[Acupuncture and biological resilience].

This paper systematically elaborates the theories of stress, biological resilience and hormesis. Mild low-dose stress induces organismic overcompensation via preconditioning, activates nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant, autophagy, DNA repair and other pathways, and presents an inverted U-shaped biphasic dose-response relationship. Preconditioning is categorized into local and remote types, which exerts long-term protective effects on systemic organs via metabolic reprogramming as well as neuro-humoral and immune pathways, with 2 distinct protective time wi

Lack of promoting effects of phenobarbital at low dose on diethylnitrosamine-induced hepatocarcinogenesis in TGF-alpha transgenic mice.

Phenobarbital (PB), a rodent non-genotoxic carcinogen, showed hormesis, biphasic effects on rat liver carcinogenesis. To test the hypothesis that the hormesis earlier observed for PB induced hepatocarcinogenesis might also exist in the TGF-alpha transgenic mice model, one which is highly susceptible to carcinogenesis, the carcinogenic or promotion effects of a wide range of phenobarbital (PB) concentrations were investigated. Two weeks after a single i.p. dose of 5 mg /kg bw of diethylnitrosamine (DEN) to 15 day old mice, animals were treated with diet containing PB at doses of 0, 2, 15 or 500

Quorum-sensing gene regulates hormetic effects induced by sulfonamides in Comamonadaceae.

Antibiotics can induce dose-dependent hormetic effects on bacterial cell proliferation, i.e., low-dose stimulation and high-dose inhibition. However, the underlying molecular basis has yet to be clarified. Here, we showed that sulfonamides play dual roles as a weapon and signal against Comamonas testosteroni that can modulate cell physiology and phenotype. Subsequently, through investigating the hormesis mechanism, we proposed a comprehensive regulatory pathway for the hormetic effects of Comamonas testosteroni low-level sulfonamides and determined the generality of the observed regulatory mod

Background radiation impacts human longevity and cancer mortality: reconsidering the linear no-threshold paradigm.

The current linear no-threshold paradigm assumes that any exposure to ionizing radiation carries some risk, thus every effort should be made to maintain the exposures as low as possible. We examined whether background radiation impacts human longevity and cancer mortality. Our data covered the entire US population of the 3139 US counties, encompassing over 320 million people. This is the first large-scale study which takes into account the two major sources of background radiation (terrestrial radiation and cosmic radiation), covering the entire US population. Here, we show that life expectanc

Cytotoxicity models of Huntington's disease and relevance of hormetic mechanisms: A critical assessment of experimental approaches and strategies.

This paper assesses in vivo cytotoxicity models of Huntington's disease (HD). Nearly 150 agents were found to be moderately to highly effective in mitigating the pathological sequelae of cytotoxic induction of HD features in multiple rodent models. Typically, rodents are treated with a prospective HD-protective agent before, during, or after the application of a chemical or transgenic process for inducing histopathological and behavioral symptoms of HD. Although transgenic and knockout rodent models (1) display relatively high construct and face validity, and (2) are ever more routinely employ

Sublethal chemical stimulation of arthropod parasitoids and parasites of agricultural and environmental importance.

An increasing number of studies have reported stimulation of various organisms in the presence of environmental contaminants. This has created a need to critically evaluate sublethal stimulation and hormetic responses of arthropod parasitoids and parasites following exposure to pesticides and other contaminants. Examining this phenomenon with a focus on arthropods of agricultural and environmental importance serves as the framework for this literature review. This review shows that several pesticides, with diverse chemical structures and different modes of action, applied individually or in co

A theoretical framework informing research about the role of stress in the pathophysiology of bipolar disorder.

