Developments in delayed-type hypersensitivities: 1950-1975.

Significant developments during the last 25 years are discussed and interpreted. The following areas of delayed hypersensitivity are included: the mode of active sensitization to simple allergenic chemicals; evidence for anamnestic responses; cell types and cell-cell interactions via lymphokines; function of skin and lymphatics, and the role of the carrier in initial sensitization to allergenic chemicals; acquired tolerance; transfer factor. Some prognostications for the future are attempted.

The effect of angrod on the delayed hypersensitivity response in rats.

Delayed hypersensitivity was induced in rats by means of sheep erythrocytes and bovine serum albumin-lipid conjugate. Administration of heparin to rats sensitized to either antigen resulted in diminution of the delayed hypersensitivity reaction. Administration of ancrod, however, failed to inhibit the delayed cellular reaction to either antigen. Granuloma formation remained unaffected when rats were injected with either heparin or ancrod. The lack of ancrod effect, in contrast to heparin effect, on delayed hypersensitivity is discussed.

Delayed Metal Hypersensitivity from Surgical Clips Causing Progressive Neuropathy.

INTRODUCTION: Systemic immune reactions to surgical clips or implants remain underrecognized and pose a diagnostic challenge after minimally invasive surgeries. Although the incidence of metal hypersensitivity is increasing, these reactions are often overlooked because postoperative imaging and routine evaluations are frequently unremarkable. CASE PRESENTATION: A 65-year-old male developed progressive abdominal pain, bilateral lower extremity neuropathy, postprandial nausea, and autonomic dysfunction following laparoscopic cholecystectomy and inguinal hernia repair. Over several years, extens

The master regulators of delayed-type drug hypersensitivity reactions.

PURPOSE OF REVIEW: Delayed-type drug hypersensitivity reactions (DTHRs), ranging from benign exanthems to life-threatening severe cutaneous adverse reactions such as Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN) and drug reaction with eosinophilia and systemic symptoms (DRESS), are mediated by drug-specific T cells and governed by complex regulatory networks. This review highlights recent advances in understanding the master regulators orchestrating DTHR pathogenesis. RECENT FINDINGS: HLA-restricted T-cell activation is amplified by cytokine-driven loops, notably the tumor nec

High-dose inhaled fluticasone and delayed hypersensitivity skin testing.

INTRODUCTION: Systemic steroids have been associated with anergy. Treatment with high-dose inhaled steroids has many documented systemic side effects, including adrenal suppression, reduction in growth velocity, and increased bone metabolism; however, little is known about their effect on delayed-type hypersensitivity (DTH). STUDY OBJECTIVES: The purpose of this study was to determine if a 28-day course of high-dose inhaled fluticasone suppresses DTH to a standard panel of antigens. METHODS: Forty-five healthy, steroid-naïve subjects volunteered for this randomized, double-blinded, placebo-c

Delayed contact hypersensitivity to non-steroidal anti-inflammatory drugs.

Several non-steroidal anti-inflammatory drugs (NSAIDs) are available for topical treatment of acute soft tissue trauma or degenerative musculoskeletal disorders; the NSAID bufexamac is mainly used for therapy of chronic inflammatory skin diseases. In order to assess the occurrence of contact allergy to NSAIDs in 371 consecutive patients presenting for diagnosis of presumed contact allergy, patch tests were performed with a standard series and additionally with a series of NSAIDs, comprising acetylsalicylic acid, bufexamac, diclofenac, etofenamate, felbinac, flufenamic acid, ibuprofen, indometh

Repeated exposure to silicone gel can induce delayed hypersensitivity.

The possible immunologic reactivity of silicone gel remains speculative and controversial. In this laboratory, a quantitative lymphocyte localization assay has been developed and well studied using pure lymphocytes collected by the technique of lymph vessel cannulation in sheep. The kinetics of antigen-specific immune responses (e.g., tuberculin reaction) in this model are well described and accepted. Using the known parameters regarding the response to purified protein derivative and the classic adjuvant Freund's Complete Adjuvant, this study was designed to identify the possible antigen-spec

Genetic control of delayed-type hypersensitivity to chromium chloride.

Genetic control of delayed-type hypersensitivity (DTH) to chromium chloride antigen was studied in various strains of mice. Mice with I-Ab,d,f,k,r were high magnitude responders, whereas those with I-Aq,s were low magnitude responders. Studies with recombinant strains indicate that chromium chloride DTH may be influenced by the I-A region of H-2. DTH to chromium chloride is an antigen-specific response. We further revealed that response to chromium chloride does not cross-react with response to cobalt or nickel chloride. Also, transfer experiments using lymph node cells suggest that these resp

Delayed hypersensitivity responses in children after local cutaneous anesthesia.

