The method of horseradish peroxidase retrograde axonal transport was used to study the connections between neurons of respiratory nuclei and reticular formation of medulla oblongata and descending fibre pathways from these structures in the spinal cord of cats. The rate of reactions of a basic horseradish peroxidase adsorbed on the carboxymethylcellulose membrane was measured spectrophotometrically by monitoring the state of the enzyme. We have investigated the effects of mutations at residues His-42, Arg-38 and Phe-41 in the distal haem pocket of horseradish peroxidase on the changes in protonation state that accompany redox- and ligand-linked changes to the haem group. A conjugate of alpha-bungarotoxin with horseradish peroxidase was used to visualize alpha-bungarotoxin binding sites at the fine structural level in isolated axonal membrane vesicles from lobster walking leg nerve. Achieving a single-molecule level for RNA detection remains a significant challenge in point-of-care settings. We present CCHP, a Cas13-Csm6-horseradish peroxidase three-step enzymatic cascade assay that enables amplification-free RNA detection with a simple colorimetric readout at 450 nm within 1 h under standard conditions.
