The transient receptor potential ankyrin 1 (TRPA1) has been identified as a relevant target for the development of novel analgesics. Gallic acid (GA) is a polyphenolic compound commonly found in green tea and various berries and possesses a wide range of biological activities. The goal of this study was to identify GA as a TRPA1 antagonist and observe its antinociceptive effects in different pain models. First, we evaluated the ability of GA to affect cinnamaldehyde-induced calcium influx. Then, we observed the antinociceptive and antiedematogenic effects of GA (3-100 mg/kg) oral administratio

Xanthine oxidase is a flavoprotein enzyme which catalyzes the oxidative hydroxylation of purine substrates. Because of its availability, it has become a model for structural molybdoenzymes in general. The enzyme is a well-established target of drugs against gout and hyperuricemia and exists in two forms: oxidase and deshydrogenase. In some pathologies, its level increases in oxidase form, being the source of free radicals which can cause damage to surrounding tissues. It is important to understand the mechanisms of the enzyme inhibition to help in the search of new inhibitors. The main active

OBJECTIVE: To investigate the effect of gallic acid (GA) monomer on the cytotoxic function of tumour-infiltrating CD8+ T cells in gastric cancer and explore the underlying mechanisms.
METHODS: Tumour-infiltrating CD8+ T cells were isolated from tumour tissues of patients with gastric adenocarcinoma using fluorescence-activated cell sorting. The purified CD8+ T cells were then co-incubated with GA at five different concentrations (0, 1, 2, 4, and 8 μM) for two incubation periods (24 and 48 h). Subsequently, the cytotoxic activity of tumour-infiltrating CD8+ T cells against gastric cancer cells

Experimental evidence has shown that gallic acid (GA), a naturally occurring polyphenolic compound, and docetaxel (DTX), a taxane chemotherapeutic agent, each possess antitumor activity against multiple cancer types. Although both compounds have been investigated individually, their combined effects in triple-negative breast cancer (TNBC) have received limited attention, and the molecular basis of their interaction remains unclear. The present study examined the in vitro effects of GA and DTX in MDA-MB-231 TNBC cells while simultaneously assessing their comparative cytotoxicity in HaCaT human

Ulcerative colitis (UC), a chronic inflammatory bowel disease, exhibits increasing global prevalence. However, there remains a paucity of evidence on whether gallic acid (GA) can be used to mitigate acute and chronic colitis. In this study, the underlying mechanism of GA in alleviating UC was thoroughly investigated. Our results showed that GA protected mice from acute and chronic colitis, evidenced by relieved pathological symptoms of UC, strengthened the tight junction, and repaired the damaged intestinal barrier. Moreover, GA regulated the balance of microbiota in the gut and increased the

Gentamicin (GEN), commonly used to treat gram-negative bacterial infections, can cause testicular damage, maybe by increasing oxidative stress. Gallic acid (GA) a polyphenolic acid, has antioxidant, anti-bacterial, anti-viral, and anti-allergic activities. The study aimed to investigate the effects of GA on GEN-induced testicular toxicity. Thirty Wistar rats were randomly divided into five groups: Control, which received only normal saline, and GEN, which received 100 mg/kg/day of GEN intra-peritoneally for six days. The rats in groups 3, 4, and 5 received GA by gavage at 60, 90, and 120 mg/kg

Replicative senescence presents a significant challenge in mesenchymal stem cell (MSC) expansion due to high reactive oxygen species (ROS) levels generated during culture. Elevated ROS levels lead to oxidative stress, cellular damage, and senescence, limiting the biomedical applications of MSCs. In this study, a supramolecular thermo-reversible hydrogel composed of the natural polyphenolic compound gallic acid (GA) and polyvinyl alcohol (PVA) was designed to scavenge ROS and mitigate MSC senescence. The PVA-GA hydrogel, stabilized by strong hydrogen bonding forces, exhibited an elastic modulus

Due to the large amount of antibiotics used for human therapy, agriculture, and even aquaculture, the emergence of multidrug-resistant Streptococcus suis (S. suis) led to serious public health threats. Antibiotic-assisted strategies have emerged as a promising approach to alleviate this crisis. Here, the polyphenolic compound gallic acid was found to enhance sulfonamides against multidrug-resistant S. suis. Mechanistic analysis revealed that gallic acid effectively disrupts the integrity and function of the cytoplasmic membrane by dissipating the proton motive force of bacteria. Moreover, we f

