Extraction of gunshot residues from the larvae of the forensically important blowfly Calliphora dubia (Macquart) (Diptera: Calliphoridae).

Whole body concentrations of lead (Pb), barium (Ba) and antimony (Sb) were determined in larvae of the blowfly Calliphora dubia (Macquart) (Diptera: Calliphoridae) removed from a piece of beef shot and contaminated with gunshot residue and compared with the concentrations detected within larvae feeding on a control piece of beef. Whole larvae were taken into solution and analysed using inductively coupled plasma-mass spectrometry (ICP-MS). Significantly higher concentrations of Pb, Ba and Sb were detected within the larvae feeding on the shot piece of beef compared with larvae that were feedin

Experimental effects of paracetamol overdose on the development of forensic insects.

In forensic entomology, postmortem interval (PMI) estimation relies on the age, development, and succession patterns of insects on decomposing remains; however, the presence of drugs in the body can disrupt insect development and compromise the accuracy of these estimates. Paracetamol, a commonly used over-the-counter drug in Southern Africa, is often implicated in both intentional and accidental overdose cases. This study investigates the effects of paracetamol on insect development using spiked pig carcasses as experimental models to simulate human overdose. Three pigs received paracetamol a

Faunistic Analysis of the Families Calliphoridae and Mesembrinellidae (Diptera) at Jardim Botânico do Rio de Janeiro, Brazil.

Flies of the family Calliphoridae play a variety of ecological roles. They carry various pathogens and cause myiasis in humans and livestock, but they are useful to forensic entomology and in larval therapy. Mesembrinellidae flies, formerly classified in the family Calliphoridae, are good bioindicators of human interference in natural environments. In this study, we carried out an inventory of the Calliphoridae and Mesembrinellidae at four collecting sites within the Jardim Botânico do Rio de Janeiro. Eight traps were set, four in the arboretum (sites A and B), where the public is allowed to v

Molecular identification of forensically important blowfly species (Diptera: Calliphoridae) from Germany.

Forensic entomology applies knowledge about the behaviour and ecology of insects associated to corpses to homicide investigations. It is possible to calculate a minimum post-mortem interval by determining the age of the oldest blowfly larvae feeding on a corpse. The growth rate of the larvae is highly dependent on temperature and also varies between the different blowfly species infesting a corpse. It is, thus, crucial to correctly identify the species collected from a crime scene. To increase the quality of species identification, molecular methods were applied to 53 individuals of six differ

Genetic approaches for studying myiasis-causing flies: molecular markers and mitochondrial genomics.

"Myiasis-causing flies" is a generic term that includes species from numerous dipteran families, mainly Calliphoridae and Oestridae, of which blowflies, screwworm flies and botflies are among the most important. This group of flies is characterized by the ability of their larvae to develop in animal flesh. When the host is a live vertebrate, such parasitism by dipterous larvae is known as primary myiasis. Myiasis-causing flies can be classified as saprophagous (free-living species), facultative or obligate parasites. Many of these flies are of great medical and veterinary importance in Brazil

COI barcode reference dataset for forensic identification of Silphidae (Coleoptera) in Korea.

In forensic entomology, coleopteran evidence can provide important supplementary information when mPMI estimation based primarily on Diptera is limited; however, molecular reference data for Korean Silphidae remain scarce. This study aimed to establish a COI sequence-based reference dataset for forensically important Korean Silphidae and to support DNA barcoding-based species identification. Adult silphids were collected during pig decomposition experiments and identified morphologically. COI sequencing was performed for 38 adult specimens representing seven Silphidae species. The combined analysis of hair samples with fly larvae for the forensic identification of skeletonized human remains.

PURPOSE: In the present work, we report the combined use of hair and fly larvae samples as a valuable approach for obtaining additional toxicological evidence in cases of skeletonized corpses. In highly decomposed human remains, the absence of conventional biological specimens (e.g. blood, urine, and organs) requires alternative matrices to base the forensic investigation upon. However, it is challenging to find the most suitable medico-legal approach in these cases. METHODS: Hair and fly larvae samples were collected from two independent cases and processed by solid phase extraction using a

Standardization of histological procedures for the detection of toxic substances by immunohistochemistry in Dipteran larvae of forensic importance.

Immunohistochemistry (IHC) technique is an alternative toxicological analysis to detect drugs in insects of forensic importance, but it requires thorough histological procedures. In this study, we tested different fixatives--phosphate-buffered paraformaldehyde 4% (PP), Carnoy's fluid (CF), Kahle's solution (KS), ethanol in different concentrations, and ethanol associated to PP and CF, time of fixation and histological processes for dipteran larvae's tissue, aiming to develop a sample preparation protocol for IHC application. A suitable fixation was achieved using PP for 12 and 24 h, CF for 3 h

Proteomic profiling of bone for the estimation of post-mortem interval and post-mortem submersion interval: a systematic review.

Accurate estimation of the Post-Mortem Interval (PMI) and Post-Mortem Submersion Interval (PMSI) remains a persistent challenge in forensic science, especially when traditional morphological and entomological methods fail due to advanced decomposition or in aquatic environments. Proteomic profiling of bone tissues has recently emerged as a promising approach, leveraging the predictable degradation patterns of bone proteins to estimate time since death more reliably. This systematic review, conducted in accordance with PRISMA guidelines, analyzed 24 peer-reviewed studies focusing on the applica

Diversity and dynamics of bacteria at the Chrysomya megacephala pupal stage revealed by third-generation sequencing.

