Metabolic Regulation Mechanism of Ilex verticillata in Response to Drought Stress: "First Inhibition Then Promotion" of Lipids and the Key Mediation of Glycerophospholipid Metabolic Pathway.

BACKGROUND: Ilex verticillata belongs to the genus Ilex of the family Aquifoliaceae. It has extremely high ornamental value, and its drought tolerance is crucial for expanding its cultivation range. Therefore, studying the metabolic regulation mechanism of I. verticillata in response to drought stress at different stages under natural drought conditions provides a basis for the expanded cultivation of I. verticillata and important theoretical support for the drought-resistant breeding of woody plants. METHODS: Two-year-old cutting seedlings (0.3~0.4 m high) of I. verticillata were used as exp

Mass Spectrometry Analysis of Wild-Type and Knock-in Q140/Q140 Huntington's Disease Mouse Brains Reveals Changes in Glycerophospholipids Including Alterations in Phosphatidic Acid and Lyso-Phosphatidic Acid.

BACKGROUND: Huntington's disease (HD) is a neurodegenerative disease caused by a CAG expansion in the HD gene, which encodes the protein Huntingtin. Huntingtin associates with membranes and can interact directly with glycerophospholipids in membranes. OBJECTIVE: We analyzed glycerophospholipid profiles from brains of 11 month old wild-type (WT) and Q140/Q140 HD knock-in mice to assess potential changes in glycerophospholipid metabolism. METHODS: Polar lipids from cerebellum, cortex, and striatum were extracted and analyzed by liquid chromatography and negative ion electrospray tandem mass sp

Submicron Particulate Matter (PM1)-Induced Pulmonary Injury and Epigallocatechin-3-Gallate Intervention: Modulation of Disorders in Amino Acid, Glycerophospholipid, and Energy Metabolism.

Emerging evidence indicates that submicron particulate matter (PM1) poses greater respiratory health risks than coarser particles (e.g., PM2.5). Yet its specific toxicological mechanisms and effective mitigation strategies remain insufficiently understood. This study demonstrated that PM1 exposure induced significant dose-dependent pulmonary oxidative stress and inflammatory response in vivo and in vitro. Metabolomic analysis revealed that PM1 triggered more complex metabolic alterations than those induced by PM2.5, including disorders in amino acid metabolism, glycerophospholipid metabolism,

Occupational dust exposure is associated with cardiovascular risk via glycerophospholipid metabolism and IL-6-mediated endothelial inflammation: Insights from multi-omics integration.

Occupational dust is a widespread environmental contaminant associated with cardiovascular disease risk, yet its underlying molecular mechanisms remain insufficiently elucidated. This study investigated the associations between occupational respirable dust exposure (CDE) and early indicators of cardiovascular risk in 8662 participants from the China-Northern Anhui Coal Workers Cohort (C-NACWC). To explore potential mechanisms, we employed an integrative multi-omics approach, combining serum lipidomics from these participants with publicly available single-cell RNA sequencing data (GSE159677) f

Chrysanthemum 3,5-O-dicaffeoylquinic acid ameliorates mouse alcoholic liver injury by suppressing the TLR-4/NF-κB pathway and restoring glycerophospholipid homeostasis.

3,5-O-Dicaffeoylquinic acid (3,5-DCQA) from edible Gongju chrysanthemum has shown protective effects against alcoholic hepatocyte injury in our previous study. However, the specific effects and mechanisms of 3,5-DCQA in a mouse model of liver injury remain underexplored. This study aimed to examine the efficacy and primary mechanism of 3,5-DCQA from Gongju chrysanthemum in ameliorating acute alcoholic liver injury in mice. The results indicated that 3,5-DCQA effectively attenuated inflammation and apoptosis by regulating the content of key factors (e.g. MyD88, NF-κB) in the TLR-4/NF-κB pathway

Loss of G0/G1 switch gene 2 (G0S2) promotes disease progression and drug resistance in chronic myeloid leukaemia (CML) by disrupting glycerophospholipid metabolism.

