Effects of psoriasis and psoralen exposure on the somatic mutation landscape of the skin.

Somatic mutations are hypothesized to play a role in many non-neoplastic diseases. We performed whole-exome sequencing of 1,182 microbiopsies dissected from lesional and nonlesional epidermis from 111 patients with psoriasis to search for evidence that somatic mutations in keratinocytes may influence the disease process. Lesional skin remained highly polyclonal, showing no evidence of large-scale spread of clones carrying potentially pathogenic mutations. The mutation rate of keratinocytes was similarly only modestly affected by the disease. We found evidence of positive selection in previousl

Targeted photochemotherapy in alopecia areata.

BACKGROUND/PURPOSE: Alopecia areata (AA) is a common cause of localized non-scarring alopecia. Usage of targeted UVA after the topical application of 8-methoxypsoralen (8-MOP) is one of the rising treatment modalities for AA. Our aim was to assess the efficacy and safety of topical 8-MOP plus targeted UVA phototherapy in the treatment of patchy AA. METHODS: Seven patchy AA patients were treated by topical 8-MOP application to the lesions followed by UVA irradiation 3 times a week, with 15 to 24 sessions in total. At the end of the treatment all patients were evaluated for response on a four-p

Biosynthesis and heterologous production of furanocoumarins: perspectives and current challenges.

Covering: up to October 2020 Furanocoumarins are plant secondary metabolites used to treat several skin disorders, such as psoriasis and vitiligo, and also with other potential therapeutic activities. Furanocoumarins are extracted from plants where they accumulate in low amounts over long growth periods. In addition, their extraction and purification are difficult in an environmentally unfriendly and expensive process. Hence, new sustainable and greener production schemes able to overcome such limitations ought to be developed. While the heterologous production of simple coumarins has been dem

The evolution of photochemotherapy with psoralens and UVA (PUVA): 2000 BC to 1992 AD.

The therapeutic uses of naturally occurring psoralens in modern-day medicine (8-methoxypsoralen (8-MOP), 5-methoxypsoralen (5-MOP), 4,5',8-trimethylpsoralen, and a few other synthetic psoralens) have evolved through five stages of development. (1) In the historical period (2000 BC to 1930 AD), the pigment-stimulating properties of naturally occurring plants containing psoralens were described anecdotally. (2) The second period (1930-1960) dealing with the chemistry of psoralens involved extraction, identification of their structure, synthesis, and the relationship between chemical structure an

Photosensitizing effects of Cymopterus watsonii and Cymopterus longipes in chickens and turkey poults.

Two-week-old straight-run white broiler chicks and 2-week-old straight-run white turkey poults were fed seed or whole plants Cymopterus watsonii or seed of Cymopterus longipes at 0.4, 0.8, and 1.6% of body weight, and then placed in sunlight for 5 hours each day for 7 days. All plant material produced moderate to severe photosensitivity in chicks. Toxic signs included photophobia; red discoloration of beak, comb, and feet; loss of feathers in the periorbital area; dried serous fluid on comb and edge of beak; keratoconjunctivitis; and multiple lesions on feet and legs. Turkey poults showed a sy

Bath-PUVA still represents a valuable treatment option for the subsets of psoriatic patients who are not eligible to or rejecting systemic treatments and are not responsive to NB-UVB phototherapy.

BACKGROUND: Photochemotherapy with bathwater delivery of psoralens plus UVA exposures (bath-PUVA) is mainly used for those psoriatic patients who are not responsive to narrowband (NB)-UVB phototherapy and oral-PUVA therapy and belong to two categories (1) patients with psoriasis without systemic comorbidities who do not need long-term continuous treatment and (2) patients who have contraindications to immunosuppressive drugs and oral-PUVA or refuse systemic drugs, including oral ingestion of psoralens, for personal reasons. However, it is not known how many patients belong to the second group

Isolation of the new minor constituents dihydropyranochromone and furanocoumarin from fruits of Peucedanum alsaticum L. by high-speed counter-current chromatography.

A preparative high-speed counter-current chromatography (HSCCC) method was successfully used for isolation of two new minor compounds--alsaticol and alsaticocoumarin A. A two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water (1:1:1:1) was developed. Compounds were obtained from the dichloromethane extract of Peucedanum alsaticum fruits and their identification was performed with NMR and MS methods. Optimized HSCCC offers a rapid method of obtaining new natural compounds.

Photocarcinogenicity of psoralens used in PUVA treatment: present status in mouse and man.

