Iflaviruses in arthropods: when small is mighty.

Iflaviruses are arthropod-infecting viruses classified in the family Iflaviridae. The most well-studied iflaviruses are responsible for deleterious overt (symptomatic) infections in economically important insect species like honeybees and silkworms. The fact that iflaviruses are also found in mass reared insects such as crickets and flies, makes their study relevant for the food and feed industries and for biological control applications (e.g., mass reared insects for sterile insect techniques). In recent years it was found that iflaviruses are not restricted to insects, but are widely spread

Visualizing the ribonucleoprotein content of single bunyavirus virions reveals more efficient genome packaging in the arthropod host.

Bunyaviruses have a genome that is divided over multiple segments. Genome segmentation complicates the generation of progeny virus, since each newly formed virus particle should preferably contain a full set of genome segments in order to disseminate efficiently within and between hosts. Here, we combine immunofluorescence and fluorescence in situ hybridization techniques to simultaneously visualize bunyavirus progeny virions and their genomic content at single-molecule resolution in the context of singly infected cells. Using Rift Valley fever virus and Schmallenberg virus as prototype tri-se

Understanding the role of insects in the acquisition and transmission of antibiotic resistance.

Antibiotic resistance (AR) is a global healthcare threat that requires a comprehensive assessment. Poorly regulated antibiotic stewardship in clinical and non-clinical settings has led to a horizontal dissemination of AR. A variety of often neglected elements facilitate the circulation of AR from antibiotic sinks like concentrated animal feeding operations and healthcare settings to other environments that include healthy human communities. Insects are one of those elements that have received underwhelming attention as vectors of AR, despite their well-known role in transmitting clinically rel

Begomoviruses: what is the secret(s) of their success?

Begomoviruses constitute an extremely successful group of emerging plant viruses transmitted by whiteflies of the Bemisia tabaci complex. Hosts include important vegetable, root, and fiber crops grown in the tropics and subtropics. Factors contributing to the ever-increasing diversity and success of begomoviruses include their predisposition to recombine their genomes, interaction with DNA satellites recruited throughout their evolution, presence of wild plants as a virus reservoir and a source of speciation, and extreme polyphagia and continuous movement of the insect vectors to temperate reg

De novo genome assembly of Bradysia cellarum (Diptera: Sciaridae), a notorious pest in traditional special vegetables in China.

Bradysia cellarum (Diptera: Sciaridae) is a destructive vegetable insect pest infesting more than 30 species of host plants from seven families in Asia and Europe. B. cellarum causes grave problems in Chinese chive, which originated in China and is cultivated widely in East Asia. The B. cellarum infestation results in economic losses and subsequent severe food safety problems in farm productions, insecticide resistance and environmental pollution. The genomic and molecular information of B. cellarum to delineate the biological features, insecticide resistance, evolution remains poorly understo

Insect immune systems: same same but different but still same.

Insects are the most diverse group of animals in nature, occupying nearly every ecological niche and playing central roles as pollinators, pests, and disease vectors. Despite this vast diversity, insects rely on a set of conserved yet evolutionarily adaptable immune pathways to defend against pathogens. Early studies in insect immunity have laid the foundation for human immunology, and recent advances in genomic and transgenic technologies have renewed interest in understanding how immune responses vary across insect orders. Insects are highly diverse in their immune systems; each species has

Functional diversity of the Osiris gene family in the brown planthopper.

Osiris is a large insect-specific gene family with multiple functions. Owing to highly conserved sequences and similar expression patterns across insect species, the Osiris gene family is believed to play specific roles in the insect kingdom. However, current studies on Osiris gene functions have focused primarily on several holometabolous insects, such as Drosophila melanogaster and Bombyx mori, with few systematic studies on hemimetabolous insects. In this study, we identified 20 Osiris genes by searching the genome and transcriptome databases of the brown planthopper Nilaparvata lugens. The

First Description of Loreto Virus in Three Culicidae Species from the Atlantic Forest, Bahia, Brazil.

Loreto virus (LORV) is an insect-specific virus classified into the proposed taxon Negevirus. It was originally described in Iquitos, Peru, in 1977. Here, we describe three novel LORV genomes obtained from the isolates of three pooled samples of Trichoprosopon digitatum, Aedes (Ochlerotatus) fulvus, and Limatus durhamii collected in Ilhéus-Bahia, 2014. Samples were submitted to RNA sequencing on the Illumina platform to recover the LORV genome. The genomes presented, on average, 81.5% nucleotide identity and 92.6% global amino acid identity with the LORV reference genome (NC_034158). Subsequen

Two ABC transporters are differentially involved in the toxicity of two Bacillus thuringiensis Cry1 toxins to the invasive crop-pest Spodoptera frugiperda (J. E. Smith).

BACKGROUND: The fall armyworm Spodoptera frugiperda is a major agricultural pest that has invaded the East Hemisphere since 2016, generating a serious threat to food security worldwide including Africa and Asia. The Cry toxins produced by Bacillus thuringiensis (Bt) have been shown to be effective against this insect pest. In different insect ABC transporters (ABCC2 or ABCC3) have been shown to be involved as receptors of some Cry1 toxins. Here we analyzed the role of SfABCC2 and SfABCC3 in the toxicity of Cry1Fa and Cry1Ab toxins in this insect pest. RESULTS: Two S. frugiperda SfABCC2 and Sf

Genome-Wide Identification and functional characterization of chitinase genes in the sorghum aphid (Melanaphis sacchari).