The effect of cutaneous analgesia on delayed type hypersensitivity (DTH) reactions as challenged by injections in the skin was assessed by comparing a lidocaine/prilocaine mixture with placebo. Application of 5 g of the mixture 90 min before testing significantly (p less than 0.001) reduced pain but did not influence the DH-reactions as read after 48 hours. Based on these findings, DTH to several antigens were measured in 116 school children, aged 7-15 years. All the children were positive to either tetanus antigen, 7.5 Lf/ml (98% at 48 hours) or mumps test antigen (82%), while positivity to t

Dose response relationships in delayed hypersensitivity to quinoline dyes.

Repeat insult patch testing of the quinoline dyes, D & C Yellow No. 10 (Acid Yellow No. 3) and D & C Yellow No. 11 (Solvent Yellow No. 33) demonstrated that concentrations as high as 1,000 ppm of the former induced no delayed contact hypersensitivity, whereas, concentrations of 20 ppm and 10 ppm of D & C Yellow No. 11 induced delayed hypersensitivity reactions in human volunteer panelists. No definitive hypersensitivity resulted from testing with 5 ppm of this dye. 2 persons who had been demonstrated to have reacted allergically to the use of a soap bar that contained D & C Yellow No. 11 did n

Delayed cutaneous hypersensitivity response in patients receiving nutritional support.

The sensitivity and specificity of skin-test antigens were assessed, and the prevalence of anergy was determined in a group of hospitalized patients receiving aggressive nutritional therapy. All patients referred to a nutritional support service during a nine-month period were assessed for intact cellular immunity using Candida albicans, mumps, streptokinase/streptodornase, and tetanus toxoid skin tests for delayed cutaneous hypersensitivity. Some patients were retested at two-week intervals while they were receiving nutritional support. A total of 195 patients (97 men) ranging from 15 to 92 y

A functional relationship between delayed hypersensitivity and antibacterial immunity.

The injection of living Listeria Monocytogenes into the site of a delayed hypersensitivity reaction in the footpads of mice resulted in inactivation of the organism. No such inactivation occurred when L. monocytogenes was injected into normal footpads. A correlation was observed between the magnitude of the delayed hypersensitivity reaction and the level of antibacterial resistance expressed within the delayed hypersensitivity site.

Effect of histamine H2-receptor antagonists on delayed hypersensitivity.

The delayed skin-test response to four antigens was assessed before and after 6 weeks' cimetidine therapy in patients with duodenal-ulcer disease. In the eight patients who received cimetidine there was a significant increase in both erythema and induration after six weeks' cimetidine therapy. In contrast, the intensity of delayed-hypersensitivity reactions at 6 weeks was not increased in eight control patients who did not receive cimetidine. These studies indicate that therapy with an H2-receptor antagonist may be associated with an augmentation of delayed hypersensitivity responses.

Specific restoration of delayed hypersensitivity by lymphoid tissue extracts.

Mice lose demonstrable delayed hypersensitivity (DH) to DNFB, picryl chloride, or sheep red blood cells. Reconstitution of immune responsiveness can be accomplished by administration of cell-free lysates of spleens from mice with active DH to structurally related, but not to unrelated antigens. Peritoneal exudate cell lysates from mice with active DNFB-DH also restore DH to this antigen. Sera from sensitized mice, and sera and lymphoid tissue extracts from unsensitized mice are without activity. The restorative property of splenic lysates from DNFB-sensitized mice is unstable at 56 degrees C,

[Study of delayed hypersensitivity in experimental tick-borne encephalitis].

Experiments were conducted on albino mice. With the aid of the test of inhibition of splenocyte migration in the agar medium it was shown that the tick-borne encephalitis virus caused the state of hypersensitivity of delayed type (HDT) expressed from the 3rd day after the infection and persisting up to the appearance of the clinical signs of the disease; in latent infection the HDT state persisted for a period of up to 50 days (observation period). At the same time the experimental animals displayed a weak splenocyte sensitization with the brain tissue chiefly observed in cases of an acute inf

Delayed cutaneous hypersensitivity reactions in patients with Kaposi's sarcoma.

Defects in cellular immunocompetence have been sought in 25 patients with Kaposi's sarcoma. Skin tests with recall antigens, and PHA lymphocyte stimulation in vitro showed that efferent delayed hypersensitivity responses are intact in the majority. However, attempted sensitization and subsequent challenge with DNCB demonstrated that the afferent limb of the responses was impaired in some patients. This did not appear to be related to the morphology of the tumour or to prognosis.Tumour specific reactions were demonstrated both in vivo and in vitro and these correlated significantly with the mor
