Nano drug delivery systems: a new paradigm for treating metal toxicity.

INTRODUCTION: The standard medical treatment for metal toxicity is chelation therapy. Chelating agents work by forming less toxic complexes with the toxic metal ions which are readily excreted from the body. These compounds, based on their hydrophilic/lipophilic property, can either remove toxic metal ions from extracellular sites or can penetrate the intracellular compartments to facilitate the removal of toxic metal ions. However, there are various disadvantages associated with this kind of therapy, notably, selectivity. Other problems and challenges are that the therapy regime is expensive,

Association and mediation analyses among multiple metal exposure, mineralocorticoid levels, and serum ion balance in residents of northwest China.

Toxic metals are vital risk factors affecting serum ion balance; however, the effect of their co-exposure on serum ions and the underlying mechanism remain unclear. We assessed the correlations of single metal and mixed metals with serum ion levels, and the mediating effects of mineralocorticoids by investigating toxic metal concentrations in the blood, as well as the levels of representative mineralocorticoids, such as deoxycorticosterone (DOC), and serum ions in 471 participants from the Dongdagou-Xinglong cohort. In the single-exposure model, sodium and chloride levels were positively corre

Molecular Mechanisms of Cellular Injury and Role of Toxic Heavy Metals in Chronic Kidney Disease.

Chronic kidney disease (CKD) is a progressive disease that affects millions of adults every year. Major risk factors include diabetes, hypertension, and obesity, which affect millions of adults worldwide. CKD is characterized by cellular injury followed by permanent loss of functional nephrons. As injured cells die and nephrons become sclerotic, remaining healthy nephrons attempt to compensate by undergoing various structural, molecular, and functional changes. While these changes are designed to maintain appropriate renal function, they may lead to additional cellular injury and progression o

Novel oral detoxification of mercury, cadmium, and lead with thiol-modified nanoporous silica.

We have developed a thiol-modified nanoporous silica material (SH-SAMMS) as an oral therapy for the prevention and treatment of heavy metal poisoning. SH-SAMMS has been reported to be highly efficient at capturing heavy metals in biological fluids and water. Herein, SH-SAMMS was examined for efficacy and safety in both in vitro and in vivo animal models for the oral detoxification of heavy metals. In simulated gastrointestinal fluids, SH-SAMMS had a very high affinity (Kd) for methyl mercury (MeHg(I)), inorganic mercury (Hg(II)), lead (Pb(II)), and cadmium (Cd(II)) and was superior to other SA

Use of the molecular adsorbent recirculating system (MARS™) for the management of acute poisoning with or without liver failure.

INTRODUCTION: There is an increasing interest in recent developments in bioartificial and non-bioartificial devices, so called extracorporeal liver assist devices, which are now used widely not only to increase drug elimination, but also to enhance the removal of endogenous substances in acute liver failure. Most of the non-bioartificial techniques are based on the principle of albumin dialysis. The objective is to remove albumin-bound substances that could play a role in the pathophysiology of acute liver failure by dialysing blood against an albumin-containing solution across a high flux per

Toxicology of cadmium and its damage to mammalian organs.

The detrimental health effects of cadmium (Cd) were first described in the mid 19th century. As part of industrial developments, increasing usage of Cd has led to widespread contamination of the environment that threatens human health, particularly today. Rather than acute, lethal exposures, the real challenge in the 21st century in a global setting seems to be chronic low Cd exposure (CLCE), mainly from dietary sources. Ubiquity of Cd makes it a serious environmental health problem that needs to be thoroughly assessed because it already affects or will affect large proportions of the world's

Polysaccharides as Protective Agents against Heavy Metal Toxicity.

Polysaccharides are functional foods or drugs that can be used to alleviate heavy metal poisoning by cadmium, lead, mercury, and arsenic. Industries generate substantial quantities of toxic heavy metal wastes, such as wastewater discharges, paints, electronic waste, batteries, pigments, and plastics, into the environment that pose a risk to human health. Therefore, it is imperative to eliminate accumulated heavy metal ions from the body and the environment. Heavy metal toxicity can lead to decreased energy levels and impair the functioning of vital organs, such as the brain, lungs, kidneys, li

Successful treatment of acute oral copper sulfate poisoning: a case report.

The incidence of copper sulfate poisoning varies across different regions. It is relatively rare in Western countries but more prevalent in South Asian nations, accounting for approximately 40% of all heavy metal poisoning cases. The majority of affected individuals are rural residents, with a mortality rate ranging from 14% to 36%. Copper sulfate poisoning often leads to gastrointestinal corrosion, intravascular hemolysis, hemolytic anemia, methemoglobinemia, and acute multi-organ dysfunction, often resulting in death due to delayed treatment. In June 2024, we successfully managed a case of a

Practical screening of mercury contamination in fish tissue.

The increased release of heavy metals over the last century poses an unknown detriment to our ecosystem. Already. poisoning by toxic heavy metals has been documented in a number of species, including man. Estimation of the toxicological context of this release requires screening methods that rapidly process large numbers of samples with minimal cost, effort and ecological impact. We now describe a practical colorimetric kit to quantify mercuric ion in tissue, and demonstrate its application to screen fish. Advantageously, this test can easily be amended for field use and catch-and-release prog

Susceptibility to Environmental Heavy Metal Toxicity among Americans with Kidney Disease.

