Elucidating Molecular Mechanisms Governing TNF-Alpha-Mediated Regulation of Amyloid Beta 42 Uptake in Blood-Brain Barrier Endothelial Cells.

UNLABELLED: Cerebrovascular inflammation is prevalent in a majority of Alzheimer's patients. Inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-alpha), circulating in the plasma have been shown to cause the inflammation of blood-brain barrier (BBB) endothelium lining the cerebral microvasculature. The BBB inflammation has been implicated in the increase of toxic Aβ accumulation within Alzheimer's disease (AD) brain. TNF-alpha in the peripheral circulation can aggravate the accumulation of amyloid-beta (Aβ) peptides in Alzheimer's disease brain. In the current study, we have shown

HSF-1 Regulates Autophagy to Govern Motor Function and Facilitate Toxic Protein Clearance in a C. elegans Model of Amyotrophic Lateral Sclerosis.

Heat shock factor-1 (HSF-1) plays a crucial role in orchestrating stress responses across diverse organisms and disease conditions. Here, we investigate how the HSF-1 signaling pathway influences the degradation of toxic proteins and neuropathological changes in the Caenorhabditis elegans model of amyotrophic lateral sclerosis (ALS). We found that overexpressing HSF-1 improves locomotor ability and increases the survival rate of ALS C. elegans. Moreover, we observed a deceleration of motor neuron degeneration, demonstrating the protective effect of HSF-1 on neurodegenerative processes. Transcr

Mitochondrial ROS-ER Stress Axis Governs IL-10 Production in Neutrophils and Regulates Inflammation in Murine Chlamydia pneumoniae Lung Infection.

Neutrophils are among the first cells to be recruited to the lungs during Chlamydia pneumoniae infection in mouse models; however, their regulatory functions are not yet fully understood. This study examined the mechanisms and significance of IL-10-producing neutrophils throughout C. pneumoniae pulmonary infection in C57BL/6 mice. Our findings revealed that infection with C. pneumoniae induces IL-10 secretion in bone marrow-derived neutrophils, depending on Toll-like receptor 2 (TLR2) activation. This process involves TLR2-dependent mitochondrial reactive oxygen species (ROS) production, which

Integrated multi-omics profiling unveils a network of key signaling pathways governing ovarian function and systemic regulation in high-prolificacy goats.

BACKGROUND: Prolificacy is a crucial economic trait in goat production, yet its underlying molecular mechanisms remain incompletely understood due to its polygenic nature. While previous studies have identified several candidate genes, a comprehensive understanding of the local and systemic regulatory networks is lacking. This study aims to dissect the complex molecular basis of high prolificacy in goats through an integrated multi-omics approach. RESULTS: We conducted transcriptomic, proteomic, and metabolomic profiling of ovarian tissues from high-fecundity (HF, n = 3) and low-fecundity (LF

DR5 Governs Compound Exocytosis in Colonic Goblet Cells via TATA-Box Binding Protein-Dependent Bestrophin-2 Transcriptional Regulation.

The colonic mucus barrier, dependent on goblet cell-secreted mucin2 (Muc2), prevents microbial invasion. Compound exocytosis enables the rapid, high-volume release of mucus from goblet cells; its underlying mechanisms, however, remain unclear. Here, death receptor 5 (DR5) is identified as a critical regulator of this process. Knockout of DR5 exhibits a thinner mucus layer despite elevated Muc2 and enlarged granules, causing mild dysbiosis and enhances susceptibility to Citrobacter rodentium. DR5 knockout and its ligand TRAIL knockdown inhibits carbachol-triggered goblet cell compound exocytosi

Cullin-associated and neddylation-dissociated 1 protein (CAND1) governs cardiac hypertrophy and heart failure partially through regulating calcineurin degradation.

Pathological cardiac hypertrophy is a process characterized by significant disturbance of protein turnover. Cullin-associated and Neddylation-dissociated 1 (CAND1) acts as a coordinator to modulate substrate protein degradation by promoting the formation of specific cullin-based ubiquitin ligase 3 complex in response to substrate accumulation, which thereby facilitate the maintaining of normal protein homeostasis. Accumulation of calcineurin is critical in the pathogenesis of cardiac hypertrophy and heart failure. However, whether CAND1 titrates the degradation of hypertrophy related protein e

COVID-19 water, sanitation, and hygiene response: Review of measures and initiatives adopted by governments, regulators, utilities, and other stakeholders in 84 countries.

The COVID-19 pandemic has shone a light on handwashing as an inexpensive, widely applicable response measure. In consequence, most governments have taken action to promote access to water and sanitation services for all. This paper documents an overview of initiatives and interventions that countries have implemented during the first months of the COVID-19 response. Initiatives have been identified across 84 countries worldwide, and categorized into those that aimed at securing water, sanitation, and hygiene (WASH) for all, and those that sought to provide technical and financial support to se

The ferric Iron uptake transcriptional regulator (Fur) governs virulence and stress responses in Listeria monocytogenes and serves as a target for novel food safety interventions.

Listeria monocytogenes is a major foodborne pathogen whose environmental persistence and virulence pose significant threats to food safety. The ferric uptake regulator (Fur) is essential for iron homeostasis; however, its role in the acid and oxidative stress responses, pathogenicity and its potential as a target for food safety interventions remain poorly understand. In this study, a Δfur mutant was constructed to investigate these questions. Deletion of fur severely impaired survival under acidic stress and disrupted iron homeostasis, resulting in intracellular Fe2+ accumulation. The mutant

Regulation of Ptbp1-controlled alternative splicing of pyruvate kinase muscle by liver kinase B1 governs vascular smooth muscle cell plasticity in vivo.

