Antibodies against the "core" glycolipid of Enterobacteriaceae (2-keto, 3-deoxyoctonate-Lipid A) have been associated with protection against the sequelae of gram-negative rod bacteremia. To investigate the nature of this protection, dogs and rabbits were immunized with purified glycolipid prepared by phenol-chloroform-petroleum ether extraction of the "Re" mutant of Salmonella minnesota 595 and opsonophagocytic and bactericidal tests were carried out using lapine peritoneal granulocytes and serum factors. Whereas 1-4 mug/kg of glycolipid i.v. produced hypotension in dogs and 8 mug/kg i.v. was

Mimicking cell membrane glycocalyx, saccharide modification of nanoparticles offers a potent means to regulate their in vivo fate. Here, we investigate how glycosylation (i.e., glucose, galactose, fructose, mannose, and N-acetylglucosamine) regulates liposomal nanoparticle interactions with plasma proteins and immune cells, which further determine their biodistribution and therapeutic efficacy. While fructose conferred the greatest enhancement in tumor cell uptake in vitro, N-acetylglucosamine-modified nanoparticles achieved the highest tumor accumulation and markedly attenuated systemic clear

Candida albicans is a leading fungal pathogen in humans, responsible for infections that span from mucosal surfaces to severe systemic diseases. This study aimed to investigate potential ability of yeast-derived glycolipids from Meyerozyma guilliermondii as an antifungal against Candida albicans biofilms. Glycolipid extract (64 µg/mL) reduced metabolic activity by 50% in both immature and mature biofilms, while biofilm mass was reduced at higher concentrations of 128 and 256 µg/mL, respectively. Adhesion, a key step in biofilm formation, decreased by over 50% when cells were treated with glyco

Tpp80Aa1 from Bacillus thuringiensis is a Toxin_10 family protein (Tpp) with reported action against Culex mosquitoes. Here, we demonstrate an expanded target range, showing Tpp80Aa1 is also active against the larvae of Anopheles gambiae and Aedes aegypti mosquitoes. We report the first crystal structure of Tpp80Aa1 at a resolution of 1.8 Å, which shows Tpp80Aa1 consists of two domains: an N-terminal β-trefoil domain resembling a ricin B lectin and a C-terminal putative pore-forming domain sharing structural similarity with the aerolysin family. Similar to other Tpp family members, we observe

Mycobacteriosis and nocardiosis in cultured fish caused by infections with acid-fast bacteria, are responsible for large economic losses globally. In this study, we suggest a novel adjuvant using glycolipids that activates host immune systems. The immune response to glycolipids stimulation was investigated using ginbuna crucian carp. Ginbuna vaccinated with FKC (formalin-killed cells) + glycolipids isolated from Mycobacterium sp., upregulated inflammatory- and Th1-related cytokines, and a DTH (delayed-type hypersensitivity) response was confirmed only in ginbuna vaccinated with FKC + glycolipi

A 19-year-old male student with Guillain-Barré syndrome (GBS) was described. Neurologic examination revealed severe and moderate degrees of weakness in the distal and proximal muscles, respectively, in both upper and lower limbs. He was unable to walk even with support. Both superficial and deep sensations were normal. Serial electrophysiologic studies indicated that the predominant pathologic process of the peripheral nerve was axonal degeneration of motor nerves. Enzyme-linked immunosorbent assay showed that serum from the patient had markedly high titer of IgG antibody to Ga1NAc-GD1a (1:204

Lipid nanoparticles (LNPs) formulated with a neutral helper lipid can deliver RNA to the liver in humans. However, clinically relevant delivery to other tissues has remained challenging. To avoid the liver, scientists often add antibodies or helper lipids with a permanent charge. Here, we report an alternative approach: antibody- and charge-independent liver detargeting. Using DNA barcoding to test 109 chemically distinct LNPs in vivo, we found that replacing a neutral helper lipid with a neutral glycolipid reduced liver delivery and increased splenic delivery. Consistent with this differentia

INTRODUCTION: Diseases triggered by glucose and lipid metabolic disorders, such as hyperglycemia and hyperlipidemia, have become a global health threat. According to statistics, diabetic patients have exceeded 463 million worldwide, and the prevalence of hyperlipidemia is also continuously rising. These glycolipid metabolic diseases not only significantly increase the risk of complications such as cardiovascular disease, stroke, and kidney disease but also impose a huge economic burden on the global healthcare system. Despite the continuous emergence of treatment methods for glucose and lipid

Coronary microvascular disease (CMVD) is associated with abnormalities in glucose-lipid metabolism. And the triglyceride to high density lipoprotein (HDL) (TG/HDL) ratio can be used to characterize levels of glycolipid metabolism. Therefore, it is hypothesized that increased TG/HDL may trigger CMVD. This study enrolled patients with angina pectoris but negative coronary angiograms to explore inflammatory factor-mediated disorder of glycolipid metabolism triggers CMVD. Logistics regression model and subgroup analysis were constructed to explore the associations between TG/HDL and CMVD. Restrict

Discovering new antibacterial agents is crucial to addressing the increasing risk of bacterial infections induced by antimicrobial resistance in food and agricultural industries. Here, biocompatible acidic-type sophorolipids (ASLs) and glucolipids (GLs) prepared via chemical modification of natural sophorolipids from fermentation were functionalized via amide modification for use as potential antibacterial agents. It was found that the arginine methyl ester derivative of GLs (GLs-d-Arg-OMe) showed excellent antibacterial activity, killing more than 99.99% of Escherichia coli at 200 mg/L. The s

