Map of retinal position onto the cross section of the optic pathway of goldfish.

The position of a retinal cell is defined by the two polar coordinates: r, the distance from the optic disc, and theta, the angular (or clock-face) position. Axons of similar theta value were labeled by the punctate application of horseradish peroxidase (HRP) to optic axons in the retina, and axons of similar r-value were labeled by the application of this same marker to a tectal fascicle. Labeled axons were traced in serial transverse sections of the optic pathway from the retina to the tectum to learn the map of the retinal surface onto the cross section of the pathway. Retinas were flat-mou

Activity and the formation of ocular dominance patches in dually innervated tectum of goldfish.

This study tested (1) whether blocking impulse activity in both eyes of fish with one tectum prevents the formation of ocular dominance patches, (2) whether areas receiving a high density of innervation from one eye receive a low density from the other, and (3) whether there is an electrophysiological correlate to the anatomical patches. One tectum was removed in goldfish so that the optic nerve fibers from both eyes would compete for synaptic space in the remaining tectum. The terminal arbors from the two projections initially overlapped but by 50 to 60 days segregated into ocular dominance p

Influence of GABA on the spectral and spatial coding of goldfish retinal ganglion cells.

The responses of center red ON and center red OFF ganglion cells of the isolated goldfish retina to a flickering stimulus light diminish by GABA. Without stimulation GABA blocks the maintained activity of the OFF cells. With a green flickering spot in the receptive field center, the green ON-response of the OFF cells becomes an OFF-response during GABA administration. It is concluded that, besides the common antagonistic green input, the receptive field center of these cells also has a synergistic green input, which is mediated by a pathway other than the center opponent red and green inputs.

Ganglioside changes in the regenerating goldfish optic system: comparison with glycoproteins and phospholipids.

Axonally transported radioactivity in sialoglycoconjugates, labeled by intraocular injection of [3H]N-acetylmannosamine, increased significantly during regeneration of goldfish optic axons at 30 degrees C. Ganglioside radioactivity showed the largest increase--approximately eightfold--in the optic nerve tract at 8 days after optic nerve crush while sialoglycoprotein radioactivity increased fourfold under the same conditions. As regeneration proceeded the magnitude of the increase in the nerve tract diminished for both glycoconjugates. In the optic tectum, however, transported radioactivities r

Effects of monosodium L-glutamate on pituitary innervation in goldfish, Carassius auratus.

Monosodium L-glutamate was injected intraperitoneally into goldfish at a dosage of 2.5 mg/g body wt. The first noticeable effect in the pituitary, 30 min postinjection, was marked swelling of pituicytes in the neurohypophysis (NH). Eighteen hours postinjection a large number of degenerating type B fibers were found in the NH of the proximal pars distalis (PPD), while all type A fibers, and type B fibers in other parts of the pituitary, remained unaffected. Because glutamate causes a lesion in the nucleus lateral tuberis (NLT), from the anterior margin of the pituitary stalk through to the post

Epigenetic and post-transcriptional repression support metabolic suppression in chronically hypoxic goldfish.

Goldfish enter a hypometabolic state to survive chronic hypoxia. We recently described tissue-specific contributions of membrane lipid composition remodeling and mitochondrial function to metabolic suppression across different goldfish tissues. However, the molecular and especially epigenetic foundations of hypoxia tolerance in goldfish under metabolic suppression are not well understood. Here we show that components of the molecular oxygen-sensing machinery are robustly activated across tissues irrespective of hypoxia duration. Induction of gene expression of enzymes involved in DNA methylati

Goldfish Response to Chronic Hypoxia: Mitochondrial Respiration, Fuel Preference and Energy Metabolism.

Hypometabolism is a hallmark strategy of hypoxia tolerance. To identify potential mechanisms of metabolic suppression, we have used the goldfish to quantify the effects of chronically low oxygen (4 weeks; 10% air saturation) on mitochondrial respiration capacity and fuel preference. The responses of key enzymes from glycolysis, β-oxidation and the tricarboxylic acid (TCA) cycle, and Na+/K+-ATPase were also monitored in various tissues of this champion of hypoxia tolerance. Results show that mitochondrial respiration of individual tissues depends on oxygen availability as well as metabolic fuel

Sex chromosome and sex locus characterization in goldfish, Carassius auratus (Linnaeus, 1758).

