Interleukin-37 does not confer protection against stone formation or kidney injury at the peak of disease in a murine calcium oxalate model.

Calcium oxalate (CaOx) stones are the most prevalent form of nephrolithiasis in adults. While murine models of CaOx stone disease are established, many lack translational features for therapeutic evaluation, including in vivo assessment of kidney function and molecular profiling of renal inflammation. We refined the glyoxylate‑induced CaOx mouse model by incorporating non‑invasive transdermal glomerular filtration rate (tGFR) measurements to enable longitudinal assessment of kidney function. This platform was used to investigate the renoprotective role of the anti‑inflammatory cytokine interle

Adaption and Degradation Strategies of Methylotrophic 1,4-Dioxane Degrading Strain Xanthobacter sp. YN2 Revealed by Transcriptome-Scale Analysis.

Biodegradation of 1,4-dioxane (dioxane) contamination has gained much attention for decades. In our previous work, we isolated a highly efficient dioxane degrader, Xanthobacter sp. YN2, but the underlying mechanisms of its extraordinary degradation performance remained unresolved. In this study, we performed a comparative transcriptome analysis of YN2 grown on dioxane and citrate to elucidate its genetic degradation mechanism and investigated the transcriptomes of different dioxane degradation stages (T0, T24, T48). We also analyzed the transcriptional response of YN2 over time during which th

Three-Phasic Flow Bioreactor for Obtaining 5-Formyl-2-Furancarboxylic Acid Catalyzed by an Immobilized Laccase Using Copper Sulfate as a Stabilizing Agent.

The effect of CuSO4 on the stability of an immobilized laccase during the oxidation of HMF to FFCA was determined in a batch bioreactor and in a triphasic flow bioreactor. The enzyme was immobilized on a support functionalized with an epoxide and glyoxyl group; the latter achieved the highest immobilization yield by activity (87.5%). In batch oxidation of HMF catalyzed by laccase immobilized on a support with glyoxyl groups reached the highest conversion percentages (100%). In the biocatalyst stability, the addition of 20 mM CuSO4 allowed achieving 50% HMF conversion at the end of the third cy

The acetylcholinesterase reaction and catecholamine fluorescence in the glomus cells of rat carotid body.

The presence of both acetylcholinesterase reaction and glyoxylic acid-induced fluorescence of catecholamines in the same glomus cells of rat carotid body was demonstrated using combined histochemical methods. A suggestion is made that the glomus cells have both excitatory and inhibitory effects on the chemosensory nerve via acetylcholine and catecholamines, respectively.

Endogenous oxalate synthesis in mouse models of metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) may lead to increased urinary oxalate excretion, a risk factor for calcium oxalate kidney stone formation, and progression of chronic kidney disease. We used mouse models of diet-induced obesity to examine the impact of MASLD on endogenous oxalate synthesis and urinary oxalate excretion. Mice were either fed a high-fat diet to induce mild MASLD, or a high-fat, sucrose, and cholesterol diet (Western diet) to produce metabolic dysfunction-associated steatohepatitis (MASH), a more severe form of MASLD. Liver tissue was collected to

The catabolism of ethylene glycol by Rhodococcus jostii RHA1 and its dependence on mycofactocin.

Ethylene glycol (EG) is a widely used industrial chemical with manifold applications and also generated in the degradation of plastics such as polyethylene terephthalate. Rhodococcus jostii RHA1 (RHA1), a potential biocatalytic chassis, grows on EG. Transcriptomic analyses revealed four clusters of genes potentially involved in EG catabolism: the mad locus, predicted to encode mycofactocin-dependent alcohol degradation, including the catabolism of EG to glycolate; two GCL clusters, predicted to encode glycolate and glyoxylate catabolism; and the mft genes, predicted to specify mycofactocin bio

Trichloroethylene oxidation by toluene dioxygenase.

