the range of 1.88-9.07 μM. A series of substituted 2-ami

Pueraria flavones attenuate DSS-induced colitis by regulating the microbiota-bile acid-FXR/TGR5 axis and suppressing mtDNA-cGAS-STING signaling.

Ulcerative colitis (UC) is characterized by inflammation and impaired barrier function. Pueraria flavones (PFs) possess anti-inflammatory properties, though their underlying mechanisms remain poorly understood. This study assessed PFs in a dextran sulfate sodium (DSS)-induced colitis mouse model. PFs alleviated colitis symptoms, including weight loss and colon shortening, in a dose-dependent manner, while restoring barrier integrity and reducing pro-inflammatory cytokines. Mechanistically, PFs inhibited cGAS-STING activation by decreasing mitochondrial DNA (mtDNA) release, thereby limiting cyt

Polymethoxylated Flavones Target Cancer Stemness and Improve the Antiproliferative Effect of 5-Fluorouracil in a 3D Cell Model of Colorectal Cancer.

Polymethoxylated flavones (PMFs) from citrus fruits are reported to present anticancer potential. However, there is a lack of information regarding their effect on cancer stem cell (CSC) populations, which has been recognized as responsible for tumor initiation, relapse, and chemoresistance. In this study, we evaluated the effect of an orange peel extract (OPE) and its main PMFs, namely, nobiletin, sinensetin, tangeretin, and scutellarein tetramethylether in targeting cell proliferation and stemness using a 3D cell model of colorectal cancer composed of HT29 cell spheroids cultured for 7 days

Therapeutic Implications of a Polymethoxylated Flavone, Tangeretin, in the Management of Cancer via Modulation of Different Molecular Pathways.

Chemotherapeutics can induce oxidative stress, inflammation, apoptosis, mitochondrial dysfunction, and abnormalities in neurotransmitter metabolism leading to toxicity. Because there have been no therapeutic strategies developed to target inflammation and oxidative stress, there is a continuing need for new and improved therapy. As a result, there has been increasing interest in complementary and alternative medicine with anticancer potential. Studies have shown that the antioxidant activities and anti-inflammatory effects of citrus fruits are promising natural phytochemicals in the developmen

Chrysin (5,7-di-OH-flavone), a naturally-occurring ligand for benzodiazepine receptors, with anticonvulsant properties.

Chrysin (5,7-di-OH-flavone) was identified in Passiflora coerulea L., a plant used as a sedative in folkloric medicine. Chrysin was found to be a ligand for the benzodiazepine receptors, both central (Ki = 3 microM, competitive mechanism) and peripheral (Ki = 13 microM, mixed-type mechanism). Administered to mice by the intracerebroventricular route, chrysin was able to prevent the expression of tonic-clonic seizures induced by pentylenetertrazol. Ro 15-1788, a central benzodiazepine receptor antagonist, abolished this effect. In addition, all of the treated mice lose the normal righting refle

Flavone acetic acid (LM-975; NSC-347512) activation to cytotoxic species in vivo and in vitro.

Flavone acetic acid (FAA; LM 975; NSC 347512) is a new anticancer agent with unprecedented, broad antitumor activity in murine models. Although FAA is very effective in vivo against solid tumors, including colon 38 adenocarcinoma, it was not cytotoxic in vitro against colon 38 cells and human colon adenocarcinoma cells HCT116 at pharmacologically achievable concentrations and exposure times. For example, a concentration of 300 micrograms/ml for a 10-day exposure time was required to obtain less than 1 log cell kill. After the administration of an effective FAA dose (180 mg/kg, i.v.) to mice, p

Actions of flavonoids and the novel anti-inflammatory flavone, hypolaetin-8-glucoside, on prostaglandin biosynthesis and inactivation.

