Distinct expression of perforin and granzyme B in lip and oral cavity squamous cell carcinoma.

BACKGROUND: Perforin and granzyme B (GB) are the main constituents of cytotoxic T-lymphocyte granules, and they have important roles in preventing the initiation and progression of cancer. METHODS: The aim of this study was to compare the expression of CD8(+) /perforin(+) double-staining and GB(+) cells, by immunohistochemistry, in primary oral cavity squamous cell carcinoma (OCSCC), lip squamous cell carcinoma (LSCC), non-dysplastic leukoplakia (LK), dysplastic LK, actinic cheilitis (AC), oral lichen planus (LP) and normal oral mucosa. RESULTS: Our results showed a higher expression of CD8(

Granzyme B produced by human plasmacytoid dendritic cells suppresses T-cell expansion.

Human plasmacytoid dendritic cells (pDCs) are crucially involved in the modulation of adaptive T-cell responses in the course of neoplastic, viral, and autoimmune disorders. In several of these diseases elevated extracellular levels of the serine protease granzyme B (GrB) are observed. Here we demonstrate that human pDCs can be an abundant source of GrB and that such GrB(+) pDCs potently suppress T-cell proliferation in a GrB-dependent, perforin-independent manner, a process reminiscent of regulatory T cells. Moreover, we show that GrB expression is strictly regulated on a transcriptional leve

The granzyme B inhibitor proteinase inhibitor 9 (PI9) is expressed by human mast cells.

The activity of granzyme B, a main effector molecule of cytotoxic T lymphocytes (CTL) and natural killer cells, is regulated by the human intracellular serpin proteinase inhibitor 9 (PI9). This inhibitor is particularly expressed by CTL and dendritic cells, in which it serves to protect these cells against endogenous and locally released granzyme B. Moreover, PI9 expression by neoplastic cells may constitute one of the mechanisms for tumors to escape immune surveillance. Here we show that PI9 is also expressed by human mast cells. In immunohistochemical studies using a PI9-specific monoclonal

Granzyme B and TIA-1 expression in chronic and acute on chronic renal allograft rejection.

Although active inflammation may be deleterious and indicate immunologic activation in chronically rejected grafts, the underlying mechanism of tissue destruction has been little studied. Twenty-four cases of chronic rejection (CR) with or without acute rejection (AR) were stained with antibodies against CD3, CD8, CD68, granzyme B and TIA-1, and the number of positive cells were counted. Eleven cases of AR served as controls. The number of CD3 and CD8 positive cells increased in the acute on CR group compared to the CR group. About a half of CD3 positive T cells were CD8 positive in both group

Cytotoxic T lymphocyte-inspired nanotherapeutics chaperoning granzyme B for tumor immune remodeling.

T cell-based cancer immunotherapies have transformed oncology; however, their clinical benefit remains limited in immunologically cold tumors, where poor T cell infiltration and impaired effector function weaken endogenous tumor cell killing. Here, we developed GrB@A-nChap, a cytotoxic T lymphocyte (CTL)-inspired nanotherapeutic that chaperones granzyme B (GrB) into the tumor cell cytosol to supplement deficient cytotoxic effector activity in "cold" tumors. Mechanistically, GrB@A-nChap protects GrB under physiological conditions, undergoes partial protonation in the mildly acidic tumor microen

Distribution of granzyme-expressing NK cells in tuberculosis reflects subset and compartment-specific remodeling.

INTRODUCTION: Natural killer (NK) cells contribute to immunity against Mycobacterium tuberculosis (Mtb), yet their granzyme expression and subset distribution in TB remain poorly defined. METHODS: NK cell subsets and the expression of granzymes (GZMA, GZMB, and GZMK) and CCR5 were analyzed by multiparametric flow cytometry in peripheral blood from healthy controls, individuals with latent TB infection, active TB patients, and treated TB patients, as well as in paired pleural fluid samples. RESULTS: In peripheral blood from active TB patients, NK cells exhibited reduced co-expression of GZMA,

Granzyme B secreted by T cells is involved in anti-bacterial immune response of tilapia.

Secreted by natural killer cells and cytotoxic T lymphocytes, Granzyme B is involved in regulating the adaptive immune response in vertebrates and plays a pivotal role in resisting virus invasion and removing pathogens. Although it had been extensively studied in mammals, the involvement of Granzyme B in adaptive immune response of early vertebrates remained elusive. In this study, we investigated the Granzyme B in Oreochromis niloticus (OnGrB), found that its function domain was conserved. Additionally, OnGrB was widely expressed in various tissues and could respond to T-cell activation in vi

Cytotoxic T Cell-Derived Granzyme B Is Increased in Severe Plasmodium Falciparum Malaria.

In Plasmodium falciparum malaria, CD8+ T cells play a double-edged role. Liver-stage specific CD8+ T cells can confer protection, as has been shown in several vaccine studies. Blood-stage specific CD8+ T cells, on the other hand, contribute to the development of cerebral malaria in murine models of malaria. The role of CD8+ T cells in humans during the blood-stage of P. falciparum remains unclear. As part of a cross-sectional malaria study in Ghana, granzyme B levels and CD8+ T cells phenotypes were compared in the peripheral blood of children with complicated malaria, uncomplicated malaria, a

Impairment of Granzyme B-Producing Regulatory B Cells Correlates with Exacerbated Rheumatoid Arthritis.

