Tamoxifen inhibits histidine kinases of M. tuberculosis two-component signaling systems.

Two-component signaling systems (TCSSs) serve as primary signal transduction pathways in bacteria, regulating essential processes including growth, physiology, and virulence, making them attractive drug targets. In Mycobacterium tuberculosis (M. tb), the PhoPR system plays a critical role in pathogenesis, with the PhoR histidine kinase (HK) functioning at the intersection of cognate and noncognate signaling networks. Using PhoR as a prototypical HK, we hypothesized that targeting this system would compromise M. tb's adaptive capacity. We developed and optimized a high-throughput screen of phar

Histidine 375 Modulates CD4 Binding in HIV-1 CRF01_AE Envelope Glycoproteins.

UNLABELLED: The envelope glycoproteins (Envs) from human immunodeficiency virus type 1 (HIV-1) mediate viral entry. The binding of the HIV-1 gp120 glycoprotein to CD4 triggers conformational changes in gp120 that allow high-affinity binding to its coreceptors. In contrast to all other Envs from the same phylogenetic group, M, which possess a serine (S) at position 375, those from CRF01_AE strains possess a histidine (H) at this location. This residue is part of the Phe43 cavity, where residue 43 of CD4 (a phenylalanine) engages with gp120. Here we evaluated the functional consequences of repla

Acute hyponatremia after cardioplegia by histidine-tryptophane-ketoglutarate--a retrospective study.

BACKGROUND: Hyponatremia is the most common electrolyte disorder in hospitalized patients and is known to be associated with increased mortality. The administration of antegrade single-shot, up to two liters, histidine-tryptophane-ketoglutarate (HTK) solution for adequate electromechanical cardiac arrest and myocardial preservation during minimally invasive aortic valve replacement (MIAVR) is a standard procedure. We aimed to determine the impact of HTK infusion on electrolyte and acid-base balance. METHODS: In this retrospective analysis we reviewed data on patient characteristics, type of s

Chlamydophila (Chlamydia) pneumoniae induces histidine decarboxylase production in the mouse lung.

Chlamydophila (Chlamydia) pneumoniae (C. pneumoniae) is the third most common cause of community-acquired pneumonia and is probably involved in the development of certain chronic inflammatory diseases, including atherosclerosis and adult-onset asthma. Histamine, synthesized by histidine decarboxylase (HDC) from L-histidine, plays an essential role in allergic and inflammatory processes and in cell differentiation. The effect of C. pneumoniae infection on the expression of HDC has not been examined. In the present study, normal Balb/c mice and HDC knockouts, and control mice with a CD1 backgrou

Gastric anti-ulcer activity of (+)-cyanidanol-3, a histidine decarboxylase inhibitor.

The gastric anti-ulcer activity of (+)-cyanidanol-3, a specific histidine decarboxylase inhibitor was studied on various types of experimentally induced ulcers in rats viz., pylorus-ligated and restraint ulcers, gastric mucosal damage induced by non-steroid anti-inflammatory drugs like aspirin, phenylbutazone, indomethacin and ibuprofen and by reserpine. Guinea pigs subjected to histaminee challenge were also used in the study. (+)-Cyanidanol-3 possesses significant gastric anti-ulcer activity in all these models. However, the ED50 values against pylorus-ligated and restraint ulcers and agains

Formation of gizzard erosion-inducing substance in heated casein-histidine mixture.

Formation of a toxic substance inducing gizzard erosion (GE) was examined by using mainly a casein-histidine mixture as a simple model of fish meal. When a casein-histidine mixture was heated at 135 degrees C for three or five hours or at 160 degrees C for one hour, it induced severe GE. Changes in pH of histidine solution seemed to have little effect on the production of the toxic substance. Addition of a heated mixture of 30 mg of histidine and 150 g of casein (per kg of diet) failed to induce GE. A mixture of 750 mg of histidine and 150 g of casein was enough to produce a maximal toxicity.

Correlation between serum gastrin concentration and rat stomach histidine decarboxylase activity.