There is good evidence that 8-methoxypsoralen is photocarcinogenic in psoriatic patients undergoing long-term photochemotherapy (PUVA) in the U.S.A. However, this conclusion has not been supported by two major European studies which have indicated that PUVA is a tumour promoter of damage initiated by other agents. Variation in PUVA treatment protocols in the U.S.A. and Europe may partly account for the different conclusions. There is much interest in the therapeutic potential of monofunctional psoralens. It is hoped that these may reduce long-term risk. Monofunctional and cross-linking psorale

[Phytophotodermatitis. Some photobiological aspects of psoralens].

The frequency of plant dermatitis, in general, depends on the type of plants, the climate, occupation and lifestyle of the local inhabitants. Many plants produce adverse effects on the skin like irritation, allergic reactions and phototoxicity. Generally, allergic contact dermatitis is emphasized but it is necessary to give more attention to phototoxicity produced by plants. The patients may be exposed not only to the phototoxic plant itself but also to its derivatives in perfumes, cosmetics and industry.

Identification of psoralen, 8-methoxypsoralen, isopimpinellin, and 5,7-dimethoxycoumarin in Pelea anisata H. Mann.

5,7-dimethoxycoumarin and isopimpinellin, together with the well-known phototoxic, photo-irritant furanocoumarins psoralen and 8-methoxypsoralen, were isolated and identified from leaves and fruits of Pelea anisata H. Mann, a plant whose fruit are used in the construction of mohikana leis used in parts of the Hawaiian Islands. The presence of phototoxic furanocoumarins would explain the occurrence of photodermatitis in the wearers of the leis.

Design, synthesis, and evaluation of new series of Imperatorin analogs with potential vasodilatory activity.

Two series of imperatorin analogs were synthesized based on our previous research and evaluated for their vasodilatation activities on in vitro rat mesenteric artery, basilar artery, and renal artery ring models. Target compounds were characterized by infrared, 1H NMR, and mass spectra. Most derivatives possessed significant vasodilatory activity on the mesenteric artery, and compound 3a exhibited favorable and broad vasodilatation activities on three kinds of rat artery ring models. The pharmacological results indicated that introducing nitrogen-contained ring in side chain or large steric hi

Phytophotodermatitis From Lime Margaritas on a Mexico Vacation.

Phytophotodermatitis is a type of contact dermatitis that occurs upon skin exposure to certain plant chemicals, known as furocoumarins, along with simultaneous sun exposure. This case details a 34-year-old patient who presented to the office with an asymptomatic, irregularly shaped, and hyperpigmented patch located on the left inferior middle back that had been present since a recent beach vacation in Mexico. Upon gathering the history, clinicians should inquire about recent sunlight exposure while consuming and/or touching phytotoxic plant derivatives found in most citrus plants. The history

Citrus Essential Oils: A Review of Chemical Composition, Bioactivity, Safe Use, and Potential Applications.

The genus Citrus (Rutaceae) produces essential oils (EOs) widely utilized in food, pharmaceutical, cosmetic, and biomaterial industries due to their distinct organoleptic properties. These EOs are primarily composed of monoterpenes (e.g., limonene), sesquiterpene hydrocarbons, and oxygenated derivatives, which exhibit anti-inflammatory, antioxidant, antibacterial, antineoplastic, and insecticidal activities. Principal extraction methods include cold pressing (CP), solvent extraction, distillation, ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), and supercritical flui

Five Cases of Phytophotodermatitis Caused by Fig Leaves and Relevant Literature Review.

Phytophotodermatitis is a condition caused by sequential exposure to photosensitizing substances present in plants followed by ultraviolet light. Several plants (e.g., limes, celery, fig, and wild parsnip) contain furocoumarin compounds (psoralens). It is important for dermatologists to be aware of phytophotodermatitis because it may be misdiagnosed as cellulitis, tinea, or allergic contact dermatitis. We present five patients with a sharply defined erythematous swollen patch with bullae on both feet. They described soaking their feet in a fig leaves decoction to treat their underlying dermato

Synthesis and photobiological properties of acetylpsoralen derivatives.

In an investigation to find monofunctional reactants for DNA which can act as new agents in the photochemotherapy of psoriasis, we have synthesized and studied some methylpsoralen derivatives which contain an acetyl group at one of the two reactive sites of the furocoumarin skeleton (at the 3 or 5' positions). The compounds do not react easily with DNA; their photobiological properties (e.g. the lack of an ability to inhibit DNA synthesis in Ehrlich ascites tumour cells, to induce T2 phage sensitization and to induce erythema in guinea-pig skin) are exactly in line with this behaviour.
