Expression of human minor histocompatibility antigen on cultured kidney cells.

Incompatibility of human minor histocompatibility (hmH) antigens can induce rejection of grafts in organ transplantation and graft-versus-host reactions in bone marrow transplantation. In spite of their importance in clinical transplantation, hmH antigens are not well studied. Previous studies have demonstrated the expression of hmH antigens on T and B cells, hematopoietic progenitor cells and keratinocytes. We have for the first time demonstrated the expression of hmH antigens on cultured kidney cells using HLA-B35-restricted, hmH antigen-specific cytotoxic T lymphocyte (CTL) clones, which we

Major histocompatibility complex (MHC) antigen expression in HIV encephalitis.

In order to study possible immunopathogenic mechanisms in Human Immunodeficiency Virus (HIV) encephalitis, immunocytochemical localization of Class I and Class II major histocompatibility complex (MHC) antigens was studied in formalin-fixed tissue sections from the brains of 10 individuals who had died with this disorder. Using the avidin biotin peroxidase technique and monoclonal antibodies to these antigens, increased expression of Class I antigens was found in five out of 10 and of Class II antigens in six out of 10 cases of HIV encephalitis. This contrasted with results obtained with the H

Modulation of class II histocompatibility antigen expression by maternal serum.

Human maternal serum has been shown to down-regulate the expression of MHC class II antigen in three distinct circumstances. Cord blood mononuclear cells, incubated in the mother's own serum, showed significant modulation of class II antigen expression. This was also the case for unrelated donor lymphocytes, incubated in pooled maternal serum. One neoplastic line (Daudi) was further shown to down-regulate class II antigen expression. In this last case, the down-regulating effect persisted over a 10-day period during which maternal serum was renewed regularly. Retroplacental serum was more MHC

Human histocompatibility antigen associations in subacute cutaneous lupus erythematosus.

We have identified a clinically distinct subset of lupus erythematosus patients marked by the presence of a histologically proven, nonscarring variety of cutaneous LE (subacute cutaneous LE) in which there is a very high frequency of the human leukocyte antigens (HLA) B8 and DR3. Differences in the configuration of their skin lesions allowed a separation of the patients into two clinical subgroups; annular and papulosquamous. HLA-B8 was increased in the annular subgroup (81%, corrected P (Pc) < 0.007) and combined group (65%, Pc < 0.004). HLA-DR3 was present in all 11 of the annular patients (

High-throughput cloning reveals diverse properties of T-cell receptors targeting minor histocompatibility antigens.

Hematopoietically restricted minor histocompatibility antigens (miHAs) presented on HLA-class I molecules are ideal targets for adoptive T-cell immunotherapy in the context of an allogeneic stem cell transplant (alloSCT). This is because CD8 cells that recognize these miHAs can mediate potent antileukemia effects with a low risk for graft-versus-host disease (GVHD). A barrier to translating this concept broadly to the clinic has been the difficulty and expense of cloning potentially high-value T-cell receptors (TCRs). Here, we describe the isolation of anti-miHA TCRs using a novel high-through

High-throughput characterization of 10 new minor histocompatibility antigens by whole genome association scanning.

Patients with malignant diseases can be effectively treated with allogeneic hematopoietic stem cell transplantation (allo-SCT). Polymorphic peptides presented in HLA molecules, the so-called minor histocompatibility antigens (MiHA), play a crucial role in antitumor immunity as targets for alloreactive donor T cells. Identification of multiple MiHAs is essential to understand and manipulate the development of clinical responses after allo-SCT. In this study, CD8+ T-cell clones were isolated from leukemia patients who entered complete remission after allo-SCT, and MiHA-specific T-cell clones wer

Influence of histocompatibility antigens in recurrent spontaneous abortion and its relevance to leukocyte immunotherapy.