The newly identified plant-derived flavone-glycoside hypolaetin-8-glucoside, which has anti-inflammatory and gastric ulcer protective properties, and its corresponding aglycone, hypolaetin, were tested for effects on prostaglandin biosynthesis and degradation. They were compared with four other flavonoids, viz. rutin and its corresponding aglycone, quercetin, and the aglycones isoscutellarein and kaempferol. Over the range 10-1000 microM the glycosides rutin and hypolaetin-8-glucoside stimulated prostaglandin formation by sheep seminal vesicle microsomes incubated with radiolabelled arachidoni

Total flavones of Rhododendron protect against BBB injury following cerebral ischemia/reperfusion via promoting astrocytic polarization towards A2 subtype.

This study investigates the role and underlying mechanisms of total flavones of Rhododendron (TFR) in blood-brain barrier (BBB) disruption following transient bilateral common carotid artery occlusion-induced cerebral ischemia/reperfusion (I/R), focusing on whether A2 astrocyte polarization mediates the protective effects of TFR. RNA sequencing analysis was performed to elucidate the molecular mechanisms underlying the differential effects of A1 and A2 astrocytes on endothelial function. TFR treatment significantly reduced Evans blue (EB) extravasation and attenuated the downregulation of clau

Antiplatelet aggregation and antithrombotic benefits of terpenes and flavones from hawthorn leaf extract isolated using the activity-guided method.

Hawthorn is a well-known functional food; at present, increasing attention has been given to hawthorn leaf due to its numerous functional and nutritional properties. In this study, the antithrombotic properties of hawthorn leaves were evaluated using the activity-guided isolation process and high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (HPLC-QTOF-MS). A crude extract prepared in 75% ethanol was fractionated using macroporous resin D101 and polyamide chromatography to obtain three active fractions (Fr.C, Fr.C-1 and Fr.C-2). Thereafter, the inhibitory activi

Design, synthesis, and structure-activity relationships of novel flavone derivatives as tankyrase inhibitors against colorectal cancer.

Colorectal cancer (CRC) is a lethal malignancy driven by Wnt/β-catenin pathway hyperactivation. Tankyrase (TNKS) is a promising therapeutic target for CRC; however, existing inhibitors suffer from poor subtype selectivity and severe intestinal toxicity. Guided by structure-based drug design (SBDD), we developed a novel series of flavone-based TNKS inhibitors using 4'-hydroxyflavone, a plant-derived hit, as the structural scaffold. By systematically introducing diverse aliphatic amines and fluorine atoms onto the flavone B-ring, we designed and synthesized 37 derivatives. Structure-activity rel

A Novel Series of Flavone-Cinnamic Acid Hybrid Derivatives as Hypoglycemic Agents: Design, Synthesis, and Activity Studies.

In this work, α-glucosidase was chosen as the main therapeutic target. Novel flavone-cinnamic acid hybrid derivatives were designed, synthesized, and structurally identified based on flavone prototypes with intrinsic α-glucosidase inhibitory effects. Considering the multi-target pathological features of diabetes, α-glucosidase inhibition, lipase inhibition, DPPH radical scavenging capacities, and insulin-resistant HepG2 cell assays were systematically evaluated to clarify their hypoglycemic mechanisms. Most acacetin-cinnamic acid hybrid derivatives exerted strong dual inhibitory effects on the

Janus particle-type behavior of a model flavone derivative with distinct solvent-dependent competitive photoinduced processes.

In this article, Janus particle-type behavior, i.e. bi-faces character with respect to photoinduced charge transfer (CT) and proton transfer (PT) reactions, has been explored in the custom designed molecule 3-hydroxy-2-(4-(piperidin-1-yl)phenyl)-4H-chromen-4-one (HPPC), a molecule with both CT and PT sites. Solvent dependent distinct excited state CT and PT processes in HPPC have been explored using steady state and time resolved fluorescence spectroscopic measurements and quantum chemical calculations. The molecule HPPC shows only an excited state PT process in non-polar solvents like n-hepta

Metabolomic effects of total flavone of Abelmoschus manihot (L.) medik. on patients with radiation-induced heart disease.