BACKGROUND: The consequences of low levels of environmental heavy metal exposure, as found widely in the United States, in those with impaired renal function remain underexplored. METHODS: We examined the cross-sectional association of indices of renal function with lead and cadmium levels in blood and urine among National Health and Nutrition Examination Survey (NHANES) participants. We used the 1999-2002 cycle, which included measures of cystatin C, in order to quantify renal function most precisely and defined chronic kidney disease (CKD) as an estimated glomerular filtration rate (eGFR) <

A Review on Coordination Properties of Thiol-Containing Chelating Agents Towards Mercury, Cadmium, and Lead.

The present article reviews the clinical use of thiol-based metal chelators in intoxications and overexposure with mercury (Hg), cadmium (Cd), and lead (Pb). Currently, very few commercially available pharmaceuticals can successfully reduce or prevent the toxicity of these metals. The metal chelator meso-2,3-dimercaptosuccinic acid (DMSA) is considerably less toxic than the classical agent British anti-Lewisite (BAL, 2,3-dimercaptopropanol) and is the recommended agent in poisonings with Pb and organic Hg. Its toxicity is also lower than that of DMPS (dimercaptopropane sulfonate), although DMP

Metallic mercury vapour poisoning revisited.

Mercury poisoning was once common in the 19th century. With its declining use, now clinicians and the public in general are often unaware and unsuspecting of mercury toxicity. A 40-year-old woman and her two children were hospitalized with a 1-week history of a generalized lichenoid eruption. Clinical improvement occurred without a diagnosis; however, on returning home, features of acrodynia with digital gangrene developed in the woman, leading to suspicion of heavy metal poisoning. There was no recurrence after moving from their contaminated house.

Soil criteria to protect terrestrial wildlife and open-range livestock from metal toxicity at mining sites.

Thousands of hard rock mines exist in the western USA and in other parts of the world as a result of historic and current gold, silver, lead, and mercury mining. Many of these sites in the USA are on public lands. Typical mine waste associated with these sites are tailings and waste rock dumps that may be used by wildlife and open-range livestock. This report provides wildlife screening criteria levels for metals in soil and mine waste to evaluate risk and to determine the need for site-specific risk assessment, remediation, or a change in management practices. The screening levels are calcula

Severe Abdominal Pain as a Presentation of Lead Poisoning: A Case Presentation.

BACKGROUND: Lead poisoning is a rare but serious disease. The clinical manifestations of lead poisoning are various and nonspecific, such as abdominal pain, headache, dizziness, nightmare, fatigue and so on. Rapid diagnosis of lead poisoning is challenging because it does not have special symptoms and the morbidity is very low. CASE PRESENTATION: A 31-year-old woman presented with epigastric discomfort without any obvious cause. The patient was diagnosed with lead poisoning, as the blood levels of heavy metals were detected and the lead was 463.17 µg/L, which was very high (normal value was l

Chronic arsenic poisoning following ayurvedic medication.

BACKGROUND: Ayurveda, Indian traditional system of medicine, is practiced commonly in South East Asia and in many parts of the world. Many ayurvedic drugs contain heavy metals and may lead to metal toxicity. Of these, chronic lead poisoning is the most common. Chronic arsenic poisoning following the use of ayurvedic medication, though reported, is rare. CASE REPORTS: We describe three patients who presented with features of chronic arsenic poisoning following prolonged ayurvedic medication use. The diagnosis of chronic arsenic poisoning was confirmed by high arsenic levels in the blood, urine

Industrial chemicals and the horse.

Poisoning resulting from exposure to a wide variety of industrial chemicals is not a common occurrence in horses, but it does happen on occasion. A wide range of toxicosis can occur from a wide range of industrial pollutants, such as dioxin, carbon tetrachloride, and tetrachloroethylene, to heavy metals, such as cadmium and zinc. The equine practitioner must consider industrial chemical toxicosis in differential diagnoses and work with a reputable veterinary diagnostic laboratory to confirm or rule out industrial chemical poisoning.

Research Progress and Prospects on Poisoning Mechanism and Anti-Poisoning Modification of Cerium-Based NH3-SCR Denitrification Catalysts.

Ammonia selective catalytic reduction (NH3-SCR) has become the mainstream core technology for denitrification of industrial sintering flue gas, owing to its high denitrification efficiency and mild reaction conditions. Cerium-based catalysts, with CeO2 as the primary component, have been identified as a promising system for replacing traditional vanadium-based and noble metal catalysts. These catalysts rely on the reversible Ce3+/Ce4+ redox cycle and exhibit favorable oxygen storage-release capacity derived from lattice oxygen migration. However, the presence of multiple impurities in industri

Sources, effects and present perspectives of heavy metals contamination: Soil, plants and human food chain.

Heavy metal (HM) poisoning of agricultural soils poses a serious risk to plant life, human health, and global food supply. When HM levels in agricultural soils get to dangerous levels, it harms crop health and yield. Chromium (Cr), arsenic (As), nickel (Ni), cadmium (Cd), lead (Pb), mercury (Hg), zinc (Zn), and copper (Cu) are the main heavy metals. The environment contains these metals in varying degrees, such as in soil, food, water, and even the air. These substances damage plants and alter soil characteristics, which lowers crop yield. Crop types, growing circumstances, elemental toxicity,