Trichloroethylene was oxidized by purified toluene dioxygenase obtained from recombinant E. coli strains. The major oxidation products were formic acid and glyoxylic acid. Other potential products, dichloroacetic acid, chloral, phosgene, carbon monoxide, and carbon dioxide, were not detected. [14C]trichloroethylene became covalently attached to protein components and NADPH suggesting non-specific alkylation by reactive products. Oxidation of deuterated trichloroethylene yielded 50.2% deuterated formate. Oxidation of trichloroethylene in D2O yielded 43.7% deuterated formate. These data indicate

Glyoxalase enzyme system in human muscular dystrophy.

Glyoxalase I and glyoxalase II activities were determined in skeletal muscle of control subjects and of patients with Duchenne dystrophy, other major forms of muscular dystrophies and certain neuromuscular disorders. The glyoxalase I activity was normal in all diseases examined except in Duchenne and limb girdle types of muscular dystrophy, where it showed a significant moderate decrease. The glyoxalase II activity in normal human muscle was very low, and the activity was unaltered in muscle of patients with Duchenne and other major forms of muscular dystrophies and spinal muscular atrophy. Enol ethers as carbonyl surrogates in a modification of the Povarov synthesis of 3-aryl quinolines and their anti-Toxoplasma activity.

A novel method for the preparation of 2-carboxyl-3-aryl quinoline derivatives from anilines, ethyl glyoxalate and enol ethers as phenylacetaldehyde surrogates is reported. The three-component coupling reaction occurs rapidly under mild conditions in dichloromethane catalysed by TFA. The method allows a more direct access to 3-aryl quinolines, sidestepping issues encountered with phenylacetaldehyde derivatives. This chemistry was used to prepare quinolines with 3-diarylether functionality that showed low micromolar efficacy (IC50 range: 5-26 μM) against in vitro Toxoplasma gondii coupled with l

Genome Analysis of a Variant of Streptomyces coelicolor M145 with High Lipid Content and Poor Ability to Synthetize Antibiotics.

Streptomyces coelicolor M145 is a model strain extensively studied to elucidate the regulation of antibiotic biosynthesis in Streptomyces species. This strain abundantly produces the blue polyketide antibiotic, actinorhodin (ACT), and has a low lipid content. In a process designed to delete the gene encoding the isocitrate lyase (sco0982) of the glyoxylate cycle, an unexpected variant of S. coelicolor was obtained besides bona fide sco0982 deletion mutants. This variant produces 7- to 15-fold less ACT and has a 3-fold higher triacylglycerol and phosphatidylethanolamine content than the origina

Mutants of Saccharomyces cerevisiae with defects in acetate metabolism: isolation and characterization of Acn- mutants.

The two carbon compounds, ethanol and acetate, can be oxidatively metabolized as well as assimilated into carbohydrate in the yeast Saccharomyces cerevisiae. The distribution of acetate metabolic enzymes among several cellular compartments, mitochondria, peroxisomes, and cytoplasm makes it an intriguing system to study complex metabolic interactions. To investigate the complex process of carbon catabolism and assimilation, mutants unable to grow on acetate were isolated. One hundred five Acn- ("ACetate Nonutilizing") mutants were sorted into 21 complementation groups with an additional 20 sing

Primary hyperoxaluria: insights into its clinical presentation, genetic mutations, and transplantation outcomes in a pediatric population in a tertiary care center.

BACKGROUND: Primary hyperoxaluria (PH) is a rare inherited disorder characterized by excessive oxalate accumulation in blood and urine due to defects in glyoxylate metabolism, leading to significant clinical consequences. As genetic and phenotypic heterogeneity contribute to the morbidity of PH, this study examined the phenotypes and genotypes of PH among confirmed pediatric patients (< 18 years) diagnosed with PH at a tertiary care center in Saudi Arabia between 2014 and 2023. RESULTS: Twenty-one patients from 14 families, including 10 boys (47.6%) and 11 girls (52.4%), were included. Six fa

STAT6 promoting oxalate crystal deposition-induced renal fibrosis by mediating macrophage-to-myofibroblast transition via inhibiting fatty acid oxidation.

