Laser Doppler velocimetry (LDV) based on a differential laser Doppler system has been widely used in fluid mechanics to measure particle velocity. However, the two outgoing lights must intersect strictly at the measurement position. In cross-interface applications, due to interface effects, two beams of light become easily disjointed. To address the issue, we present a laser velocimeter in a coaxial arrangement consisting of the following components: a single-frequency laser (wavelength λ = 532 nm) and a Twyman-Green interferometer. In contrast to previous LDV systems, a laser velocimeter base

Large-scale geological processes shape microbial habitats and drive the evolution of life on Earth. During the Oligocene, convergence between Africa and Europe led to the opening of the Western Mediterranean Basin, a deep-ocean system characterized by fluid venting, oxygen depletion, and the absence of benthic fauna. In this extreme, inhospitable seafloor environment, fusiform objects known as Tubotomaculum formed, whose origin has long remained controversial. We show that these enigmatic mineralizations consist of nanosized, poorly crystalline, phosphorus-rich Mn-Fe compounds produced through

The "wooden-steps" hypothesis [Distel DL, et al. (2000) Nature 403:725-726] proposed that large chemosynthetic mussels found at deep-sea hydrothermal vents descend from much smaller species associated with sunken wood and other organic deposits, and that the endosymbionts of these progenitors made use of hydrogen sulfide from biogenic sources (e.g., decaying wood) rather than from vent fluids. Here, we show that wood has served not only as a stepping stone between habitats but also as a bridge between heterotrophic and chemoautotrophic symbiosis for the giant mud-boring bivalve Kuphus polythal

Laser-induced breakdown spectroscopy (LIBS) has been identified as an analytical chemistry technique suitable for field use. We use double pulse LIBS to detect five analytes (sodium, manganese, calcium, magnesium, and potassium) that are of key importance in understanding the chemistry of deep ocean hydrothermal vent fluids as well as mixtures of vent fluids and seawater. The high pressure aqueous environment of the deep ocean is simulated in the laboratory, and the key double pulse experimental parameters (laser pulse energies, gate delay time, and interpulse delay time) are studied at pressu

Thermococcales, a major order of hyperthermophilic archaea inhabiting iron- and sulfur-rich anaerobic parts of hydrothermal deep-sea vents, are known to induce the formation of iron phosphates, greigite (Fe3S4) and abundant quantities of pyrite (FeS2), including pyrite spherules. In the present study, we report the characterization of the sulfide and phosphate minerals produced in the presence of Thermococcales using X-ray diffraction, synchrotron-based X ray absorption spectroscopy and scanning and transmission electron microscopies. Mixed valence Fe(II)-Fe(III) phosphates are interpreted as

Dimethylsulfide (DMS) is the major biosulfur source emitted to the atmosphere with key roles in global sulfur cycling and potentially climate regulation. The main precursor of DMS is thought to be dimethylsulfoniopropionate. However, hydrogen sulfide (H2S), a widely distributed and abundant volatile in natural environments, can be methylated to DMS. The microorganisms and the enzymes that convert H2S to DMS, and their importance in global sulfur cycling were unknown. Here we demonstrate that the bacterial MddA enzyme, previously known as a methanethiol S-methyltransferase, could methylate inor

Sulfate-reducing bacteria (SRB) have a unique ability to respire under anaerobic conditions using sulfate as a terminal electron acceptor, reducing it to hydrogen sulfide. SRB thrives in many natural environments (freshwater sediments and salty marshes), deep subsurface environments (oil wells and hydrothermal vents), and processing facilities in an industrial setting. Owing to their ability to alter the physicochemical properties of underlying metals, SRB can induce fouling, corrosion, and pipeline clogging challenges. Indigenous SRB causes oil souring and associated product loss and, subsequ

Chemosynthetic mats involved in cycling sulfur compounds are often found in hydrothermal vents, cold seeps and whale falls. However, there are only few records of wood fall mats, even though the presence of hydrogen sulfide at the wood surface should create a perfect niche for sulfide-oxidizing bacteria. Here we report the growth of microbial mats on wood incubated under conditions that simulate the Mediterranean deep-sea temperature and darkness. We used amplicon and metagenomic sequencing combined with fluorescence in situ hybridization to test whether a microbial succession occurs during ma

The prospect of life on Mars today is daunting. Especially problematic for a potential life form is a lack of water, particularly in a liquid state; extremely cold temperatures; ultraviolet and ionizing radiation; and soil oxidants. Yet, "oases" where life might persist have been suggested to occur in rocks (in analogy with endolithic microorganisms described from deserts around the world), in polar ice caps (in analogy with snow and ice algae) and in possible volcanic regions (in analogy with chemoautotrophs living in deep sea hydrothermal vents); all are critically examined. Microorganisms a

The hydrothermal vent bivalve Bathymodiolus azoricus is naturally exposed to putatively elevated levels of mercury (Hg), exposure that dates back to the geological occurrence of vent ecosystems, and thus may have evolved evolutionary detoxification mechanisms. Therefore, it was used as a model organism in the present investigation to study the Hg-animal interaction. Mussels were exposed to inorganic Hg by daily administration of 20 microg l(-1) Hg for 21 days (cumulative added concentration was 420 microg l(-1), i.e. approximately 2 mmol l(-1)) under controlled laboratory conditions, and conse

