Glutamine improves intestinal barrier function in experimental biliary obstruction.

OBJECTIVE: To determine the effects of enteral administration of glutamine on intestinal barrier function in experimental biliary obstruction. BACKGROUND: Extrahepatic biliary obstruction is associated with the failure of intestinal barrier function, allowing bacteria and other substances from the intestine to enter the circulation and initiate a systemic inflammatory response, causing impairment of multiple organs. The amino acid glutamine has been shown to improve intestinal barrier function in other conditions, but its effects in biliary obstruction have not been fully examined. METHODS:

Growth hormone, glutamine, and modified diet for intestinal adaptation.

Many patients who undergo extensive resection of the gastrointestinal tract develop intestinal failure from short-bowel syndrome that results in significant malabsorption of fluid, electrolytes, and other nutrients. This may result in dependence on long-term parenteral nutrition. It has been almost a decade since Byrne and colleagues published their research demonstrating enhanced absorption of nutrients, improved weight gain, and reduction in parenteral nutrition requirements with the administration of a combination of growth hormone, glutamine, and a modified diet. Other researchers have con

Role of L-glutamine in critical illness: new insights.

PURPOSE OF REVIEW: This review will attempt to summarize recent clinical and experimental data on glutamine's use in critical illness. It will try to present the concept of glutamine as a 'drug' or 'nutraceutical', given in addition to standard nutritional support. RECENT FINDINGS: Glutamine, traditionally considered to be a nonessential amino acid, is now considered as 'conditionally essential' following critical illness and injury. States of critical illness lead to significant decreases in plasma levels of glutamine and when this decrease is severe it has been correlated with increased mor

Spinocerebellar ataxia type 6: channelopathy or glutamine repeat disorder?

Spinocerebellar ataxia type 6 (SCA6) is due to small expansions of a CAG repeat at the 3' end of the CACNA1A gene, coding for the alpha(1A) subunit of voltage-gated calcium channels type P/Q, expressed in the cerebellar Purkinje and granule cells. It is one of three allelic disorders, the other two being episodic ataxia type 2 (EA2), due mostly to protein truncating mutations, and familial hemiplegic migraine, associated with missense mutations. The latter disorders, due to point mutations altering the P/Q channel activity, clearly belong to the group of channelopathies. For SCA6, due to CAGn

Effect of oral glutamine supplementation during bone marrow transplantation.

BACKGROUND: Because all patients receiving bone marrow transplant (BMT) and peripheral blood progenitor cell transplant (PBPCT) experience gastrointestinal (GI) toxicity from the preparative regimen of chemotherapy, with or without radiation, oral glutamine was administered during the preparatory regimen and after transplant to maintain GI structure and function. METHODS: To evaluate effects of oral glutamine on nutritional status and overall outcome, a prospective, randomized, double-blinded study was performed on 58 autologous and allogeneic BMT patients. Patients received 30 g of oral glut

Parenteral glutamine protects hepatic function during bone marrow transplantation.

Hepatic veno-occlusive disease (VOD) of the liver is a common complication following high-dose cytotoxic therapy for bone marrow transplantation (BMT). The major pathological changes are seen in centrilobular (zone 3) hepatocytes and adjacent endothelium. Glutathione (GSH) becomes depleted following chemotherapy and experimental evidence suggests reduced levels predispose to centrilobular hepatocyte and endothelial cell injury. Animal studies have shown that glutamine infusions can maintain GSH levels and protect against free radical injury. We have prospectively studied the effect of glutamin

Glutamine Metabolism Supports α cell Mass and Glucagon Secretion.

The liver-α cell axis is a finely tuned biological rheostat that regulates whole body amino acid availability. Pancreatic α cells secrete glucagon that regulates amino acid catabolism through gluconeogenesis and ureagenesis, yet the mechanisms linking amino acid levels to α cell growth and function are not fully understood. Here, we identify glutaminase, the enzyme that catalyzes glutamine catabolism, as a critical α cell regulator. Glutaminase is highly enriched in α cells across species. α cell expression of glutaminase is required for nutrient-dependent mTORC1 activation, suppression of AMP

Glutamate-Glutamine metabolism and transport, implications in Parkinson's disease.

Parkinson's disease (PD) is a common neurodegenerative condition caused by the selective loss of dopaminergic neurons in the substantia nigra (SN) and the formation of Lewy bodies due to α-Synuclein (α-Syn) aggregation in the midbrain. Glutamate (Glu) is an excitatory neurotransmitter that plays an important role in the normal functioning of the basal ganglia circuit. It has been shown recently that Glu receptors are involved in the regulation of neurotransmitter release, neuron excitability and long-term plasticity and the altered mechanisms in Parkinson's disease. Glu is produced in the cyto

Restoration of Cementoblast Mineralization by Glutamine Under Inflammatory Conditions.

