Ganglionic blocking effect of some antibiotics on isolated rat sympathetic ganglion.

It was shown on the isolated superior cervical ganglion of the rat that some antiobiotics exerted a blocking effect nearly as potent as that exhibited by hexamethonium or d-turbocurarine. The ganglionic blockade thus induced proved to be non-specific and due to the local anaesthetic effect of the tested drugs.

Effect of an ET(B)-selective and a mixed ET(A/B) endothelin receptor antagonist on venomotor tone in deoxycorticosterone-salt hypertension.

OBJECTIVES: Because the ET(B) receptor is important in venoconstriction, we examined the effects of a selective ET(B) receptor antagonist (A-1 92621) and a mixed ET(A/B) receptor antagonist (A-182086) on endogenous endothelin-1 (ET-1) contributions to elevated venomotor tone in deoxycorticosterone acetate-salt (DOCA-salt) hypertension. METHODS: Changes in venomotor tone were assessed using repeated measurements of mean circulatory filling pressure (MCFP) in awake, uninephrectomized, DOCA-salt-treated rats and uninephrectomized sham rats following intravenous (i.v.) injections of the ET(B) ant

Gastric secretion during anaesthesia and its inhibition by metiamide and other drugs.

The histamine H2-receptor antagonist, metiamide, inhibits the acid gastric secretion produced by chloralose-urethane anaesthesia in dogs carrying gastric cannulae chronically. This secretion is also prevented by atropine and hexamethonium. "Spontaneous" gastric secretion of vagal origin in conscious dogs is also blocked by metiamide.

Basal and spontaneous exocrine pancreatic secretion in conscious and anaesthetized chickens, Gallus domesticus.

Basal exocrine pancreatic secretion was studied in anaesthetized and conscious chickens and spontaneous secretion was studied in anaesthetized chickens. Results are compared with those of other vertebrates to estimate the specific pattern of this secretion in birds. The flow of pancreatic juice was greater in conscious chickens than when anaesthetized (Table 1). Amylase activity was greater than that of the other species and was of the same order in anaesthetized and conscious birds (Table 1). A spontaneous exocrine pancreatic secretion was seen to remain after eliminating a large part of the

Angiotensin II in the paraventricular nucleus stimulates sympathetic outflow to the cardiovascular system and make vasopressin release in rat.

The hypothalamic paraventricular nucleus (PVN) plays essential roles in neuroendocrine and autonomic functions, including cardiovascular regulation. It was shown that microinjection of angiotensin II (AngII) into the PVN produced a pressor response. In this study, we explored the probable mechanisms of this pressor response. AngII was microinjected into the PVN and cardiovascular responses were recorded. Then, the responses were re-tested after systemic injection of a ganglionic blocker, Hexamethonium, or a vasopressin V1 receptor blocker. Hexamethonium pretreatment (i.v.) greatly and signific

Renal hemodynamics in canine DOCA-salt hypertension: effect of calcium channel blockade.

Systemic arterial blood pressure (BP), renal blood flow (RBF), and renal vascular resistance (RVR) were followed for 3-4 wks during the progression of DOCA-salt hypertension in the conscious dog. Accompanying the gradual increase in BP was an increase in RBF; however, RVR was unchanged. The hypertension was totally reversed 5-7 days after cessation of DOCA-salt treatment, but the increase in RBF persisted, presumably as a result of renal hypertrophy. Renal adrenoceptor blockade with prazosin (6 dogs) and prazosin plus idazoxan (4 dogs) caused a comparable decrease in BP in the normotensive and

Nicotine effects in mouse hippocampus are blocked by mecamylamine, but not other nicotinic antagonists.

Previous data indicated that bath-application of nicotine to mouse hippocampal slices resulted in a concentration-dependent increase in the amplitude of the orthodromic population spike and the appearance of multiple population spikes in the CA1 pyramidal cell layer. d-Tubocurarine (4-100 microM), alpha-bungarotoxin (10-160 microM), and atropine (40-200 microM) had similar effects, although for alpha-bungarotoxin these excitatory effects were transient. Mecamylamine (1.6-3.2 mM) inhibited the population spike, while hexamethonium (3.2 mM) had no effect. These cholinergic antagonists were teste

Effect of cholinergic antagonists on sympathetic ganglionic transmission of vasomotor reflexes from the carotid baroreceptors and chemoreceptors of the dog.