Glycolipids are a group of sugar-containing lipids with versatile functions. In this study, a natural glycolipid product was obtained from soy lecithin, and its emulsifying, oil-gelling, antibacterial and antiviral properties were investigated. A silica-based extraction method on a preparative scale was used to recover the glycolipid product (GLP) from soy lecithin. The GLP consisted of three different glycolipid classes: acylated sterol glucoside (64.16%), sterol glucoside (25.57%) and cerebroside (6.71%). As an emulsifier, the GLP was able to form a stable water-in-oil emulsion. The GLP exhi

BACKGROUND: Recent investigations indicate that schistosome infection is closely associated with aberrant glycolipid metabolism. However, the actual glycolipid metabolism gene expression, as well as the possible pathways that regulate glycolipid metabolism in the schistosome-infected liver, has not been extensively explored.
METHODS: In this study, we evaluated the dynamic expression of glycolipid metabolism-associated genes and proteins in the livers from mice infected with Schistosoma japonicum at the indicated time points using real-time PCR and immunofluorescence. Then, cultures of macrop

The cellular entry of the bacterial Shiga toxin and the related verotoxins has been scrutinized in quite some detail. This is due to their importance as a threat to human health. At the same time, the study of Shiga toxin has allowed the discovery of novel molecular mechanisms that also apply to the intracellular trafficking of endogenous proteins at the plasma membrane and in the endosomal system. In this review, the individual steps that lead to Shiga toxin uptake into cells will first be presented from a purely mechanistic perspective. Membrane-biological concepts will be highlighted that a

CD1 proteins present microbial lipids to T cells. Germline-encoded mycolyl lipid-reactive (GEM) T cells with conserved αβ T cell receptors (TCRs) recognize CD1b presenting mycobacterial mycolates. As the molecular basis underpinning TCR recognition of CD1b remains unknown, here we determine the structure of a GEM TCR bound to CD1b presenting glucose-6-O-monomycolate (GMM). The GEM TCR docks centrally above CD1b, whereby the conserved TCR α-chain extensively contacts CD1b and GMM. Through mutagenesis and study of T cells from tuberculosis patients, we identify a consensus CD1b footprint of TCRs

X-linked adrenomyeloneuropathy (AMN) is an inherited neurometabolic disorder caused by malfunction of the ABCD1 gene, characterized by slowly progressing spastic paraplegia affecting corticospinal tracts, and adrenal insufficiency. AMN is the most common phenotypic manifestation of adrenoleukodystrophy (X-ALD). In some cases, an inflammatory cerebral demyelination occurs associated to poor prognosis in cerebral AMN (cAMN). Though ABCD1 codes for a peroxisomal transporter of very long-chain fatty acids, the molecular mechanisms that govern disease onset and progression, or its transformation to

Pseudozyma antarctica is one of the best producers of the glycolipid biosurfactants known as mannosylerythritol lipids (MELs), which show not only excellent surface-active properties but also versatile biochemical actions. In order to obtain a variety of producers, all the species of the genus were examined for their production of MELs from soybean oil. Pseudozyma fusiformata, P. parantarctica and P. tsukubaensis were newly identified to be MEL producers. Of the strains tested, P. parantarctica gave the best yield of MELs (30 g L(-1)). The obtained yield corresponded to those of P. antarctica,

BACKGROUND: We studied whether the level of anti-skeletal muscle glycolipid antibodies (AGA), a marker of acute rejection in heart transplantation, may be associated with an adverse prognosis in unstable angina.
METHODS AND RESULTS: The in-hospital evolution of 50 patients with unstable angina (Braunwald class III B) was assessed. We determined the incidence of death, myocardial infarction, and refractory angina. Blood was collected at admission and 24 hours later for determination of AGA levels by enzyme-linked immunosorbent assay. Twenty-three patients showed a decrease in the AGA level at

Immunostaining of human, bovine and rodent unfixed nervous tissue sections was performed in order to characterize the structures recognized by anti-glycolipid antibodies. Four human sera from patients, two with M-IgM and motor neuron syndrome or motor neuropathy and two with motor neuropathy and polyclonal IgG antibody activity against gangliosides (GL; i.e. GM1, GD1b, GD1a), were utilized. Serum from a patient with sensory neuropathy and M-IgM immunoglobulins with antibody activity against sulfatide (SUL) was included in this series. This study shows that polyclonal and monoclonal anti-glycol

The protective antigen (PAg), a glycolipid substance, was extracted from Leptospira interrogans serovar lai strain 017 with a chloroform-methanol-water (1:2:0.8 [vol/vol/vol]) solution and partially purified by silica gel column chromatography. The PAg was not detected by Coomassie brilliant blue staining in sodium dodecyl sulfate-polyacrylamide gel electrophoresis but was observed as a smearlike band, which corresponded to a 24- to 30-kilodalton standard protein, by silver staining. The outer envelope (OE) fraction showed the same band, suggesting that the PAg was one of the chemical componen

Surfactant lipids in the alveolar space are believed to play an important role in normal respiratory function. Although the surface-active phospholipids have been extensively studied, the possible role of glycolipids in the surfactant remains to be explored. We have studied the glycolipid composition of cell-free bronchoalveolar lavage from healthy subjects and from adult patients with respiratory distress syndrome. Glycolipids were barely detectable in bronchoalveolar lavage from healthy subjects. However, in adult respiratory distress syndrome, the amount of glycolipid relative to phospholip
