Chromium chloride increases insulin-stimulated glucose uptake in the perfused rat hindlimb.

AIM: To determine the effect of chromium chloride (CrCl3 ) on healthy skeletal muscle glucose uptake in the absence and presence of submaximal insulin using the rat hindlimb perfusion technique. METHODS: Sprague-Dawley rats were randomly assigned to an experimental group: basal (Bas), chromium chloride (Cr), submaximal insulin (sIns) or chromium chloride plus submaximal insulin (Cr-sIns). RESULTS: Insulin significantly increased [H(3)]-2 deoxyglucose (2-DG) uptake in the gastrocnemius muscles. Additionally, Cr-sIns displayed greater rates of 2-DG uptake than sIns (Cr-sIns 6.86 ± 0.74 μmol g

The effect of induced hindlimb lameness on thoracolumbar kinematics during treadmill locomotion.

REASON FOR PERFORMING STUDY: There are no detailed studies describing a relationship between hindlimb lameness and altered motion of the back. OBJECTIVES: To quantify the effect of induced subtle hindlimb lameness on thoracolumbar kinematics in the horse. METHODS: Kinematics of 6 riding horses were measured during walk and trot on a treadmill before and during application of pressure on the sole of the left hindlimb using a well-established sole pressure model. Reflective markers were located at anatomical landmarks on the limbs, back, head and neck for kinematic recordings. Ground reaction

Orthostatic response in rats after hindlimb unloading: effect of transcranial electrical stimulation.

INTRODUCTION: Orthostatic hypotension is a commonly observed phenomenon after exposure to microgravity and in various forms of autonomic failure. It has been suggested that insufficient activation of supraspinal structures responsible for descending sympathetic drive could play a significant role in this disorder. We examined the effect of transcranial electrical stimulation (TES) of autonomic nuclei within the brain on the orthostatic hypotension induced by exposure to simulated microgravity using a hindlimb unloading model. METHODS: There were 20 male Wistar rats that were suspended by thei

Hindlimb withdrawal reflexes evoked by Er:YAG laser and scalpel incisions in rats.

OBJECTIVE: In this study, we assessed the magnitudes of nociceptive withdrawal reflexes evoked by skin and muscle incisions made with steel scalpel and erbium:yttrium-aluminum-garnet (Er:YAG) laser. BACKGROUND DATA: A few studies have suggested that laser incisions would be less painful than conventional scalpel incisions. METHODS: Twenty adult male Sprague-Dawley rats were used. Under light barbiturate anesthesia, graded incisions were made into the plantar surface of the animals' hindpaws. Electromyographic (EMG) recordings from a hamstring muscle were used to estimate the nociceptive resp

Motor control of locomotor hindlimb posture in the American alligator (Alligator mississippiensis).

Crocodilians are unusual among quadrupedal tetrapods in their frequent use of a wide variety of hindlimb postures, ranging from sprawling to a more erect high walk. In this study, we use synchronized kinematic videos and electromyographic recordings to test how the activity patterns of hindlimb muscles in American alligators (Alligator mississippiensis Daudin) differ between sprawling and more upright postures. Previous force platform analyses suggested that upright posture in alligators would require greater activation by hindlimb extensors to counter increases in the flexor moments exerted a

Long-term (6-wk) hindlimb suspension inhibits spermatogenesis in adult male rats.

The International Space Station will allow extended habitation in space and long-term exposure to microgravity (microG). A concern is the impact of long-term microG exposure on the ability of species to reproduce. The model often used to simulate microG is rat hindlimb suspension (HLS), where the hindlimbs are elevated above the cage floor with a tail harness. Experiments described here are the first to examine the effect of long-term HLS on testicular function in adult male rats. Free-roaming (controls), animals with only the tail harnessed but hindlimbs in contact with the cage floor (TO), a

Effect of trotting speed, load and incline on hindlimb stance-phase kinematics.