BACKGROUND: The staggering illness burden associated with Bipolar Disorder (BD) invites the need for primary prevention strategies. Before preventative strategies can be considered in individuals during a pre-symptomatic period (i.e., at risk), unraveling the mechanistic steps wherein external stress is transduced and interacts with genetic vulnerability in the early stages of BD will be a critical conceptual necessity. METHODS: Herein we comprehensively review extant studies reporting on stress and bipolar disorder. The overarching aim is to propose a conceptual framework to inform research

Naturally occurring target-site EPSPS mutations and moderate EPSPS over-expression confer glyphosate resistance in Lolium perenne ssp. multiflorum biotypes from Argentina.

BACKGROUND: Glyphosate resistance in Lolium perenne ssp. multiflorum (Italian ryegrass), a major weed in temperate cropping systems, threatens effective weed control and crop productivity. This study characterized the glyphosate response and target-site glyphosate resistance mechanisms in three biotypes (PA, MG and HE) from northwest Pampas region. RESULTS: The resistant biotypes exhibited significantly higher glyphosate median lethal dose (LD50) and dose to reduce growth rate by 50% (GR50) values, with resistance indices ranging from 5.5 to 9.3. Both resistant biotypes (MG and HE) displayed

Dynamin inhibitor dynasore modulates longitudinal bone growth in a hormetic manner.

BACKGROUND: Longitudinal bone growth occurs via endochondral ossification, involving a complex interplay of chondrocyte proliferation, differentiation, and matrix remodeling. As with all mammalian cells, chondrocytes require dynamin for mitochondrial fission, to shuttle vesicles from the Golgi apparatus, and for both clathrin- and caveolin-mediated endocytosis. Here, we aimed to test the functions of dynamin on bone growth. To do so, we applied dynasore-a small molecule that is a reversible dynamin inhibitor-to mouse metatarsal bones cultured ex vivo. We assessed gross changes using bone lengt

Hormetic dose-responses for silver antibacterial compounds, quorum sensing inhibitors, and their binary mixtures on bacterial resistance of Escherichia coli.

Silver antibacterial compounds (SACs) and quorum sensing inhibitors (QSIs), as the potential antibiotic substitutes, have been recommended to prevent and treat microbial infections for the purpose of controlling the increasingly serious bacterial resistance induced by the abuse of antibiotics. However, there is little information regarding the resistance risk of these compounds, especially their mixtures. In this study, bacterial mutation and RP4 plasmid conjugative transfer among bacteria were used to characterize the bacterial endogenous and exogenous resistance, respectively. The effects of

A hormetic approach to understanding antidepressant effectiveness and the development of antidepressant tolerance - A conceptual view.

Antidepressants such as the selective serotonin reuptake inhibitors (SSRIs) have complex temporal effects. They may worsen symptoms during early treatment, they may reduce depressive symptoms over several weeks of treatment, and they may lose effectiveness over more prolonged treatment or after repeated treatment trials. Conceptually, these effects fall within the domain of hormesis, which refers to a biphasic or multiphasic response to a drug or toxin. Hormetic effects are commonly triggered when a drug interacts with homeostatic mechanisms. We develop and evaluate a theoretical framework for

Classic dose-response and time postinoculation models for leptospira.

Leptospirosis is a preeminent zoonotic disease concentrated in tropical areas, and prevalent in both industrialized and rural settings. Dose-response models were generated from 22 data sets reported in 10 different studies. All of the selected studies used rodent subjects, primarily hamsters, with the predominant endpoint as mortality with the challenge strain administered intraperitoneally. Dose-response models based on a single evaluation postinfection displayed median lethal dose (LD50 ) estimates that ranged between 1 and 10(7) leptospirae depending upon the strain's virulence and the peri

Demonstration of an adaptive response to preconditioning Frankliniella occidentalis (Pergande) to sublethal doses of spinosad: a hormetic-dose response.

