A CCaMK-CYCLOPS-DELLA Complex Activates Transcription of RAM1 to Regulate Arbuscule Branching.

Intracellular arbuscular mycorrhiza symbiosis between plants and glomeromycotan fungi leads to formation of highly branched fungal arbuscules that release mineral nutrients to the plant host. Their development is regulated in plants by a mechanistically unresolved interplay between symbiosis, nutrient, and hormone (gibberellin) signaling. Using a positional cloning strategy and a retrotransposon insertion line, we identify two novel alleles of Lotus japonicus REDUCED ARBUSCULAR MYCORRHIZA1 (RAM1) encoding a GRAS protein. We confirm that RAM1 is a central regulator of arbuscule development: arb

High-throughput quantitative analysis of phytohormones in sorghum leaf and root tissue by ultra-performance liquid chromatography-mass spectrometry.

Plant development, growth, and adaptation to stress are regulated by phytohormones, which can influence physiology even at low concentrations. Phytohormones are chemically grouped according to both structure and function as auxins, cytokinins, abscisic acid, jasmonates, salicylates, gibberellins, and brassinosteroids, among others. This chemical diversity and requirement for highly sensitive detection in complex matrices create unique challenges for comprehensive phytohormone analysis. Here, we present a robust and efficient quantitative UPLC-MS/MS assay for 17 phytohormones, including jasmona

Genome-Wide Identification and Expression Profiling of the GATA Gene Family Reveals Candidate Genes Associated with GA3-Induced Shoot Elongation in Citrus grandis 'Wanbai'.

Plant GATA transcription factors are zinc-finger proteins involved in signal transduction, growth and development. However, their potential involvement in plant height and gibberellin-associated shoot growth in pummelo insufficiently characterised. In this study, members of the CgGATA gene family were identified genome-wide and characterised through analyses of physicochemical properties, chromosomal distribution, phylogenetic relationships, gene structures, conserved motifs, collinearity and predicted cis-acting elements in putative upstream regulatory regions. RT-qPCR was used to examine exp

N-terminal truncated RHT-1 proteins generated by translational reinitiation cause semi-dwarfing of wheat Green Revolution alleles.

The unprecedented wheat yield increases during the Green Revolution were achieved through the introduction of the Reduced height (Rht)-B1b and Rht-D1b semi-dwarfing alleles. These Rht-1 alleles encode growth-repressing DELLA genes containing a stop codon within their open reading frame that confers gibberellin (GA)-insensitive semi-dwarfism. In this study, we successfully took the hurdle of detecting wild-type RHT-1 proteins in different wheat organs and confirmed their degradation in response to GAs. We further demonstrated that Rht-B1b and Rht-D1b produce N-terminal truncated proteins throug

Transcriptome and metabolome analyses reveal that GA3ox regulates the dwarf trait in mango (Mangifera indica L.).

BACKGROUND: Mango is a tropical fruit with high economic value. The selection of suitable dwarf mango varieties is an important aspect of mango breeding. However, the mechanisms that regulate mango dwarfing remain unclear. RESULTS: In this study, we compared the transcriptomes and metabolomes of mango varieties Guiqi (a dwarfed variety) and Jinhuang (an arborized variety). A total of 4,954 differentially expressed genes and 317 differentially abundant metabolites were identified between the two varieties, revealing the molecular mechanism of the gibberellin 3β-hydroxylase gene GA3ox in regula

Characterization of the molecular mechanism underlying the dwarfism of dsh mutant watermelon plants.

Developing dwarf watermelon is a major objective among breeders. The dsh dwarf watermelon germplasm developed in our laboratory is genetically stable. We previously produced preliminary evidence that Cla010726, which encodes a gibberellin 20-oxidase-like protein, is the primary gene controlling dwarfism in watermelon. However, the underlying genetic mechanism was unknown. In this study, we characterized the spontaneous recessive mutant dsh, which is a gibberellin (GA)-deficient mutant. Many of the phenotypic traits of dsh plants are similar to those of known GA-deficient mutants. The dsh plants

Roles of Hormones in Elevated pH-Mediated Mitigation of Copper Toxicity in Citrus sinensis Revealed by Targeted Metabolome.

