Hepatic mitochondrial oxidative metabolism and lipid peroxidation in experimental hexachlorobenzene-induced porphyria with dietary carbonyl iron overload.

Both human porphyria cutanea tarda and experimental hexachlorobenzene-induced porphyria are associated with hepatic injury and are potentiated by excess hepatic iron. The mechanisms whereby cellular injury occurs and the synergistic role of iron overload are unknown. In the present experiments, we studied hepatic mitochondrial function and lipid peroxidation in rats with hexachlorobenzene-induced porphyria in which iron loading was achieved by dietary carbonyl iron supplementation. Female rats were treated for 8 weeks, receiving a chow diet supplemented with hexachlorobenzene (0.2%, w/w), carb

Threats to water resources from hexachlorobenzene waste at Kalush City (Ukraine)--a review of the risks and the remediation options.

The production of chlorinated solvents such as tetrachloroethylene and tetrachloromethane has resulted in large stockpiles of unintentionally produced persistent organic pollutants (POPs) including high content of hexachlorobenzene (HCB waste). HCB waste of 15,000 t arising from the production of chlorinated solvents at the Kalush factory in Ukraine was landfilled. In 2008, it was discovered that HCB and other pollutants were escaping from the landfill into local environment including the Sapogi-Limnytsia Rivers, tributaries of the Dniester River. This showed that the HCB waste was not appropr

Effects of ozone, hexachlorobenzene, and bis(tri-n-butyltin)oxide on natural killer activity in the rat lung.

The respiratory tract is a major route of exposure to noxious agents as well as pathogens such as viruses. Natural killer (NK) activity is an important first line of defense to virally infected cells as well as certain neoplasms; therefore, testing the effects of exposure to toxic compounds on this activity is important in understanding the immunotoxic potential of the compound. Lymphoid cell suspensions, obtained after enzymatic dispersion of rat lungs and purification over nylon wool columns, showed in vitro natural killer activity toward YAC lymphoma cells. Validation of the test with well-

Enhanced withdrawal from chickens of hexachlorobenzene (HCB) and pentachlorophenol (PCP) by colestipol, mineral oil, and/or restricted feeding.

Young chickens fed hexachlorobenzene (HCB) or pentachlorophenol (PCP) for 14 d at 10 ppm in the diet contained body burdens of 573 or 362 micrograms, respectively. These diets were withdrawn (d 0) and replaced for 21 d with diets containing 5% mineral oil (MO), or 5% colestipol (CO), a bile-acid-binding resin, or the chickens were restricted in feed intake to 50% of controls (50-RF), fed MO plus 50-RF, or CO plus 50-RF. Without any treatment during withdrawal, body burdens were reduced to 63% and 70% of the d 0 values for HCB and PCP, respectively. MO, CO, or 50-RF reduced body burdens of HCB

The environmental pollutant hexachlorobenzene causes eosinophilic and granulomatous inflammation and in vitro airways hyperreactivity in the Brown Norway rat.

Based on observations that the persistent environmental pollutant hexachlorobenzene (HCB) induces inflammatory skin lesions and eosinophilic and granulomatous lung pathology as well as in vivo airways hyperresponsiveness to methacholine in the BN/SsNOlaHsd rat (Michielsen et al., Toxicol Appl Pharmacol 172:11-20, 2001), which are features of human Churg-Strauss syndrome (CSS), we have investigated whether HCB induced other features of CSS such as asthma and systemic vasculitis involving the heart and kidneys in this strain of rat. To this end, BN/SsNOlaHsd rats received control feed or feed su

Hexachlorobenzene-induced eosinophilic and granulomatous lung inflammation is associated with in vivo airways hyperresponsiveness in the Brown Norway rat.

We investigated whether the eosinophilic and granulomatous lung pathology that develops in Brown Norway (BN/SsNOlaHsd) rats upon feeding hexachlorobenzene (HCB) is associated with nonspecific in vivo airways hyperresponsiveness (AHR) to methacholine. To this end, female BN/SsNOlaHsd rats were exposed to diets with no supplementation or diets supplemented with 450 mg HCB per kg feed. On days 7 or 21 of exposure in vivo airways hyperresponsiveness to increasing concentrations of methacholine was assessed both by whole body plethysmography and by visual scoring. In addition, lungs were lavaged to

Effects of hexachlorobenzene feeding and iron overload on enzymes of haem biosynthesis and cytochrome P 450 in rat liver.