INTRODUCTION AND AIMS: Cementoblasts are critical for cementum formation and periodontal regeneration during inflammatory diseases such as periodontitis and apical periodontitis (AP). Bacterial infection leads to metabolic changes in cementoblasts and reduces mineralization capability. This study aimed to fully elucidate the metabolic changes of cementoblasts in inflammatory mineralization and explore therapeutic strategies targeting metabolism. METHODS: Mouse cementoblasts cell line OCCM-30 were infected with Porphyromonas gingivalis (P.g) to model microbial infection stress. Metabolic profi

L-Glutamine-Induced Acute Kidney Injury: A Clinical Observation.

INTRODUCTION: L-Glutamine is increasingly used as a dietary supplement and its use is, as is the case with other amino acids, considered safe. L-Glutamine is the most abundant amino acid in the human body and is involved in many metabolic reactions. Within the kidney L-Glutamine has an important role in the generation of ammonia and bicarbonate. CASE PRESENTATION: We report a case of acute kidney injury (AKI) as a result of tubular damage in a patient who used 18 grams of L-Glutamine on a daily basis. Possible mechanisms are proposed of which increased single nephron ammonia production and to

Prion diseases disrupt glutamate/glutamine metabolism in skeletal muscle.

In prion diseases (PrDs), aggregates of misfolded prion protein (PrPSc) accumulate not only in the brain but also in extraneural organs. This raises the question whether prion-specific pathologies arise also extraneurally. Here we sequenced mRNA transcripts in skeletal muscle, spleen and blood of prion-inoculated mice at eight timepoints during disease progression. We detected gene-expression changes in all three organs, with skeletal muscle showing the most consistent alterations. The glutamate-ammonia ligase (GLUL) gene exhibited uniform upregulation in skeletal muscles of mice infected with

Amino Acid Starvation-Induced Glutamine Accumulation Enhances Pneumococcal Survival.

Bacteria are known to cope with amino acid starvation by the stringent response signaling system, which is mediated by the accumulation of the (p)ppGpp alarmones when uncharged tRNAs stall at the ribosomal A site. While a number of metabolic processes have been shown to be regulatory targets of the stringent response in many bacteria, the global impact of amino acid starvation on bacterial metabolism remains obscure. This work reports the metabolomic profiling of the human pathogen Streptococcus pneumoniae under methionine starvation. Methionine limitation led to the massive overhaul of the pn

L-glutamine Induces Expression of Listeria monocytogenes Virulence Genes.

The high environmental adaptability of bacteria is contingent upon their ability to sense changes in their surroundings. Bacterial pathogen entry into host poses an abrupt and dramatic environmental change, during which successful pathogens gauge multiple parameters that signal host localization. The facultative human pathogen Listeria monocytogenes flourishes in soil, water and food, and in ~50 different animals, and serves as a model for intracellular infection. L. monocytogenes identifies host entry by sensing both physical (e.g., temperature) and chemical (e.g., metabolite concentrations)

Oral Glutamine Supplementation Protects Female Mice from Nonalcoholic Steatohepatitis.

BACKGROUND: Genetic factors, a diet rich in fat and sugar, and an impaired intestinal barrier function are critical in the development of nonalcoholic steatohepatitis (NASH). The nonessential amino acid glutamine (Gln) has been suggested to have protective effects on intestinal barrier function but also against the development of liver diseases of various etiologies. OBJECTIVE: The effect of oral Gln supplementation on the development of Western-style diet (WSD)-induced NASH in mice was assessed. METHODS: Female 6- to 8-wk-old C57BL/6J mice were pair-fed a control (C) diet or a WSD alone or

Parenteral glutamine supplementation in critical illness: a systematic review.

INTRODUCTION: The potential benefit of parenteral glutamine (GLN) supplementation has been one of the most commonly studied nutritional interventions in the critical care setting. The aim of this systematic review was to incorporate recent trials of traditional parenteral GLN supplementation in critical illness with previously existing data. METHODS: All randomized controlled trials of parenterally administered GLN in critically ill patients conducted from 1997 to 2013 were identified. Studies of enteral GLN only or combined enteral/parenteral GLN were excluded. Methodological quality of stud

Glutamine preserves gut glutathione levels during intestinal ischemia/reperfusion.

Intestinal ischemia/reperfusion (I/R) causes formation of reactive oxygen intermediates (ROI) which lead to mucosal cell injury. Glutathione (GSH), an ROI scavenger, protects tissues from ROI-mediated cell injury. Since GSH biosynthesis is partially dependent on glutamine (Gln) levels, we tested the hypothesis that intravenous Gln infusion will assist in maintaining mucosal cell GSH levels and decrease membrane lipid peroxidation during intestinal I/R. The external jugular vein of male Sprague-Dawley rats was cannulated and infused with normal saline (NS) at 2 cc/hr. After 3 days, matched pair