The effects of copper (Cu)-pH interactions on the levels of hormones and related metabolites (HRMs) in Citrus sinensis leaves and roots were investigated. Our findings indicated that increased pH mitigated Cu toxicity-induced alterations of HRMs, and Cu toxicity increased low-pH-induced alterations of HRMs. Increased pH-mediated decreases in ABA, jasmonates, gibberellins, and cytokinins, increases in (±)strigol and 1-aminocyclopropanecarboxylic acid, and efficient maintenance of salicylates and auxins homeostasis in 300 μM Cu-treated roots (RCu300); as well as efficient maintenance of hormone

Stereoselective Synthesis and Antimicrobial Studies of Allo-Gibberic Acid-Based 2,4-Diaminopyrimidine Chimeras.

Background: Gibberellins (GAs) are a family of tetracyclic ent-kaurenoid diterpenes found widely in several commonly used plants. Besides agricultural applications, gibberellins play an important role in the synthesis of bioactive compounds, especially those with antiproliferative and antibacterial activity. Methods: A series of gibberellic acid-based 2,4-diaminopyrimidines was designed and synthesized from commercially available gibberellic acid. The antimicrobial activity of the prepared compounds was also explored in B. subtilis, S. aureus, E. coli, and P. aeruginosa bacteria, as well as in

CRISPR/Cas9-mediated enhancement of semi-dwarf glutinous traits in elite Xiangdaowan rice (Oryza sativa L.): targeting SD1 and Wx genes for yield and quality improvement.

In rice cultivation, the traits of semi-dwarfism and glutinous texture are pivotal for optimizing yield potential and grain quality, respectively. Xiangdaowan (XDW) rice, renowned for its exceptional aromatic properties, has faced challenges due to its tall stature and high amylose content, resulting in poor lodging resistance and suboptimal culinary attributes. To address these issues, we employed CRISPR/Cas9 technology to precisely edit the SD1 and Wx genes in XDW rice, leading to the development of stable genetically homozygous lines with desired semi-dwarf and glutinous characteristics. A novel lily anther-specific gene encodes adhesin-like proteins associated with exine formation during anther development.

The anther-specific gene LLA1271 isolated from lily (Lilium longiflorum Thunb.) anthers is novel and exists in two forms. The protein encoded by LLA1271 may represent an adhesin-like protein first found in higher plants. The protein contains a typical N-terminal signal peptide followed by a highly conserved repeat domain. The LLA1271 gene is temporally expressed at the phase of microspore development. RNA blot and RNA in situ hybridization analyses demonstrated that the gene was expressed both in the tapetum and in the microspore. The gene is endo- and exogenously induced by gibberellin. Studi

Root phenotypes of dwarf and "overgrowth" SLN1 barley mutants, and implications for hypoxic stress tolerance.

Gibberellins are central to the regulation of plant development and growth. Action of gibberellins involves the degradation of DELLA proteins, which are negative regulators of growth. In barley (Hordeum vulgare), certain mutations affecting genes involved in gibberellin synthesis or coding for the barley DELLA protein (Sln1) confer dwarfism. Recent studies have identified new alleles of Sln1 with the capacity to revert the dwarf phenotype back to the taller phenotypes. While the effect of these overgrowth alleles on shoot phenotypes has been explored, no information is available for roots. Her

Modulating red: Far-red ratios accelerates speed breeding efficiency by coordinating photosynthesis and flowering in winter wheat (Triticum aestivum L.) under controlled environments.

The ratio of red to far-red light (R/FR) is well known to regulate plant photosynthesis and flowering. However, the molecular and physiological mechanisms underlying these coordinated responses in winter wheat remain largely unclear. In a controlled-environment growth chamber, we investigated flowering behavior, photosynthetic performance, genome-wide transcriptional responses, and breeding efficiency in winter wheat under four R/FR light treatments: 10:1, 2:1, 1:1, and 1:2. Compared with an R/FR ratio of 10:1, an R/FR ratio of 2:1 significantly enhanced breeding efficiency, enabling up to 4.7

An Integrative morphology-hormone-transcriptome analysis of uniconazole induced internode shortening in Paeonia lactiflora.