A total of 108 couples with recurrent spontaneous abortion (RSA) were studied to investigate the possible influence of histocompatibility antigens (HLA) on their condition and its management. HLA-B18 was shown to be at a higher frequency in RSA women, but not significantly so after statistical correction. Just over half the RSA women shared two or more HLA-A, B or DR antigens with their partners (P < 0.01), but this group did not differ from the others in clinical or laboratory features, nor in subsequent pregnancy success rate. Leukocyte immunotherapy in which the donor shared at least one HL

Immunohistologic characterization of major histocompatibility antigens and inflammatory cellular infiltrate in human breast cancer.

The presence of major histocompatibility antigens [HLA-A,B,C; beta 2 microglobulin (beta 2)m; la-like] on tumor cells and the nature and distribution of mononuclear cellular infiltrates in benign and malignant human breast tumors were studied by the use of monoclonal antibodies and immunoperoxidase techniques on frozen tissue sections. Although all benign tumors expressed HLA-A,B,C or beta 2m, 20 of 53 primary breast cancers did not react with anti-HLA-A,B,C or anti-beta 2m antibodies. Expression of la-like antigens was rarely encountered on tumor cells. The intensity of the inflammatory monon

[Antibody response to histocompatibility antigens in hemodialysis patients subjected to planned whole-blood transfusions].

In hemodialysis patients planned whole blood transfusions from single donors, induce the formation of anti HLA A, B, C and DR cytotoxic antibodies. The percentage of immunization, however, is lower than that observed in healthy subjects immunized following a similar transfusional schedule. This observation may reflect the existence in some patients of an impairment of the immune response to histocompatibility antigens. The production of lymphocytotoxic antibodies appear to be associated with severe graft rejections while anti HLA antibodies detected only by an indirect rosette assai don't seem

Delivery of exogenous protein antigens to major histocompatibility complex class I pathway in cytosol.

A fragment of anthrax lethal factor possesses the interesting function of delivering recombinant protein antigens through the classical major histocompatibility complex (MHC) class I pathway. This region of the lethal factor lacks the domain associated with anthrax cytotoxicity and functions independently of its binary partner, protective antigen. Experiments that used inhibitors at different steps of the MHC class I pathway supported this hypothesis. Application of this discovery to current T cell assays allows for the measurement of cytotoxic T lymphocyte function without resorting to live v

Histocompatibility antigens and polymyalgia rheumatica in a Japanese patient with insulin-dependent diabetes mellitus.

A 30-year-old Japanese female developed insulin-dependent diabetes mellitus (IDDM). She later complained of muscle pains at the age of 37. Erythrocyte sedimentation rate and C-reactive protein were abnormal, with negative antinuclear antibody and rheumatoid factor tests. The diagnosis of polymyalgia rheumatica (PMR) was made. She had HLA phenotypes including A2 and without DR4, consistent with common types of Japanese PMR. Her DNA typing included DQB1*0303 which is positively associated with Japanese IDDM. It seems likely that she suffered from these diseases at a young age on the basis of hav

Influence of histocompatibility antigens in recurrent spontaneous abortion couples and on their reproductive performances.

PROBLEM: To determine if human leukocyte antigens (HLA) play any role in the aetiology of recurrent spontaneous abortion (RSA), a substantial group of RSA couples were studied, and their reproductive performances in a 3-year follow-up recorded. METHODS: HLA typing was performed for HLA-A, -B, and DR antigens in both partners of 75 couples with unexplained RSA, and compared with a control group of 30 fertile couples that never experienced abortion. A further 57 couples of this group were studied for their reproductive performance in a 3-year follow-up, and subdivided into three subgroups: 1) c

Autoantibodies in chronic arthritis of childhood: relations with each other and with histocompatibility antigens.

BACKGROUND: Studies have shown the presence of either antibodies to histone or anticardiolipin antibodies in some forms of childhood chronic arthritis. The relation between these autoantibodies has not been previously reported, however, and the immunogenetics of their association with childhood arthritis has not been studied. METHODS: The interrelation of fluorescent antinuclear antibodies, antibodies to histone, and anticardiolipin antibodies and their associations with histocompatibility antigens (HLA) were studied in 114 children with chronic arthritis (45 children with pauciarticular onse

Effect of mismatches for major histocompatibility complex and minor antigens on corneal graft rejection.

