Guiding precision medicine strategy for intravenous ginsenosides via pharmacokinetic-informed bioinformatic approaches: a study on XueShuanTong.

BACKGROUND: Intravenous ginsenosides, derived from Panax species, are widely used in China. XueShuanTong injection, enriched with ginsenosides Rb1 and Rg1, is recommended for unstable angina treatment. Although effective, it may cause adverse effects, especially in patients with renal or hepatic impairment, as these organs are vital in ginsenosides' systemic exposure. PURPOSE: This investigation aimed to inform precision medicine by employing a physiologically based pharmacokinetic (PBPK) model to evaluate transporter-mediated interactions between both ginsenosides and their systemic exposure

Ginsenosides: a potential natural medicine to protect the lungs from lung cancer and inflammatory lung disease.

Lung cancer is the malignancy with the highest morbidity and mortality. Additionally, pulmonary inflammatory diseases, such as pneumonia, acute lung injury, chronic obstructive pulmonary disease (COPD), and pulmonary fibrosis (PF), also have high mortality rates and can promote the development and progression of lung cancer. Unfortunately, available treatments for them are limited, so it is critical to search for effective drugs and treatment strategies to protect the lungs. Ginsenosides, the main active components of ginseng, have been shown to have anti-cancer and anti-inflammatory activitie

Estrogen receptor-β of microglia underlies sexual differentiation of neuronal protection via ginsenosides in mice brain.

AIMS: Streptococcus pneumoniae infection in acute bacterial meningitis can lead to widespread brain damage and mortality. Inflammatory responses by immune cells in the brain are thought to determine the degree of brain injury. Yet, the mechanisms underlying host responses to pneumococcal meningitis are largely unknown. To explore host responses as a potential therapeutic target for preventing brain injury after pneumococcal meningitis. METHODS: We evaluated signaling mechanisms that minimize neuronal damage caused by pneumococcal infection; specifically, we assessed pathways related to neuron

A novel β-glucosidase from Lacticaseibacillus rhamnosus: Catalytic specificity and efficient production of high-value rare ginsenosides.

Rare ginsenosides, especially compound K (CK), have attracted interest due to their notable bioactivities and potential applications in pharmaceuticals and functional foods. Here, a novel β-glucosidase (Lrbgl) was identified from Lacticaseibacillus rhamnosus. Lrbgl exhibited optimal activity at weakly acidic pH (5.0-7.0) and remained stable at temperatures up to 40 °C. The enzyme remained stable in the presence of organic solvents such as isopropanol and ethanol, and its catalytic activity was significantly enhanced by the addition of 0.2-1 M NaCl. Site-directed mutagenesis confirmed aspartate

Ameliorative effect of total ginsenosides from heat-treated fresh ginseng against cyclophosphamide-induced liver injury in mice.

This study evaluated the effect of heat treatment on the conversion of ginsenoside and the ameliorative effect of heat-treated total ginsenoside (HG) from fresh ginseng on cyclophosphamide (CTX)-induced liver injury. LC-MS analysis revealed that the content of rare ginsenosides increased markedly after heat treatment. HG significantly attenuated CTX-induced hepatic histopathological injury in mice. Western blotting analysis showed that untreated total ginsenoside (UG) and HG regulated the Nrf2/HO-1 and TLR4/MAPK pathways. Importantly, these results may be relevant to the modulation of the inte

Improvement of tissue-specific distribution and biotransformation potential of nicotinamide mononucleotide in combination with ginsenosides or resveratrol.

Decreased Nicotinamide adenine dinucleotide (NAD+ ) level has received increasing attention in recent years since it plays a critical role in many diseases and aging. Although some research has proved that supplementing nicotinamide mononucleotide (NMN) could improve the level of NAD+ , it is still uncertain whether the NAD+ level in specific tissues could be improved in combination with other nutrients. So far, a variety of nutritional supplements have flooded the market, which contains the compositions of NMN coupled with natural products. However, the synergy and transformation process of N

Selected ginsenosides interfere efficiently with hepatitis B virus mRNA expression levels and suppress viral surface antigen secretion.

Ginsenosides are a class of natural steroid glycosides and triterpene saponins found in Panax ginseng. After screening of a commercial ginsenoside compound library for low cellular cytotoxicity and the ability to mediate efficient reductions in hepatitis B virus (HBV) mRNA expression levels in HepG2.2.15 cells, three ginsenosides (Rg6, Rh4, and Rb3) are selected. Thereafter, using the same cellular model, all three ginsenosides are shown to mediate efficient, selective inhibition of HBV mRNA expression levels, and also interfere with the secretion of both HBV particles and hepatitis B surface

Combination of the ginsenosides Rb3 and Rb2 exerts protective effects against myocardial ischemia reperfusion injury in rats.

