[Cryptosporidium and Giardia as water contaminant pathogens in Hungary].

INTRODUCTION: Many species of Cryptosporidium, and two assemlages of Giardia duodenalis cause typically acute diaorrhoea in human. The oocysts and cysts of these parasites excreted in faeces are capable of infecting other hosts and those are environmentally stable. AIM: The aims of the study were to evaluate the prevalence and genotypes of Cryptosporidium and Giardia species from different water sources as well as to monitor and characterize the (oo)cyst contamination sources in watersheds. In addition, an epidemiological study was performed in three selected settlements. METHOD: Wide range

Mechanisms of adaptation in the intestinal parasite Giardia lamblia.

Giardia lamblia, a parasite of humans, is a major source of waterborne diarrhoeal disease. Giardia is also an excellent system to study basic biochemical processes because it is a single-celled eukaryote with a small genome and its entire life cycle can be replicated in vitro. Giardia trophozoites undergo fundamental changes to survive outside the intestine of their host by differentiating into infective cysts. Encystation entails the synthesis, processing, transport, secretion and extracellular assembly of cyst wall components. To survive within the intestine, Giardia undergoes antigenic vari

Cryptosporidium and Giardia in wild otters (Lutra lutra).

A total of 437 faecal samples from wild otter (Lutra lutra) were collected from 161 sites in Galicia (northwest Spain) and were analysed by a direct immunofluorescence antibody test (IFAT). Cryptosporidium sp. oocysts and Giardia sp. cysts were detected in 17 (3.9%) and 30 (6.8%) samples, respectively. The results demonstrate that otters may contribute to the contamination of watercourses, although further studies are required to establish which species or genotypes of Cryptosporidium and Giardia infect these animals and also the significance in terms of public health.

[Are domestic Cryptosporidium and Giardia infections in Norway underdiagnosed?].

BACKGROUND: Cryptosporidium and Giardia are recognised as common causes of waterborne disease in several countries. In order to describe investigative practices for these protozoan parasites in Norway, we surveyed medical microbiology laboratories nationwide for faecal screening policies and methods used for detection of Cryptosporidium and Giardia. MATERIAL AND METHODS: All medical microbiology laboratories in Norway received questionnaires on laboratory methods, indications for screening, and numbers of samples investigated over the 1998-2002 period. RESULTS: Of the 22 laboratories that re

The prevalence of Giardia and Cryptosporidium in Taiwan water supplies.

Giardia and Cryptosporidium have emerged as waterborne pathogens of concern. Thirty-one water samples were collected from nine potable water treatment plants in Taiwan and investigated for the presence of Giardia cysts and Cryptosporidium oocysts. The immunofluorescence assay was used for the simultaneous detection of cysts and oocysts. The frequency of occurrence of cysts was 77.8% for Giardia and 72.2% for Cryptosporidium in 18 raw water samples. Ten out of 13 samples collected from treated water samples showed the presence of cysts, while in 5 out of 13 treated water samples oocysts were de

Possible role of calmodulin in excystation of Giardia lamblia.

The protozoan Giardia lamblia initiates infection when trophozoites emerge from a cyst in the hosts by the excystation process. Although this process is crucial to the initiation of infection by G. lamblia, little is known about its regulation. To study the possible involvement of calmodulin (CaM) in excystation we tested the effect of several CaM antagonists (TFP, W-7, and W-5) on this cellular function. Except for W-5 the rest of these compounds inhibited excystation. The protein kinase C inhibitor H-7 had no effect on excystation, suggesting that CaM antagonists acted by selectively inhibit

Giardia and Cryptosporidium in dairy calves in British Columbia.

A study was undertaken to determine the prevalence of Giardia infections in dairy calves and to compare Giardia and Cryptosporidium infections in calves of different ages. Fresh fecal samples were collected from 386 male and female Holstein calves (newborn to 24 wk) in 20 dairies located in the lower Fraser river valley area of British Columbia. Giardia intestinalis, Cryptosporidium parvum, and Cryptosporidium muris were enumerated in each sample after concentration by sucrose gradient centrifugation and immunofluorescent staining. Giardia was identified at all farm locations. The overall prev

Protozoocidal activity of Stemona collinsiae against Giardia duodenalis.

Giardia duodenalis is a major pathogen of the gastrointestinal tract, and infections impact both human health and the economy. The ongoing issues with drug resistance and the side effects of current anti-Giardia treatments highlight the urgent need for new therapeutic options. This study focused on investigating the in vitro efficacy of crude extracts of Stemona collinsiae from Thailand against G. duodenalis. S. collinsiae was initially extracted with hexane, followed sequentially by dichloromethane, ethanol, and water. The extracts were tested for growth inhibition against G. duodenalis. Dich

Giardia duodenalis triggered neutrophil extracellular traps in goats.

Giardia duodenalis is a globally distributed zoonotic parasite primarily transmitted through the fecal-oral route, infecting various vertebrates, and the infection of which is prevalent in goats. Immune cells play a crucial role in pathogens invasion, and neutrophil extracellular traps (NETs) released by neutrophils serve as a non-specific defense mechanism against pathogens including parasites. In this study, we investigated the characteristics, components, and molecular mechanisms of goat NETs upon stimulation with G. duodenalis trophozoites. This study demonstrates that G. duodenalis trigge

Intestinal pseudo-obstruction caused by Giardia lamblia infection.