Radiation-induced heart disease (RIHD) is a severe complication of thoracic radiotherapy. Total flavone of Abelmoschus Manihot (L.) Medik. (TFA) demonstrates therapeutic potential on RIHD. However, its metabolic mechanisms remain elusive. This study aims to elucidate the change of serum metabolic profile of RIHD treated with TFA and identify potential metabolic pathways for mitigating irradiation damage. We randomly divided 100 RIHD patients into two groups, (1) TFA group: patients receiving TFA intervention (n = 50) and (2) non-TFA group: the other not receiving intervention (n = 50). The ser

Antagonizing astrocytic platelet activating factor receptor-neuroinflammation for total flavone of epimedium in response to cuprizone demyelination.

Demyelinating diseases of the central nervous system are characterized by recurrent demyelination and progressive neurodegeneration, but there are no clinical drugs targeting myelin regeneration or improving functional disability in the treatment of multiple sclerosis. Total flavone of Epimedium (TFE) is the main active components of Epimedium, which exhibits the beneficial biological activities in the treatment of diseases, but there is no report in the treatment of demyelinating disorder. The purpose of this study was to explore the therapeutic potential and possible mechanism of TFE in the

Plant-derived flavone Apigenin: The small-molecule with promising activity against therapeutically resistant prostate cancer.

Prostate cancer is the second leading cause of cancer related deaths in men in the United States. Mounting evidences suggest that in the pathophysiology of prostate cancer epigenetic modifications play a considerable role. Histone deacetylases (HDACs) have strong crosstalk with prostate cancer progression as they regulate various genes meant for tumour suppression. HDACs are emerging as striking molecular targets for anticancer drugs and therapy as their aberrant expression has been implicated in several cancers. Histone deacetylase inhibitors (HDACi), the small molecules interfering HDACs are

Dual effects of nobiletin, a citrus polymethoxy flavone, on catecholamine secretion in cultured bovine adrenal medullary cells.

Nobiletin, a compound of polymethoxy flavones found in citrus fruits, possesses a wide range of pharmacological activities. Here we report the effects of nobiletin on catecholamine secretion in cultured bovine adrenal medullary cells. Nobiletin (1.0-100 microM) concentration-dependently stimulated catecholamine secretion and (45)Ca(2+) influx. Its stimulatory effect of nobiletin on catecholamine secretion was abolished by deprivation of extracellular Ca(2+) and partially inhibited by specific inhibitors of voltage-dependent Ca(2+) channels and Na(+)/Ca(2+) exchangers. On the other hand, nobile

Site-specific DNA cleavage by mammalian DNA topoisomerase II induced by novel flavone and catechin derivatives.

Four naturally occurring flavones (baicalein, quercetin, quercetagetin and myricetin) and two novel catechins [(-)-epicatechin gallate and (-)-epigallocatechin gallate, from the tea plant Camellia sinensis], which are known inhibitors of reverse transcriptase, were shown to induce mammalian topoisomerase II-dependent DNA-cleavage in vitro. The flavones differed from the catechins in causing unwinding of duplex DNA, but both classes of compound induced enzymic DNA breakage at the same sites on DNA. Moreover, the cleavage specificity was the same as that for the known intercalator 4'-(acridin-9-

Induction of natural killer cell activity by the antitumour compound flavone acetic acid (NSC 347 512).

Flavone acetic acid (FAA), an antitumour drug at present undergoing clinical trial for cancer treatment, has been found to activate natural killer cell activity in spleen cells of mice following in vivo treatment at doses of 45-330 mg/kg. The activity of FAA was measured using a 51Cr-release assay, employing the YAC-1 lymphoma line as a target cell population, and was found to be comparable to that of the interferon inducer polyinosinic acid-polycytidylic acid. The induction of activity was blocked by anti-asialo GM1 antibody. The time course of induction of activity by FAA was similar to that