Sublethal doses of some insecticides have been reported to either stimulate or reduce the survival and fecundity of insects. Many sublethal-effect studies have been conducted after exposure of only one generation to sublethal insecticides, and there is little information about the sublethal effects on insects after long-term exposure to sublethal insecticides. In this study, changes in biological characteristics were investigated in spinosad-susceptible (Spin-S) and sublethal-spinosad-treated (Spin-Sub) strains of Frankliniella occidentalis (Pergande) after exposure to their corresponding subl

Imidacloprid decreases energy production in the hemolymph and fat body of western honeybees even though, in sublethal doses, it increased the values of six of the nine compounds in the respiratory and citric cycle.

BACKGROUND: Neonicotinoids, including imidacloprid (IM), cause harm to Apis mellifera in a number of ways, among others by impairing body maintenance, resistance and immunity. Energy resources are important to preventing this, as we hypothesized, not only in the hemolymph but particularly in the fat body, the insufficiently investigated, as yet, equivalent of the mammalian liver and pancreas. Both suppression and hormesis (diaphasic stressor response) of energy supply was reported in the energy-dependent traits of bees exposed to sublethal doses of imidacloprid. Therefore, our goal was to answ

[Cancer incidence and mortality after low-dose radiation exposure: epidemiological aspects].

Current recommendations for limiting exposure to ionizing radiation are based on the linear-no-threshold (LNT) model for radiation carcinogenesis under which every dose, no matter how low, carries with it some cancer risk. In this review, epidemiological evidences are discussed that the LNT hypothesis is incorrect at low doses. A large set of data was accumulated that showed that cancer risk after ordinarily encountered radiation exposure (natural background radiation, medical X-rays, etc.) is much lower than projections based on the LNT model. The discovery of the low-level radiation hormesis

[Mild stress as a means to modulate aging: from fly to human?].

Hormesis is the phenomenon by which adaptive responses to low doses of otherwise harmful conditions improve the functional ability of organisms. Some mild stresses have beneficial effects on longevity, aging and resistance to strong stresses (heat or cold shocks, infection) in Drosophila flies. Studies on rodents are indeed scarce but mild stress seems to be effective in humans because, for instance, patients suffering from angina have a higher survival when confronted with a heart attack. A few studies, in less tragic situations however, suggest that mild stress could have positive effects in

Adaptation strategies of cyanobacteria under trace tetracycline exposure: A comparative study of toxigenic and non-toxigenic strains.

Trace antibiotics exert unclear ecological impacts on cyanobacteria, especially the differential responses between toxigenic and non-toxigenic strains. To address this, we established a paired-strain comparative framework exposing toxigenic Microcystis aeruginosa and non-toxigenic Synechocystis sp. to environmentally relevant tetracycline (0.2, 2, 20 μg/L) for 14 days. Results showed that TC induced hormetic growth in both strains, with final cell density increases of 2.47-9.79% (M. aeruginosa) and 4.98-8.59% (Synechocystis sp.) relative to controls, but exhibited divergent adaptation strategi

The Role of GDF15 as a Myomitokine.

Growth differentiation factor 15 (GDF15) is a cytokine best known for affecting systemic energy metabolism through its anorectic action. GDF15 expression and secretion from various organs and tissues is induced in different physiological and pathophysiological states, often linked to mitochondrial stress, leading to highly variable circulating GDF15 levels. In skeletal muscle and the heart, the basal expression of GDF15 is very low compared to other organs, but GDF15 expression and secretion can be induced in various stress conditions, such as intense exercise and acute myocardial infarction,

Stimulatory effect of xenobiotics on oxidative electron transport of chemolithotrophic nitrifying bacteria used as biosensing element.

Electron transport chain (ETCh) of ammonium (AOB) and nitrite oxidizing bacteria (NOB) participates in oxidation of ammonium to nitrate (nitrification). Operation of ETCh may be perturbed by a range of water-soluble xenobiotics. Therefore, consortia of nitrifying bacteria may be used as a biosensor to detect water contamination. A surprising feature of this system is an increase of oxygen consumption, detected in the presence of certain inhibitors of ETCh. Thus, to shed light on the mechanism of this effect (and other differences between inhibitors) we monitored separately respiration of the b
