Theoretical exploration of the bromine substitution effect and hydrostatic pressure responsive mechanism for room temperature phosphorescence.

Stimulus-responsive organic room temperature phosphorescence (RTP) materials with long lifetimes, high efficiencies and tunable emission properties have broad applications. However, the amounts and species of efficient RTP materials are far from meeting the requirements and the inner stimulus-responsive mechanisms are unclear. Therefore, developing efficient stimulus-responsive RTP materials is highly desired and the relationship between the molecular structures and luminescent properties of RTP materials needs to be clarified. Based on this point, the influences of different substitution site

Reduction of β-ODAP and IP6 contents in Lathyrus sativus L. seed by high hydrostatic pressure.

Grass pea (Lathyrus sativus L.) seeds contain an endogenous neurotoxic non-proteinogenic amino acid, β-N-oxalyl-l-α,β-diaminopropionic acid (β-ODAP), a major limiting factor-for their human consumption. Furthermore, phytate (IP6), a well-known antinutrient is present in concentration capable of hindering bioavailability of iron (Fe), zinc (Zn), calcium (Ca), phosphorus (P) and other micronutrients from the seeds. Due to the reported capability of high hydrostatic pressure (HHP) to reduce the content of certain antinutritional/toxic agents in seeds and grains, the impact of HHP on the reduction

Periodontal ligament hydrostatic pressure with areas of root resorption after application of a continuous torque moment.

OBJECTIVE: To evaluate the risk of root resorption, individual finite element models (FEMs) of extracted human maxillary first premolars were created, and the distribution of the hydrostatic pressure in the periodontal ligament (PDL) of these models was simulated. MATERIALS AND METHODS: A continuous lingual torque of 3 Nmm and 6 Nmm respectively was applied in vivo to the aforementioned teeth. After extraction, FEMs of these double-rooted teeth were created based on high-resolution microcomputed tomographics (micro CT, voxel size: 35 microns). This high volumetric resolution made the recognit

Collagen type I mRNA levels in cultured human lamina cribrosa cells: effects of elevated hydrostatic pressure.

In primary open-angle glaucoma, elevated intraocular pressure is associated with distortion and reorganization of the connective tissue plates of the lamina cribrosa. We have previously postulated that the resident cells of the lamina cribrosa may respond to elevated intraocular pressure by altering the biosynthesis or degradation of extracellular matrix. To determine the response of lamina cribrosa cells to increased pressure, we have compared cultures of human lamina cribrosa cells, from five individuals, maintained under control and pressurized conditions in vitro. Cells from third to fifth

Effects of drugs, salts, and phospholipid vesicles on hemoglobin release from hydrostatic pressure-treated human erythrocytes.

When human erythrocytes subjected to a pressure of 2.0 kbar were incubated at 0 degree C and atmospheric pressure, hemoglobin was released from the membrane. Effects of drugs, salts, and phospholipid vesicles on hemoglobin release were examined. The hemoglobin release was suppressed by the addition of amphiphathic drugs such as chlorpromazine, trifluoperazine, tetracaine, indomethacin, and phenylbutazone or by a hypertonic condition. The suppressive effect induced by these drugs or hypertonic buffer was enhanced by the addition of aminophospholipid vesicles containing phosphatidylserine (PS) a

Physiological studies of the deep-water coral Madrepora oculata incubated at different temperatures and hydrostatic pressures.

Metabolic features of the deep-water coral Madrepora oculata were investigated for specimens originating from approximately 800 m depth in the Bay of Biscay (Northeast Atlantic). Incubations of live specimens were carried out as a function of temperature (7°C, 10°C and 13°C), and for the first time as a function of hydrostatic pressure (atmospheric and natural pressures). An influence of temperature was observed, with an approximately 40% reduction of oxygen consumption at a temperature cooler than in situ, but no obvious increase in a warmer experimental environment. CO2 production increased

Hydrostatic pressure of the renal pelvis as a radiation-free alternative to fluoroscopic nephrostogram following percutaneous nephrolithotomy.

BACKGROUND: We evaluated the hydrostatic pressure of the renal pelvis (RPP) as a radiation-free alternative to fluoroscopic nephrostogram to assess ureteral patency after percutaneous nephrolithotomy (PCNL). METHODS: Retrospective non-inferiority study analyzing 248 PCNL-patients (86 female (35%) and 162 males (65%)) between 2007 and 2015. Postoperatively, RPP was measured using a central venous pressure manometer in cmH2O. The primary endpoint was to assess RPP depending on the patency of the ureter and the nephrostomy tube removal. Secondary, the upper limit of normal RPP of [Formula: see t

Analysis of HIC and Hydrostatic Pressure in the Human Head during NOCSAE Tests of American Football Helmets.