Psoriasis and atopic dermatitis (AD) are characterized by enhanced skin inflammation, which results in hyperproliferation and the recruitment of immune cells into the skin. For that reason, it is needed a chemical capable to reduce cell proliferation and the recruitment of cells. The search for new molecules for therapeutic skin treatment mainly focuses on the antioxidant and anti-inflammatory properties, highlighting the rheological properties of polymeric polypeptides. We studied L-arginine (L-Arg) grafted (-g-) to enzymatic poly(gallic acid) (PGAL). The latter is a multiradical antioxidant

It is widely held that polyphenols accumulate in soil due to limited phenol oxidase activity, thereby inhibiting resident microbes and promoting carbon stabilization. Contradicting this paradigm, we demonstrate that bioreduced Fe(II) from iron minerals may be enriched under anaerobic conditions, which accelerates phenol decomposition. Using gallic acid (GA) as a model, the biodegradation rate was 1.6-2.2 times higher under anaerobic conditions (k = 0.021-0.26 h-1) compared to aerobic environments in bacteria (Bacillus megaterium, Lactoccus Lactis, or Shewanella putrefaciens) cocultured with ir

Gallic acid (GA), also known as 3,4,5-trihydroxybenzoic acid, is a polyphenolic bioactive component in pomegranates, grapes, tea leaves, and gallnuts. This study employed LC-MS/MS to systematically analyze the pharmacokinetic and tissue distribution of GA in rats. Our findings demonstrated that GA accumulated most prominently in kidney and prostate tissues. Based on these targeted distribution characteristics, chronic bacterial prostatitis (CBP) was chosen as a target tissue inflammatory model to explore the anti-inflammatory effects of GA. The results showed that GA exerted a direct anti-infl

Eight gallic acid alkyl esters (1−8) were synthesized via Fischer esterification and evaluated for their trypanocidal and leishmanicidal activity using bloodstream forms of Trypanosoma brucei and promastigotes of Leishmania major. The general cytotoxicity of the esters was evaluated with human HL-60 cells. The compounds displayed moderate to good trypanocidal but zero to low leishmanicidal activity. Gallic acid esters with alkyl chains of three or four carbon atoms in linear arrangement (propyl (4), butyl (5), and isopentyl (6)) were found to be the most trypanocidal compounds with 50% growth

INTRODUCTION: Ketamine (KET) is an anesthetic agent widely used in human and veterinary medicine. According to studies, KET is associated to direct neutorotoxic damages due to its capacity to induce oxidative stress. Because of the free radical generation in the organism and its relation with diseases' development, there is a growing interest to study antioxidant molecules, such as gallic acid (GA), a natural phenolic compound.
AIM: Evaluate the GA antioxidant potential for the prevention of oxidative damage in the brain and liver tissue of rats exposed to acute KET administration.

Knee osteoarthritis (KOA) is a degenerative disease associated with chronic pain, cartilage degradation, subchondral bone sclerosis, and synovial inflammation. The search for new molecules capable of reducing the release of pro-inflammatory cytokines has led to the investigation of poly (gallic acid) (PGAL), an enzyme-mediated polyoxidized form of gallic acid, due to its relatively high stability and antioxidant activity. Experimental evidence suggests that PGAL (100 ppm) is capable to decrease IL-6, TNF-α, and IL-1β levels in human osteoarthritic synoviocytes. Additionally, a combination of P

Glioblastoma (GBM) conventional treatment is not curative, and it is associated with severe toxicity. Thus, natural compounds with anti-cancer properties and lower systemic toxicity, such as gallic acid (GA), have been explored as alternatives. However, GA's therapeutic effects are limited due to its rapid metabolism, low bioavailability, and low permeability across the blood-brain barrier (BBB). This work aimed to develop poly (lactic-co-glycolic acid) (PLGA) nanoparticles (NPs) modified with folic acid (FA), as its receptor is overexpressed in BBB and GBM cells, for GA delivery to enhance it
