The role of leptin in glucose homeostasis.

The fat-derived hormone, leptin, is well known to regulate body weight. However, there is now substantial evidence that leptin also plays a primary role in the regulation of glucose homeostasis, independent of actions on food intake, energy expenditure or body weight. As such, leptin might have clinical utility in treating hyperglycemia, particularly in conditions of leptin deficiency, such as lipodystrophy and diabetes mellitus. The mechanisms through which leptin modulates glucose metabolism have not been fully elucidated. Leptin receptors are widely expressed in peripheral tissues, includin

Blood sphingolipids in homeostasis and pathobiology.

Sphingolipids have emerged as key signaling molecules involved in the regulation of a variety of cellular functions including cell growth and differentiation, proliferation and apoptotic cell death. Sphingolipids in blood constitute part of the circulating lipoprotein particles (HDL, LDL and VLDL), carried by serum albumin and also present in blood cells and platelets. Recent lipidomic and proteomic studies of plasma lipoproteins have provided intriguing data concerning the protein and lipid composition of lipoproteins in the context of disease. Sphingolipids have been implicated in several di

[The homeostasis assessment by hemorrhagic shock].

The comparative analysis of different protocols of infusion therapy of the hemorrhagic shock stage III was performed. The infusion of the colloid solution of hydroxiethylstarch 200/0.5 and non-balanced crystalloid 0.9% solution of natrium chloride leads to the development of negative changes in homeostasis. Whereas infusion of the 4% solution of the modified gelatin and balanced crystalloid solition (sterofundin) allows to avoid the registered changes in electrolyte and alkaline balance.

[Calcium homeostasis in pregnancy and lactation].

The daily required dose of calcium is greatly increased during pregnancy. It is caused by the transition of about 30 g to the fetus and by enhanced urinary excretion. The normal calcium homeostasis is maintained by parathormone (PTH), calcitriol and PTH-related peptide (PTHrP). The concentration of calcitriol is augmented as well as the absorptive surface of the gut. PTHrP is involved in the regulation of placental calcium transport and also in lactation and initiation of labor. Bone turnover is enhanced during pregnancy and sometimes may evoke osteoporosis, especially in bones which form the

The genetic basis of glucose homeostasis.

Genetic and environmental determinants play critical roles in insulin resistance and beta-cell function. A model of the complex feedback system for maintenance of glucose tolerance has been developed that reflects the constraint of glycemia within narrow physiologic limits. The "glucose homeostasis" model is described by insulin sensitivity, glucose effectiveness, acute insulin response to glucose, and disposition index (DI). Relatively little is known about the genetic basis of glucose homeostasis phenotypes or their relationship to risk of diabetes and atherosclerotic cardiovascular disease.

The role of the intestine in bone homeostasis.

The intestine is critical for calcium absorption and skeletal mineralization. Nutritional factors affect the dietary intake of calcium and its bioavailability in the intestine. Individuals differ considerably in their ability to absorb calcium but the reasons for this physiological variation are not fully understood although much is now known about the effects of vitamin D metabolites and the molecular steps in calcium absorption. Bone changes are found in many intestinal disorders including lactose intolerance, coeliac disease, short bowel syndrome, small intestinal bacterial overgrowth and C

A genetic view of iron homeostasis.

Inherited disorders of iron metabolism are invariably disorders of iron balance or distribution. This review describes the proteins known to be involved in establishing and maintaining iron balance, and discusses regulation of iron homeostasis in the context of three cell types: intestinal enterocytes, reticuloendothelial macrophages, and hepatocytes. It emphasizes information gleaned from the use of genetic analyses, particularly in mice, and poses new questions to help advance our understanding of iron balance.

Disturbances of potassium homeostasis in poisoning.

Unless renal function is impaired or rhabdomyolysis is severe, hyperkalemia is a relatively uncommon metabolic complication of poisoning. In contrast, marked hypokalemia is a more common problem and may have serious sequelae. Most potassium disturbances in acute poisoning are due to disruption of extra-renal control mechanisms, notably the activity of Na+/K+ ATPase and K+ channels. Hypokalemia occurs because of increased Na+/K+ ATPase activity (e.g. beta 2 agonist, theophylline or insulin poisoning), competitive blockade of K+ channels (e.g. barium or chloroquine poisoning), gastrointestinal l

Interrelationships of magnesium and potassium homeostasis.