Tyrosine kinase inhibitors (TKIs) targeting BCR::ABL1 have turned chronic myeloid leukaemia (CML) from a fatal disease into a manageable condition for most patients. Despite improved survival, targeting drug-resistant leukaemia stem cells (LSCs) remains a challenge for curative CML therapy. Aberrant lipid metabolism can have a large impact on membrane dynamics, cell survival and therapeutic responses in cancer. While ceramide and sphingolipid levels were previously correlated with TKI response in CML, the role of lipid metabolism in TKI resistance is not well understood. We have identified dow

Omega 6 to omega 3 fatty acid imbalance early in life leads to persistent reductions in DHA levels in glycerophospholipids in rat hypothalamus even after long-term omega 3 fatty acid repletion.

Failure to provide omega 3 fatty acids in the perinatal period results in alterations in nerve growth factor levels, dopamine production and permanent elevations in blood pressure. The present study investigated whether changes in brain (i.e., hypothalamus) glycerophospholipid fatty acid profiles induced by a diet rich in omega 6 fatty acids and very low in alpha-linolenic acid (ALA) during pregnancy and the perinatal period could be reversed by subsequent feeding of a diet containing ALA. Female rats (6 per group) were mated and fed either a low ALA diet or a control diet containing ALA throu

Spatially and temporally probing distinctive glycerophospholipid alterations in Alzheimer's disease mouse brain via high-resolution ion mobility-enabled sn-position resolved lipidomics.

Dysregulated glycerophospholipid (GP) metabolism in the brain is associated with the progression of neurodegenerative diseases including Alzheimer's disease (AD). Routine liquid chromatography-mass spectrometry (LC-MS)-based large-scale lipidomic methods often fail to elucidate subtle yet important structural features such as sn-position, hindering the precise interrogation of GP molecules. Leveraging high-resolution demultiplexing (HRdm) ion mobility spectrometry (IMS), we develop a four-dimensional (4D) lipidomic strategy to resolve GP sn-position isomers. We further construct a comprehensiv

Expression of AGPAT2, an enzyme involved in the glycerophospholipid/triacylglycerol biosynthesis pathway, is directly regulated by HIF-1 and promotes survival and etoposide resistance of cancer cells under hypoxia.

Hypoxia inducible factor-1 (HIF-1) supports survival of normal cells under low oxygen concentration and cancer cells in the hypoxic tumor microenvironment. This involves metabolic reprogramming via upregulation of glycolysis, downregulation of oxidative phosphorylation and, less well documented, effects on lipid metabolism. To investigate the latter, we examined expression of relevant enzymes in cancer cells grown under hypoxia. We show that expression of acylglycerol-3-phosphate acyltransferase 2 (AGPAT2), also known as lysophosphatidic acid acyltransferase β (LPAATβ), was upregulated under h

Unravelling lumbar disc herniation severity beyond MRI : integrated transcriptomic and metabolomic analyses highlight glycerophospholipid metabolism and inform a machine-learning diagnostic model: a pilot study.

AIMS: While MRI serves as a tool for assessing the severity of lumbar disc herniation (LDH), it has been observed that imaging diagnoses do not always align with clinical symptoms in nearly half of patients. The absence of dependable prognostic biomarkers impedes the early and accurate diagnosis of LDH, which is critical for the development of further treatment approaches. Thus, the aim of this study was to elucidate the molecular mechanisms that determine pain and LDH severity. METHODS: We conducted a pilot study with 55 patients, employing transcriptomic and metabolomic analyses on blood sa

Lactiplantibacillus plantarum KLDS1.0344 ameliorates depressive-like behavior in CUMS mice: integrative multi-omics evidence linking gut microbiota alterations to sphingolipid and glycerophospholipid metabolic pathways.