The objective was to understand how the stance-phase kinematics of the hindlimb of trotting horses change with speed under 3 conditions (level, loaded and incline), to compare our results with the predictions of the spring-mass model and to help focus our future studies of muscle function. Video recordings were made of 5 Arabian horses trotting on a treadmill. Five consecutive strides were digitised and averaged for each trial. The angle-time diagrams were qualitatively similar to those reported previously. As speed increases, the range of motion of the hindlimb increases, as predicted by the

Physiological assessment of augmented vascularity induced by VEGF in ischemic rabbit hindlimb.

This study was designed to assess the physiological consequences of augmented vascularity induced by administration of vascular endothelial growth factor (VEGF), an endothelial cell-specific mitogen, in a rabbit model of hindlimb ischemia. Ten days after excision of the common and superficial femoral arteries from one hindlimb of 24 New Zealand White rabbits, VEGF (n = 15) or saline (control; n = 9) was selectively injected into the ipsilateral internal iliac artery. Limb perfusion was evaluated immediately pre-VEGF (baseline) and again at days 10 and 30. A Doppler guide wire was advanced to t

Nerve and muscle blood flow during hindlimb ischemia and reperfusion in rats.

Animal models of peripheral nerve ischemia have yielded variable results. The question of whether post-ischemia re-establishment of blood flow to the nerves augments injury has not been examined. To study this question, the ipsilateral common iliac and femoral arteries were occluded with arterial snares for 3 hours in rats; 14C-butanol tissue distribution was then used to measure blood flow in both sciatic and posterior tibial nerve trunks and in both biceps femoris muscles during occlusion and reperfusion. Clinical limb function was graded serially, with the undisturbed contralateral limb ser

[Analytical study of Xenopus hindlimb regenerate with special reference to muscle regeneration].

Amputated hindlimbs of Xenopus laevis, develop various types of regenerates in relation with amputation level as well as stage development. The present experiments is an attempt to study the histological characteristics of Xenopus regenerations, i.e., rational changes of tissue components along the length of the regenerated part with special emphasis on the degree of muscle regeneration. Four types of regenerates were studied viz; a 4th toe obtained from a completely restored regenerated limb at 126 days after amputation of limb at base level in stage 51. Response of large hindlimb veins of dog to aortic arch chemoreceptor stimulation.

Experiments were performed in chloralose-anesthetized dogs to examine the reflex effects on tone in the saphenous vein of stimulating the aortic arch chemoreceptors. The saphenous vein was perfused with cooled (30 degrees C) aortic blood at constant flow, and perfusion pressure was measured. In 13 animals the aortic arch chemoreceptors were stimulated by injection of cyanide (0.1 mg/kg) into the root of the aorta. Cyanide caused a decrease (20 +/- 2 mmHg) in saphenous vein perfusion pressure and an increase (44 +/- 4) in aortic pressure. These changes that occurred subsequent to aortic root cy

Protocol for modeling blood flow recovery and angiogenesis in response to hindlimb ischemia.

Here, we present a protocol that models peripheral artery disease using the surgical induction of hindlimb ischemia (HLI) in mice. We list the steps to surgically excise the femoral artery and assess blood flow recovery through laser Doppler imaging. We then detail procedures for visualizing immune cell recruitment and blood vessel growth to explore the role of inflammatory macrophages in angio/arteriogenesis. For complete details on the use and execution of this protocol, please refer to Mantsounga et al.1.

The interaction of in vivo muscle operating lengths and passive stiffness in rat hindlimbs.

The operating length of a muscle is a key determinant of its ability to produce force in vivo. Muscles that operate near the peak of their force-length relationship will generate higher forces whereas muscle operating at relatively short length may be safe from sudden lengthening perturbations and subsequent damage. At longer lengths, passive mechanical properties have the potential to contribute to force or constrain operating length with stiffer muscle-tendon units theoretically being restricted to shorter lengths. Connective tissues typically increase in density during aging, thus increasin

Collateral Development and Arteriogenesis in Hindlimbs of Swine After Ligation of Arterial Inflow.

