Progress in breeding low phytate crops.

Populations that depend on grains and legumes as staple foods consume diets rich in phytic acid (myo-inositol-1,2,3,4,5,6-hexkisphosphate), the storage form of phosphorus in seeds. This compound binds tightly to important mineral nutrients such as iron and zinc, forming salts that are largely excreted. This phenomenon can contribute to mineral depletion and deficiency. As one approach to solving this and environmental problems associated with seed-derived dietary phytic acid, the U. S. Department of Agriculture and others have isolated cereal and legume low-phytic acid mutations and have used

Tracking and Treating Fungal Contamination in Indoor-Growing Barley Sprouts.

The industrialised production of barley sprouts is a nutritional replenish for livestock, whereas it is being threatened by fungal contamination derived from the closed and humid environment. This study investigates the fungal communities in barley seeds and sprouts and explores the utilisation of ozone water as a mould control method. In barley seeds of 10 cultivars, Alternaria alternata, Phoma epicoccina and Fusarium cerealis were the most abundant fungal species and varied between barley cultivars. A significant transformation in fungal communities after seed germination was observed, featu

Phytosiderophore pathway response in barley exposed to iron, zinc or copper starvation.

Efficient micronutrient acquisition is a critical factor in selecting micronutrient dense crops for human consumption. Enhanced exudation and re-uptake of metal chelators, so-called phytosiderophores, by roots of graminaceous plants has been implicated in efficient micronutrient acquisition. We compared PS biosynthesis and exudation as a response mechanism to either Fe, Zn or Cu starvation. Two barley (Hordeum vulgare L.) lines with contrasting micronutrient grain yields were grown hydroponically and PS exudation (LC-MS) and root gene expression (RNAseq) were determined after either Fe, Zn, or

Dynamics changes in physicochemical properties, volatile metabolites, non-volatile metabolites, and physiological functions of barley juice during Bifidobacterium infantis fermentation.

The purpose of this research was to explore the potential of Bifidobacterium infantis fermentation to modify the composition and physiological properties of barley juices. B. infantis JFM12 showed a potent capability to decrease the total sugar contents from 0.39 ± 0.01 mg/mL to 0.35 ± 0.01 mg/mL within 24 h of fermentation. The volatile metabolite profiles were enriched after B. infantis JFM12 fermentation, leading to the changes of 13 aldehydes, 11 ketones, 10 acids, 7 alcohols, and 6 esters. A total of 98 key non-volatile metabolites were identified in the barley juice between before and af

Characterization of Siccibacter sp. Strain C2 a Novel Rhizobacterium that Enhances Tolerance of Barley to Salt Stress.

Plant growth promoting rhizobacteria (PGPR) arouse an increasing interest as an eco-friendly solution for improving crop tolerance to environmental stresses. In this study, we report the characterization of a novel halotolerant PGPR strain (named C2) identified in a screen of rhizospheric bacterial isolates from southeast of Tunisia. Phylogenetic analysis showed that strain C2 is most likely affiliated to the genus Siccibacter with Siccibacter turicensis as the closest species (98.19%). This strain was able to perform phosphate solubilization and production of indole acetic acid (IAA), siderop

Exogenous Melatonin Enhances the Low Phosphorus Tolerance of Barley Roots of Different Genotypes.

Melatonin (N-acetyl-5-methoxytryptamine) plays an important role in plant growth and development, and in the response to various abiotic stresses. However, its role in the responses of barley to low phosphorus (LP) stress remains largely unknown. In the present study, we investigated the root phenotypes and metabolic patterns of LP-tolerant (GN121) and LP-sensitive (GN42) barley genotypes under normal P, LP, and LP with exogenous melatonin (30 μM) conditions. We found that melatonin improved barley tolerance to LP mainly by increasing root length. Untargeted metabolomic analysis showed that me

Combined effect of nano-CuO and nano-ZnO in plant-related system: From bioavailability in soil to transcriptional regulation of metal homeostasis in barley.

The co-existence of engineered nanoparticles (ENPs) in the environment is an emerging issue remaining poorly investigated. The present study aimed at analyzing the fate of binary mixtures of CuO and ZnO ENPs in a soil-plant system. The ENPs were singly or jointly dosed into soil at 300 mg kg-1 and aged for 7 and 30 days. To evaluate nano-specific effects, individual and combined treatments of metal salts were also applied. Interactions between ENPs and soil-grown barley Hordeum vulgare were determined in terms of biomass, plant mineral composition as well as expression of genes regulating meta

Assessing the Health Loss from Kashin-Beck Disease and Its Relationship with Environmental Selenium in Qamdo District of Tibet, China.