1. The effect of hexachlorobenzene feeding on liver delta-aminolaevulinate synthase, uroporphyrinogen decarboxylase and cytochrome P 450 was studied at various time-intervals in siderotic and non-siderotic rats. 2 In the non-siderotic group hexachlorobenzene feeding led to a progress decrease in liver uroporphyrinogen decarboxylase activity, accompanied by a progressive increase in delta-aminolaevulinate synthase activity. Cytochrome P 450 concentrations were above normal throughout but fell toward the end of the experiment. 3. Similar but more marked changes were found in the siderotic animal

Simultaneous LC/MS analysis of hexachlorobenzene and pentachlorophenol by atmospheric pressure chemical ionization (APCI) and photoionization (APPI) methods.

The simultaneous LC/MS analyses of hexachlorobenzene (HCB) and pentachlorophenol (PCP), two dioxin precursors were studied by both atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization mass spectrometry (APPI/MS). The optimum LC/MS conditions of the simultaneous analysis were determined for these two compounds with different polarity. Ionization by APPI was found to be more effective than by APCI, and is thus a better ionization method for analysis of the two compounds. Using LC/APPI/MS we can achieve the simultaneous analysis of HCB and PCP at ppb levels.

Relationship of lead, mercury, mirex, dichlorodiphenyldichloroethylene, hexachlorobenzene, and polychlorinated biphenyls to timing of menarche among Akwesasne Mohawk girls.

BACKGROUND: Children are commonly exposed at background levels to several ubiquitous environmental pollutants, such as lead and persistent organic pollutants, that have been linked to neurologic and endocrine effects. These effects have prompted concern about alterations in human reproductive development. Few studies have examined the effects of these toxicants on human sexual maturation at levels commonly found in the general population, and none has been able to examine multiple toxicant exposures. The aim of the current investigation was to examine the relationship between attainment of men

Degradation behaviors and accumulative effects of coexisting chlorobenzene congeners on the dechlorination of hexachlorobenzene in soil by nanoscale zero-valent iron.

It is well known that many chlorinated organic pollutants can be dechlorinated by nanoscale zero-valent iron. However, in the real chlorinated organic compounds contaminated soil, the congeners of high- and low-chlorinated isomer often coexist and their dechlorination behaviors are poorly known, such as hexachlorobenzene (HCB). In this work, the degradation behaviors of three coexisting chlorobenzene congeners pentachlorobenzene (PeCB), 1,2,4,5-tetrachlorobenzene (1,2,4,5-TeCB) and 1,2,4-trichlorobenzene (1,2,4-TCB) and the influence of initial pH and reaction temperature on the dechlorination

Augmenting atrazine and hexachlorobenzene degradation under different soil redox conditions in a bioelectrochemistry system and an analysis of the relevant microorganisms.

Soil microbial fuel cells (MFCs) are a sustainable technology that degrades organic pollutants while generating electricity. However, there have been no detailed studies of the mechanisms of pollutant degradation in soil MFCs. In this study, the effects of external resistance and electrode effectiveness on atrazine and hexachlorobenzene (HCB) degradation were evaluated, the performance of soil MFCs in the degradation of these pollutants under different soil redox conditions was assessed, and the associated microorganisms in the anode were investigated. With an external resistance of 20Ω, the d

Solid-phase microextraction using octadecyl-bonded silica immobilized on the surface of a rotating disk: determination of hexachlorobenzene in water.

Solid-phase microextraction of hexachlorobenzene from water was implemented for the first time on a rotating disk coated with an octadecyl-bonded silica (C(18)) sorptive phase. The results indicate that the sorption performance of this phase for the model analyte selected is similar to that observed using a rotating disk containing PDMS. In both cases, equilibrium is achieved within approximately 120 min for samples volumes of 50 mL and decreases to 20-30 min when the sample volume is decreased to 10 mL. The comparable behavior observed for the sorption of HCB in both phases is consistent with

Chromosomal linkage analysis of porphyria in mice induced by hexachlorobenzene-iron synergism: a model of sporadic porphyria cutanea tarda.

Genetic susceptibility to toxic chemicals is of major importance but most studies concentrate on candidate genes and searches for unknown susceptibility genes are uncommon. Human sporadic porphyria cutanea tarda is usually precipitated by alcohol, oestrogens, hepatitis viruses, HIV or haemodialysis. The mechanism is not known but there is a role for iron metabolism and an underlying genetic predisposition is suspected. A similar porphyria in humans has also been caused by hexachlorobenzene. These human porphyrias can be modelled in iron-loaded mice exposed to hexachlorobenzene, in which C57BL/

A comparison of the effects of hexachlorobenzene, beta-naphthoflavone, and phenobarbital on cytochrome P-450 and mixed-function oxidases in Japanese quail.