Ginsenoside Rb3 (G‑Rb3) has been demonstrated to alleviate myocardial ischemia reperfusion injury (MIRI); however, it is difficult to separate G‑Rb2 from its isomer G‑Rb3. The current study aimed to compare the cardioprotective effects of G‑Rb3 and the concomitant use of G‑Rb3 and G‑Rb2 (G‑Rb3/Rb2) on MIRI in rats. A rat model of MIRI was established by ligation of the left anterior descending coronary artery and the rats were randomly divided into five groups. Prior to MIRI, G‑Rb3/Rb2 (20 mg/kg), G‑Rb3 (20 mg/kg) and diltiazem (DLZ; 20 mg/kg, as a positive control) were orally administered to

Ginsenoside Rd: molecular mechanisms, pharmacological activities, and therapeutic potential.

Ginsenoside Rd (Rd), a major protopanaxadiol saponin from Panax species, is a pleiotropic candidate with potential relevance for treating ischemic, inflammatory, metabolic, and degenerative diseases. Over the past two decades, evidence from cellular systems, animal models, and early clinical studies indicates that Rd provides multi-level protection by modulating oxidative stress, inflammatory signaling, calcium dysregulation, mitochondrial dysfunction, and regulated cell death. Mechanistic studies implicate interconnected pathways, including PI3K/Akt, ERK, NF‑κB, Nrf2‑mediated antioxidant prog

Ginsenoside Rg1: A bioactive therapeutic agent for diverse liver diseases.

Diverse liver diseases are characterised by late diagnosis and rapid progression and have become one of the major threats to human health. To delay the transition from benign tissue lesions to a substantial organ injury, scientists have gradually applied natural compounds derived from plants as a complementary therapy in the field of hepatology. Ginseng (Panax ginseng C. A. Meyer) is a tonic traditional Chinese herbal medicine, and natural products, including ginsenoside Rg1 (G-Rg1), which is a kind of 20(S)-protopanaxatriol saponin with a relatively high biological activity, can be isolated f

Ginsenoside Rb1 Alleviated the AKI to CKD Transition by Targeting VEGFR2.

The decrease of peritubular capillaries, a pathological feature of acute kidney injury (AKI), is a critical part that promotes the transition of AKI to chronic kidney disease (CKD). Ginsenoside Rb1 (Rb1) has various pharmacological effects on multiple systemic diseases. However, whether Rb1 delays the transition of AKI to CKD and the mechanism are unclear. Here, we discovered that Rb1 could alleviate kidney pathological damage in mice with unilateral ischaemia/reperfusion injury (uIRI), and it enhanced kidney function, reduced renal fibrosis while increasing microvessel density. Ginsenoside RB1 Influences Macrophage-DPSC Interactions in Inflammatory Conditions.

INTRODUCTION AND AIMS: Unresolved inflammation and tissue destruction are supposed to underlie the failure of dental pulp repair. As crucial regulators of the injury response, dental pulp stem cells (DPSCs) play a key role in pulp tissue repair and regeneration. M2 macrophages have been demonstrated to induce osteogenic/odontogenic differentiation of DPSCs. Ginsenoside Rb1 (GRb1) is the major component of ginseng and manifested an anti-inflammatory role by promoting M1 macrophage polarised into M2 macrophage in inflammatory disease. However, whether GRb1 facilitates odontogenic differentiation

Effect of Ginsenoside-Rh2 and Curcurbitacin-B on Cryptosporidium parvum in vitro.

Ginsenoside-Rh2 and cucurbitacin-B (CuB) are secondary metabolites of Ginseng (Panax ginseng) and Cucurbitaceae plants respectively. We assessed the anticryptosporidial activity of these two functional compounds in a cell culture model of cryptosporidiosis. The highest concentration of each compound that was not toxic to the host cells was used to assess the activity against C. parvum during infection/invasion and growth in HCT-8 cell monolayers. Monolayers were infected with pre-excysted C. parvum oocysts. Infected monolayers were incubated at 37 °C for 24 h and 48 h in the presence of differ

Minor ginsenoside F1 improves memory in APP/PS1 mice.

Ginseng has been shown to produce a cognitive improvement effect. The key molecular components in ginseng that produce pharmacological effects are ginsenosides. Previous studies reported a memory improvement effect of a few major ginsenosides. However, the identity of specific minor ginsenosides mediating such function remains unknown. Here, we report that a minor ginsenoside F1 improves memory function in APPswe/PSEN1dE9 (APP/PS1) double-transgenic Alzheimer's disease (AD) model mice. After 8-wk oral administration of F1 jelly, we observed that spatial working memory, but not context-dependen