After prolonged fasting the activity of histidine decarboxylase in the oxyntic mucosa of the rat stomach is low. Feeding or injection of gastrin or insulin rapidly raises the enzyme activity. It was earlier suggested that all enzyme-activating agents act through release of gastrin. This view has found experimental support in studies which show that in antrectomized rats the enzyme is activated by gastrin but not by gastrin-releasing stimuli like feeding or vagal excitation (insulin hypoglycemia). In the present investigation rats were subjected to a variety of treatments and serum gastrin conc

Ligand binding represses bacterial histidine kinase activity by inhibiting its dimerization.

Two-component systems (TCS) mediate bacterial signal transduction in response to specific environmental conditions. The two components are the sensor kinase (SK), which senses the signal and autophosphorylates on a histidine residue, and a response regulator (RR), which is phosphorylated by the kinase and modifies gene expression. Despite intensive study, the mechanisms of signal sensing by sensor kinases remain incompletely defined, and it is yet unclear how these kinases sense multiple ligands. Mycobacterium tuberculosis PdtaS/PdtaR is a soluble TCS pair that participates in the Rip1 signal

A gut-secreted histidine methyltransferase enforces cross-kingdom catalytic antifungal defense.

Mucosal immunity deploys diverse defenses against fungal pathogens, yet the evolution of fungal resistance demands new antifungal strategies. Here, we uncover that intestinal epithelial cells secrete the host histidine methyltransferase METTL9 as a cross-kingdom, catalytic antifungal effector. Proteomic profiling revealed that exposure to Candida albicans induces robust METTL9 secretion into the intestinal lumen. Extracellular METTL9 directly binds the fungal zinc-scavenging protein PRA1, catalyzing histidine methylation of this zincophore to disrupt zinc acquisition-an essential micronutrient

A novel histidine-acetate buffer for freeze-dried monoclonal antibody formulations.

Histidine (His)-hydrochloride (HCl) is widely used in freeze-dried monoclonal antibody (mAb) formulations, but alternative buffers are required when chloride ions are undesirable. This study evaluates His-acetate (Ac) as a substitute and its impact on the stability of two model mAbs with distinct physicochemical properties- infliximab (mAb-1) and a humanized anti-ricin mAb (mAb-2). pH shifts during freeze-drying were compared among His-Ac, His-HCl, and sodium Ac buffers, confirming strong buffering capacity of His-Ac within pH 5.5-6.5. Conformational and colloidal stability assessments reveale

Histidine Tag-Specific PEGylation Improves the Circulating Half-Life of TIMP2.

An overarching limitation of therapeutic biologics is the limited half-life these proteins often exhibit once in circulation. PEGylation, the chemical conjugation of proteins to poly(ethylene glycol) (PEG), is a common strategy to improve protein pharmacokinetics (PK) by enhancing stability, reducing immunogenicity, and decreasing renal clearance. Tissue Inhibitor of Metalloproteinases 2 (TIMP2) is a 22 kDa matrisome protein that exhibits therapeutic potential across a range of human disease models yet possesses a short serum half-life. To advance the therapeutic development of recombinant His

The histidine kinase NahK regulates pyocyanin production through the PQS system.

Many bacterial histidine kinases work in two-component systems that combine into larger multi-kinase networks. NahK is one of the kinases in the GacS Multi-Kinase Network (MKN), which is the MKN that controls biofilm regulation in the opportunistic pathogen Pseudomonas aeruginosa. This network has also been associated with regulating many virulence factors P. aeruginosa secretes to cause disease. However, the individual role of each kinase is unknown. In this study, we identify NahK as a novel regulator of the phenazine pyocyanin (PYO). Deletion of nahK leads to a fourfold increase in PYO prod

CHANGES OF HISTIDINE-RICH GLYCOPROTEIN LEVELS IN CRITICALLY ILL SEPTIC PATIENTS.

Background: Histidine-rich glycoprotein (HRG), a potential prognostic factor in sepsis, lacks clarity regarding its relevance in septic-induced shock, disseminated intravascular coagulation (DIC), and acute respiratory distress syndrome (ARDS) pathogenesis. This study investigated the association between HRG concentrations and these critical conditions. Methods: Blood samples were collected from 53 critically ill patients on days 1, 3, 5, and 7 after ICU admission at the Kyushu University Hospital. Daily clinical and laboratory data were recorded, and patient survival was assessed 28 days afte

Abnormal histidine metabolism promotes macrophage lipid accumulation under Ox-LDL condition.