Depression is a common and debilitating psychiatric disorder, and current pharmacotherapies are often limited by delayed onset, variable efficacy, and adverse side effects. Therefore, safer and more effective adjunctive or alternative strategies are needed. In the present study, a chronic unpredictable mild stress-induced murine model was used to evaluate the antidepressant potential of Lactiplantibacillus plantarum KLDS1.0344 by integrating behavioral assessments, neurochemical analyses, 16S rRNA sequencing, and untargeted metabolomics of intestinal contents. Parallel metabolomic and transcriptomic profiling reveals the cytotoxic mechanism of protein-bound uremic toxin p-cresol sulfate via disruption of glutathione and glycerophospholipid metabolism in MC3T3-E1 osteoblasts.

BACKGROUND: Chronic kidney disease-mineral bone disorder (CKD-MBD), a common complication of chronic kidney disease (CKD), leads to vascular calcification, osteoporosis, and electrolyte disturbances, impacting patient survival and quality of life. Conventional dialysis poorly removes protein-bound uremic toxins like p-cresyl sulfate (PCS), which are linked to CKD-MBD. This study combined metabolomics and transcriptomics to explore PCS's cytotoxic mechanisms in CKD-MBD for better clinical management. METHODS: Using MC3T3-E1 osteoblasts treated with or without PCS, metabolic changes were analyz

A UHPLC-Orbitrap-MS Metabolomics Strategy Reveals Glycerophospholipid Metabolic Remodeling Is Associated with the Anti-Arthritic Effect of Glycyrrhiza Protein-Paeoniflorin Nanoparticles via PI3K/AKT/NLRP3 Axis.

Rheumatoid arthritis involves chronic synovitis and immune-metabolic dysregulation, highlighting a need for multi-target therapies that jointly modulate metabolism and inflammation. We developed glycyrrhiza protein-paeoniflorin self-assembled nanoparticles (GP-PF NPs) and investigated their anti-arthritic mechanism in adjuvant-induced arthritis (AIA) mice, using UHPLC-Orbitrap-MS-based metabolomics. Male C57BL/6 mice (n = 42) were assigned to the control, model, GP-PF NPs, paeoniflorin, glycyrrhiza protein, physical mixture, and celecoxib groups. All groups except controls received complete Fr

Impaired embryonic development in glucose-6-phosphate dehydrogenase-deficient Caenorhabditis elegans due to abnormal redox homeostasis induced activation of calcium-independent phospholipase and alteration of glycerophospholipid metabolism.

Glucose-6-phosphate dehydrogenase (G6PD) deficiency is a commonly pervasive inherited disease in many parts of the world. The complete lack of G6PD activity in a mouse model causes embryonic lethality. The G6PD-deficient Caenorhabditis elegans model also shows embryonic death as indicated by a severe hatching defect. Although increased oxidative stress has been implicated in both cases as the underlying cause, the exact mechanism has not been clearly delineated. In this study with C. elegans, membrane-associated defects, including enhanced permeability, defective polarity and cytokinesis, were

Ganglioside changes in the regenerating goldfish optic system: comparison with glycoproteins and phospholipids.

Axonally transported radioactivity in sialoglycoconjugates, labeled by intraocular injection of [3H]N-acetylmannosamine, increased significantly during regeneration of goldfish optic axons at 30 degrees C. Ganglioside radioactivity showed the largest increase--approximately eightfold--in the optic nerve tract at 8 days after optic nerve crush while sialoglycoprotein radioactivity increased fourfold under the same conditions. As regeneration proceeded the magnitude of the increase in the nerve tract diminished for both glycoconjugates. In the optic tectum, however, transported radioactivities r

Cellular membranes of the related fission yeast species differ in their dipole potential.

The dielectric environment and membrane dipole potential are key electrostatic properties of biological membranes, which modulate the function of membrane-associated proteins. If and how these properties are shaped by species-specific cellular lipid landscapes are fundamental questions. Here, we use two related fission yeast species, Schizosaccharomyces pombe (S. pombe) and Schizosaccharomyces japonicus (S. japonicus), which exhibit remarkable differences in membrane lipid composition, to address this problem. S. pombe synthesizes membranes from largely unsaturated glycerophospholipids with 18
