Recently, it has been well-established that the glymphatic or glial-lymphatic system plays a vital role in the pathophysiology of various neurological compromise, especially hydrocephalus (HCP). Till now, the complete pathway is not yet fully understood, and little evidence is available from the literature that links hydrocephalus to disorders of the glymphatic system. Most published molecular studies and animal research have shown that, in models with hydrocephalus, the drainage of cerebrospinal fluid (CSF) via the glymphatic system is disrupted. This is strongly observed in normal pressure a

OBJECTIVES: Premenstrual dysphoric disorder (PMDD), which is an emotional disorder characterized by symptoms of irritability, depression, anxiety, and sleep disturbances during the luteal phase of the menstrual cycle. Advanced neuroimaging findings provided insights into the neurobiological underpinnings of PMDD. However, the role of the glymphatic system and hypothalamus-related functional connectivity (FC) remains unclear. We aimed to investigate the abnormalities of glymphatic system and hypothalamus-related FC in PMDD patients.
METHODS: In this study, 23 PMDD patients and 27 healthy contr

Sleep is increasingly recognized as a fundamental regulator of brain homeostasis, yet the mechanisms linking sleep to neurological health have only recently begun to emerge. The glymphatic system, a brain-wide perivascular transport network, has provided a mechanistic framework connecting sleep physiology with brain health and disease. Accumulating evidence indicates that glymphatic activity is markedly enhanced during sleep through coordinated regulation of vasomotion, norepinephrine oscillations, aquaporin-4 polarization, extracellular space expansion, respiration, and meningeal lymphatic dr

Ischemic brain injury not only results in significant neurological, motor, and cognitive impairment but also contributes to the accumulation of toxic solutes and proinflammatory cytokines in the infarction region, exacerbating ischemic brain damage. The glymphatic system, which is crucial for brain waste clearance and homeostasis, is impaired by ischemic injury, highlighting the importance of developing therapeutic strategies for poststroke complications. Herein, a novel hybrid electro-optical stimulation device is proposed that integrates near-infrared micro-light-emitting diode with transpar

The glymphatic system, a network of perivascular channels in the brain, clears toxins and metabolic products such as amyloid-β (Aβ), supporting cognitive function and preventing neurodegenerative diseases. The gut microbiome also affects brain health and cognitive functions by producing anti-inflammatory metabolites and neurotransmitters. In this narrative review study, recently published articles from reputable scientific databases were collected and analyzed. Characteristics related to the gut microbiome, its metabolites, and their impact on the glymphatic system and ultimately cognitive fun

BACKGROUND: Blood-brain barrier damage has traditionally been considered to determine the occurrence and development of poststroke brain edema, a devastating and life-threatening complication. However, no treatment strategy targeting blood-brain barrier damage has been proven clinically effective in ameliorating brain edema.
METHODS: In mice with stroke models induced by transient middle cerebral artery occlusion (MCAO), the changes in glymphatic system (GS) function impairment were detected by ex vivo fluorescence imaging, 2-photon in vivo imaging, and magnetic resonance imaging within 1 wee

Alpha-synuclein deposits in the brain have been suspected to cause Parkinson's disease and dementia with Lewy bodies (DLB). It was recently revealed that the glymphatic system is largely responsible for the removal of alpha-synuclein. We investigated changes in the glymphatic system's activity by determining the DTI‑ALPS (diffusion tensor image analysis along the perivascular space) index in DLB patients. Twenty-six patients with DLB and 43 healthy subjects underwent diffusion tensor imaging (DTI) scanning at our hospital during the period April 2013 to March 2023. We retrospectively computed

OBJECTIVES: To investigate the recovery of human glymphatic system (GS) function in patients with temporal lobe epilepsy (TLE) after successful anterior temporal lobectomy (ATL) using diffusion tensor image analysis along the perivascular space (DTI-ALPS).
METHODS: We retrospectively analysed DTI-ALPS index in 13 patients with unilateral TLE before and after ATL, and compared the index with 20 healthy controls (HCs). Two-sample t tests and paired t tests were performed to analyse differences in the DTI-ALPS index between patients and HCs. The Pearson correlation analysis was used to observe t

Fluid transport in the perivascular space by the glia-lymphatic (glymphatic) system is important for the removal of solutes from the brain parenchyma, including peptides such as amyloid-beta which are implicated in the pathogenesis of Alzheimer's disease. The glymphatic system is highly active in the sleep state and under the influence of certain of anaesthetics, while it is suppressed in the awake state and by other anaesthetics. Here we investigated whether light sheet fluorescence microscopy of whole optically cleared murine brains was capable of detecting glymphatic differences in sleep- a

INTRODUCTION: A significant proportion of the astronauts who spend extended periods in microgravity develop ophthalmic abnormalities, including optic disc edema, optic nerve sheath distention, globe flattening, chorioretinal folds, hyperopic refractive error shifts, and nerve fiber layer infarcts. A constellation of these neuro-ophthalmic findings has been termed spaceflight-associated neuro-ocular syndrome. An increased understanding of factors contributing to this syndrome is one of the top priorities for ESA and NASA because the length of missions is expected to increase substantially in th

Postoperative cognitive dysfunction (POCD) is a common complication in surgical patients, particularly those with pre-existing chronic inflammation. Although impaired glymphatic clearance, a brain waste drainage system dependent on astrocytic aquaporin-4 (AQP4) polarization, is implicated in neurodegenerative disorders, its role in POCD pathogenesis and interaction with neuroinflammation remains unknown. Here, we investigated whetherglymphatic dysfunctiondrives postoperative neuroinflammation and cognitive deficits using a "dual-hit inflammation"model. Our results revealed glymphatic influx/eff

BACKGROUND AND PURPOSE: Cognitive impairment is a core deficit in anti-LGI1 encephalitis, yet its underlying mechanisms remain incompletely understood. The glymphatic system (GS), a brain waste clearance pathway, is implicated in cognitive dysfunction in other neuroinflammatory conditions. However, its role in anti-LGI1 encephalitis is unknown. We hypothesized that GS dysfunction is a key mechanism contributing to cognitive deficits in this disease. This study aimed to investigate GS function using diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) and other MRI indices,

BACKGROUND: Although the diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) method has been widely applied to gauge glymphatic function in temporal lobe epilepsy (TLE), glymphatic microstructure remains underexplored. Furthermore, the choroid plexus and ventricles are key elements of the brain clearance network. This study aimed to comprehensively evaluate changes in ALPS indices alongside hippocampal, choroid plexus, and lateral ventricular volumes in unilateral TLE. METHODS: We recruited 14 healthy controls (HCs) and 30 TLE patients, including 16 left TLE (LTLE) and 14

OBJECTIVES: The choroid plexus produces cerebrospinal fluid, which is crucial for glymphatic system function. Evidence suggests that changes in the volume of the choroid plexus may be associated with glymphatic system function. Therefore, this study aimed to investigate alterations in choroid plexus volume in patients with migraines compared with healthy controls.
METHODS: We enrolled 59 patients with migraines (39 and 20 with episodic and chronic migraines, respectively) and 61 healthy controls. All participants underwent brain magnetic resonance imaging, including three-dimensional T1-weigh

Defined by the World Health Organization as a neurological disorder, Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) is a disabling illness, affecting millions of people worldwide. First reported in the early nineteenth century, ME/CFS is uniquely characterized by a wide array of symptoms, including fatigue, brain fog, post-exertional malaise (PEM), sleep dysfunction, and orthostatic intolerance (OI). Despite decades of extensive research, there are no effective medical treatments or simple diagnostics for ME/CFS, with an estimated 90% of patients remaining undiagnosed. The recentl