Distinct macrophage populations exert highly heterogeneity and perform various functions, among which, a crucial function of lipid metabolism is highlighted. However, the role of histidine metabolism disorder in macrophage lipid metabolism remains elusive. Addressed this question, we sorted and cultured the bone marrow-derived macrophages (BMDMs) of histidine decarboxylase (Hdc) knockout (Hdc-/-) mice with an in vitro oxidized low-density lipoprotein (ox-LDL) model, and detected the intracellular lipids by Oil Red O staining as well as lipid probe staining. Revisiting histidine-dependent acid phosphatases: a distinct group of tyrosine phosphatases.

Although classical protein tyrosine phosphatase (PTP) superfamily members are cysteine-dependent, emerging evidence shows that many acid phosphatases (AcPs) function as histidine-dependent PTPs in vivo. These AcPs dephosphorylate phospho-tyrosine substrates intracellularly and could have roles in development and disease. In contrast to cysteine-dependent PTPs, they utilize histidine, rather than cysteine, for substrate dephosphorylation. Structural analyses reveal that active site histidine, but not cysteine, faces towards the substrate and functions as the phosphate acceptor. Nonetheless, dur

Protective effects from carnosine and histidine on acetaminophen-induced liver injury.

Protective effects of carnosine or histidine against acetaminophen-induced hepatotoxicity in Balb/cA mice were examined. Each compound, at 0.5, 1, or 2 g/L, was added into the drinking water for 4 wk. Acute liver injury was induced by acetaminophen treatment intraperitoneally (i.p. 350 mg/kg body weight). Acetaminophen treatment significantly depleted hepatic GSH and ascorbic acid levels, increased hepatic level of malonyldialdehyde (MDA), reactive oxygen species (ROS), and oxidized glutathione (GSSG), as well as decreased hepatic activity of glutathione peroxidase (GPX), catalase, and superox

Cyanamide mediated syntheses of peptides containing histidine and hydrophobic amino acids.

Using the model of a primitive earth evaporation pond, the synthesis of three histidyl peptides in yields up to 11% was demonstrated when aqueous solutions of histidine, leucine, ATP, cyanamide, and MgCl2 were evaporated and heated for 24 h at 80 degrees C. In addition, peptides were formed in yields of up to 56%, 35%, and 21%, respectively for phenylalanine, leucine, and alanine when aqueous solutions of the appropriate amino acid were evaporated and heated with cyanamide and one or more of the following components: ATP, AMP, 4-amino-5-imidazole carboxamide, or MgCl2. Non-mast cell histamine levels in rat tissues after histidine loading.

The i.p. administration of L-histidine in doses of 500 and 1000 mg/kg, caused prolonged high levels of histidine but did not influence the levels of histamine in the non-mast cell tissues such as the stomach, lungs and liver in the rat. After polymyxin B or 48/80 treatments as well as in anaphylaxis, the levels of histamine in the lungs and liver were greatly reduced but histidine administration failed to alter noticeably the concentrations of histamine in these organs. Similarly, the low contents of histamine in the stomach of 48/50-treated or polymyxin B-treated rats remained unchanged in th

Apomorphine-induced turning behavior in 6-hydroxydopamine lesioned rats is increased by histidine and decreased by histidine decarboxylase, histamine H1 and H2 receptor antagonists, and an H3 receptor agonist.

The role of histamine and its receptors in basal ganglia neurocircuitry was assessed in apomorphine-induced turning behavior. Rats with unilateral 6-hydroxydopamine lesions of the substantia nigra pars compacta and medial forebrain bundle were administered histaminergic agents, and apomorphine-induced turning behavior was tested on Days 7 and 14 post-lesion. Compared with saline-treated rats, histidine (500 mg/kg, i.p.), a precursor of histamine, increased turning behavior (p<0.05), while alpha-fluoromethylhistidine (alpha-FMH, 25 microg, i.c.v.), an irreversible inhibitor of histidine decarbo