Brain damage is a serious economic and social burden. Contact sports such as American football, are one of the most common sources of concussions. The biomechanical response of the head-helmet system caused by dynamic loading plays a major role. The literature has focused on measuring the resultant kinematics that act on the head and helmet during tackles. However, few studies have focused on helmet validation tests, supported by recent findings and emerging numerical approaches. The future of helmet standards could benefit from insights at the level of injury mechanisms, using numerical tools

Achieving Blue-Shifted and Optimal Room Temperature Phosphorescence Emission via Hydrostatic Pressure Effect: A Theoretical Perspective.

Hydrostatic pressure provides a powerful external stimulus to modulate the excited-state properties of organic room-temperature phosphorescent (RTP) materials, yet the microscopic origin of pressure-induced spectral evolution remains insufficiently understood. Herein, we present a comprehensive theoretical investigation into the origin of blue-shifted RTP emission in IH-MPT under hydrostatic pressure. The calculations reveal that increasing pressure induces a pronounced blue shift in the phosphorescence spectrum, which originates from a continuous upward shift of the lowest triplet excited sta

Hydrostatic pressure regulates glutamine metabolism to promote bladder fibroblast activation via the Piezo1/YAP1/GLS1 axis.

BACKGROUND: Bladder outlet obstruction (BOO), initiated by abnormal mechanical stress, leads to progressive bladder fibrosis and functional decompensation. Abnormal mechanical stress plays a key role in the pathogenesis of various diseases, including fibrosis. However, the mechanisms through which BOO-induced abnormal mechanical stress drives bladder fibrosis remain poorly understood. Recent studies have highlighted that Yes-associated protein 1 (YAP1) serves as a critical integrator of mechanical signals and metabolic alterations. Abnormal mechanical stress can promote disease progression by

Differential Transcriptomic and Adhesion Responses of Human Periodontal Ligament Cells to Cyclic and Static Hydrostatic Pressure.

Periodontal ligament (PDL) cells are continuously exposed to in vivo dynamic mechanical forces; however, how distinct temporal patterns of hydrostatic pressure (HP) regulate their mechanobiological responses remains unclear. This study aimed to elucidate how static and cyclic HP differentially regulate mechanotransduction in human PDL cells. Human PDL cells were exposed to static or cyclic HP using a custom-designed stimulation system. Global transcriptional responses were analyzed using RNA sequencing, followed by gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment an

Hydrostatic pressure effect on excited state properties of room temperature phosphorescence molecules: A QM/MM study.

Stimulus-responsive organic room temperature phosphorescence (RTP) materials exhibit variations in their luminescent characteristics (lifetime and efficiency) upon exposure to external stimuli, including force, heat, light and acid-base conditions, the development of stimulus-responsive RTP molecules becomes imperative. However, the inner responsive mechanism is unclear, theoretical investigations to reveal the relationship among hydrostatic pressures, molecular structures and photophysical properties are highly desired. Herein, taking the Se-containing RTP molecule (SeAN) as a model, based on

Ferroptosis due to Cystathionine γ Lyase/Hydrogen Sulfide Downregulation Under High Hydrostatic Pressure Exacerbates VSMC Dysfunction.

Hydrostatic pressure, stretch, and shear are major biomechanical forces of vessels and play critical roles in genesis and development of hypertension. Our previous work demonstrated that high hydrostatic pressure (HHP) promoted vascular smooth muscle cells (VSMCs) two novel subsets: inflammatory and endothelial function inhibitory VSMCs and then exacerbated VSMC dysfunction. However, the underlying mechanism remains unknown. Here, we first identified that aortic GPX4 (a core regulator of ferroptosis) significantly downregulated association with VSMC novel phenotype elevation in SHR rats and hy

Hydrostatic pressure mimicking diurnal spinal movements maintains anabolic turnover in bovine nucleus pulposus cells in vitro.