Insect chitinases are integral to a spectrum of vital physiological processes, including growth, development, molting, and metamorphosis. Consequently, chitin metabolic enzymes are potential targets for eco-friendly insecticide development. This study employed bioinformatics approaches to analyze the chitinase gene family in the sorghum aphid(Melanaphis sacchari), identifying seven MsCht genes. Phylogenetic distribution analysis revealed that the chitinases of the sorghum aphid are distributed across multiple clades, and the presence of several orthologous relationships with the pea aphid sugg

A chromosome-level reference genome for the common bed bug, Cimex lectularius, with identification of sex chromosomes.

The common bed bug, Cimex lectularius, is a globally distributed pest insect of medical, veterinary, and economic importance. Previous reference genome assemblies for this species were generated from short-read sequencing data, resulting in a ~650 Mb composed of thousands of contigs. Here, we present a haplotype-resolved, chromosome-level reference genome, generated from an adult Harlen strain female specimen. Using PacBio long read and Omni-C proximity sequencing, we generated a 540 Mb genome with 15 chromosomes (13 autosomes and 2 sex chromosomes-X1X2) with an N50 > 30 Mb and BUSCO > 90%. Pr

A chromosome-level genome assembly of wild silkmoth, Bombyx mandarina.

The wild silk moth, Bombyx mandarina, is the closest relative of the domesticated silk moth, Bombyx mori. National BioResource Project of Japan (NBRP) maintains a B. mandarina strain derived from individuals captured at Sakado (Saitama, Japan) in 1982. Now, NBRP has developed chromosome replacement strains of B. mori. In each strain, one autosome of B. mori was replaced with the corresponding chromosome of B. mandarina. To facilitate the use of chromosome replacement strains and B. mandarina itself, we constructed a chromosome-level genome assembly of B. mandarina. Furthermore, we performed fu

Cloning and identification of an oxytocin/vasopressin-like receptor and its ligand from insects.

More than 20 years ago, an oxytocin/vasopressin-like peptide, CLITNCPRGamide, was isolated from the locust, Locusta migratoria [Proux JP, et al. (1987) Identification of an arginine vasopressin-like diuretic hormone from Locusta migratoria. Biochem Biophys Res Commun 149:180-186]. However, no similar peptide could be identified in other insects, nor could its prohormone be cloned, or its physiological actions be established. Here, we report that the recently sequenced genome from the red flour beetle Tribolium castaneum contains a gene coding for an oxytocin/vasopressin-like peptide, identical

Comparative genomics identifies male accessory gland proteins in five Glossina species.

Accessory gland proteins (ACPs) are important reproductive proteins produced by the male accessory glands (MAGs) of most insect species. These proteins are essential for male insect fertility, and are transferred alongside semen to females during copulation. ACPs are poorly characterized in Glossina species (tsetse fly), the principal vector of the parasite that causes life-threatening Human African Trypanosomiasis and Animal trypanosomiasis in endemic regions in Africa. The tsetse fly has a peculiar reproductive cycle because of the absence of oviposition. Females mate once and store sperm in

Complete sequences of mitochondria genomes of Aedes aegypti and Culex quinquefasciatus and comparative analysis of mitochondrial DNA fragments inserted in the nuclear genomes.

We present complete sequences of the mitochondrial genomes for two important mosquitoes, Aedes aegypti and Culex quinquefasciatus, that are major vectors of dengue virus and lymphatic filariasis, respectively. The A. aegypti mitochondrial genome is 16,655 bp in length and that of C. quinquefasciatus is 15,587 bp, yet both contain 13 protein coding genes, 22 transfer RNA (tRNA) genes, one 12S ribosomal RNA (rRNA) gene, one 16S rRNA gene and a control region (CR) in the same order. The difference in the genome size is due to the difference in the length of the control region. We also analyzed in

The evolution of the Anopheles 16 genomes project.

We report the imminent completion of a set of reference genome assemblies for 16 species of Anopheles mosquitoes. In addition to providing a generally useful resource for comparative genomic analyses, these genome sequences will greatly facilitate exploration of the capacity exhibited by some Anopheline mosquito species to serve as vectors for malaria parasites. A community analysis project will commence soon to perform a thorough comparative genomic investigation of these newly sequenced genomes. Completion of this project via the use of short next-generation sequence reads required innovatio

Characterizing the symbiotic relationship between Wolbachia (wSpic) and Spodoptera picta (Lepidoptera: Noctuidae): From genome to phenotype.

Wolbachia is a genus of symbiotic bacteria prevalent in arthropods, with diverse effects on host reproduction and fecundity; however, it is unclear how Wolbachia modulates the host reproductive system. In this study, a novel Wolbachia strain, wSpic, was identified in the Noctuid moth Spodoptera picta and its effect on the reproduction of this host was investigated. We sequenced and annotated the 1,339,720 bp genome of wSpic. We identified a total of five WO phage regions in the genome and found no evidence of any plasmids associated with wSpic. Evolutionary analysis revealed that wSpic belongs
