Furosemide inhibits in vitro histamine release induced by antigens and anti-IgE.

We used 56 samples of heparinized blood of patients with asthma and/or allergic rhinitis, which were stimulated with Dermatophagoides pteronyssinus or gramineous pollen and anti-IgE in order to study the effect of furosemide on histamine release in basophils. At the same time, blood samples incubated in furosemide in different concentrations (2.5 microM to 1.49 mM) were also subjected to the same stimuli. Results showed a linear and dose-dependent inhibition of histamine release for antigens (r = 0.96; p < 0.01) and anti-IgE (r = 0.95; p < 0.05), reaching 95% at maximum inhibition. These resul

Histamine release from human basophils by the insect allergen Chi t I.

Hemoglobins of the Diptera species Chironomus thummi thummi (Chi t I) are potent inhalant allergens. Chi t I-specific histamine release was measured by a new radioimmunoassay in whole human blood taken from 20 sensitized patients, 11 exposed nonsensitized probands, and 11 nonexposed controls. The sensitized patients, who all had positive skin tests and radioallergosorbent test results with Chi t I, showed a significantly higher histamine release than the two other groups. However, within the patient group, the percentage of released histamine did not correlate with the intensity of the skin te

Histamine release, pulmonary blood shunts, and rapid, shallow breathing in the dog.

Trimetaphan (Arfonad), which acts as a histamine-releasing agent in dogs, was observed to increase the shunt of blood through the lungs. Although external examination of the lungs showed no appreciable collapse, more detailed examination of lungs fixed by perfusion showed that the shunt could be explained on the basis of alveolar collapse and there was no need to postulate the opening of anatomical shunts. Histamine-releasing agents or histamine produced rapid, shallow breathing, which persisted after inflation of the lungs sufficient to restore the elastic and non-elastic resistance of the lu

Correlations of in vivo mediator release with late cutaneous allergic responses in humans. I. Kinetics of histamine release.

To evaluate the contribution of mast cell-derived mediators in the late cutaneous allergic response, the duration and quantity of antigen-induced histamine release was compared to the intensity of the antigen-induced skin reactions in atopic volunteers. Chambers containing either pollen extract or buffer were appended to denuded bases for 1 hr and were replaced hourly with buffer for 3 additional hr. These were compared to the extinction dilution skin test titer and to the mean diameters of the 20-minute wheal and induration at 6 and 8 hr after intradermal injection of antigen. Chamber-fluid h

Potent histamine-releasing activity of atrahagin, a novel snake venom metalloproteinase.

Poisonous snakebite wound is a popular disease worldwide. However, the pathogenesis remains unclear. In the present study, a novel metalloproteinase atrahagin in Chinese cobra (Naja atra) snake venom was purified, using heparin-sepharose followed by Superdex 75 gel filtration chromatography. Apart from its alpha-fibrinogenase activity, atrahagin potently activated human colon, lung and tonsil mast cells with the net histamine release being 25.9+/-4.4, 17.0+/-1.9, 13.2+/-3.6%, respectively. Time course studies revealed that the peak histamine release induced by atrahagin occurred at 12, 12 and

Potentiation of vancomycin-induced histamine release by muscle relaxants and morphine in rats.

The intravenous injection of vancomycin sometimes causes anaphylactoid reactions, in which histamine release may play a major role. These reactions are more frequently manifested when vancomycin is injected into anesthetized patients. We examined the vancomycin-induced histamine release and the interaction of vancomycin with muscle relaxants or opioid in rats. In an in vitro study with rat peritoneal mast cells, treatment with vancomycin at concentrations of greater than 1.25 mM produced significant histamine release. Tubocurarine, vecuronium, pancuronium, succinylcholine, and morphine up to c

Chronic idiopathic urticaria: possible contribution of histamine-releasing factor to pathogenesis.

BACKGROUND: Histamine-releasing factor was recently shown to be clinically relevant in allergic rhinitis and asthma. HRF could also be involved in the pathogenicity of chronic idiopathic urticaria (CU). The purpose of this study was to investigate the role of HRF in the pathophysiology of CU. METHODS: Blisters were induced on lesional and nonlesional skin of 12 patients with CU and on normal skin of five control subjects. HRF activity and histamine content were measured in all samples recovered from each skin site. RESULTS: Significantly more HRF was found in blister fluids from lesional ski

[Histamine-releasing action of x-ray contrast media in rats in vivo].

