Correction of Huntington's Disease Phenotype by Genistein-Induced Autophagy in the Cellular Model.

Huntington's disease (HD) is a monogenic disorder, caused by mutations in the HTT gene which result in expansion of CAG triplets. The product of the mutated gene is misfolded huntingtin protein that forms aggregates leading to impairment of neuronal function, neurodegeneration, motor abnormalities and cognitive deficits. No effective cure is currently available for HD. Here we studied effects of genistein (trihydroxyisoflavone) on a HD cellular model consisting of HEK-293 cells transfected with a plasmid bearing mutated HTT gene. Both level of mutated huntingtin and number of aggregates were s

In vivo study on splenomegaly inhibition by genistein in Plasmodium berghei-infected mice.

Spleen plays an important role in removing old and damaged red blood cells and malaria-infected erythrocytes. When malaria parasites invade the spleen and induce splenomegaly, splenic function tends to be impaired. Thus, the inhibition of splenomegaly is strongly required to protect the spleen. In this study, malaria-induced splenomegaly is inhibited by injecting genistein into a Plasmodium berghei-infected ICR mouse. To explain this phenomenon, the effect of genistein in spleen and liver of malaria-infected mice was evaluated by histological examination. Malaria parasites disrupted splenic ar

Genistein reduces glycosaminoglycan levels in a mouse model of mucopolysaccharidosis type II.

BACKGROUND AND PURPOSE: Mucopolysaccharidoses (MPS) are lysosomal storage disorders resulting from a deficit of specific lysosomal enzymes catalysing glycosaminoglycan (GAG) degradation. The typical pathology involves most of the organ systems, including the brain, in its severe forms. The soy isoflavone genistein has recently attracted considerable attention as it can reduce GAG synthesis in vitro. Furthermore, genistein is able to cross the blood-brain barrier in the rat. The present study was undertaken to assess the ability of genistein to reduce urinary and tissue GAG levels in vivo. EXP

Relaxation of isolated guinea pig trachea by genistein via inhibition of phosphodiesterase.

We investigated the mechanisms of the relaxant action of genistein, an isoflavone, phytoestrogen and non-specific protein tyrosine kinase inhibitor. Changes in tension of guinea pig tracheal segments were isometrically recorded on a polygraph. Genistein concentration-dependently relaxed histamine (30 microM)-, carbachol (0.2 microM)-, KCl (30 mM)- and leukotriene D4 (10 nM)-induced precontractions and inhibited cumulative histamine- and carbachol-induced contractions in a non-competitive manner. Genistein also concentration-dependently and non-competitively inhibited the cumulative, Ca2+-induc

Epithelial cells treated with genistein inhibit adhesion and endocytosis of Paracoccidioides brasiliensis.

Paracoccidioidomycosis is caused by Paracoccidioides brasiliensis, which although not formally considered an intracellular pathogen, can be internalized by epithelial cells in vitro and in vivo. The mechanisms used by P. brasiliensis to adhere to and invade non-professional phagocytes have not been identified. The signal-transduction networks, involving protein tyrosine kinase (PTK) and protein phosphatase activities, can modulate crucial events during fungal infections. In this study, the involvement of PTK has been investigated in P. brasiliensis adherence and invasion in mammalian epithelia

Response to genistein: assaying the activation status and chemotaxis efficacy of isolated neutrophils.

Neutrophil (PMN) activation and chemotaxis toward inflammatory stimuli play critical roles in host defense and tissue inflammation. To determine the molecular mechanisms that regulate PMN function, many studies currently employ in vitro PMN activation and transmigration assays using freshly isolated peripheral PMN or PMN isolated from bone marrow. However, due to the highly sensitive nature of PMN, cell activation or priming can occur during isolation, which demands assay(s) that ensure the consistency of isolated PMN prior to using them in subsequent experiments. Here, we introduce a simple s

Inhibition of prostate specific antigen expression by genistein in prostate cancer cells.