BACKGROUND: Development of collateral vasculature is key in compensating for arterial occlusions in patients with peripheral artery disease (PAD). We aimed to examine the development of collateral pathways after ligation of native vessels in a porcine model of PAD. METHODS: Right hindlimb ischemia was induced in domestic swine (n = 11) using two versions of arterial ligation. Version 1 (n = 6) consisted of ligation with division of the right external iliac, profunda femoral, and superficial femoral arteries. Version 2 (n = 5) consisted of the ligation of version 1 with additional ligation wit

Effect of eplerenone on endothelial progenitor cells and oxidative stress in ischemic hindlimb.

BACKGROUND: We have demonstrated that angiotensin II receptor blocker (ARB) improved endothelial progenitor cells (EPCs) dysfunction through the antioxidative mechanism. Therefore, we investigate whether the selective mineralocorticoid receptor (MR) antagonist eplerenone improves EPCs function in rat hindlimb ischemia. METHODS: Unilateral hindlimb ischemia was surgically induced in Wistar rats. After induced ischemia, rats received eplerenone (30 mg/kg/day), valsartan (3 mg/kg/day), or vehicle for 3 weeks. Peripheral blood mononuclear cells were isolated, subjected to flow cytometric analysis

Effects of L1 blockade on sensory axon outgrowth and pathfinding in the chick hindlimb.

In the developing chick hindlimb, sensory axons, which grow together in bundles as they extend distally, and the motoneuron axons they encounter express the cell adhesion molecule L1. Following injection of function-blocking anti-L1 antibodies into the limb at stage 25, some sensory axons choose inappropriate peripheral nerves even though motoneuron pathfinding is unaffected. Here, to further elucidate L1's role, we assessed the effects of this perturbation using pathway tracing, immune labeling, confocal microscopy, and electron microscopy. After L1 blockade, sensory axons were still bundled

Healing of tibial fractures is not impaired after acute hindlimb ischemia in rats.

The influence of transient circulatory arrest on the healing of closed tibial fractures was investigated in rats by the use of a hindlimb tourniquet technique. Twenty-four animals were randomly divided into three groups. In all animals, the left lower leg was fractured and fixed with an intramedullary nail system. In the ischemic group, complete acute transient ischemia for 4.5 h and neurapraxia of the sciatic and femoral nerves were induced prior to fracture. In the neurapraxia group, the sciatic and femoral nerves were crushed with forceps before fracture. In the control group, no other inte

Neuronal and muscular inclusions in rats with hindlimb dysfunction after treating with difluorobenzhydrylpiperadine.

Rats showing an ataxic gait induced by 20 wk of treatment with 0, 30, or 60 mg/kg of difluorobenzhydrylpiperadine (DFBP), a detriazinyl metabolic of almitrine, were examined by light microscopy and transmission electron microscopy. Vacuolar degeneration associated with lamellar inclusions was observed in musculus soleus and m. interossei of the hindlimbs in DFBP-treated rats. The inclusions were also produced within sensory neurons, satellite and Schwann cells, and vascular endothelial cells of thoracic and lumbar dorsal root ganglia as well as muscle spindles of affected muscles. Eperythrozoon infection in young bulls with scrotal and hindlimb edema, a herd outbreak.

Young (9 to 10 mo) Aberdeen-Angus bulls (n = 5) in excellent body condition and pastured with approximately 35 other bulls of similar age on a farm in southeastern Alabama had fever, harsh pulmonary sounds, increased respiratory rate and variable amounts of hindlimb and scrotal edema. Bulls had mild microcytic, normochromic anemia. Numerous eperythrozoon organisms were identified on blood smears. Indirect hemagglutination inhibition test results for Eperythrozoon suis antibodies were negative either because E. suis antigens do not cross react with cattle eperythrozoon organism antibodies or bl
