MicroRNA-124-3p attenuates the development of nerve injury-induced neuropathic pain by targeting early growth response 1 in the dorsal root ganglia and spinal dorsal horn.

Emerging evidence indicates the early growth response 1 (Egr1) plays an important role in the pathogenesis of chronic pain. However, the regulation of Egr1 expression in the DRG and spinal cord in neuropathic pain remains unclear. In the current study, the neuropathic pain was conducted by lumber 5 spinal nerve ligation (SNL) in rats. The role of miR-124-3p in Egr1 expression was examined. Our results showed that the SNL led to a significant increase in the expression of Egr1 mRNA and protein in the DRG and dorsal horn. This increased expression of Egr1 correlated with a reduction of miR-124-3

Melatonin Mitigates Central Sensitization and Nociplastic Pain in Spinal Cord and Dorsal Root Ganglia of FM Rat Model: Modulation of SIRT1/PGC-1α/MAPK/NF-κB Signaling.

Fibromyalgia (FM) is a central sensitization syndrome characterized by neuroinflammation and synaptic hyperexcitability, which amplifies pain signals in the spinal cord (SC) and dorsal root ganglia (DRG). Melatonin (MEL) has demonstrated analgesic and antinociceptive properties in experimental models, supporting its clinical application in various pathological conditions. This study aimed to elucidate the impact of Melatonin on neuroinflammation and glutamatergic dysregulation in the spinal cord and DRG using a Reserpine-induced fibromyalgia model (RIFM). RIFM was induced in female rats by adm

Expression of vascular endothelial growth factor and its fetal liver kinase-1 receptor in spinal cord and dorsal root ganglia after neurotomy of sciatic nerve in rats.

OBJECTIVE: To investigate the expression and pattern of vascular endothelial growth factor (VEGF) and its fetal liver kinase-1 (Flk-1) receptor in spinal cord and dorsal root ganglia after neurotomy of sciatic nerve in rats. METHODS: Forty-five adult male Wistar rats were divided randomly into a control group (n=5) and an experimental group (n=40). The bilateral sciatic nerves of the rats in the experimental group underwent neurotomy and the L4-L6 spinal cord and the corresponding dorsal root ganglia were harvested respectively at 8 hours, and 1, 3, 5, 7, 10, 14 and 21 days (8 subgroups with

Differential regulation of fibroblast growth factor (FGF)-2 and FGF receptor 1 mRNAs and FGF-2 isoforms in spinal ganglia and sciatic nerve after peripheral nerve lesion.

To study the functional role of endogenous basic fibroblast growth factor-2 (FGF-2) during degeneration and regeneration of the sensory system, we have determined the expression and regulation of FGF-2 and FGF receptor (FGFR)-1 mRNAs in spinal ganglia and sciatic nerve during experimental transection and crush injury of the sciatic nerve. In contrast to levels of the FGFR-1 transcript, which is not altered, the level of FGF-2 mRNA is dramatically up-regulated in spinal ganglia after injury. In the proximal and distal nerve stumps both transcript levels are significantly elevated, albeit at dif

Peri-sciatic administration of recombinant rat TNF-alpha induces mechanical allodynia via upregulation of TNF-alpha in dorsal root ganglia and in spinal dorsal horn: the role of NF-kappa B pathway.

Previous studies have shown that tumor necrosis factor-alpha (TNF-alpha) and TNF receptor 1 (TNFR1) in dorsal root ganglia (DRG) and in spinal dorsal horn are upregulated after nerve injury and that many TNF-alpha-containing neurons overexpress TNFR1. In the present study, we found that peri-sciatic administration of rat recombinant TNF-alpha (rrTNF) at the concentrations of 10, 100 and 1000 pg/ml (daily for 2 days) induced mechanical allodynia in bilateral hindpaws, lasting for about 20 days. The immunoreactivity (IR) of TNF-alpha and TNFR1 in the ipsilateral (but not in the contralateral) L4

Clarification of the Innervation of the Bladder, External Urethral Sphincter and Clitoris: A Neuronal Tracing Study in Female Mongrel Hound Dogs.

Many studies examining the innervation of genitourinary structures focus on either afferent or efferent inputs, or on only one structure of the system. We aimed to clarify innervation of the bladder, external urethral sphincter (EUS) and clitoris. Retrograde dyes were injected into each end organ in female dogs. Spinal cord, mid-bladder, and spinal, caudal mesenteric, sympathetic trunk and pelvic plexus ganglia were examined for retrograde dye-labeled neurons. Neurons retrogradely labeled from the bladder were found primarily in L7-S2 spinal ganglia, spinal cord lateral zona intermedia at S1-S

Distribution of the cells of primary afferent fibers to the cat auricle in relation to the innervated region.