1. In anaesthetized dogs the reflex vascular resistance changes in a perfused hind limb were studied following carotid baroreceptor or chemoreceptor stimulation. 2. The observed rises in resistance were sympathetically mediated and thus provided a means of studying the action of the cholinergic antagonists on the sympathetic ganglion transmission. 3. The reflex response to carotid baroreceptor stimulation produced by lowering the pressure in the carotid sinuses was abolished by hexamethonium bromide but not reduced by hyoscine methyl bromide. 4. The reflex response to carotid body chemorecepto

Physiological effects of muscarinic vs nicotinic ACh antagonists upon ganglion cell activity in the frog retina.

The intravitreal administration of ACh agonists (eserine, carbachol, oxotremorine) or that of ACh muscarinic antagonists (scopolamine, atropine) provoked a reduction of the On-Off ganglion cell discharges. The agonists depressed the Off discharges more than the On discharges, while the ACh muscarinic antagonists depressed the On- more than the Off discharges. These drugs did not modify the ganglion cell receptive field area; thus, the muscarinic cholinergic system seems not to be involved in the spatial organization of the On-Off ganglion cells, but rather seems to play an important part in th

Sodium and volume depletion activates neurogenic mechanisms in renal hypertensive dogs.

The acute response to ganglionic blockade (hexamethonium bromide, 30 mg/kg, i.v.) was used to evaluate the neurogenic contributions to mean arterial pressure maintenance in the conscious one-kidney, one clip hypertensive dog. Approximately 2 hours (112 minutes) after ganglionic blockade, captopril (10 mg/kg, i.v.) was given to block the renin-angiotensin system. Hypertensive animals were studied 3 days after clipping (group 2) or 2 to 4 weeks after clipping (groups 3 and 4). Groups 2 and 3 were fed a regular sodium diet, but group 4 animals were sodium and volume depleted. Normotensive control

[Pharmacoprophylaxis of an experimental neurogenic pancreatic injury].

Experimental neurogenous injury to the rabbit pancreas was used to show that mechanical traumatization of the pyloroduodenal region leads to a considerable decrease in the creatine phosphate content and to the rise in the level of lactic acid in the gland tissue. Pharmacological analysis of the changes obtained demonstrated the reflex nature of the injury under consideration. High doses of noradrenaline were also found to cause pronounced metabolic alterations in the pancreatic tissue.

Changes in nasal airway resistance and secretory response in the guinea pig after nasal challenge with capsaicin and histamine.

We studied serial changes of nasal airway resistance (NAR) and secretory response after topical stimulation with either capsaicin or histamine, since both are known to stimulate a chemosensitive sensory C-fiber ending. After topical capsaicin stimulation, a dose-related increase of NAR was noted, with a peak response occurring at 10-20 min. These NAR responses were completely abolished following systemic pretreatment with capsaicin. However, they were not affected by atropine, diphenhydramine or hexamethonium pretreatment. After topical histamine stimulation, an atropine-resistant increase of

Extrinsic and intrinsic neural control of pyloric sphincter pressure in the dog.

1. In chloralose-urethane-anaesthetized dogs a manometric assembly was inserted via a gastrostomy to monitor pyloric pressure with a sleeve sensor. Antral and duodenal contractions were monitored with both manometric side holes and serosal strain gauges. 2. Subserosal silver wire electrodes were placed in the antrum 5 cm orad and the duodenum 3 cm aborad to the pylorus to facilitate field stimulation of intramural nerves. 3. The pylorus exerted spontaneous tone (10.8 +/- 4.8 mmHg) with phasic contractions occurring at a rate varying from 1-5 min-1 and, at times, with a superimposed higher freq

Vagally mediated insulin secretion by stimulation of brain cholinergic neurons with neostigmine in bilateral adrenalectomized rats.