A woman in her 40s presented with malaise, nausea, reduced appetite, abdominal distention, loose stools and weight loss. Symptoms had started 6 months earlier and worsened in the last 2 weeks. CT enterography showed hypotonic dilated small bowel loops in absence of any mechanical obstruction. Endoscopic examinations including capsule endoscopy did not reveal any obstructing lesion, but a delayed small bowel transit time of the capsule. Duodenal histology revealed Marsh 3a villous atrophy. Secondary causes of intestinal pseudo-obstruction and villous atrophy were investigated. Giardia lamblia t

Giardia duodenalis in colony stray cats from Italy.

Giardia duodenalis is the most common intestinal protozoan in humans and animals worldwide, including eight morphologically identical assemblages, infecting pets, livestock, wildlife and human beings. Assemblages A and B are those with the higher zoonotic potential, and they have been detected in several mammals other than humans; the others (C to H) show a higher host specificity. Cats can harbour both the specific Assemblage F and the zoonotic ones A and B. Several studies have been carried out on G. duodenalis genotypes in cats; however, the role of this species in the epidemiology of giard

New multilocus genotypes of Giardia lamblia human isolates.

Giardia lamblia is considered a species complex, whose members show little differences in their morphology, but have remarkable genetic variability. The aim of this study was to identify inter- and intra-assemblage genetic variation in G. lamblia among patients in Rio de Janeiro. The parasitological study was performed on faeces, and DNA was extracted from the samples which tested positive for G. lamblia. The genetic assemblages and subtypes were determined via multilocus sequence typing (MLST) using β-giardin, triose phosphate isomerase and glutamate dehydrogenase gene loci. Fourteen assembla

Giardia duodenalis Infection and Pachymeningitis in a Patient With Agammaglobulinemia.

X-linked agammaglobulinemia (XLA) is a rare immunodeficiency caused by a mutation in Bruton's tyrosine kinase, resulting in profoundly low immunoglobulin levels and susceptibility to recurrent infections. We report a 14-year-old patient with XLA who presented with persistent fever, bloody diarrhea, and headache suggestive of pachymeningitis. The patient had chronic giardiasis. His clinical condition deteriorated with worsening thrombocytopenia, hepatitis, and minimal diffuse dural thickening on brain magnetic resonance imaging, interpreted as pachymeningitis. Empirical immunosuppressive therap

Regarding the Zoonotic Transmission of Giardia duodenalis Infection.

Giardia duodenalis, a member of the genus Giardia, is a common intestinal protozoan parasite in humans and some farm, companion, and wild animals. Due to the uniform morphology of the parasites sampled from a wide range of hosts, the taxonomy of the genus Giardia has been the subject of debate for more than a century. The confusion generated by this controversy, and the technological limitations to carry out the necessary research to clarify the matter, led to an insufficient understanding of the epidemiology of the infection, mainly in aspects related to host specificity and zoonotic transmis

Clues on the dynamics of DNA replication in Giardia lamblia.

Genomic replication is a critical, regulated process that ensures accurate genetic information duplication. In eukaryotic cells, strategies have evolved to prevent conflicts between replication and transcription. Giardia lamblia, a binucleated protozoan, alternates between tetraploid and octaploid genomes during its cell cycle. Using single-molecule techniques like DNA combing and nanopore-based sequencing, we investigated the spatio-temporal organization of DNA replication, replication fork progression and potential head-on replication-transcription collisions in Giardia trophozoites. Our fin

A Detailed Gene Expression Map of Giardia Encystation.

Giardia intestinalis is an intestinal protozoan parasite that causes diarrheal infections worldwide. A key process to sustain its chain of transmission is the formation of infectious cysts in the encystation process. We combined deep RNAseq of a broad range of encystation timepoints to produce a high-resolution gene expression map of Giardia encystation. This detailed transcriptomic map of encystation confirmed a gradual change of gene expression along the time course of encystation, showing the most significant gene expression changes during late encystation. Few genes are differentially expr

Giardia duodenalis in humans and animals - Transmission and disease.

Giardia duodenalis is a protozoan parasite infecting the upper intestinal tract of humans, as well as domestic and wild animals worldwide. Transmission of giardiasis occurs through the faecal-oral route, and may be either direct (i.e., person-to-person, animal-to-animal or zoonotic) or indirect (i.e., waterborne or foodborne). While asymptomatic infections are common in both humans and animals, a wide range of enteric symptoms have been reported, along with extra-intestinal and post-infectious complications. A definitive diagnosis of giardiasis is generally made by detection of cysts in stool

Host specificity in the Giardia duodenalis species complex.

Giardia duodenalis is a unicellular flagellated parasite that infects the gastrointestinal tract of a wide range of mammalian species, including humans. Investigations of protein and DNA polymorphisms revealed that G. duodenalis should be considered as a species complex, whose members, despite being morphologically indistinguishable, can be classified into eight groups, or Assemblages, separated by large genetic distances. Assemblages display various degree of host specificity, with Assemblages A and B occurring in humans and many other hosts, Assemblage C and D in canids, Assemblage E in hoof

[Giardia duodenalis and its involvement in skin diseases].

BACKGOUND: Over the last thirty years, the scientific community has become increasingly interested in the intestinal flora, whether commensal or pathogenic, and its impact on other organs. In dermatology, the correlation between intestinal microbial agents and cutaneous lesions is well established. Giardia duodenalis, an intestinal parasite, has been particularly widely studied. The aim of this work is to provide a review of studies demonstrating the involvement of G. duodenalis in various forms of dermatosis. PATIENTS AND METHODS: The data were obtained by an English-language literature sear