Plant height represents a critical ornamental trait in Paeonia lactiflora. While uniconazole has been demonstrated to effectively modulate plant height in this species-enhancing its ornamental value and supporting commercial production-the molecular mechanisms responsible for uniconazole-induced dwarfism remain largely elusive. In this study, we employed an integrated approach combining morphological, physiological, cytological, and molecular analyses to elucidate the dwarfing mechanism in P. lactiflora 'Dafugui'. Our results indicate that uniconazole treatment significantly suppressed interno

Metabolic Engineering for Gibberellic Acid Production in Fusarium fujikuroi: Advances and Perspectives.

Gibberellic acids (GAs) are a class of tetracyclic diterpene carboxylic acid compounds produced by green plants, fungi, and bacteria, which have a wide range of applications in agricultural production and food ingredients processing. Owing to the continuously growing market demand, enhancing GA yield has become imperative. The biosynthesis of GAs is a multi-enzymatic synergistic process that can be enhanced through genetic and metabolic engineering strategies. In this review, we first summarize recent advances in GA production by Fusarium fujikuroi. We then highlight key metabolic engineering

MdCo31 interacts with an RNA polymerase II transcription subunit 32 to regulate dwarf growth with short internodes in columnar apple.

The dominant Co locus controls the columnar growth phenotype of apple (Malus × domestica) trees. Candidate gene MdCo31, encoding 2-oxoglutarate-dependent dioxygenase, causes dwarf growth with short internodes in transgenic plants by reducing the abundance of biologically active gibberellin. However, the pathway regulating MdCo31 in the dwarfism of apple trees remains unclear. In this study, expression of MdCo31 was proved to be negatively correlated with internode length in F1 populations created by crossing columnar parents, and with dwarfism in transgenic apple plantlets. Yeast (Saccharomyce

Epigenetic Regulation of Cotton Plant Height by Histone Acetyltransferase GhHAT11 via Hormonal Pathway Modulation.

Histone acetyltransferases (HATs) regulate transcription epigenetically through histone acetylation and play crucial roles in plant development. In this study, we identified a cotton HAT gene, GhHAT11, associated with plant height and investigated its regulatory functions and underlying mechanisms. Tissue-specific expression analysis showed that GhHAT11 was predominantly expressed in stems. Virus-induced gene silencing (VIGS) of GhHAT11 significantly reduced cotton plant height and internode cell length, whereas heterologous overexpression of GhHAT11 in Arabidopsis thaliana increased plant hei

A domestication gene links plant architecture and nitrogen metabolism to enhance yield in foxtail millet.

Foxtail millet (Setaria italica) has lagged in yield compared with major cereals, largely due to suboptimal architecture. Here, we report a telomere-to-telomere (T2T) genome assembly of the dwarf, multi-tillering cultivar Dungu and identify SiSD1 (Semi Dwarf 1) via map-based cloning. This gene encodes GA20ox2, the rice SD1 ortholog involved in gibberellin biosynthesis. The loss-of-function sisd1 reduces bioactive gibberellins and remodels nitrogen metabolism, conferring semi-dwarfism, increased tillering, and higher yield. Critically, the semi-dominantly heterozygous SiSD1/sisd1 in F1 hybrids

Identification of GASA Protein Family Expression Levels in Cotton (Gossypium hirsutum L.) Affected by Verticillium wilt.

Verticillium wilt, caused by Verticillium dahliae, is a major constraint on cotton production. The GASA (Gibberellic Acid-Stimulated in Arabidopsis) proteins are cysteine-rich peptides induced by gibberellin signaling; however, their systemic role in upland cotton (Gossypium hirsutum) resistance to Verticillium wilt remains elusive. Here, we identified 40 GhGASA members from the G. hirsutum genome, which were clustered into three subfamilies. All members possessed a typical gibberellin-regulated domain, and segmental duplication was the primary driver of family expansion. Transcriptomic analys

Gibberellic acid maintains postharvest quality of Agaricus bisporus mushroom by enhancing antioxidative system and hydrogen sulfide synthesis.

The application of gibberellic acid (GA3 ) treatment to the postharvest quality maintenance of white button mushroom (Agaricus bisporus) was investigated. The optimum concentration of exogenous GA3 was 100 mg/L. At this concentration, the color change was inhibited, the firmness was maintained, and the weight loss and respiratory rates were reduced. The GA3 group had significantly lower malonaldehyde (MDA) content and membrane permeability. Reactive oxygen species accumulation was reduced due to the regulation of polyphenol oxidase (PPO), peroxidase (POD), and superoxide dismutase (SOD) enzyme