The origin and peripheral distribution of the primary afferent fibers to the cat auricle were studied by the horseradish peroxidase (HRP) method. Following HRP injection into the whole region of the auricle, HRP-labeled cells were found ipsilaterally in the trigeminal ganglion, geniculate ganglion, superior ganglion of the vagus nerve and in the C1 to C4 spinal ganglia. In addition to the ganglia of cranial and spinal nerves, labeled cells were also observed in the facial nerve trunk and in the dorso-lateral portion of the superior cervical ganglion. In the case of HRP injection into the centr

Cholecystokinin in mammalian primary sensory neurons and spinal cord: in situ hybridization studies in rat and monkey.

The peptide cholecystokinin (CCK) has been suggested to be involved in nociception, but its exact localization at the level of the spinal cord and in spinal ganglia has been a controversial issue. Therefore the distribution of messenger RNA (mRNA) for CCK was studied by in situ hybridization using oligonucleotide probes on sections of adult rat lumbar dorsal root ganglia following unilateral section of the sciatic nerve and on sections of untreated monkey trigeminal ganglia, spinal cord and spinal ganglia from all levels. For comparison, calcitonin gene-related peptide (CGRP) mRNA was also stu

Immunoneutralization of nerve growth factor in lumbosacral spinal cord reduces bladder hyperreflexia in spinal cord injured rats.

PURPOSE: We investigated the effects of intrathecal application of nerve growth factor (NGF) antibodies (Ab) on bladder hyperreflexia in chronic spinalized rats. MATERIALS AND METHODS: In adult female rats an intrathecal catheter was implanted at the level of the L6 to S1 spinal cord, followed by complete transection of the Th8 to 9 spinal cord. At 10 days after spinalization the intrathecal catheter was connected to an osmotic pump for continuous delivery of vehicle or NGF Ab (10 microg daily) for 2 weeks. Awake cystometry was then performed. NGF levels in the L5 to S1 dorsal root ganglia, L

Mu-Opioid Receptors in Ganglia, But Not in Muscle, Mediate Peripheral Analgesia in Rat Muscle Pain.

BACKGROUND: Previous studies have demonstrated the participation of peripheral μ-opioid receptors (MOR) in the antinociceptive effect of systemically administered morphine and loperamide in an orofacial muscle pain model, induced by hypertonic saline, but not in a spinally innervated one, in rats. In this study, we determine whether this peripheral antinociceptive effect is due to the activation of MOR localized in the muscle, ganglia, or both. METHODS: To determine the local antinociceptive effect of morphine and loperamide, 2 models of acute muscle pain (trigeminal and spinal) were used. Al

Acetylcholinesterase activity and its molecular forms in selected regions of the dog's nervous system.

Acetylcholinestrase (AChE, EC 3.1.1.7) activity and its molecular forms were studied in the cerebral cortex, basal ganglia, thalamus, cerebellar cortex, medulla oblongata, the various parts of spinal cord, the spinal ganglia and the sciatic nerve in intact adult dogs. The number and the proportion of different molecular forms of AChE in these regions of the dog's nervous system varied considerably. The highest AChE activity was found in the basal ganglia, where only one molecular forms was detected. Activity was also high in the cerebellar cortex (2 isoenzymes), the thalamus (4 isoenzymes) and

Membrane leakage in spinal ganglion nerve cells induced by experimental whiplash extension motion: a study in pigs.

Nerve cells in the cervical and upper thoracic spinal ganglia were examined for possible plasma membrane leakage, as revealed by their ability to exclude a dye-protein complex, after experimentally induced whiplash in a pig model system. The rationale for this approach is found in the fact that the interstitium of spinal ganglia differs from most other parts of the nervous system in that it lacks a barrier, allowing blood constituents to gain access. The dye Evans blue, which rapidly conjugates with blood proteins, is found in the interstitium of normal spinal ganglia after intravenous injecti

The dorsal root ganglion of the American alligator (Alligator mississippiensis).

The peripheral nervous system of vertebrates develops through the merger of neuritic outgrowths from the central nervous system, and derivatives of the neural crest cells (ncc), including the spinal (or dorsal root) ganglia. Before the neural crest cells differentiate into neural tissue, they migrate through the vertebrate embryo. Relatively little comparative work has been done on the potential relationship between differential ncc migration and interspecific variation in the spinal ganglia. In Alligator mississippiensis, the sensory ganglia are located on the dorsal root, immediately distal

Posttraumatic activity of signal pathways of nuclear factor κB in mature sensory neurons.

The aim of this study was to clear out whether injury to the peripheral nerve leads to activation of nuclear factor κB in mature spinal ganglia. Analysis of matrix RNA of nuclear factor κB-dependent genes (monocyte chemoattractant protein MCP-1 and inhibitor of nuclear factor κB IκBα) showed different levels of expression of these genes in the spinal ganglia in vivo after axotomy and in vitro after TNF-α stimulation. On the other hand, DNA-binding activity of nuclear factor κB increased in the spinal ganglia 6 h after axotomy and after 10-min incubation of sensory neuron culture with TNF-α. Th