The effect of roentgen contrast media (RCM)--50% bilignost, 76% triombrast, 80% iodamide-380, on degranulation of peritoneal mast cells in rats was studied. It was found that 20 minutes after intravenous injections of RCM the content of intracellular histamine significantly decreased. This effect depended on the drug dose and increased in the order: triombrast, iodamide, triiotrast, bilignost. Different sensitivity of animals to the histamine-releasing effect of RCM was detected by using the nonparametric method of mathematical statistics. The number of "sensitive" animals was 50-75%.

Purification of serum proteins with inhibitory activity on the histamine release in vitro and/or in vivo.

Recently, we described a factor in rat and human serum with inhibitory activity on the 2-hour passive cutaneous anaphylactic (PCA) reaction in rat skin. The Ca-ionophore-induced histamine release from rat peritoneal mast cells was inhibited as well. In human serum, inhibitory activity for the in vitro histamine release from rat mast cells was shown to be present within two serum fractions. One molecule was purified by 50-70% ammonium sulfate precipitation and subsequent DEAE chromatography at pH 5.4. The main component showed complete identity with transferrin. TAZ deficiency exacerbates psoriatic pathogenesis by increasing the histamine-releasing factor.

BACKGROUND: Transcriptional coactivator with PDZ-biding motif (TAZ) is widely expressed in most tissues and interacts with several transcription factors to regulate cell proliferation, differentiation, and death, thereby influencing organ development and size control. However, very little is known about the function of TAZ in the immune system and its association with inflammatory skin diseases, so we investigated the role of TAZ in the pathogenesis of psoriasis. RESULTS: Interestingly, TAZ was expressed in mast cells associated, particularly in lysosomes, and co-localized with histamine-rele

Effects of ibrutinib on proliferation and histamine release in canine neoplastic mast cells.

The Bruton's tyrosine kinase (BTK) inhibitor ibrutinib is effective in the treatment of human chronic lymphocytic leukaemia and mantle cell lymphoma. Recent data have shown that ibrutinib also blocks IgE-dependent activation and histamine release in human basophils (BAs) and mast cells (MCs). The aim of this study was to investigate whether BTK serves as a novel therapeutic target in canine mast cell tumours (MCTs). We evaluated the effects of ibrutinib on two canine MC lines, C2 and NI-1 and on primary MCs obtained from canine MCTs (n = 3). Using flow cytometry, we found that ibrutinib suppre

Induction of microvascular leakage and histamine release by promutoxin, an Arg49 phospholipase A2.

It has been recognized that phospholipase A(2) (PLA(2)) is a crucial factor of snake venom induced inflammation. Recently, promutoxin, a novel member of minor subgroup of snake venom PLA(2) (R49 PLA(2)) has been characterized in our laboratory, but its roles in induction of inflammation remain uninvestigated. Using highly purified promutoxin, we found this enzymatically inactive PLA(2) provoked a dose-dependent increase in microvascular leakage in the skin of rats. Pretreatment of rats with compound 48/80 diminished promutoxin-induced skin reaction and reduced mast cell numbers in rats. Cyproh

Characteristics of histamine-releasing activity in the sera of patients with chronic idiopathic urticaria.

BACKGROUND: The serum histamine-releasing activity (HRA) found in a sizable percentage of patients with chronic idiopathic urticaria (CIU) has been partially characterized. However, the variable effect of individual HRA+ sera in basophils of different donors and the relationship of HRA to the clinical course require further investigation. OBJECTIVE: The study was performed to characterize the HRA found in sera of some members of a sizable group of carefully evaluated patients with CIU. METHODS: Sera of 70 patients with CIU, evaluated with a standard protocol, were screened for increased HRA.

Influence of seasonal variations on histamine release and other immunological parameters in pollinosis.

One hundred and fifty-six pollen-sensitive patients with sensitization to grass pollen only were chosen for this study; 65 patients were allergic to Dermatophagoides, while 45 were non-allergic subjects with no atopic history. The following tests were carried out on all patients: histamine release of total blood, basal histamine, total histamine, determination of total IgE and specific IgE (RAST) serum levels, serum hemagglutinating antibodies and skin tests. Seasonal increase of IgE was observed, with post-seasonal fall; 50% of the cases reached normal values. However, RAST as a semiquantitat

Histamine release as the basis for morphine action on bile duct and sphincter of Oddi.

We have investigated the mechanism by which morphine contracts hog bile duct and sphincter of Oddi. Morphine contraction is antagonized by naloxone, competitively on the sphincter, noncompetitively on the bile duct. Diphenhydramine at low concentration (3.4 X 10(-6)M) also antagonizes both actions of morphine. Histamine has a very potent contracting action on the sphincter and bile duct and this is antagonized by diphenhydramine. Burimamide only weakly antagonizes the actions of morphine or histamine. Compound 48/80 causes a pronounced contraction of sphincter and bile duct following which mor
