G-quadruplexes (G4s) are non-canonical nucleic acid structures formed by guanine-rich sequences. Within the human genome, beyond telomeres, G4 motifs have also been found in promoters, implying a potential role in gene regulation that could be achieved through interactions with small-molecule ligands. In this field, anthraquinone represented a privileged scaffold for the design of G4-targeting molecules. Additionally, emerging evidence proved that compounds bearing two aromatic cores can offer enhanced affinity and selectivity toward G4s due to cooperative binding in a tweezer-like fashion. Th

Viral genomes are enriched with G-quadruplexes (G4s), non-canonical structures formed in DNA or RNA upon assembly of four guanine stretches into stacked quartets. Because of their critical roles, G4s are potential antiviral targets, yet their function remains largely unknown. Here, we characterize the formation and functions of a conserved G4 within the polymerase coding region of orthoflaviviruses of the Flaviviridae family. Using yellow fever virus, we determine that this G4 promotes viral replication and suppresses host stress responses via interactions with hnRNPH1, a host nuclear protein

BACKGROUND: Pathogenic mutations in WRN are a cause of premature aging disease Werner syndrome (WS). Besides accelerated aging phenotypes and cancer predisposition, patients with WS also display underdevelopment in the skeletal system, characterized by short stature, light body weight and unusually thin extremities. The reasons for these developmental defects are not completely understood and the underlying molecular mechanism remains to be elucidated.
RESULTS: In this study, WRN was found to modulate transcription of short stature homeobox gene SHOX. Loss of WRN resulted in insufficient expr

G-quadruplexes are four-stranded nucleic acid structures involved in multiple cellular pathways including DNA replication and telomere maintenance. Such structures are formed by G-rich DNA sequences typified by telomeric DNA repeats. Whilst there is evidence for proteins that bind and regulate G-quadruplex formation, the molecular basis for this remains poorly understood. The budding yeast telomeric protein Rap1, originally identified as a transcriptional regulator functioning by recognizing double-stranded DNA binding sites, was one of the first proteins to be discovered to also bind and prom

Aptamers are valuable tools that provide great potential to develop cost-effective diagnostics and therapies in the biomedical field. Here, we report a novel DNA aptamer that folds into an unconventional G-quadruplex structure able to recognize and enter specifically into human Burkitt's lymphoma cells. We further optimized this aptamer to a highly versatile and stable minimized version. The minimized aptamer can be easily equipped with different functionalities like quantum dots, organic dyes, or even a second different aptamer domain yielding a bi-paratopic aptamer. Although the target molec

The abnormal expansion of GGGGCC (G4C2) repeats in the noncoding region of the C9orf72 gene is a major genetic cause of two devastating neurodegenerative disorders, amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). These G4C2 repeats are known to form G-quadruplex (G4) structures, which are hypothesized to contribute to disease pathogenesis. Here, we demonstrated that four DNA G4C2 repeats can fold into two structurally distinct G4 conformations: a parallel and an antiparallel topology. The high-resolution crystal structure of the parallel G4 reveals an eight-layered dimer

Helicobacter pylori, a leading human pathogen associated with duodenal ulcer and gastric cancer, presents a significant threat to human health due to increasing antibiotic resistance rates. This study investigates G-quadruplexes (G4s), which are non-canonical secondary structures form in G-rich regions within the H. pylori genome. Extensive research on G4s in eukaryotes has revealed their role in epigenetically regulating cellular processes like gene transcription, DNA replication, and oncogene expression. However, understanding of G4-mediated gene regulation in other organisms, especially bac

The mRNA SARS-CoV-2 vaccines were brought to market in response to the public health crises of Covid-19. The utilization of mRNA vaccines in the context of infectious disease has no precedent. The many alterations in the vaccine mRNA hide the mRNA from cellular defenses and promote a longer biological half-life and high production of spike protein. However, the immune response to the vaccine is very different from that to a SARS-CoV-2 infection. In this paper, we present evidence that vaccination induces a profound impairment in type I interferon signaling, which has diverse adverse consequenc

Biomimetics is defined as a "practice of making technological design that copies natural processes", with the idea that "nature has already solved the challenges we are trying to solve" (Cambridge Dictionary). The challenge we decided to address several years ago was the selective targeting of G quadruplexes (G4s) by small molecules (G4 ligands). Why? Because G4s, which are four-stranded DNA and RNA structures that fold from guanine (G)-rich sequences, are suspected to play key biological roles in human cells and diseases. Selective G4 ligands can thus be used as small-molecule modulators to g

Pt-ttpy (tolyl terpyridin-Pt complex) covalently binds to G-quadruplex (G4) structures in vitro and to telomeres in cellulo via its Pt moiety. Here, we identified its targets in the human genome, in comparison to Pt-tpy, its derivative without G4 affinity, and cisplatin. Pt-ttpy, but not Pt-tpy, induces the release of the shelterin protein TRF2 from telomeres concomitantly to the formation of DNA damage foci at telomeres but also at other chromosomal locations. γ-H2AX chromatin immunoprecipitation (ChIP-seq) after treatment with Pt-ttpy or cisplatin revealed accumulation in G- and A-rich tande