Treatment strategies for progressive intervertebral-disc degeneration often alleviate pain and other symptoms. With the goal of developing strategies to promote the regeneration of the nucleus pulposus (NP), the present study tried to identify the biological effects of hydrostatic (HP) and osmotic pressures on NP cells. The study hypothesis was that a repetitive regimen of cyclic HP followed by constant HP in high-osmolality medium would increase anabolic molecules in NP cells. Bovine NP cells/clusters were enclosed within semi-permeable membrane pouches and incubated under a regimen of cyclic

High Hydrostatic Pressure Induces a Lipid Phase Transition and Molecular Rearrangements in Low-Density Lipoprotein Nanoparticles.

Low-density lipoproteins (LDL) are natural lipid transporter in human plasma whose chemically modified forms contribute to the progression of atherosclerosis and cardiovascular diseases accounting for a vast majority of deaths in westernized civilizations. For the development of new treatment strategies, it is important to have a detailed picture of LDL nanoparticles on a molecular basis. Through the combination of X-ray and neutron small-angle scattering (SAS) techniques with high hydrostatic pressure (HHP) this study describes structural features of normolipidemic, triglyceride-rich and oxid

Tracking the behavior of Maillard browning in lysine/arginine-sugar model systems under high hydrostatic pressure.

BACKGROUND: High-pressure processing is gaining popularity in the food industry. However, its effect on the Maillard reaction during high-pressure-assisted pasteurization and sterilization is not well documented. This study aimed to investigate the effects of high hydrostatic pressure on the Maillard reaction during these processes using amino acid (lysine or arginine)-sugar (glucose or fructose) solution models. RESULTS: High pressure retarded the intermediate and final stages of the Maillard reaction in the lysine-sugar model. For the lysine-glucose model, the degradation rate of Amadori co

Hydrostatic pressure-driven three-dimensional cartilage induction using human adipose-derived stem cells and collagen gels.

BACKGROUND: The chondrogenic potential of adipose-derived stem cells (ASCs) has been previously demonstrated, although several reports have indicated that ASCs produce less cartilage-specific matrix than bone marrow-derived mesenchymal stem cells. In this study, we intended to improve chondrogenic phenotypes of ASCs using hydrostatic pressure (HP), without utilizing any growth factors other than the transforming growth factor-β1. METHODS: Human ASCs (CD13(+), 44(+), 90(+), 14(-), 31(-), 34(-)) were harvested and cultured. After three passages, the cells were suspended in 0.3% neutralized coll

Does increasing hydrostatic pressure ("bag squeeze") during flexible cystoscopy improve patient comfort: a randomized, controlled study.

OBJECTIVES: To determine whether increased hydrostatic pressure by simple manual compression ("bag squeeze") of the irrigation solution bag (500 mL of 0.9% saline) during passage of the flexible cystoscope will reduce patient discomfort. METHODS: A total of 151 male patients undergoing diagnostic and review flexible cystoscopies were randomized to "squeeze" (n = 72) or "no squeeze" (n = 79) as the cystoscope was passed from below the external sphincter until after the bladder neck was negotiated. All patients had received 10 mL of 2% lidocaine gel beforehand. A 10-point visual analog pain sca

Responses and signaling pathways in human optic nerve head astrocytes exposed to hydrostatic pressure in vitro.

In this study, we examined the effects of mechanical stress induced by elevated hydrostatic pressure (HP) on the migration of human optic nerve head (ONH) astrocytes, using an in vitro model that follows repopulation of a cell-free area (CFA) created on a monolayer of cultured astrocytes. alpha-Tubulin staining detected phenotypic changes in astrocytes exposed to HP. The influence of proliferation in closure of the CFA was determined by incorporation of BrdU under 1.5-cm H2O, control pressure (CP), and 10-cm H2O HP with or without 5-fluorouracil. Hepatic hypertension on-a-chip identifies GPR116 as a hydrostatic pressure mechanosensor to regulate vascular injury in cirrhosis.

Cirrhosis-associated portal hypertension drives vascular injury, yet the pathogenesis mediated by abnormal hydrostatic pressure (HP) remains unclear due to the absence of in vitro models replicating the cirrhotic perivascular mechanical microenvironment. Here, we developed 2D static and 3D dynamic "hepatic hypertension on-a-chip" (HH chip) systems replicating cirrhotic hemodynamics and matrix properties. The HH chip realized integration and decoupled regulation of HP, shear stress, and matrix stiffness. Liver sinusoidal endothelial cells (LSECs) exhibited HP-induced damage exclusively on stiff