BACKGROUND: Goldfish is an important model for various areas of research, including neural development and behavior and a species of significant importance in aquaculture, especially as an ornamental species. It has a male heterogametic (XX/XY) sex determination system that relies on both genetic and environmental factors, with high temperatures being able to produce female-to-male sex reversal. Little, however, is currently known on the molecular basis of genetic sex determination in this important cyprinid model. Here we used sequencing approaches to better characterize sex determination and

Cytokine gene expression profiles in goldfish (Carassius auratus) during Gyrodactylus kobayashii infection.

Monogeneans of the genus Gyrodactylus are well-known pathogens causing huge mortalities in wild and cultured fish. Cytokine expression is one of most important host defense mechanisms against parasite infections. In this study, the expression pattern of the key pro-inflammatory (IL-1β, IL-8, IFN-γ, TNF-α, IL-12 and iNOS) and anti-inflammatory cytokine genes (IL-10, TGFβ and IL-4) of Gyrodactylus kobayashii infected goldfish (Carassius auratus) were determined by real-time quantitative PCR analysis. Our results showed that G. kobayashii infection caused increased expression of the pro-inflammat

Goldfish neurokinin B: Cloning, tissue distribution, and potential role in regulating reproduction.

Neurokinin B (NKB) is a member of the tackykinin (TAC) family known to play a critical role in the neuroendocrine regulation of reproduction in mammals. However, its biological functions in teleosts are less clear. The aim of this study was to determine the role of NKB in fish reproduction using goldfish as a model. Two transcripts, TAC3a and TAC3b, which encode several NKBs, including NKBa-13, NKBa-10, NKBb-13, and NKBb-11, were cloned. Phylogenetic analysis revealed that NKBa-10 and NKBb-11 are closely related to mammalian NKB, while NKB-13s are more conserved in teleosts. Quantitative real-

Screening Medicinal Plants for Use against Dactylogyrus intermedius (Monogenea) Infection in Goldfish.

Abstract Methanol extracts of 24 traditional medicinal plants with potential anthelmintic activity against Dactylogyrus intermedius (Monogenea) in Goldfish Carassius auratus were investigated. Abrus cantoniensis, Citrus medica, Dioscorea collettii, and Polygonum multiflorum exhibited 100% activity and were selected for further evaluation by applying five solvents (petroleum ether, chloroform, ethyl acetate, methanol, and water) for the extraction of the samples, followed by an in vivo bioassay. Among the plants tested, water, methanol, and ethyl-acetate extracts of P. multiflorum showed the hi

Antiparasitic effect of (+)-catechin derived from Pseudolarix amabilis against Dactylogyrus intermedius in goldfish.

Medicinal plants are considered promising candidates for controlling parasitic pathogen in aquaculture. Our previous study demonstrated that the crude extracts of Pseudolarix amabilis exhibit promising anti-Dactylogyrus intermedius activity. However, the specific compounds responsible for the antiparasitic effects of these crude extracts remain elusive. In this study, the bioactive compounds from the ethyl acetate extract of P. amabilis were isolated by the multi-column chromatography and in vivo bioassay-guided methods. Two crystalline compounds were identified as (+)-catechin through the nuc

Molecular cloning and functional characterization of Caspase-8 in goldfish (Carassius auratus L.).

Apoptosis, a form of programmed cell death, is essential for maintaining homeostasis within the internal environment. Caspase-8, an initiator caspase, plays a pivotal role in activating the caspase cascade during the apoptotic process. This study cloned and expressed Caspase-8 from goldfish, aimed to investigate the role of Caspase-8 in the immune response of fish to bacterial infections, specifically those caused by Aeromonas hydrophila. The Gf-Casp8 gene consists of 1425 base pairs, encoding a protein of 474 amino acids with a molecular weight of 54.55 kDa. Quantitative real-time PCR analysi

POTASSIUM CHLORIDE ADMINISTERED VIA FOUR ROUTES FOR EUTHANASIA OF ANESTHETIZED GOLDFISH (CARASSIUS AURATUS).

