Activation of the complement system by pathogenic fungi.

Fungi have been studied as prototype activators of the complement cascade since the early 1900s. More recently, attention has focused on the role of the complement system in the pathogenesis of fungal infections. The interactions of Cryptococcus neoformans and Candida albicans with the complement system are the most widely characterized; however, all pathogenic fungi examined to date have the ability to initiate the complement cascade. The molecular mechanisms for initiation and regulation of the complement cascade differ from one fungus to another, most likely reflecting differences in the st

Phylogenetic spectrum of fungi that are pathogenic to humans.

Recent phylogenetic studies based on ribosomal RNA sequences have confirmed that the organisms traditionally treated as fungi include those that have evolved from several different lines (multiphyletic organisms), as has been suspected. Even organisms causing disease in humans represent at least two evolutional lines. Pythium insidiosum and Prototheca species are both believed to have evolved from one line, while the rest of the pathogens have evolved from another line. P. insidiosum is more closely related to red algae and diatoms than to fungi. Prototheca species, as has been previously post

The medically important dematiaceous fungi and their identification.

Dematiaceous fungi include a large group of organisms that are darkly pigmented (dark brown, olivaceous, or black). In most cases the pigment is melanin, and specifically, dihydroxynaphthalene melanin. The diseases produced include chromoblastomycosis, eumycotic mycetoma, and phaeohyphomycosis. Phaeohyphomycosis is a new classification for a diverse group of previously known entities grouped together on the basis of finding dematiaceous hyphal and/or yeast-like forms in tissue; tissue involvement may be superficial, cutaneous and corneal, subcutaneous, or systemic. Identification of these fung

[The role of clothing in the transmission of pathogenic fungi ].

Laboratory studies were made to examine the possibility of transmission of pathogenic fungi through textiles. 1. Dermatophytes were proven to be present in clothing and leather articles worn over areas infected by fungi. In patients known to be infected with Pityriasis versicolor, contaminated scales were obtained from their textiles, the fungi could be demonstrated. 2. Artificially infected textiles were found to harbor dermatophytes and pathogenic yeasts for a number of weeks without limiting the survival capacity of the fungi. 3. At low temperatures, fungi were not affected by various deter

Culturable Fungi Recovered from Deep-Sea Sediments: Diversity, Phylogenetic Placement, and In Vitro Antagonistic Activity Against Phytopathogenic Fungi.

Deep-sea sediment-derived fungal isolates remain underrepresented in organized living collections for phenotype-based screening. We applied 12 complementary isolation protocols-dilution plating, ethylenediaminetetraacetic acid (EDTA) pretreatment, and plate stamping-to four deep-sea sediment materials (2595-6000 m) representing three sampling units from the Mariana Trench slope region and the South China Sea. After morphotype-based purification, isolates were characterized by internal transcribed spacer (ITS) closest-match analysis and phylogenetic placement; selected representatives were furt

The legally protected fungi and lichens in the EU.

Despite decades of recommendations from the scientific community, fungi have yet to be included in any international legal instruments that regulate wildlife species conservation in the European Union (EU), such as the Bern Convention. However, several EU member states include lists of protected non-lichenized and lichenized fungal species in their national legislation. Most of these legal documents are available only in local languages and are dispersed across national legal databases. To increase the visibility of fungi protected across the EU and facilitate comparisons of these regulations,

Clinical Relevance of Fungi in Cystic Fibrosis.

In cystic fibrosis, a new era has started with the approval and use of highly effective cystic fibrosis transport regulator (CFTR) modulator therapy. As pulmonary function is increasing and exacerbation rate significantly decreases, the current meaning of fungal pulmonary diseases is questioned. During the past couple of decades, several studies have been conducted regarding fungal colonization and infection of the airways in people with cystic fibrosis. Although Aspergillus fumigatus for filamentous fungi and Candida albicans for yeasts remain by far the most common fungal species in patients

Epigenetic mechanisms of drug resistance in fungi.

The emergence of drug-resistant fungi poses a continuously increasing threat to human health. Despite advances in preventive care and diagnostics, resistant fungi continue to cause significant mortality, especially in immunocompromised patients. Therapeutic resources are further limited by current usage of only four major classes of antifungal drugs. Resistance against these drugs has already been observed in pathogenic fungi requiring the development of much needed newer antifungal drugs. Epigenetic changes such as DNA or chromatin modifications alter gene expression levels in response to cer

Not a fungi to be with: Shiitake mushroom flagellate dermatitis.

Shiitake dermatitis is a rare adverse cutaneous reaction to the ingestion of raw or undercooked shiitake mushrooms (Lentinula edodes). We report the case of a patient who developed a striking linear flagellate dermatitis without urticaria three days after returning from a trip from Tokyo where he had eaten shiitake mushrooms. The rash resolved after two weeks with topical corticosteroids and antihistamines given for symptomatic relief. Shiitake dermatitis is thought to be either a toxic or hypersensitivity reaction to lentinan, a heat-inactivated polysaccharide found in the cell walls of shiit

Bacteria that masquerade as fungi: actinomycosis/nocardia.

The order Actinomycetales includes phylogenetically diverse but morphologically similar aerobic and anaerobic bacteria that exhibit filamentous branching structures which fragment into bacillary or coccoid forms. The aerobic actinomyces are a large, diverse group of gram-positive bacteria including Nocardia, Gordona, Tsukamurella, Streptomyces, Rhodococcus, Streptomycetes, Mycobacteria, and Corynebacteria. The anaerobic genera of medical importance include Actinomyces, Arachnia, Rothia, and Bifidobacterium. Both Actinomyces and Nocardia cause similar clinical syndromes involving the lung, bone

[The role of parasites and fungi in secondary infertility].

INTRODUCTION: Parasite-host relationships can cause diminished or absent ability to conceive, ectopic pregnancy or pregnancy with undesired course. LITERATURE REVIEW: There are reports that some protozoa, helminths and fungi may impair women's reproductive capacity, causing deformities of genital tract, so that conception is impossible, or, if it does occur, normal implantation and development of placenta are impossible. Schistosoma haematobium may cause vulvar papule, swelling, tumors, irregular vaginal hemorrhage, tubular infertility and ectopic pregnancies. Patients with cirrhosis caused b

Molecular evolution of the fungi: human pathogens.

The morphological, ecological, and clinical diversity among ascomycete fungi that are pathogenic to humans suggest that the potential for pathogenicity may have arisen multiple times within these higher fungi. We have obtained 18S ribosomal DNA sequences from a diverse group of human pathogenic fungi in order to determine their evolutionary origins. The fungi studied include a skin pathogen that is confined to humans (Trichophyton rubrum) and three systemic, facultative parasites that cause histoplasmosis (Histoplasma capsulatum), blastomycosis (Blastomyces dermatitidis) and coccidioidomycosis

Acid-fastness of fungi in blastomycosis and histoplasmosis.

In 60% of the infections caused by Blastomyces dermatitidis and in 47% of the infections caused by Histoplasma capsulatum, some of the organisms are acid-fast in tissue sections. Treatment with 1N hydrochloric acid at 60 degrees C abolishes the acid-fastness of H capsulatum, which indicates that the acid-fastness may be related to mycolic acid. Organisms of B dermatitidis, however, remain acid-fast in 83% of the sections treated with acid. The presence of acid-fastness in small cells can aid in the differentiation of B dermatitidis and H capsulatum from Cryptococcus neoformans and Candida albi
