Genetic diversity across different human populations can enhance understanding of the genetic basis of disease. We calculated the genetic risk of 102 diseases in 1,043 unrelated individuals across 51 populations of the Human Genome Diversity Panel. We found that genetic risk for type 2 diabetes and pancreatic cancer decreased as humans migrated toward East Asia. In addition, biliary liver cirrhosis, alopecia areata, bladder cancer, inflammatory bowel disease, membranous nephropathy, systemic lupus erythematosus, systemic sclerosis, ulcerative colitis, and vitiligo have undergone genetic risk d

HYPOTHESIS: That acquired cholesteatoma of the human middle ear and mastoid process can be caused by migration of squamous epithelium from the tympanic membrane.
OBJECTIVE: To provide histologic evidence in humans of the potential for medial epithelial migration toward the middle ear.
BACKGROUND: The origin of cholesteatomas of the middle ear complex of humans is still not clearly understood. The most popular theory, the medial migration of squamous epithelium from the external auditory canal/tympanic membrane through a perforation of the tympanic membrane to form a cholesteatoma, was first

Myofibroblasts play central roles in wound healing, deposition of the extracellular matrix and epithelial function. Their functions depend on migration and proliferation within the subepithelial matrix, which results in accelerated cellular metabolism. Upregulated metabolic pathways generate protons which need to be excreted to maintain intracellular pH (pH(i)). We isolated human gastric myofibroblasts (HGMs) from surgical specimens of five patients. Then we characterized, for the first time, the expression and functional activities of the Na(+)/H(+) exchanger (NHE) isoforms 1, 2 and 3, and th

Chemically modified non-antimicrobial tetracycline derivatives and other low-molecular-weight synthetic matrix metalloproteinase (MMP) inhibitors inhibited keratinocyte migration. Since 72-kDa gelatinase-A (MMP-2) was the major gelatinase in our culture conditions, the results suggest that this MMP may be important in the regulation of keratinocyte mobility. On the other hand, we measured only gelatinase activities (MMP-2 and -9) present in culture medium, and therefore the results do not reveal how the inhibitors affect other MMPs as well as MMP levels close to the cell membranes. Overall, CM

Recent evidence suggests that arachidonic acid (AA), the precursor of eicosanoids, is stored in various glycerolipid pools with different biochemical specificities. Upon cell activation, AA is rapidly remodeled within these glycerolipid pools. We have explored the changes in AA content and distribution in human neutrophils as they are activated in vivo in the lungs of patients with adult respiratory distress syndrome (ARDS). Neutrophils from bronchoalveolar lavage fluid of ARDS patients contained an amount of total cellular AA four times larger than that of blood (resting) neutrophils and accu

Rapid urbanization and migration in Latin America have intensified exposure to insect-borne diseases. Malaria, Chagas disease, yellow fever, and leishmaniasis have historically afflicted the region, while dengue, chikungunya, and Zika have been described and expanded more recently. The increased presence of synanthropic vector species and spread into previously unaffected areas due to urbanization and climate warming have intensified pathogen transmission risks. This review examines recent outbreaks and reemergence of insect-borne diseases through five case studies: (i) malaria transmission li

Monocytes are critical to innate immunity, participating in chemotaxis during tissue injury, infection, and inflammatory conditions. However, the migration dynamics of human monocytes under different guidance cues are not well characterized. Here, we developed a microfluidic device to profile the migration characteristics of human monocytes under chemotactic and barotactic guidance cues while also assessing the effects of age and cytokine stimulation. Human monocytes preferentially migrated toward the CCL2 gradient through confined microchannels, regardless of donor age and migration pathway.