Immersion overdose in tricaine methanesulfonate (MS-222) is ineffective for euthanasia of adult goldfish (Carassius auratus), and investigation of alternative techniques is warranted. This study evaluated potassium chloride (KCl) administered via four routes for goldfish euthanasia. Thirty clinically healthy adult goldfish (17.2-41.9 g) were anesthetized via immersion in buffered MS-222 (300 mg/L) for 5 min beyond anesthetic induction. Fish were then randomly administered one of five treatments: KCl (333 mg/mL, 4.5 mmol/mL, 4.5 mEq/mL) via intracardiac injection at 10 mEq/kg (IC), intracoelomi

Nocturnal melatonin increases glucose uptake via insulin-independent action in the goldfish brain.

Melatonin, a neurohormone nocturnally produced by the pineal gland, is known to regulate the circadian rhythm. It has been recently reported that variants of melatonin receptors are associated with an increased risk of hyperglycemia and type 2 diabetes, suggesting that melatonin may be involved in the regulation of glucose homeostasis. Insulin is a key hormone that regulates circulating glucose levels and cellular metabolism after food intake in many tissues, including the brain. Although cells actively uptake glucose even during sleep and without food, little is known regarding the physiologi

Different patterns of chronic hypoxia lead to hierarchical adaptive mechanisms in goldfish metabolism.

Some hypoxia-tolerant species, such as goldfish, experience intermittent and severe hypoxia in their natural habitat, causing them to develop multiple physiological adaptations. However, in fish, the metabolic impact of regular hypoxic exposure on swimming performance in normoxia is less well understood. Therefore, we experimentally tested whether chronic exposure to constant (30 days at 10% air saturation) or intermittent hypoxia (3 h in normoxia and 21 h in hypoxia, 5 days a week) would result in similar metabolic and swimming performance benefits after reoxygenation. Moreover, half of the n

Population growth of Gyrodactylus kobayashii in goldfish (Carassius auratus) associated with host density.

Host density is a key regulatory factor in parasite transmission. The goldfish (Carassius auratus)-Gyrodactylus kobayashii model was used to investigate effects of host density on population growth of gyrodactylids. A donor fish infected by five gravid gyrodactylids was mixed with 11 parasite-free goldfish at five host densities. There was a significant positive correlation between host density and mean abundance of G. kobayashii throughout the 58-day experiment. During early infection (days 15–24), mean abundance in medium high (0.5 fish L−1) and high host density groups (1 and 2 fish L−1) wa

Somatostatin-28 inhibitory action on somatolactin-α and -β gene expression in goldfish.

Somatostain (SS) is known to inhibit growth hormone (GH) and prolactin (PRL) secretion. Somatolactin (SL) is a member of the GH/PRL family, but its regulation by goldfish brain somatostatin-28 (gbSS-28) has not been examined. To this end, the structural identity of goldfish SLα was established by 5'/3'-rapid amplification of cDNA ends. As revealed by in situ hybridization and immunohistochemical staining, the expression of SL isoforms was detected in pituitary cells located in the neurointermediate lobe (NIL). The transcripts of goldfish SS receptor 5a (Sst5a) but not Sst1b, Sst2, or Sst3a wer

A hindbrain segmental scaffold specifying neuronal location in the adult goldfish, Carassius auratus.

The vertebrate hindbrain develops as a series of well-defined neuroepithelial segments or rhombomeres. While rhombomeres are visible in all vertebrate embryos, generally there is not any visible segmental anatomy in the brains of adults. Teleost fish are exceptional in retaining a rhombomeric pattern of reticulospinal neurons through embryonic, larval, and adult periods. We use this feature to map more precisely the segmental imprint in the reticular and motor basal hindbrain of adult goldfish. Analysis of serial sections cut in three planes and computer reconstructions of retrogradely labeled