AIMS: Vascular smooth muscle cell (VSMC) plasticity is a state in which VSMCs undergo phenotypic switching from a quiescent contractile phenotype into other functionally distinct phenotypes. Although emerging evidence suggests that VSMC plasticity plays critical roles in the development of vascular diseases, little is known about the key determinant for controlling VSMC plasticity and fate. METHODS AND RESULTS: We found that smooth muscle cell-specific deletion of Lkb1 in tamoxifen-inducible Lkb1flox/flox;Myh11-Cre/ERT2 mice spontaneously and progressively induced aortic/arterial dilation, an

The accumulation of organotins in adult and seed oysters from selected estuaries prior to the introduction of U.K. regulations governing the use of tributyltin-based antifouling paints.

Between April and November, 1985, a study of the levels of organotins in adult and seed Crassostrea gigas grown in the southwest and southeast of England was carried out. This work corresponds with a similar study of organotin water levels (reported separately). The analytical procedure used was directly coupled high performance liquid chromatography-flame atomic absorption spectrometry. Individual adult oysters and groups of 20-40 seed oysters were taken for analysis on a monthly basis. The profiles of tributyltin (TBT), dibutyltin (DBT) and monobutyltin (MBT) are discussed in relation to eac

Fission Yeast SCYL1/2 Homologue Ppk32: A Novel Regulator of TOR Signalling That Governs Survival during Brefeldin A Induced Stress to Protein Trafficking.

Target of Rapamycin (TOR) signalling allows eukaryotic cells to adjust cell growth in response to changes in their nutritional and environmental context. The two distinct TOR complexes (TORC1/2) localise to the cell's internal membrane compartments; the endoplasmic reticulum (ER), Golgi apparatus and lysosomes/vacuoles. Here, we show that Ppk32, a SCYL family pseudo-kinase, is a novel regulator of TOR signalling. The absence of ppk32 expression confers resistance to TOR inhibition. Ppk32 inhibition of TORC1 is critical for cell survival following Brefeldin A (BFA) induced stress. Treatment of

HIV-infection and cocaine use regulate semen extracellular vesicles proteome and miRNAome in a manner that mediates strategic monocyte haptotaxis governed by miR-128 network.

BACKGROUND: Extracellular vesicles (EVs) are regulators of cell-cell interactions and mediators of horizontal transfer of bioactive molecules between cells. EV-mediated cell-cell interactions play roles in physiological and pathophysiological processes, which maybe modulated by exposure to pathogens and cocaine use. However, the effect of pathogens and cocaine use on EV composition and function are not fully understood. RESULTS: Here, we used systems biology and multi-omics analysis to show that HIV infection (HIV +) and cocaine (COC) use (COC +) promote the release of semen-derived EVs (SEV)

Elucidating molecular mechanisms governing tumor necrosis factor-alpha-mediated regulation of amyloid beta 42 uptake in blood-brain barrier endothelial cells.

Cerebrovascular inflammation is prevalent in a majority of patients with Alzheimer disease. Elevated levels of inflammatory cytokines, such as tumor necrosis factor-α (TNF-α), circulating in the plasma have been shown to cause the inflammation of blood-brain barrier (BBB) endothelium lining the cerebral microvasculature in Alzheimer disease. The BBB inflammation triggered by exposure of TNF-α in the peripheral circulation can aggravate the accumulation of Aβ peptides in Alzheimer disease brain. In the current study, we have shown that infusion of wild-type mice with TNF-α led to an increase in

A modern reinterpretation of the "kidney governs bone" theory: systemic regulation of the neuro-endocrine-immune network via the brain-bone axis and traditional chinese medicine intervention strategies.

The traditional Chinese medicine (TCM) theory of "Kidney Governs Bone' has been progressively validated in modern medical research. However, a purely endocrinological perspective cannot fully explain its regulatory implications under complex conditions such as stress, inflammation, and nerve injury. As bone metabolism research shifts toward a systemic regulatory paradigm, the "brain-bone axis' provides a new framework for reinterpreting "Kidney Governs Bone.' Based on current evidence, the core scientific proposition of this article is that we propose the hypothesis that the therapeutic effect

Regulation of somatic stem cell development through positional and proliferative signals during Drosophila melanogaster pupal ovary development resembles the framework governing adult stem cell behavior.

Follicle stem cells (FSCs) in the Drosophila melanogaster ovary are maintained through independent regulation of division and differentiation to become proliferative follicle cells (FCs) to their posterior or quiescent escort cells (ECs) to their anterior. These behaviors are guided by graded extracellular Hedgehog (Hh) and Wnt signals emanating from cells anterior to FSCs and an inverse gradient of JAK-STAT pathway activity. Here, we used lineage analyses to investigate regulation of the development of ECs, FSCs, and FCs from a common set of precursors during pupation. Previous studies found

The Key Role of Experiential Uncertainty when Dealing with Risks: Its Relationships with Demand for Regulation and Institutional Trust.

The results of a survey and an experiment show that experiential uncertainty-people's experience of uncertainty in risk contexts-plays a moderating role in individuals' risk-related demand for government regulation and trust in risk-managing government institutions. First, descriptions of risks were presented to respondents in a survey (N = 1,017) and their reactions to questions about experiential uncertainty, risk perception, and demand for government regulation were measured, as well as levels of risk-specific knowledge. When experiential uncertainty was high, risk perceptions had a positiv