Hexachlorobenzene (HCB), beta-naphthoflavone (BNF), or phenobarbital (PB) was administered to Japanese quail to determine their effects on hepatic porphyrin levels and drug-metabolizing enzymes. While HCB increased porphyrin levels, PB slightly reduced them, and BNF had no effect. HCB was an excellent inducer in quail, increasing the specific content of cytochrome P-450 to levels similar to those produced by BNF. Additional similarities between HCB- and BNF-treated quail included a comparable hypsochromic absorption shift in the CO-reduced difference spectra of cytochrome P-450 and similar eff

Effects of pentachlorophenol on rat liver changes induced by hexachlorobenzene, with special reference to porphyria, and alterations in mixed function oxygenases.

Hexachlorobenzene (HCB, 1000 ppm) and 500 ppm pentachlorophenol (PCP) were fed separately or in combination to female Wistar rats. A control group was provided with standard food without HCB or PCP. Subgroups of 4 rats were killed after 1, 2, 4, 6 and 8 weeks. No significant difference was found between the amounts of HCB accumulated in the livers of the HCB and HCB + PCP fed rats. Administering HCB together with PCP caused a noticeable accumulation of PCP in the liver, compared to the results after administering HCB and PCP separately. In the HCB and HCB + PCP fed groups liver weight increase

Hexachlorobenzene, a dioxin-type compound, increases malic enzyme gene transcription through a mechanism involving the thyroid hormone response element.

Hexachlorobenzene (HCB) is a dioxin-type chemical that acts mainly through the aryl hydrocarbon receptor. Chronic exposure of rats to HCB increases the activity of malic enzyme (ME). In this report, we show that this increase is correlated with an induction of ME messenger RNA (mRNA) levels, with the maximal HCB effect achieved after 9 days of intoxication. This effect is specific for ME, as other liver enzymes, such as glyceraldehyde-3-phosphate dehydrogenase, phosphoenol pyruvate carboxykinase, and mitochondrial alpha-glycerol-3-phosphate dehydrogenase, are not affected by HCB. The induction

Measurement of PCBs, DDE, and hexachlorobenzene in cord blood from infants born in towns adjacent to a PCB-contaminated waste site.

There are limited data on the concentrations of common contaminants--polychlorinated biphenyls (PCBs), dichlorodiphenyl dichloroethene (pp'-DDE) and hexachlorobenzene (HCB)--in umbilical cord blood. Cord blood provides the primary direct measure of prenatal exposure to these contaminants, the key determinant of PCBs' neurodevelopmental toxicities. The objective of this study was to characterize cord blood levels of PCBs, pp'-DDE, and HCB among 751 infants who were born between 1993 and 1998 to mothers residing adjacent to a PCB-contaminated harbor in New Bedford, Massachusetts, and for whom th

Monitoring bioaccumulation and toxic effects of hexachlorobenzene using the polyurethane foam unit method in the microbial communities of the Fuhe River, Wuhan.

Hexachlorobenzene (HCB) is a chlorinated aromatic hydrocarbon that was widely used for seed dressing in prevention of fungal growth on crops, and also as a component of fireworks, ammunition, and synthetic rubbers. Because of its resistance to degradation and mobility, HCB is widely distributed throughout the environment and is accumulated through food chains in different ecosystems. In this study, a preliminary investigation was carried out on the bioaccumulation and the toxic effects of HCB in the microbial (protozoan in particular) communities in the Fuhe River, Wuhan, a water body receivin

Aryl hydrocarbon receptor-ligand hexachlorobenzene promotes immunosuppression leading to mammary tumor growth and metastasis in a HER2-positive syngeneic mouse model.

Pesticide exposure has been established as a risk factor for breast cancer. Hexachlorobenzene (HCB) is an aryl hydrocarbon receptor (AhR) ligand. Mice bearing LM3 HER2(+) breast tumors were exposed to HCB (35 and 15 days). Tumor growth and lung metastases were evaluated, and key molecules were analyzed by western blot, immunofluorescence and RT-qPCR. Splenic and tumor infiltrating lymphocytes (TILs) were quantified by flow cytometry. Results showed that HCB (35 days) accelerates tumor growth and promotes lung metastases development. HCB upregulates AhR, CYP1A2, IDO1, COX-2 and VEGF expression.