Characterization of the microbial community is essential for understanding the symbiotic relationships between microbes and host insects. Chrysomya megacephala is a vital resource, a forensic insect, a pollinator, and a vector for enteric bacteria, protozoa, helminths, and viruses. However, research on its microbial community is incomprehensive, particularly at the pupal stage, which comprises approximately half of the entire larval development stage and is important entomological evidence in forensic medicine. For the first time, this study investigated the bacterial communities of C. megacep

First contribution of mites (Acari) to the forensic analysis of hanged corpses: a case study from Spain.

This case study from North Spain, highlights the importance of the collection of mites in addition to insects, from crime scenes or corpses subjected to environmental constraints that reduce or minimise insect activity, such as hanged corpses. In addition, this analysis highlights the relevance of arthropods' collection in the field, even after the corpse has been moved away for autopsy. Four species of mites, phoretic on carrion (Silphidae) and rove (Staphylinidae) beetles, complemented and reinforced the autopsy analysis as well as the scarce information provided by insect activity. Confocal Laser Scanning Microscopy as a Method for Identifying Variation in Puparial Morphology and Establishing Characters for Taxonomic Determination.

Calliphoridae, or blow flies, are of much ecological and practical importance given their roles in decompositional ecology, medical and veterinary myiasis, and forensic entomology. As ephemeral and rapidly developing species, adults are frequently not present for identification, but puparia (the remaining outer integument of the third instar larvae) are frequently found. These heavily sclerotized remains are stable in the environment but they are of a conservative character. Historically, scanning electron microscopy (SEM) has been used for characterization, a technique which is not only time-

Ultrastructure of immature stages of Cochliomyia macellaria (Diptera: Calliphoridae), a fly of medical and veterinary importance.

Cochliomyia macellaria (Diptera: Calliphoridae) is known as the secondary screwworm because it causes secondary or facultative myiasis when the larvae feed on necrotic tissues. This fly has a significant medical and veterinary importance since it has been reported to transport eggs of Dermatobia hominis (human botfly), which can cause significant economic losses to livestock. Since this screwworm has been collected colonizing both pig carcasses and human cadavers, it is considered one of the most important species for forensic entomology studies. Scanning electron microscopy (SEM) gives detail

Selection and evaluation of reference genes for RT-qPCR normalization across developmental stages and tissues in Dermestes maculatus (Coleoptera: Dermestidae).

Real-time quantitative polymerase chain reaction (RT-qPCR) is widely used for gene expression quantification, and stably expressed reference genes are essential for accurate data normalization. However, validated reference genes are not yet available for the hide beetle, Dermestes maculatus De Geer, 1774 (Coleoptera: Dermestidae), an economically important stored-product pest and a forensically relevant necrophagous beetle. In this study, 8 candidate reference genes (RPL4, RPL13A, RPL27, RPS3, RPS18, RPS28b, GAPDH, and EF1α) were systematically evaluated across 8 developmental stages, 6 tissue

Alteration of expirated bloodstain patterns by Calliphora vicina and Lucilia sericata (Diptera: Calliphoridae) through ingestion and deposition of artifacts.

Bloodstain pattern analysis can provide insight into a sequence of events associated with a violent crime. However, bloodstain pattern analysis can be confounded by the feeding activity of blow flies. We conducted two laboratory experiments to investigate the relationships between Lucilia sericata (green bottle fly) and Calliphora vicina (blue bottle fly), expirated bloodstains, and pooled bloodstains on a range of surfaces (linoleum, wallpaper, textured paint). C. vicina and L. sericata changed bloodstain pattern morphology through feeding and defecation. They also deposited artifacts in room

Serial estimation of gene expression of cardiac troponin I (cTnI) and autophagy gene HMGB1 to determine postmortem interval.

The estimation of accurate post mortem interval (PMI) is a crucial question in forensic medicine. Several approaches have been used to determine the PMI including physical, metabolic, autolytic, entomological, physiochemical and biochemical methods over time. For estimation of PMI, RNA degradation after death is reported to be an important tool. This study aimed to analyse the pattern of gene expression by serial estimation of cardiac specific cardiac troponin I (cTnI) gene and autophagy gene HMGB1 for determining PMI at room temperature by using housekeeping gene GAPDH. Pupal age estimation of forensically important Megaselia spiracularis Schmitz (Diptera: Phoridae).

In addition to causing myiasis in humans, Megaselia spiracularis Schmitz has also been reported as a forensically important fly. The determination of a minimum post-mortem interval (PMI) often relies on the determination of the age of the larvae and pupae. The pupal stage represents about 50% of the immature development time and the pupal age may therefore serve as an important tool in entomological PMI estimation. The present study focusses on the key developmental processes during the metamorphosis of M. spiracularis pupae at different constant temperatures (21, 24, 27, 30, 33 and 36 °C). Th

A BASIC algorithm for calculating the postmortem interval from arthropod successional data.

A computer algorithm, implemented in the BASIC language, is presented for calculating the postmortem interval (PMI) from arthropod successional data. Entomology-assisted determination of the PMI promises to be a reliable technique in cases of homicide, suicide, accidental death, and unattended death due to natural causes. The program requires, as input, the identity of arthropod taxa recovered from human remains in a death scene investigation and machine-readable data on carrion-associated arthropod taxa and their known successional patterns of activity for the same geographical area. The prog

Effects of bleach on carcass postmortem microbiomes and necrophagous beetle carcass attendance and their internal microbiomes.

Forensic entomology, and potentially microbiology in the future, can be used during death investigations to corroborate an estimated time since death. Instances where products are introduced to a cadaver, either in attempt to conceal or by accident, can affect decomposition and succession of associated decomposer taxa (i.e., insects and bacteria). In this study, we tested the effect of two bleach (sodium hypochlorite) submersions on the outdoor decomposition and succession of carcass, soil, and beetle postmortem microbiomes using replicate (n = 5 per treatment) stillborn swine carcasses.