Hyperactivated B cells have been demonstrated the contribution to the development of rheumatoid arthritis (RA). While the recognition of the negative regulatory function of B cells further promoted our understanding of their pathogenic role in RA. Recently, a new population of granzyme B (GrB)-producing B cells was identified, which was proved to be involved in cancer and infectious diseases. However, their characteristics and roles in RA remain to be elucidated. In the present study, we aim to further characterize whether B cells could produce GrB and reveal their potential role in the pathog

The role of serpinb9/serine protease inhibitor 6 in preventing granzyme B-dependent hepatotoxicity.

UNLABELLED: Virally infected hepatocytes are resistant to cytotoxic lymphocyte killing by perforin-dependent and granzyme-dependent effector mechanisms. The present studies were designed to examine the role of serine protease inhibitor 6 (SPI-6) in limiting granzyme B-dependent cytotoxic effector mechanisms in the liver. SPI-6-specific small interfering RNA (siRNA) administration to C57Bl/6J (B6) mice elicited transient alanine aminotransferase (ALT) elevations that were not observed in either granzyme B-deficient B6 (B6.gzmb(-/-)) or natural killer (NK) cell-depleted B6 mice. When SPI-6 expre

Differential regulation of granzyme and perforin in effector and memory T cells following smallpox immunization.

Primary immunization of healthy adults with vaccinia virus induces a local vesicle or "take" in the majority of vaccinees that previously has been shown to correlate with protection against smallpox. However, the immunologic mechanisms underlying this protective response in humans are not well characterized. We have studied human CD8+ T cells for the expression patterns of phenotypic markers and cytolytic effector molecules before and after primary smallpox immunization using nine-color polychromatic flow cytometry. One month after immunization, vaccinees developed vaccinia virus-specific CD8+

Immunoexpression of perforin and granzyme B on infiltrating lymphocytes in human renal acute allograft rejection.

Graft destruction can be effected by direct cell-to-cell contact between activated effector T cell and a target graft resulting in delivery of cytotoxic molecules. Perforin and granzyme B can be used as activation markers for cytotoxic cells in allograft tissue. The aim of the study was to determine the immunoexpression of perforin and granzyme B by immune cells infiltrating renal tissue during acute allograft rejection and to evaluate any correlation between the phenotype of infiltrating lymphocytes and cells expressing cytotoxic granules as well as the severity of graft damage as defined by

Perforin and granzyme B lytic protein expression during chronic viral and autoimmune hepatitis.

BACKGROUND: Cytotoxic T lymphocytes and Kupffer cells are essential components of the immune response during liver diseases. Recent studies have highlighted the role of cytotoxic T lymphocytes using Fas and its ligand in induced hepatocyte death during acute and chronic hepatitis. METHODS: In the present work, the main purpose was to investigate perforin and granzyme B expression in liver biopsies of patients with chronic hepatitis (10 HBV, 14 HCV and 10 autoimmune hepatitis) using immunohistochemistry. The liver biopsies of two normal individuals were also studied in the same conditions. RE

Granzyme A is critical for recovery of mice from infection with the natural cytopathic viral pathogen, ectromelia.

Cytolytic lymphocytes are of cardinal importance in the recovery from primary viral infections. Both natural killer cells and cytolytic T cells mediate at least part of their effector function by target cell lysis and DNA fragmentation. Two proteins, perforin and granzyme B, contained within the cytoplasmic granules of these cytolytic effector cells have been shown to be directly involved in these processes. A third protein contained within these granules, granzyme A, has so far not been attributed with any biological relevance. Using mice deficient for granzyme A, we show here that granzyme A

Tissue Levels of MMP-9 and Granzyme B in Patients With Bullous Pemphigoid: A Case-Control Study.

BACKGROUND: Proteolytic enzymes, including matrix metalloproteinase-9 (MMP-9) and granzyme B, contribute to dermal-epidermal separation, but their expression in human skin lesions is not fully quantified. We aimed to quantify tissue levels of MMP-9 and granzyme B in bullous pemphigoid (BP) lesional skin compared to healthy controls. METHODS: In a case-control study at the National Hospital of Dermatology and Venereology in Hanoi, Vietnam, between June 2024 and June 2025, lesional skin from BP patients and normal skin from healthy controls were analyzed. Matrix metalloproteinase-9 (MMP-9) and

Granzyme A-expressing Terminal Effector T Cells Dedifferentiate into Long-lived Memory T Cells.

Memory T cells are critical for vaccines and T-cell-based therapies. However, the differentiation path of memory T cells is not completely understood. Early bulk cell population-based studies support a linear model, in which effector T cells lose the effector program and dedifferentiate into memory T cells. Recent research supports a bifurcation model, where T cells choose one of the two paths, either the effector or the memory path, at an early stage of T cell responses. Terminal effector T cells lose the capacity to become memory T cells. Here, we first demonstrate that the expression of gra

Granzyme B as a potential biological target in toxic encephalopathy: A big data-based exploratory analysis.

Toxic encephalopathy is a central nervous system disorder caused by endogenous or exogenous toxic substances. Granzyme B (GZMB), a key serine protease, plays a crucial role in immune regulation and the progression of various diseases. Abnormal expression of GZMB may contribute to the development and progression of toxic encephalopathy; however, its specific mechanisms remain unclear. This study aims to preliminarily explore the association between granzyme B and toxic encephalopathy through big data. This study retrieved the toxic encephalopathy dataset GSE253309 from the gene expression omnib