The interrelationships of magnesium (Mg) and potassium (K) homeostasis are reviewed. Evidence from clinical and experimental studies including whole animal and cell culture experiments indicate that (1) homeostasis of Mg and K are closely related in the whole organism, (2) deficiencies of Mg and K frequently co-exist with gastrointestinal and especially renal losses from diuretic and nephrotoxic drug treatment being mainly responsible, and (3) Mg is required for maintenance of normal cellular K. Evidence from many laboratories indicate that Mg has direct effects at a cellular level on K transp

[Calcium homeostasis in the newborn infant].

The Calcium homeostasis in the perinatal period depends on the changes of the Calcium balance and on the influence of the three Calcium-regulating hormones parathyroid hormone, Calcitonin and 1,25-Dihydroxycholecalciferol. The most common disorder is neonatal hypocalcemia which occurs either as early type (frequent, mainly in prematures) or as late type (rare, probably due to transient hypoparathyroidism).

Disruption of Dopamine Homeostasis by Psychostimulants.

Dopamine is a catecholamine that acts as a modulatory neurotransmitter in the central nervous system. Although dopamine levels are kept within appropriate ranges through various mechanisms, they can be disrupted by multiple factors, including external psychostimulants such as cocaine and methamphetamine. Disruption contributes to cognitive deficits and emotional dysregulation that is associated with psychostimulant-use disorders (PSUD). These stimulants interfere with the dopamine system through various mechanisms that affect its synthesis, storage, release, reuptake, and degradation. Such cha

Rethinking Complement in Homeostasis and Disease.

The complement system, traditionally viewed as a central component of immune defense, is now recognized to play diverse roles in morphogenesis, tissue regeneration, cellular metabolism, and intercellular communication. This expanded perspective highlights the spatially distributed and context-dependent nature of complement activity. Consequently, our understanding of complement-mediated diseases has evolved from a model centered on excessive activation to one in which chronic inflammatory processes displace protective, complement-regulated homeostatic functions. In this review, we examine seve

Lysosomal homeostasis at the crossroads of neurodegeneration.

Lysosomes function as metabolic control centers that integrate degradation, nutrient sensing, and stress signaling. In neurons, which must maintain proteostasis and energetic balance throughout life, lysosomal homeostasis determines cellular resilience. Emerging evidence identifies lysosomal injury and defective repair as common denominators across neurodegenerative diseases. Damage to the lysosomal membrane caused by oxidative stress, lipid imbalance, or genetic mutations triggers a hierarchical quality control cascade. Early lesions recruit the endosomal sorting complex required for transpor

Macrophages of the heart: Homeostasis and disease.

Cardiac macrophages (CMs) are the most abundant immune cell type in the heart. They are critical for maintaining cardiac homeostasis and in the orchestration of immune responses to ischemic and non-ischemic cardiomyopathies. Their functions are highly heterogeneous and regulated by their tissue microenvironment. CMs have high plasticity, which allows them to perform various functions in the myocardium to bring about homeostasis within the cardiovascular system (CVS). CMs also play critical roles in coronary development and angiogenesis, tissue repair and remodeling, cardiac conduction and in t

Role of Neutrophils in Homeostasis and Diseases.

Neutrophils, constituting a predominant subset of innate immune cells in mammalian systems, play pivotal roles in pathogenic clearance and homeostatic maintenance. In the progressive development of cancer, neutrophils exert dual roles in both anticancer and procancer processes through their heterogeneity. In recent years, research into the role of neutrophils in cancer and various nontumor diseases has been continuously deepening. However, current research in this area remains incomplete. This review comprehensively summarizes the tissue homing dynamics, lifespan regulation, and physiological

Glucose Homeostasis and Derangement in Birds.

Birds (class Aves) have 1.5 to 2 times higher blood glucose concentrations than mammals of comparable sizes. The reasons for this have been studied and are believed to be multifactorial. There is low expression of insulin receptors, decreased sensitivity of the pancreatic β-cells to glucose, an absent or dysfunctional glucose transporter type 4 pathway, and increased blood glucagon concentrations. Glucagon and somatostatin appear to play a greater role than insulin in glucose homeostasis in birds. Severe hyperglycemia in birds can be attributed to diabetes mellitus, necessitating therapy to pr