This study investigated the relationship between central cholinergic neurons and insulin secretion in bilateral adrenalectomized fed rats. Neostigmine (a cholinesterase inhibitor, 5 x 10(-8) mol) administered into the third cerebral ventricle produced significant increases in hepatic venous plasma insulin and glucose concentrations, whereas i.v. injection of the same dose of neostigmine did not. Prior acute subdiaphragmatic vagotomy or i.p. pre-injection with methylatropine (10(-8) mol) completely prevented the neostigmine-induced rise in plasma insulin concentration. Intraperitoneal pretreatm

Indirect excitatory actions of histamine on isolated chicken oesophagus.

The histamine-induced contraction on chicken oesophagus was antagonised non-competitively by atropine, hexamethonium, cocaine, methysergide, indomethacin, theophylline and verapamil. Physostigmine slightly potentiated the excitatory action of histamine. These results indicate that histamine excreted its excitatory action by involving a number of mechanisms as suggested in guinea pig oesophagus.

Autonomic nervous system activity in Macaca fascicularis.

This study was performed to determine the contributions of the sympathetic and parasympathetic nervous systems to cardiovascular control. Hexamethonium was administered to block the autonomic ganglia, propranolol to block beta adrenergic receptors of the sympathetic nervous system, and methylatropine to block the parasympathetic nervous system. The results of this study indicate high sympathetic tone and low parasympathetic tone in resting Macaca fascicularis. These findings are different from those in man but are similar to other nonhuman primates.

On the character of the electrical activity of the stomach, pyloric sphincter and duodenum of cats.

In cats with chronically implanted electrodes in the muscle wall of the gastro-intestinal tract, the electrical activity of the stomach (S) pyloric sphincter (PS) and duodenum (D) is characterized by cycles consisting of two phases: active--whereby 2 to 3 slow potential changes (SPC) are accompanied by spike potentials (SP) and a pause during which for 60-90 s only SPC are recorded. After bilateral transthoracic vagotomy there is a disruption of the cyclic character of the electrical activity in the investigated regions of the gastro-intestinal tract--SP from S and PS are recorded with each SP

[Effect of spasmolytics on cathartic activity of pilocarpine, 5-HT and prostaglandlin E2 in mice (author's transl)].

Effects of autonomic, anti-histaminic, anti-5HT drugs, papaverine and morphine on cathartic activity of spasmogens such as pilocarpine, 5-HT and prostaglandin E2 (PGE2) were tested by giving these drugs to mice subcutaneously. Mice used were male dd strain weighing 20 +/- 2 g and the catharatic effect was evaluated by the all or none method. The cathartic effect by pilocarpine was inhibited markedly by anti-cholinergic drugs such as atropine and diphenhydramine, but not by hexamethonium and methysergide. Therefore, the action of pilocarpine was found to be direct on the acetylcholine receptors

A COMPARISON OF THE GANGLION POTENTIALS AND BLOCK PRODUCED BY ACETYLCHOLINE AND TETRAMETHYLAMMONIUM.

Potentials recorded from the surface of the superior cervical ganglion of cats after an intra-arterial injection of acetylcholine were characterized by a complex waveform which depended on the amount of drug administered. Small doses of acetylcholine evoked a potential consisting of low amplitude hyperpolarization followed by low amplitude depolarization. Somewhat larger doses of acetylcholine caused a triphasic potential containing an initial period of depolarization in addition to the periods of hyperpolarization and delayed depolarization. Still larger doses of acetylcholine produced usuall

Changes in preganglionic sympathetic nerve function following chronic morphine treatment in rats.

1. The effects of acute or chronic morphine treatment on the changes in blood pressure and pulse rate in response to ganglionic stimulation or blockade and to vagal stimulation, and of isolated atria to field stimulation or noradrenaline, were studied. 2. In pithed rats, intravenously injected hexamethonium significantly depressed the blood pressure responses to sympathetic nerve stimulation. The ganglionic blocking effects of hexamethonium were significantly greater in chronically morphine-treated rats, but were not significantly affected by acute morphine administration in naive animals. 3.