The importance of minor histocompatibility genes in corneal graft rejection was investigated using a model that simulates the major histocompatibility complex (MHC) and minor mismatches of the human allograft more accurately than previous animal models. DA(RT1a) x LEW(RT1(1]F1 hybrid rats were backcrossed to LEW, and the backcross generation were used as corneal graft recipients. Female DA(RT1a) strain animals were used as donors throughout. As in humans, the MHC disparity (a to 1) between each donor-recipient pair could be controlled; minor mismatches were variable and unknown. The MHC haplot

Major histocompatibility antigens and lymphocyte subsets during experimental allergic neuritis in the Lewis rat.

Major histocompatibility antigens were identified in frozen sections of normal Lewis rat peripheral nerve tissue with monoclonal antibodies and an avidin-biotin-peroxidase complex system. Class I antigen is normally required for cytotoxic/suppressor T lymphocyte function and class II antigen for activation of helper T lymphocytes. In the sciatic nerves class I antigen was expressed diffusely by most endoneurial and perineurial cells but class II antigen only by a minority. In the cauda equina class I antigen was expressed by all arachnoid and some endoneurial cells, while class II antigen was

Histocompatibility antigens and dermatitis herpetiformis with special reference to jejunal abnormalities and acetylator phenotype.

Sixty-one patients with dermatitis herpetiformis (DH) were studied. Of these fifty-three (87%) had the histocompatibility (HL-A) antigen HL-A8 as compared with 17% of a control population; the difference is significant. This incidence of HL-A8 among patients with DH is higher than that reported earlier for such patients and is similar to that reported for patients with adult coeliac disease. The incidence of HL-A1 was also significantly greater in the patient group, and was attributed to the increased frequency of haplotype HL-A1, 8. The frequency of HL-A8 was about equal in patients in whom D

Chloroquine treatment affects T-cell priming to minor histocompatibility antigens and graft-versus-host disease.

Graft-versus-host disease (GVHD) caused by T-cell recognition of minor histocompatibility (MiHC) antigens is a major complication of bone marrow transplantation. GVHD therapy has focused on removal or suppression of donor T cells, but modulation of MiHC antigen presentation to CD4+ T cells may represent an alternative approach. Chloroquine is known to inhibit major histocompatibility complex (MHC) class II presentation of antigen in vitro by affecting invariant chain dissociation from MHC class II. The goal of this study was to evaluate the role of chloroquine in abrogating T-cell priming to M

Epithelial expression of major histocompatibility complex (MHC) antigens in normal rat salivary and lacrimal glands.

The presence and distribution of MHC class I and class II antigens within these glands of Wistar and Lewis rats was studied using monoclonal antibodies and an indirect immunoperoxidase technique. In rats of both strains and sexes, MHC class 1 antigens were expressed by ductal epithelium in addition to vascular endothelium and other connective tissue elements. Except for a small proportion of secretory cells within lacrimal glands, acini were uniformly negative for class I antigen. MHC class II immune response-associated (Ia) antigens were expressed by stromal dendritic cells in all glands. Gla

Expression of major histocompatibility complex antigens in the brains of patients with progressive multifocal leukoencephalopathy.

Progressive multifocal leukoencephalopathy (PML) is caused by JC virus (JCV) infection of the central nervous system (CNS) in immunosuppressed patients. The immunopathogenesis of this chronic encephalitis is unknown. Because major histocompatibility (MHC) class I and class II antigens are important in modulating the immune response and viral clearance, we examined the tissue expression of MHC molecules in relation to CNS damage and presence of virus. By immunocytochemical staining, both MHC class I and class II antigens were expressed at high levels within PML lesions. Beta-2 microglobulin (be
