Growth and development of a new subspecialty: pediatric hepatology.

Several major forces converged to catalyze the formal emergence of a body of knowledge and an organized focus on disorders of the liver in early life. Attendant to the development of a focused clinical subspecialty the pace of patient- and laboratory-based research in the field quickened in parallel to decipher the consequences of genetic or metabolic aberrations on immature liver structure and function. The key research observations that catalyzed the emergence and subsequent rapid growth of Pediatric Hepatology include: (1) an understanding of the dynamic events occurring during hepatobiliar

The role of iodine in human growth and development.

Iodine is an essential component of the hormones produced by the thyroid gland. Thyroid hormones, and therefore iodine, are essential for mammalian life. Iodine deficiency is a major public health problem; globally, it is estimated that two billion individuals have an insufficient iodine intake. Although goiter is the most visible sequelae of iodine deficiency, the major impact of hypothyroidism due to iodine deficiency is impaired neurodevelopment, particularly early in life. In the fetal brain, inadequate thyroid hormone impairs myelination, cell migration, differentiation and maturation. Mo

Stem cell trafficking in tissue development, growth, and disease.

Regulated movement of stem cells is critical for organogenesis during development and for homeostasis and repair in adulthood. Here we analyze the biological significance and molecular mechanisms underlying stem cell trafficking in the generation of the germline, and the generation and regeneration of blood and muscle. Comparison across organisms and lineages reveals remarkable conservation as well as specialization in homing and migration mechanisms used by mature leukocytes, adult and fetal stem cells, and cancer stem cells. In vivo trafficking underpins the successful therapeutic applicatio

Multiple roles for neurofibromin in skeletal development and growth.

Neurofibromatosis type 1 (NF1) is a prevalent genetic disorder primarily characterized by the formation of neurofibromas, café-au-lait spots and freckling. Skeletal abnormalities such as short stature or bowing/pseudarthrosis of the tibia are relatively common. To investigate the role of the neurofibromin in skeletal development, we crossed Nf1flox mice with Prx1Cre mice to inactivate Nf1 in undifferentiated mesenchymal cells of the developing limbs. Similar to NF1 affected individuals, Nf1(Prx1) mice show bowing of the tibia and diminished growth. Tibial bowing is caused by decreased stabilit

Choroid plexus cyst development and growth following ventricular shunting.

Choroid plexus cysts are typically incidental, asymptomatic cysts. They have been reported to hemorrhage and grow, causing symptoms of obstruction. However, growth and multiplication has not been reported following ventriculoperitoneal shunt procedures. A 66-year-old woman initially underwent a suboccipital retrosigmoid craniotomy for resection of a large petroclival meningioma. Preoperatively, the patient had hydrocephalus. After surgery the patient required a ventriculoperitoneal shunt. Two years after the initial shunting procedure, imaging demonstrated significant growth of new bilateral c

The effect of postnatal steroids on growth and development.

Single courses of prenatal corticosteroids have many beneficial effects including reduction in respiratory distress syndrome, intraventricular hemorrhage and neonatal mortality. Postnatal corticosteroids, however, have both beneficial and harmful effects. The beneficial effects include shortened time on the ventilator and a reduction in chronic lung disease. Dexamethasone treatment started within the first 4 days of life is associated with an increased risk of abnormal neurodevelopment including cerebral palsy. Other adverse effects include metabolic disturbances, cardiac hypertrophy, reduced

Nutrients, growth, and the development of programmed metabolic function.

For each individual, the genetic endowment at conception sets the limits on the capacity or metabolic function. The extent to which this capacity is achieved or constrained is determined by the environmental experience. The consequences of these experiences tend to be cumulative throughout life and express themselves phenotypically as achieved growth and body composition, hormonal status and the metabolic capacity for one or other function. At any time later in life the response to an environmental challenge, such as stress, infection or excess body weight is determined by an interaction among

Growth, development and differentiation: a functional food science approach.

Few other aspects of food supply and metabolism are of greater biological importance than the feeding of mothers during pregnancy and lactation, and of their infants and young children. Nutritional factors during early development not only have short-term effects on growth, body composition and body functions but also exert long-term effects on health, disease and mortality risks in adulthood, as well as development of neural functions and behaviour, a phenomenon called 'metabolic programming'. The interaction of nutrients and gene expression may form the basis of many of these programming eff

Growth and pubertal development in survivors of childhood cancer.

Linear growth and final adult stature in survivors of childhood cancer may be affected adversely by the disease itself as well as by the treatments utilized. The endocrine causes of impaired growth, which include growth hormone deficiency, primary thyroid dysfunction, and premature sexual maturation, are generally the consequence of radiation therapy. Growth failure is most prominent following craniospinal irradiation for brain tumors and total body irradiation for bone marrow transplantation. Pubertal development in both males and females is generally unaffected by chemotherapy. Leydig cell f

Glucocorticoid action on the growth and development of insects.

Cortisol increased growth and differentiation in the large milkweed insect (Oncopeltus fasciatus). The glucocorticoid significantly increased the growth of the insect as analyzed by wet and dry weights. Cortisol also stimulated the development of the insects over that of the controls during the six day bioassay.

Clinical presentation, growth, and pubertal development in Addison's disease.

The clinical course of eight boys and six girls with Addison's disease has been reviewed. Adrenal antibodies were found in five boys and five girls, and four children showed clinical evidence of other autoimmune disease (hypoparathyroidism (three); diabetes (one)). The presentation was insidious in 12 children but acute in two. On treatment, linear growth was normal and, with the exception of one girl with theca cell antibodies, pubertal development proceeded normally in the older patients.

Growth and development in sickle cell anemia. Preliminary report.

Historically, adults with sickle cell anemia were described as being short, thin, and eunuchoid in appearance with a particular body habitus. More current investigations in children have suggested decreased height, weight, and hypogonadism although Jamaican studies suggest supranormal heights in adolescence. All studies to date have evaluated children at one point in time. We evaluated children with sickle cell anemia longitudinally at six monthly intervals over 3 years to assess somatic growth and the development of sexual maturation. Our data support reduced height and weight as compared to

Nutrition and development: other micronutrients' effect on growth and cognition.

Previous studies have provided strong evidence that protein-energy malnutrition, deficiencies of iron and iodine early on in life led to compromised growth and cognitive functions in young children. Recently, a growing body of knowledge indicated that other micronutrients also play a role in young child development. Severe as well as marginal vitamin A efficiency were shown to lead to an increased risk of morbidity and mortality in children. A meta-analysis of vitamin A intervention trials indicated an overall reduction of 23% in all-cause mortality rate. Delayed growth, especially stunting, w

Growth and development of the tibia, fibula, and ankle joint.

Deformities of the leg and ankle may result from growth abnormalities of the tibia and fibula. Measurements of the distance between the proximal tibial and fibular physes, the distal tibial and fibular physes, and the angle between the long axis of the tibia and the distal tibial physis and talar dome were made to determine the changes that occur during normal skeletal development. These measurements demonstrate that early detection of growth discrepancy is possible.

Growth Hormone and IGF1 Actions in Kidney Development and Function.

Growth hormone (GH) exerts multiple effects on different organs including the kidneys, either directly or via its main mediator, insulin-like-growth factor-1 (IGF-1). The GH/IGF1 system plays a key role in normal kidney development, glomerular hemodynamic regulation, as well as tubular water, sodium, phosphate, and calcium handling. Transgenic animal models demonstrated that GH excess (and not IGF1) may lead to hyperfiltration, albuminuria, and glomerulosclerosis. GH and IGF-1 play a significant role in the early development of diabetic nephropathy, as well as in compensatory kidney hypertroph