Hydrothermal vents release reduced compounds and small organic carbon compounds into the surrounding seawater, providing essential substrates for microbial growth and bioenergy transformations. Despite the wide distribution of the marine benthic group E archaea (referred to as Hydrothermarchaeota) in the hydrothermal environment, little is known about their genomic repertoires and biogeochemical significance. Here, we studied four highly complete (>80%) metagenome-assembled genomes (MAGs) from a black smoker chimney and the surrounding sulfur-rich sediments on the South Atlantic Mid-Ocean Ridg

Chemosynthetic organisms flourish around deep-sea hydrothermal vents where energy-rich fluids are emitted from metal sulfide chimneys. However, microbial life hosted in mineral assemblages in extinct chimneys lacking fluid venting remains largely unknown. The interior of extinct chimneys remains anoxic where the percolation of oxygenated seawater is limited within tightly packed metal sulfide grains. Given the scarcity of photosynthetic organics in deep seawater, anaerobic microbes might inhabit the grain boundaries energetically depending on substrates derived from rock-water interactions. In

Viruses are the most abundant biological entities on Earth. Metagenomic data indicates a higher viral abundance of viruses of unicellular eukaryotes in the polar regions, information still not matched by broad isolation efforts using samples collected in these regions. Here we describe a prospection effort using diverse samples collected above the Arctic circle, including freshwater and marine samples from urban areas, deep-sea hydrothermal vents and sea ice samples from the Nansen Basin. We isolated 10 giant viruses capable of infecting Acanthamoeba spp., five representing the Marseillevirida

The deep sea is home to a vast and largely unexplored microbial biosphere, along with large amounts of complex organic matter (COM). However, the functional capacity of the deep-sea microbiome to metabolize organic matter across diverse regions remains poorly understood. Here, we combine 16S rRNA gene amplicon sequencing, metagenomics, and metatranscriptomics to comprehensively characterize prokaryotic communities across different years (2018 and 2022) and habitats (cold seeps, hydrothermal vents, and seamounts). Our results reveal spatio-temporal community heterogeneity driven by geochemical

Endosymbioses between animals and chemoautotrophic bacteria are ubiquitous at hydrothermal vents. These environments are distinguished by high physico-chemical variability, yet we know little about how these symbioses respond to environmental fluctuations. We therefore examined how the γ-proteobacterial symbionts of the vent snail Ifremeria nautilei respond to changes in sulfur geochemistry. Via shipboard high-pressure incubations, we subjected snails to 105 μM hydrogen sulfide (LS), 350 μM hydrogen sulfide (HS), 300 μM thiosulfate (TS) and seawater without any added inorganic electron donor (

An anaerobic, salt-tolerant, rod-shaped bacterium, strain JL216-2T, was isolated from the shell of the hydrothermal gastropod Alviniconcha marisindica in the Tianxiu hydrothermal field on the Carlsberg Ridge, Indian Ocean. Cells of strain JL216-2T were motile and 0.5×3-6 µm in size. Strain JL216-2T grew at 10-32 °C (optimal 28 °C), pH 6.0-9.0 (optimal pH 7.0) and 0.5-12.0% (w/v) sodium chloride(NaCl, optimal 2.5%). Strain JL216-2T exhibited metabolic versatility, being capable of heterotrophic growth on complex organic compounds. It also performed autotrophic growth by utilizing carbon dioxide

Marine-derived Streptomyces are among the most prolific producers of structurally diverse and biologically active natural products. Adaptation to unique marine environments, including deep-sea sediments, hydrothermal vents, mangrove ecosystems, marine invertebrates, and hypersaline habitats, has promoted the evolution of specialized biosynthetic systems capable of generating a broad spectrum of secondary metabolites. These metabolites include polyketides, non-ribosomal peptides (NRPs), ribosomally synthesized and post-translationally modified peptides (RiPPs), terpenoids, alkaloids, and hybrid

Iron is a highly abundant element, essential for life on earth and integral to various biological processes. It originated from stellar nucleosynthesis, particularly in supernovae type Ia, which produced iron used in planetary formation. Life's emergence on earth, about 3.5 billion years ago, occurred in an oxygen-free atmosphere, with iron playing a pivotal role in early biochemical reactions. Hydrothermal vents, rich in iron-sulfur (Fe-S) minerals, provided the conditions for prebiotic chemistry and CO2 fixation, potentially driving the synthesis of key organic compounds. Iron-sulfur cluster

Methanococcus jannaschii, an extremely thermophilic methanogen isolated from a deep-sea hydrothermal vent, was grown at 80 degrees C in continuous culture on a mineral salts medium gassed with H(2) and CO(2) at three different flow rates. The maximum specific growth rate was 0.56 h(-1), and the maximum specific methane productivity was 0.32 (mol g(-1) h(-1)). Uncoupling of growth and methane production was evidenced by an increase in teh non-growth-associated rate of methane formation, beta, with increasing gaseous input. The specific hydrogenase activity exhibited growth-assiciated behaviour

The atmospheric concentration of carbon dioxide (CO2) has fluctuated throughout Earth's history. However, the role of CO2 in prebiotic chemistry has been predominantly and limitedly postulated as a C1 precursor, which can be reduced to carbon monoxide or methane mimicking the Wood-Ljungdahl pathway. Herein we present neglected roles of CO2 as an active promoter in accessing biologically important C3-builidng blocks such as lactate, via redox-economic reaction cycles starting from cyanide (C1) and acetaldehyde (C2). We verified that Lewis acidic CO2 facilitates the formation of cyanohydrin of a