OBJECTIVE AND DESIGN: Kidney stones commonly occur with a 50% recurrence rate within 5 years, and can elevate the risk of chronic kidney disease. Macrophage-to-myofibroblast transition (MMT) is a newly discovered mechanism that leads to progressive fibrosis in different forms of kidney disease. In this study, we aimed to investigate the role of MMT in renal fibrosis in glyoxylate-induced kidney stone mice and the mechanism by which signal transducer and activator of transcription 6 (STAT6) regulates MMT. METHODS: We collected non-functioning kidneys from patients with stones, established glyo

Synthesis of Carbo[6]helicene Derivatives Grafted with Amino or Aminoester Substituents from Enantiopure [6]Helicenyl Boronates.

Enantiopure carbo[6]helicenyl boronates were synthesized using a photocyclization reaction as the key step. These compounds were further converted to various amino derivatives using copper-catalyzed azidation or amination and reductive alkylation of benzylazide by a helicenyl dichloroborane. Asymmetric Petasis condensation with glyoxylic acid and morpholine controlled by the helical chirality afforded the corresponding amino esters.

Desilylative activation of TMSCN in chemoselective Strecker-Ugi type reaction: functional fused imidazoles as building blocks as an entry route to annulated purines.

A pathway of desilylative activation of TMSCN as a functional isonitrile equivalent, and DABCO-THF as an appropriate system for activation in a chemoselective Strecker-Ugi type reaction, has rendered ethyl glyoxalate and various heterocyclic-2-amidines as feasible substrates, and afforded the successful synthesis of 3-amino-2-carboxyethyl substituted fused imidazoles as useful building blocks. This class of functional scaffold has provided, via construction of the fused pyrimidinone motif, the synthesis of biologically important C8-N9 annulated purines, adenines and their oxo/thio analogs. Quantitative determination of CGS 26214, a cholesterol lowering agent, in human plasma using negative electrospray ionization liquid chromatography-tandem mass spectrometry.

CGS 26214 is a synthetic cholesterol-lowering agent shown to be active in the rat, dog and monkey. The present work was conducted to develop a sensitive liquid chromatography-tandem mass spectrometry (LC-MS-MS) method for quantitative determination of the compound in human plasma following clinical doses of 10-100 microg per day. A number of analytical challenges were encountered during the development of the assay. The compound was an ester and susceptible to hydrolysis under experimental conditions. A lower limit of quantitation of 50 pg/ml was needed due to the low clinical doses. Positive

Changes in ficin specificity by different substrate proteins promoted by enzyme immobilization.

Ficin extract has been immobilized using different supports: glyoxyl and Aspartic/1,6 hexamethylenediamine (Asp/HA) agarose beads. The latter was later submitted to glutaraldehyde modification to get covalent immobilization. The activities of these 3 kinds of biocatalysts were compared utilizing 4 different substrates, casein, hemoglobin and bovine serum albumin and benzoyl-arginine-p-nitroanilide at pH 7 and 5. Using glyoxyl-agarose, the effect of enzyme-support reaction time on the activity versus the four substrates at both pH values was studied. Reaction time has been shown to distort the

Oral findings associated with primary hyperoxaluria type I.

In the present paper we report the oral findings of a patient who was diagnosed with hyperoxaluria. Hyperoxalurias can basically be classified as primary and secondary, with the first being inborn errors of metabolism and the second a result of excessive oxalate intake. Primary hyperoxalurias form a rare group of metabolic diseases that are inherited in the autosomal recessive fashion. The affected genes code for specific hepatic enzymes that are involved in glyoxylate metabolism and their deficiency results in overproduction of oxalate. Two different types are described: Primary hyperoxaluria