Recent studies have provided convincing evidence for the role of soy-isoflavones, particularly genistein, in the inhibition of prostate cancer cell growth. Prostate specific antigen (PSA) is a biological marker used to detect and monitor the treatment of prostate cancer patients. Previous studies have documented that isoflavones can inhibit the secretion of PSA in the androgen-dependent prostate cancer cell line, LNCaP, however, the effects of genistein on androgen-independent PSA expression has not been explored. In this study, we have utilized a prostate cancer cell line, VeCaP, which expres

Genistein Mediates Beauveria bassiana-Induced Resistance Against Bemisia tabaci Reproduction in Tomato.

Endophytic colonization by entomopathogenic fungi Beauveria bassiana enhances plant resistance to herbivores; however, the underlying molecular mechanisms, particularly those involving hormonal disruption of pests, remain poorly understood. This study examined whether root inoculation of tomato with B. bassiana suppresses whitefly Bemisia tabaci fitness via flavonoid modulation. Metabolomic profiling of leaf tissues showed a 3.5-fold accumulation of the isoflavonoid genistein in B. bassiana-inoculated plants. KEGG enrichment analysis indicated significant upregulation of the isoflavonoid biosy

BACE1 Inhibition by Genistein: Biological Evaluation, Kinetic Analysis, and Molecular Docking Simulation.

β-site amyloid precursor protein cleaving enzyme 1 (BACE1) plays a role in generating amyloid β (Aβ), thus playing a major part early in the pathogenesis of Alzheimer's disease (AD). BACE1 has emerged as a crucial therapeutic target for decreasing the Aβ concentration in the AD brain. To explore natural BACE1 inhibitors, the present study concentrated on isoflavones, including genistein, formononetin, glycitein, daidzein, and puerarin. In this study, in vitro anti-AD activities were assessed using BACE1 inhibition assays, as well as enzyme kinetic predictions. Molecular docking analysis was ap

Genistein improves thyroid function in Hashimoto's thyroiditis patients through regulating Th1 cytokines.

Hashimoto thyroiditis (HT) is a common autoimmune disease. Genistein is an isoflavone with immunomodulatory functions in various diseases. In this double-blind, randomized, placebo-controlled clinical study, we investigated the effects of genistein on patients with HT. A total of 218 patients were recruited. Genistein treated patients had a significant increased T4, fT4 levels, as well as reduction in serum TSH, TPOAb and TgAb levels, compared with those receiving placebo. Furthermore, Th1 related cytokine IFN-γ and IL-2 changes after genistein treatment suggest the immune modulating effect of

SPECT/CT analysis of splenic function in genistein-treated malaria-infected mice.

Spleen traps malaria-infected red blood cells, thereby leading to splenomegaly. Splenomegaly induces impairment in splenic function, i.e., rupture. Therefore, splenomegaly inhibition is required to protect the spleen. In our previous study, genistein was found to have an influence on malaria-induced splenomegaly. However, the effect of genistein in malaria-induced splenomegaly, especially on the function of spleen, has not been fully investigated. In this study, hematoxylin and eosin (H&E) staining images show that genistein partially prevents malaria-induced architectural disruption of spleen

The phytoestrogen genistein modulates lysosomal metabolism and transcription factor EB (TFEB) activation.

Genistein (5,7-dihydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one) has been previously proposed as a potential drug for use in substrate reduction therapy for mucopolysaccharidoses, a group of inherited metabolic diseases caused by mutations leading to inefficient degradation of glycosaminoglycans (GAGs) in lysosomes. It was demonstrated that this isoflavone can cross the blood-brain barrier, making it an especially desirable potential drug for the treatment of neurological symptoms present in most lysosomal storage diseases. So far, no comprehensive genomic analyses have been performed to el

Genistein stimulates electrogenic Cl- secretion via phosphodiesterase modulation in the mouse jejunum.

Previously, we demonstrated that genistein stimulated Cl(-) secretion in the mouse jejunum (Baker MJ and Hamilton KL, Am J Physiol Cell Physiol 287: C1636-C1645, 2004); however, the mode of action of genistein still remains unclear. Here, we examined the activation of Cl(-) secretion by the modulation of phosphodiesterases (PDEs) by genistein (75 microM) in the mouse jejunum with the Ussing short-circuit current (I(sc)) technique. Drugs tested included theophylline (10 mM), a nonspecific PDE inhibitor; 8-methoxymethyl-3-isobutyl-1-methylxanthine (8-MM-IBMX; 100 microM), erythro-9-(2-hydroxyl-3