Extracellular DNA (eDNA) in bacterial biofilms can form non-canonical structures like Z-DNA and G-quadruplex (G4), which enhance biofilm resilience by providing protection against mammalian DNases. However, the conformation of eDNA in dental biofilms remains unexplored. Using fluorescence immunolabeling and confocal microscopy, we examined dental biofilms from healthy and caries-active subjects, revealing B-DNA, G4-, and Z-DNA structures surrounding clusters of bacteria, with some structures directly associated with the bacterial cell surface. We demonstrated that these non-canonical DNA struc

Overexpression of c-Myc is a key factor in the development of leukemia and other malignancies, highlighting the urgent need for novel drugs to inhibit c-Myc protein levels. DNA G-quadruplexes (G4) have emerged as potential regulatory targets for c-Myc expression. Previous studies identified trovafloxacin, a topoisomerase II inhibitor, as a novel c-Myc G4 stabilizer. In this study, virtual screening based on structural similarity led to the identification of nine derivatives of trovafloxacin, among which voreloxin exhibited potent cytotoxicity in multiple myeloma cells and showed promising ther

Transthyretin Amyloidosis arises from the misfolding of monomers or oligomers of the normal transthyretin protein. Our investigation revealed that certain guanine-rich regions within the 5' UTR sequence of the transthyretin gene possess the ability to form G2-quadruplex structures, as determined through analysis with QGRS mapper. We demonstrated that small molecule ligands, including TMPyP4, Braco-19, NMM, and TO, have a significant impact on the stabilization of transthyretin G-quadruplexes. The objective of this study was to confirm the effect of ligands on transthyretin gene transcription t

G-quadruplexes (G4s) are four-stranded non-canonical nucleic acid secondary structures, formed by the stacking arrangement of guanine tetramers. They are involved in a wide range of biological processes because of their exceptionally unique and distinct structural characteristics. After the completion of the human genome sequencing project, a lot of bio-informatics algorithms were introduced to predict the active G4 regions in vitro based on the canonical G4 sequence elements, G-richness and G-skewness, as well as the non-canonical sequence features. Recently, sequencing techniques like G4-seq

G-quadruplex (G4) structures play key physiological roles and serve as functional building blocks in DNA/RNA-based aptamer biosensors. Their function critically depends on their structural stability and nonspecific interactions in cells. However, the behavior of exogenously delivered G4s in living cells remains poorly understood. Here, using 19F NMR, we monitored three representative G4 sequences, HT, hVEGFP, and TBA, in intact Xenopus laevis oocytes, HeLa cells, and derived lysates. We found that G4 structural stability, interactions, and NMR signal detectability are cell-type-dependent. In o

G-quadruplexes (G4s) are four-stranded nucleic acid structures that regulate virtually all nucleic acid-dependent cellular processes. At present, most functional studies involving G4s have focused on cancer cells. This study investigated how neurons respond to genotoxic stress induced by quarfloxin (CX-3543), a small molecule that stabilizes G4s. We found that quarfloxin treatment induced DNA damage in neurons, with double-strand breaks enriched in the nucleolus. Proteomic analysis revealed that quarfloxin promoted substantial protein changes, affecting networks associated with Alzheimer's, Pa

Conventional cancer immunotherapy suffers from insufficient immune activation, immunosuppressive tumor microenvironment (TME), and rapid CpG adjuvant degradation. To address these challenges, we developed a G-quadruplex (G4)-modular CpG nanoplatform named IONP-G4-DOX/IMT, which uses iron oxide nanoparticles (IONPs) as a stable structural and biocompatible core and G4 as a multifunctional hub. The rationally designed G4 module enables three synergistic functions encompassing enhanced CpG nuclease resistance for sustained TLR9 pathway activation, site-specific loading of doxorubicin (DOX) to tri

G-quadruplexes (GQs) are four-stranded DNA structures formed by guanine-rich sequences and can act as regulatory impediments in gene expression. The core of GQ consists of stacked layers of Hoogsteen-paired guanine bases (G-tetrads), which can arise from various topologies adopted by the backbone strands of guanine tracts (G-tracts). The polymorphic nature of GQs presents significant challenges for therapeutic targeting. An as-yet-addressed question is how, if at all, chemical moieties of different G-tetrads and G-tracts mediate distinct mechanical strengths such that the strands and layers di

The selective photodegradation of G-quadruplex DNA (GQ) was investigated using the cationic zinc phthalocyanine ZnPc as a photosensitizer. UV-Vis absorption titration suggests that electrostatic interactions contribute to ZnPc association with both duplex and G-quadruplex DNA, whereas 1H NMR analysis indicates that one major ZnPc-GQ association mode involves π-π end-stacking at the 5'-terminal G-quartet. The efficiency of singlet oxygen generation by ZnPc is similar in the presence of duplex and G-quadruplex DNA, and fluorescence-based assays indicate comparable hydroxyl radical-type reactivit

To face the increasing multidrug resistance of the malaria parasite Plasmodium falciparum, the development of new compounds that act in novel ways is urgent. G-quadruplexes (G4), Guanine-rich secondary structures of DNA and RNA sequences, have an essential role in cell survival and cell metabolism regulation, including in Plasmodium. We have recently demonstrated that the G-quadruplex ligand RHPS4, an acridine derivative, displays strong antiplasmodial activity by disrupting P. falciparum through G4 stabilization, leading to parasite death. Based on this original mode of action, we propose her