Kashin-Beck Disease (KBD) is one of major endemic diseases in China. In this study, we estimated the health loss from KBD in Qamdo district of Tibet using the years lived with disability (YLD) metric and investigated the influence of environmental selenium (Se) on it by multiple regression model. The results showed that YLD rates produced a different ranking of health loss of KBD from that produced by prevalence rates between Basu and Luolong County, with higher health loss from KBD (43.61 YLD/1000) but lower prevalence (17.86%) in Basu County. YLD rates in two counites were both highest for t

Study of the intercellular fluid of healthy Lupinus albus organs. Presence of a chitinase and a thaumatin-like protein.

Proteins in the intercellular fluid (IF) of healthy Lupinus albus leaves were characterized. Silver staining of the proteins separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed more than 30 polypeptides, with the major ones having a molecular mass lower than 36 kD. After amino-terminal amino acid sequence analysis, one of the major polypeptides, IF4, was shown to have no identity with any of the proteins present in the data bases. Two others, IF1 and IF3, showed identity with previously reported pathogenesis-related proteins, IF1 with an antifungal protein from Hord

The Effect of Bacillus pumilus 3-19 Metalloproteinase Gene Knockout on the Expression of Minor Proteases of the Strain Degradome and Its PGP Properties.

The Bacillus pumilus 3-19 strain exhibits increased secretion of various hydrolases, including proteases, and is a promising plant growth-promoting (PGP) agent. A unique secreted minor metalloproteinase, MprBp, which has no homologs among prokaryotic enzymes, has been identified in its genome. The role of the enzyme in B. pumilus cells is poorly understood. Using CRISPR/Cas9 genome editing, a deletion mutant of B. pumilus 3-19 with an inactivated mprBp gene was obtained. Comparative analysis of the mutant and native strains by quantitative reverse transcription PCR (RT-qPCR) revealed a signifi

Detection and quantitation of immunogenic epitopes related to celiac disease in historical and modern hard red spring wheat cultivars.

Celiac disease (CD) develops in genetically susceptible individuals as a result of ingesting gluten-forming proteins found in cereals, such as wheat (Triticum aestivum L.), rye (Secale cereale L.) and barley (Hordeum sativum L.). There are claims that breeding practices have changed wheat protein chemistry over the years and this has resulted in modern wheat being more antigenic in terms of CD as opposed to historical wheat. The aim of this study was to detect and quantify celiac-disease-initiating peptides of α-gliadin proteins in historical and modern spring wheat cultivars. The results indi

Effect of sulfur dioxide hydrates on cell cycle, sister chromatid exchange, and micronuclei in barley.

The effects of sulfur dioxide (SO(2)) hydrates exposure on cell cycle, sister chromatid exchange (SCE), and micronuclei (MN) were investigated in barley (Hordeum vulgare) roots. A mixture of sodium bisulfite and sodium sulfite (1:3), at various concentrations from 1x10(-5) to 3x10(-2)M, was used for the treatments. The results showed that the mixture induced the formation of SCE and MN in barley root cells with different effective concentrations and with different trends as treatment concentrations increased. At high concentrations of 0.5-30.0mM, SO(2) hydrates inhibited the mitotic activity a

CCR4, a RNA decay factor, is hijacked by a plant cytorhabdovirus phosphoprotein to facilitate virus replication.

Carbon catabolite repression 4 (CCR4) is a conserved mRNA deadenylase regulating posttranscriptional gene expression. However, regulation of CCR4 in virus infections is less understood. Here, we characterized a pro-viral role of CCR4 in replication of a plant cytorhabdovirus, Barley yellow striate mosaic virus (BYSMV). The barley (Hordeum vulgare) CCR4 protein (HvCCR4) was identified to interact with the BYSMV phosphoprotein (P). The BYSMV P protein recruited HvCCR4 from processing bodies (PBs) into viroplasm-like bodies. Overexpression of HvCCR4 promoted BYSMV replication in plants. Conversel

Remediation of contaminated agricultural soils near a former Pb/Zn smelter in Austria: batch, pot and field experiments.

Metal contaminated crops from contaminated soils are possible hazards for the food chain. The aim of this study was to find practical and cost-effective measures to reduce metal uptake in crops grown on metal contaminated soils near a former metal smelter in Austria. Metal-inefficient cultivars of crop plants commonly grown in the area were investigated in combination with in-situ soil amendments. A laboratory batch experiment using 15 potential amendments was used to select 5 amendments to treat contaminated soil in a pot study using two Barley (Hordeum vulgare L.) cultivars that differed in

Differential Infection by Barley Yellow Mosaic Disease and Its Impact on Key Agronomic Traits in Barley.

Barley yellow mosaic disease (BYMD), a soilborne viral disease, severely compromises winter barley productivity. This disease is caused by barley yellow mosaic virus (BaYMV) and the barley mild mosaic virus (BaMMV), occurring in either single or mixed infections. To elucidate pathogenesis, 10 barley varieties with differential resistance were systematically analyzed. Virus-specific RT-qPCR revealed distinct accumulation patterns of BaYMV and BaMMV in infected tissues. Crucially, transmission requires temperatures persistently below 10°C for 40 to 54 days, facilitating viral entry into roots fo