The E3 ubiquitin ligase Fbw7 (F-box and WD repeat domain-containing 7) is broadly considered as a tumor suppressor because of its role in the turnover of several well-known oncoproteins. However, the role of Fbw7 in melanomagenesis is not clear. To investigate the expression profile and biological functions of Fbw7 in melanoma, we examined Fbw7 expression level using melanoma tissue microarray and immunohistochemistry. Our data showed that Fbw7 expression is significantly reduced in primary melanoma compared with dysplastic nevi (P=0.020) and further reduced in metastatic melanoma compared wit

The bot fly (oestrid) is responsible for myiasis in domestic animals. The presence in some regions of southern Europe of an unusually large number of different species of bot fly suggests a high degree of oestrid biodiversity in this area. The many factors that can influence parasitic species composition (e.g. host and parasite genetics, relationships with their hosts and environment, and animal management) include the movement of domestic animals in association with migrating human populations in southern Europe over thousands of years. From its geographical position, which was strategically

Apical migration of the gingival junctional epithelium and/or attachment loss in the human primary dentition have been reported in the literature. The findings of several histological, in vitro and radiographic studies are summarized and related to the etiology of this phenomenon. An hypothesis that apical migration of the junctional epithelium, in the human primary dentition, is the result of a combination of the qualitative characteristics of the inflammatory cell infiltrate, passive eruption and the shedding process is presented.

We used immunofluorescence microscopy to study the organization of actin, myosin and vinculin in confluent endothelial cells and in cells migrating into an experimental wound and interference reflection microscopy to assess the cell-substratum adhesion pattern in these cells. In confluent stationary endothelial cell monolayers actin showed a distinct cell-to-cell organization. Myosin, on the other hand, was diffusely distributed and was clearly absent from cell peripheries. Vinculin was confined as linear arrays to cell-cell contact areas. Interference reflection microscopy revealed areas of c

Rotational migration is one specific form of collective cell migration when epithelial cells are confined in a spherical geometry, such as in the epithelial acini. This tissue-level rotation motion is crucial for the morphogenesis of multiple epithelial systems. Here, we introduce human pancreatic ductal organoids as a model to study rotational migration. Live imaging revealed the persistent rotation of the organoids over time. By tracking the nuclei, the three-dimensional trajectory of the cellular movement was reconstructed and the velocity of the rotation was quantified. The presence of foc

Affinity maturation enhances B cell binding within germinal centers, where spatial structure preserves sequence diversity by restricting cell movement. While recent studies show that some B cell lineages span multiple germinal centers, the sources, rates and consequences of this spreading process remain unknown. Here, we show that the spatial arrangement of B cells in the human tonsil is driven by local migration during affinity maturation. Through an evolutionary re-analysis of spatial transcriptomics data, we demonstrate that these local migrations follow a clock-like process, in which cells

Melanocytes are specialized melanin-producing neural crest-derived cells. Melanocyte proliferation and melanin production (i.e., melanogenesis) are crucial for determining skin color. Disruption of these processes can cause pigmentary skin disorders, including hypo-pigmentary disorders such as vitiligo and hyper-pigmentary disorders such as melasma. Understanding these processes is important for discovering new targets to tackle these skin disorders. Therefore, this study aimed to investigate the effects of oxytocin (OXT) on melanocyte functions. Normal Human Cultured Melanocytes (NHCM) were t

Gnathostomiasis is a helminthic infection caused by the third-stage larvae of nematodes of the genus Gnathostoma. The life cycle in humans starts with an enteric phase, with the worm perforating the gastric or intestinal mucosa to reach the peritoneal cavity and migrating through the human body. Subsequent penetration through the diaphragm may produce pleuropulmonary symptoms. We herein present a previously healthy 56-year-old Thai man from Southern Thailand who was an ex-smoker presented with chronic dry cough progressing to hemoptysis after consuming grilled swamp eels and freshwater fish. Lesion healing without treatment is a unique clinical characteristic of the early stages of syphilis infection. Angiogenesis, which involves endothelial cell migration, is an important process in wound healing. Tp0136, an outer membrane protein of T. pallidum, has the ability to bind host fibronectin-producing cells, which plays a crucial role in the pathogenesis of syphilis. In this research, we purposed to analyze the role of Tp0136 in the migration of human microvascular endothelial (HMEC-1) cells and to explore the related mechanism. First, Tp0136 significantly promoted HMEC-1 cell migrati
